Description

FIELD OF THEINVENTION

[0001] The present disclosure relates to aerosolizable formulations, methods of making aerosolizable formulations, containers containing aerosolizable formulations, devices containing aerosolizable formulations, and methods and uses of aerosolizable formulations.

[0002] Electronic aerosol delivery systems, such as e-cigarettes, generally contain a reservoir of liquid to be vaporized, typically containing nicotine. When a user inhales on the device, a heater is activated to vaporize a small amount of the liquid, which is then inhaled by the user.

[0003] Use of e-cigarettes in the UK is growing rapidly and it is now estimated that there are over one million e-cigarette users in the UK.

[0004] One challenge faced in providing such a system is to deliver an inhaled aerosol from the aerosol delivery device that provides an acceptable experience to the consumer. Some consumers may prefer e-cigarettes that generate an aerosol that "resembles" the smoke inhaled from tobacco products such as cigarettes. The aerosol from e-cigarettes and the smoke from tobacco products such as cigarettes provide the user with a complex chain of flavors in the mouth, nicotine absorption in the mouth and throat, and subsequently nicotine absorption in the lungs. These various aspects are described by users in terms of flavor, strength / quality, impact, spiciness / smoothness, and nicotine reward. Nicotine contributes to several of these factors and is strongly associated with factors such as spiciness, spiciness, and smoothness, which are easily perceived by consumers, and e-cigarettes may give consumers too much or too little of these parameters, depending on personal preferences. Nicotine reward is particularly complex because it is a result of both the amount and rate at which nicotine is absorbed through the oral mucosa (which is typically nicotine in the vapor phase) and the amount and rate at which nicotine is absorbed through the lungs (which is typically nicotine in the particle phase of the inhaled aerosol). Each of these factors, and the balance of factors, can strongly contribute to consumer acceptability of an e-cigarette. Thus, delivery measures that optimize the overall vaping experience are desirable for e-cigarette manufacturers.

[0005] A further challenge facing such systems is the ongoing demand for harm reduction: the harm caused by cigarettes and e-cigarette devices comes primarily from toxins, and therefore it is desirable to reduce or eliminate components that may produce toxins. Summary of the Invention

[0006] In one embodiment, (i) water; (ii) nicotine, (iii) at least one acid; (iv) one or more fragrances; and (v) one or more encapsulating materials that encapsulate at least one of the one or more flavorings in preference to protonated nicotine; and (a) a molar ratio of an encapsulating material encapsulating one or more flavorings to (b) a non-encapsulated flavoring is (c) a molar ratio of encapsulating material encapsulating nicotine that exceeds (d) unencapsulated nicotine; Aerosolizable formulations are provided.

[0007] In one embodiment, (i) water, (ii) nicotine, (iii) at least one acid, and (iv) one or more flavorings, and (v) one or more encapsulating materials 1. A method for producing an aerosolizable formulation comprising: (a) contacting nicotine with at least one acid to partially or fully protonate the nicotine; (b) contacting the partially or fully protonated nicotine with water, one or more flavorings, and one or more encapsulating materials; wherein the encapsulating material encapsulates at least one of the one or more flavorants in preference to protonated nicotine.

[0008] In one embodiment, (a) a container; (b) (i) water, (ii) nicotine, (iii) at least one acid, and (iv) one or more flavorings, and (v) one or more encapsulating materials that encapsulate at least one of the one or more flavorings in preference to protonated nicotine. and an aerosolizable formulation comprising: 1. A contained aerosolizable formulation comprising: (a) a molar ratio of an encapsulating material that encapsulates one or more flavorings to (b) a non-encapsulated flavoring, (c) a molar ratio of encapsulating material encapsulating nicotine that exceeds (d) unencapsulated nicotine; A contained aerosolizable formulation is provided.

[0009] In one embodiment, an electronic aerosol delivery system is provided, comprising: (a) an aerosolization device for aerosolizing the formulation for inhalation by a user of the electronic aerosol delivery system; (b) a power source comprising a cell or battery for powering the aerosolization device; and (c) (i) water, (ii) nicotine, (iii) at least one acid, and (iv) one or more flavorings, and (v) one or more encapsulating materials that encapsulate at least one of the one or more flavorings in preference to protonated nicotine. and an aerosolizable formulation comprising: Equipped with (a) a molar ratio of an encapsulating material that encapsulates one or more flavorings to (b) a non-encapsulated flavoring, (c) a molar ratio of encapsulating material encapsulating nicotine that exceeds (d) unencapsulated nicotine; An electronic aerosol delivery system is provided.

[0010] As discussed herein, in one embodiment, (i) water; (ii) nicotine, (iii) at least one acid; (iv) one or more fragrances; and (v) one or more encapsulating materials that encapsulate at least one of the one or more flavorings in preference to protonated nicotine; and (a) a molar ratio of an encapsulating material encapsulating one or more flavorings to (b) a non-encapsulated flavoring is (c) a molar ratio of encapsulating material encapsulating nicotine that exceeds (d) unencapsulated nicotine; Aerosolizable formulations are provided.

[0011] The inventors have found that it is possible to provide an advantageous system in which protonated nicotine is generated before combining nicotine with flavoring and encapsulation material. The inventors have found that protonating nicotine before combining nicotine with encapsulation material (such as cyclodextrin) results in reduced interaction of nicotine with encapsulation material, and thus reduced competition between nicotine and flavoring for interaction with encapsulation material and / or encapsulation by encapsulation material. For example, protonated nicotine has a different size than non-protonated nicotine, and this size difference can result in nicotine having reduced interaction with encapsulation material and not being taken up by encapsulation material when nicotine is protonated. The encapsulation material interacts with at least one of the one or more flavors in preference to protonated nicotine and / or encapsulates at least one of the one or more flavors in preference to protonated nicotine, and the molar ratio of (a) encapsulated flavor to (b) unencapsulated flavor exceeds the molar ratio of (c) encapsulated nicotine to (d) unencapsulated nicotine. This system provides improved compatibility of the flavor with the encapsulation material and / or improved incorporation of the flavor in the encapsulation material (as the flavor is not displaced from the encapsulation material by the addition of nicotine).

[0012] As will be appreciated by those of skill in the art, nicotine may exist in unprotonated, monoprotonated or diprotonated forms. The structures of each of these forms are shown below. [ka]

[0026] The relevant amount of nicotine present in the formulation in protonated form is specified herein. These amounts can be easily calculated by those skilled in the art. Nicotine (3-(1-methylpyrrolidin-2-yl)pyridine) is a dibasic acid with a pKa of 3.12 for the pyridine ring and 8.02 for the pyrrolidine ring. Nicotine can exist in protonated (mono- and di-) and non-protonated (free base) forms with different bioavailability depending on pH. [ka]

[0027] The distribution of protonated and unprotonated nicotine will change at various pH increments. [ka]

[0028] The proportion of unprotonated nicotine dominates at high pH levels, while lowering the pH will see an increase in the proportion of protonated nicotine (mono- or di-, depending on the pH). If the relative proportion of protonated nicotine and the total amount of nicotine in a sample are known, the absolute amount of protonated nicotine can be calculated.

[0029] The relative proportion of protonated nicotine in a formulation can be calculated by using the Henderson-Hasselbalch equation, which expresses pH as a derivation of the acid dissociation constant equation, and which is widely used in chemical and biological systems. Consider the following equilibrium: [ka]

[0030] The Henderson-Hasselbalch equation for this equilibrium is:

number

[0031] where [B] is the amount of unprotonated nicotine (i.e., the free base), [BH+] is the amount of protonated nicotine (i.e., the conjugate acid), and pKa is the reference pKa value of the pyrrolidine ring nitrogen of nicotine (pKa=8.02). The relative proportion of protonated nicotine can be derived from the alpha value of unprotonated nicotine calculated from the Henderson-Hasselbalch equation as follows:

number

[0032] Determination of the pKa values of nicotine formulations was performed using the basic method described in "Spectroscopic investigations into the acid-base properties of nicotine at different temperatures", Peter M. Clayton, Carl A. Vas, Tam TTBui, Alex F. Drake and Kevin McAdam, Anal. Methods, 2013, 5, 81-88.

[0076] In addition to the aerosolizable formulations of the present invention and systems, such as containers and electronic aerosol delivery systems, containing same, the present invention provides methods for improving the sensory characteristics of aerosolized nicotine.

[0077] Reference to improving the sensory properties of the vaporized nicotine solution may include improving the smoothness of the vaporized nicotine solution as perceived by the user.

[0078] Methods of the present invention may include additional steps either before the recited steps, after the recited steps, or between one or more of the recited steps.

[0079] The invention will now be described in more detail, by way of example only, with reference to the accompanying drawings in which: [Brief description of the drawings]

[0080] [Figure 1] 1 is a graph showing the change in psKa2 with nicotine concentration. [Diagram 2] FIG. 1 shows the relative diffusion rates of nicotine, 2-hydroxypropyl-β-cyclodextrin, menthol, and citric acid (acidified solution only) in the formulations. EXAMPLES

[0081] The invention will now be described with reference to the following non-limiting examples.

[0082] Example 1 In fact, as the nicotine content increases, the amount of encapsulated flavor decreases. In this application, the flavor loading is not sufficient at nicotine concentrations above 3.0% (w / w). The addition of a weak organic acid in situ does not sufficiently protonate the nicotine before it forms a complex molecule with the cyclodextrin, so precipitation of some hydrophobic components, such as flavor, is observed. In this case, a heating step is required to prevent the formation of a precipitate.

[0083] An aqueous solution of citric acid and nicotine was prepared using a 1:1 molar equivalent ratio. The resulting liquid is bright orange in color and shows no signs of separation. Using a desiccator, the water and any water-soluble impurities are removed from the liquid, which produces a bright orange precipitate with a gel-like consistency. This was called nicotinium citrate.

[0084] The formulation can then be prepared as described above, except that no heating or filtering steps are required. When nicotinium citrate gel is added in place of nicotine in the final formulation step, the nicotinium citrate gel dissociates into the corresponding nicotinium (+) ion and citrate (-) ion.Since nicotinium (+) is originally larger than nicotine (nicotinium (+) takes over a proton from the corresponding citrate (-) ion), hydrophobic components are now the preferred guest species, and therefore nicotinium (+) cannot interfere with the cyclodextrin complexation mechanism. This is supported by the absence of visible precipitate.

[0085] Sensorily, the formulation is more potent when prepared in this manner. Flavor delivery is optimized and no longer tapers off with increasing nicotine concentration. Nicotine delivery is also optimized since nicotine is now in the particle phase rather than in the vapor phase, and therefore can penetrate deep lungs. The presence of citrate (-) ions complements the nicotine attributes by providing an inherent bitter taste and enhanced throat catch. This effect makes it feasible to further reduce the nicotine concentration.

[0086] Example 2 NMR spectroscopy was used to evaluate the interactions of cyclodextrin with menthol and cyclodextrin with nicotine (unprotonated and monoprotonated). Formulations containing the following were studied: Formulation 1 - Water 90.0% (w / w), l-menthol 0.9% (w / w), 2-hydroxypropyl-β-cyclodextrin 9% (w / w), and nicotine 0.1% (w / w). Formulation 2 - Water 89.9% (w / w), l-menthol 0.9% (w / w), 2-hydroxypropyl-β-cyclodextrin 9% (w / w), nicotine 0.1% (w / w), and citric acid 0.1% (w / w).

[0087] During the NMR studies, the diffusion coefficients of monoprotonated nicotine, unprotonated nicotine, and menthol, and their subsequent residence times within 2-hydroxypropyl-β-cyclodextrin were evaluated. The diffusion coefficients and subsequent residence times were studied because they are understood to provide a quantitative assessment of the degree of interaction of the compound with the complexing agent with each of the components studied. Encapsulation is understood to be not a static state, but a dynamic state in which a material may be encapsulated (e.g., complexed or bound to the encapsulating agent) for a period of time, then released and subsequently encapsulated. Thus, a decrease in residence time indicates a decrease in interaction with the complexing agent.

[0088] Diffusion-Ordered NMR Spectroscopy (DOSY) experiments were selected as suitable for evaluating the diffusion times of nicotine and individual components in menthol solutions in the above formulations and formulations spiked with varying component ratios (nicotine:2-hydroxypropyl-β-cyclodextrin:menthol=1:4:1, and nicotine:2-hydroxypropyl-β-cyclodextrin:menthol:citric acid=1:4:1:1 molar ratios).

[0089] The results of this study indicate that monoprotonated nicotine has a faster diffusion coefficient compared to nicotine without the use of citric acid and suggest that unprotonated nicotine has a longer residence time in 2-hydroxypropyl-β-cyclodextrin than monoprotonated nicotine. The results of this study suggest that the residence time of menthol in 2-hydroxypropyl-β-cyclodextrin was longer than the residence times of both monoprotonated and unprotonated forms of nicotine. The results also suggest that protonating nicotine reduces its interaction with 2-hydroxypropyl-β-cyclodextrin. The data suggest that the residence time of the compound in 2-hydroxypropyl-β-cyclodextrin was longer than the residence times of both monoprotonated and unprotonated forms of nicotine. Monoprotonated nicotine < unprotonated nicotine < menthol (Figure 2). This suggests that.

[0090] The inventors have found that protonating nicotine reduces its residence time in an encapsulation material such as cyclodextrin. This modification allows flavors to be encapsulated in preference to nicotine, particularly to protonated nicotine. Thus, a system is encouraged and provided in which the molar ratio of (a) an encapsulation material encapsulating one or more flavors to (b) unencapsulated flavors exceeds the molar ratio of (c) an encapsulation material encapsulating nicotine to (d) unencapsulated nicotine.
