Abstract
Description
[Technical field]
[0001]
[0001]
[0002] The present disclosure relates to disinfectant compositions containing hydrogen peroxide and a novel solvent system that are effective against tuberculosis-containing bacteria. [Background technology]
[0002]
[0003]
[0004] Among known disinfectants and antimicrobial agents, hydrogen peroxide is a suitable choice not only because of its potential as a biocide, but also because of its low toxicity, which decomposes into non-toxic oxygen and water. Furthermore, hydrogen peroxide's inherent ability to kill a broad range of microorganisms combined with the low toxicity of the products makes it an ideal active for disinfectant formulations for non-hard, porous surfaces.
[0003]
[0005] Unfortunately, hydrogen peroxide alone is not able to kill higher levels of organisms such as Mycobacterium tuberculosis, especially when it is difficult to provide shorter kill times (preferably <5 minutes). Thus, hydrogen peroxide is not very effective against certain organisms by itself. Hydrogen peroxide is not effective against Mycobacterium tuberculosis even with high activity levels and long contact times. Furthermore, most commercial hydrogen peroxide-based products are unable to kill Mycobacterium tuberculosis (TB) on hard surfaces within short contact times.
[0004]
[0006] There is a need in the art to provide the hospital disinfectant market with a disinfectant product that is effective in killing tuberculosis-causing bacteria and that exhibits rapid disinfecting properties after application to a surface. Summary of the Invention

[0087] Killing of Mycobacterium terrae (M. terrae) was evaluated using the OECD Quantitative Method for Evaluating the Bactericidal and Mycobactericidal Activity of Microbicides. This method provides a quantitative assessment of the performance against M. terrae of liquid antimicrobials designed for hard nonporous surfaces. The method is based on a guidance document from the Organization for Economic Cooperation and Development (OECD) and provides the log reduction (LR) as a quantitative measure of the efficacy of liquid disinfectants. The log reduction of M. terrae 10 The log reduction is often used as a preliminary screening tool to determine potential efficacy against M. tuberculosis, which is generally recognized as being more difficult to kill and more challenging to work with in the laboratory. 10 Reduction was assessed using a 3 minute contact time and 5% soil.
[0076]

[0083]
[0095] The invention is illustrated without limitation by the following examples. All parts and percentages are by weight unless otherwise indicated. EXAMPLES
[0084]
[0096] Example 1. Formulation
[0097] Table 1. Formulations
[0085] [Table 1]
[0086]
[0098] All formulation solutions in Table 1 can be present in their neat (ready to use) state or can be concentrated in two basic ways (shown as an example in Table 7 using Formulation 2). The first way is, for example but not limited to, simply making a 1:4 concentrate (which will be diluted with the appropriate amounts of hydrogen peroxide and water).
[0087]
[0099] Formulation 5 in Table 1 is an acidified hydrogen peroxide formulation containing a surfactant, but the solvent blend was not included in the composition, resulting in a low log reduction of M. terrae (see Table 2). Formulation 3 contains the ingredients of formulation 5 plus a new solvent blend, and showed a dramatic increase in the log reduction of M. terrae. Formulation 4 contains the ingredients of formulation 5 plus half the diethylene of the solvent blend, and showed little log reduction of M. terrae. Formulation 1 contains a new solvent blend ratio in the formulation, and showed a dramatic increase in the log reduction of M. terrae. Formulation 2 is similar to 4, but also contains 1.5% phenoxyethanol, which increased the log reduction of M. terrae. Formulation 6 is similar to 2, but the level of phenoxyethanol (amount, concentration) was doubled along with the butyl carbitol level, which dramatically increased the log reduction of M. terrae.
[0088]
[0100] It is important to note that the solvent blends containing glycol ethers and phenoxyethanol (formulations 7 and 2) had an increasing effect on the efficacy of M. terrae and TB. What is even more unexpected is that formulation 2, despite having a lower level of the solvent blend, managed to pass TB.
[0089]
[0101] Example 2. OECD Test Method for M. terrae with 3 minute contact time
[0102] Table 2. Results of OECD test methods for M. terrae
[0090] [Table 2]
[0091]
[0103] TB was evaluated using the tuberculinicidal test of the AOAC Germicidal Spray Test. This test involves testing Mycobacterium bovis (BCG) ATCC 35743 cultured in 7H11 growth medium for 21 ± 2 days at 36 ± 1°C. According to regulatory agency (EPA) guidelines, all formulations passed, no visible growth was observed in any of the subculture broth tubes per lot, and the controls met their prescribed criteria. All formulations passed BCG when tested as described above (containing 5% organic soil load, 3 minutes contact). All test controls met the established criteria for valid testing.
[0092]
[0104] However, what was particularly surprising was the significant effect that the solvent blends containing glycol ethers and phenoxyethanol (formulations 7 and 2) had on potency enhancement of M. terrae and TB. Even more unexpectedly, formulation 2, which contained an even lower level of the solvent blend, managed to pass TB.
[0093]
[0105] Example 3. TB Evaluation (Pass / Fail)
[0106] Table 3. TB rating at 3 min contact time
[0094] [Table 3]
[0095]
[0107] Example 4. Stability test
[0108] Table 4. Physical stability tests of disinfectant compositions
[0096] [Table 4]
[0097]
[0109] Formulations 1, 7, and 2 were filled into 4 ounce (113 g) glass jars with lids and placed into various controlled temperature ovens. Samples were removed and visually evaluated at the times and temperatures listed in Table 4. At the end of the four week period, all samples were found to have acceptable physical stability.
[0098]
[0110] Example 5. Stability of Hydrogen Peroxide
[0111] Table 5. Stability of hydrogen peroxide
[0099] [Table 5]
[0100]
[0112] The same formulations tested in Table 4 were also tested for H2O2 stability at 50° C. for 4 weeks (see Table 5). All formulations had fairly good peroxide stability with no greater than 6% w / w loss in H2O2 at 50° C. for 4 weeks.
[0101]
[0113] Calculate the percent reduction of hydrogen peroxide: % H2O2 reduction = (initial amount of H2O2 - amount of H2O2 after storage) / initial amount of H2O2, multiply this result by 100.
[0114] Example 6. AOAC Germicidal Spray Method for Disinfecting Hard Surfaces
[0115] Table 6. AOAC Germicidal Spray Methods for Disinfecting Hard Surfaces
[0102] [Table 6]
[0103]
[0116] Formulation 2 was micro tested against S. enterica, S. aureus, and P. aeruginosa using the AOAC germicidal spray method for hard surface disinfection under GLP conditions with a 5% soil load (fetal bovine serum) and a 1 minute contact time (see Table 6). Formulation 2 passed against all three organisms under GLP conditions.
[0104]
[0117] Example 7. Formulation No. 2
[0118] Table 7. Comparison of Formulation No. 2 (w / w%)
[0105] [Table 7]
[0106]
[0119] All formulation solutions in Table 1 can be present in their neat (ready to use) state or can be concentrated in two basic ways (shown as an example in Table 7 using Formulation 2). The first way is, for example but not limited to, simply making a 1:4 concentrate (which will be diluted with the appropriate amounts of hydrogen peroxide and water).
[0107]
[0120] Example 8. Concentrate pack formulation of Formulation No. 2
[0121] Table 8. Comparison of pack formulations of concentrates A and B
[0108] [Table 8]
[0109]
[0122] The second method is similar to the first, but instead of just removing the hydrogen peroxide from the concentrate, two solvents are also removed (Table 7).The concentrate is then diluted with water, hydrogen peroxide, and / or a solvent pack containing diethylene glycol monobutyl ether and phenoxyethanol (Table 8).
