Abstract
Description
[Technical field]
[0001] The present invention relates to protein kinase inhibitors that inhibit mitogen-activated protein kinase 4 (MKK4), and in particular to protein kinase inhibitors that selectively inhibit MKK4 over the protein kinases JNK1 and MKK7. [Background technology]
[0002] Liver disease can be caused by infection, injury, exposure to toxic compounds such as alcohol and drugs, autoimmune processes, genetic defects, and other factors. Although the liver has a remarkable regenerative capacity, in pathological conditions this may not function properly and therefore may be insufficient to compensate for the loss of liver cells and organ function.
[0003] US Patent No. 5,999,943 describes compounds that are protein kinase inhibitors useful for the treatment of diseases and conditions associated with abnormal activity of protein kinases. These compounds are inhibitors of Raf protein kinases, particularly B-Raf and c-Raf and their mutants, and are therefore useful for the treatment of cancer. They are also said to inhibit a wide variety of other protein kinases, particularly c-Jun N-terminal kinase (JNK), in particular JNK1.
[0004] Patent Document 2 has a similar disclosure, and Patent Documents 3 and 4 disclose modified compounds having Raf protein kinase inhibitory activity. H. Vin et al. refer to two compounds in Patent Document 5 as B-Raf inhibitors that suppress apoptosis via off-target inhibition of JNK signaling.
[0005] US Patent No. 6,399,663 describes pyrazolo[3,4-b]pyridine compounds that are protein kinase inhibitors useful in the treatment of diseases and conditions mediated by Raf protein kinase, such as cancer. In addition, they are said to inhibit a wide variety of other protein kinases, particularly c-Jun N-terminal kinase (JNK), especially JNK1. US Patent No. 6,399,663 discloses several compounds that are described as inhibitors of mitogen-activated protein kinase 4 (MKK4) and are useful in the treatment of liver failure, protection of liver cells from apoptosis, and regeneration of liver cells.
[0006] Non-Patent Document 1 describes a functional genetic approach for the identification of gene targets that can be used to enhance the regenerative capacity of hepatocytes. In particular, Non-Patent Document 1 identifies the protein kinase MKK4 as a key regulator of hepatocyte regeneration, and reports that inhibition of MKK4 increased hepatocyte regeneration through compensatory upregulation of MKK7 and JNK1-dependent activation of ATF2 and ELK1. Based on the findings of the prior art, it was concluded that MKK4 and JNK1 inhibitors may be useful for the treatment of JNK1-mediated diseases.
[0007] However, there is a recognized failure to treat liver disease with such compounds in clinical practice. [Prior art documents] [Patent documents]

[0038] The present invention also includes any tautomers, crystal forms and polymorphs of the compounds of the present invention and salts thereof, and mixtures thereof.
[0039] The present invention also includes solvates such as hydrates.
[0040] The compounds of the present invention may contain one or more chiral centers and therefore exist in different optically active forms, such as enantiomers and diastereomers.
[0041] As used herein, the term "prodrug" refers to an agent that is converted into the parent drug in vivo by some physiological chemical process. An example of a prodrug is, but is not limited to, an ester form of a compound of the present invention.
[0042] Prodrugs have many useful properties. For example, they may be more water soluble than the ultimate drug, facilitating intravenous administration of the drug. They may also have a higher level of oral bioavailability than the ultimate drug. After administration, the prodrug is enzymatically or chemically cleaved, and the ultimate drug is delivered to the blood or tissues.
[0043] Exemplary prodrugs include, but are not limited to, those in which the free hydrogen is (C1-C4) alkyl, (C1-C 12 )alkanoyloxy-methyl, (C4-C9)1-(alkanoyloxy)ethyl, 1-methyl-1-(alkanoyloxy)-ethyl having 5 to 10 carbon atoms, alkoxycarbonyloxymethyl having 3 to 6 carbon atoms, 1-(alkoxycarbonyl-oxy)ethyl having 4 to 7 carbon atoms, 1-methyl-1-(alkoxycarbonyloxy)-ethyl having 5 to 8 carbon atoms, N-(alkoxycarbonyl)aminomethyl having 3 to 9 carbon atoms, 1-(N-(alkoxycarbonyl)aminomethyl having 4 to 10 carbon atoms Examples of suitable carboxylic acid substituents include N,N-(C1-C2)ethyl, 3-phthalidyl, 4-crotono-lactonyl, γ-butyrolactone-4-yl, di-N,N-(C1-C2)alkylamino(C2-C3)alkyl (e.g., β-dimethylaminoethyl), carbamoyl-(C1-C2)alkyl, N,N-di(C1-C2)-alkylcarbamoyl-(C1-C2)alkyl, and compounds having carboxylic acid substituents substituted with piperidino-, pyrrolidino-, or morpholino(C2-C3)alkyl.
[0044] Other exemplary prodrugs release alcohols of formula (I), where the free hydrogen of the hydroxyl substituent (e.g., an R group containing a hydroxyl) is (C1-C6)alkanoyloxy-methyl, 1-((C1-C6)alkanoyloxy)-ethyl, 1-methyl-1-((C1-C6)alkanoyloxy)ethyl, (C1-C 12)alkoxy-carbonyl-oxymethyl, N-(C1-C6)-alkoxy-carbonylaminomethyl, succinoyl, (C1-C6)alkanoyl, α-amino(C1-C4)alkanoyl, aryl, acyl, and α-aminoacyl or α-aminoacyl-α-aminoacyl, where the α-aminoacyl moieties are independently either naturally occurring L-amino acids found in proteins, P(O)(OH)2, -P(O)(O(C1-C6)alkyl)2, or glycosyl (the group resulting from removal of the hydroxyl of a carbohydrate hemiacetal).
[0045] The phrase "MKK4 inhibitor" means one which upon administration inhibits the kinase activity of MKK4 with an IC50 of <10 μmol / l, preferably <1 μmol / l, in particular <0.5 μmol / l. As used herein, the expression "selectively inhibits the protein kinase MKK4 over the protein kinases JNK1 and MKK7" means that the ratio of MKK7 inhibitory activity to MKK4 inhibitory activity, or the ratio of JNK1 inhibitory activity to MKK4 inhibitory activity, expressed either as a percentage of the control or as a Kd, is ≧10, as measured using KINOMEscan™.
[0046] As used herein, the expression "promoting liver regeneration or inhibiting or preventing liver cell death" means that the relative number of proliferating liver cells is increased by at least 30%, preferably at least 50%, compared to the number of proliferating cells at the start of treatment. In particular, the expression means an increase of ≧100% compared to the number of proliferating cells at the start of treatment.
[0047] In this context, experimental measurements and quantification are performed using standard methods, such as the quantification of the protein Ki67, which is precisely related to cell proliferation. For the quantification of proliferating hepatocytes in tissue slides, several immunohistochemical standard methods are available, which use a primary anti-Ki67 antibody, followed by visualization of anti-Ki67 binding, for example, using a horseradish peroxidase-conjugated secondary antibody. The amount of peroxidase activity, visualized by enzymatic conversion of a chromogenic substrate, correlates with the amount of Ki67 protein and the number of proliferating cells.
[0048] In the experiments described below, hepatocyte proliferation was quantified by Ki67 staining using the fluorophore tetramethylrhodamine containing primary polyclonal rabbit anti-Ki67 antibody from Abcam (cat. no. ab 15580, Abcam, Cambridge, USA) and secondary goat polyclonal antibody from Invitrogen (cat. no. 16101, Invitrogen / ThermoFisher).
[0049] Based on data from several preclinical mouse models, it was found that shRNA-mediated inhibition of MKK4 in a mouse model of chronic CCl4-induced liver injury increased hepatocyte proliferation from 13% to 27% (compared to control shRNA) and was associated with reduced liver damage (transaminases) and decreased liver fibrosis.
[0050] According to the definition in the previous chapter, the relative increase in proliferating cells was 108%. In a model of alcohol-induced steatohepatitis (ASH), shRNA-mediated silencing of MKK4 resulted in a 4% hepatocyte proliferation rate compared to 2% with control shRNA (relative increase: 100%). Repeated hepatocyte proliferation was associated with reduced steatosis (fatty deposits) and reduced liver damage measured by transaminases. Similarly, in a partial hepatectomy model (48 hours after surgical resection of two-thirds of the liver), shRNA-mediated MKK4 silencing increased hepatocyte proliferation from 16% (control shRNA) to 33% (relative increase: 106%). Again, increased hepatocyte proliferation was associated with improved liver regeneration and faster recovery of liver mass.
[0051] The organic moieties mentioned in the definitions of the variables above are generic to the individual lists of members of the individual groups, as is the term halogen. n -C m indicates in each case the possible number of carbon atoms in the group.
[0052] The term halogen stands in each case for fluorine, bromine, chlorine or iodine, in particular for fluorine or chlorine.
[0053] Alkyl is a linear or branched alkyl group, preferably a C1-C6-alkyl group (i.e. an alkyl group having 1 to 6 carbon atoms), more preferably a C1-C4-alkyl group. Examples of alkyl groups are methyl, ethyl, n-propyl, isopropyl, n-butyl, 2-butyl, isobutyl, tert-butyl, pentyl, 1-methylbutyl, 2-methylbutyl, 3-methylbutyl, 2,2-dimethylpropyl, 1-ethylpropyl, hexyl, 1,1-dimethylpropyl, 1,2-dimethylpropyl, 1-methylpentyl, 2-methylpentyl, 3-methylpentyl, 4-methylpentyl, 1,1-dimethylbutyl, 1,2-dimethylbutyl, 1,3-dimethylbutyl, 2,2-dimethylbutyl, 2,3-dimethylbutyl, 3,3-dimethylbutyl, 1-ethylbutyl, 2-ethylbutyl, 1,1,2-trimethylpropyl, 1,2,2-trimethylpropyl, 1-ethyl-1-methylpropyl, and 1-ethyl-2-methylpropyl.
[0054] The definition of alkyl is equally applicable to any group that contains an alkyl group.
[0055] Cycloalkyl is an alicyclic group, preferably C3-C8-cycloalkyl, i.e. a cycloalkyl group having 3 to 8 carbon atoms. In particular, 3 to 6 carbon atoms form a ring structure, such as cyclopropyl, cyclobutyl, cyclopentyl and cyclohexyl. The ring structure may be unsubstituted or may carry 1, 2, 3 or 4 C1-C4 alkyl groups, preferably one or more methyl groups.
[0056] The compounds of the present invention (including pharma- ceutically acceptable salts, prodrugs, biologically active metabolites, solvates and stereoisomers thereof) can be prepared as disclosed in WO2007 / 002433, the entirety of which is incorporated herein by reference, or by similar procedures. Acid or base addition salts are prepared in a conventional manner by mixing a free base with the corresponding acid, or by mixing a free acid with the desired base. Optionally, the reaction is carried out in a solution of an organic solvent, for example a lower alcohol such as methanol, ethanol or propanol, an ether such as methyl tert-butyl ether or diisopropyl ether, a ketone such as acetone or methyl ethyl ketone, or an ester such as EtOAc.
[0057] The compounds of the present invention are useful for promoting liver regeneration or inhibiting or preventing liver cell death while simultaneously increasing liver cell proliferation. Thus, the compounds of the present invention are useful for treating, regulating, ameliorating, or preventing diseases involving acute or chronic damage to the liver, which may be caused by infection, injury, exposure to toxic compounds, abnormal accumulation of normal substances in the blood, autoimmune processes, genetic defects, or unknown causes.
[0058] These liver diseases include all diseases in which it is believed to be useful to promote liver regeneration and inhibit or prevent liver cell death in order to achieve a potential therapeutic effect (i.e. partial or complete restoration of liver function). These diseases include: Acute and chronic liver disease or acute exacerbation of liver disease, for example acute and chronic viral hepatitis such as hepatitis B, hepatitis C, hepatitis E, Epstein-Barr virus, cytomegalovirus, herpes simplex virus and other viral hepatitis, all types of autoimmune hepatitis, primary sclerosing hepatitis, alcoholic hepatitis;
[0059] Metabolic liver diseases, such as metabolic syndrome, fatty liver such as nonalcoholic fatty liver (NAFL), nonalcoholic steatohepatitis (NASH), alcoholic steatohepatitis (ASH), Wilson's disease (Morbus Wilson), hemochromatosis, alpha1-antitrypsin deficiency, glycogen storage disease;
[0060] All types of cirrhosis, e.g. primary biliary cirrhosis, ethyl toxic cirrhosis, cryptogenic cirrhosis;
[0061] Acute (fulminant) or chronic liver failure, e.g. acetaminophen (paracetamol) induced liver failure, alpha-amanitin induced liver failure, drug induced hepatotoxicity e.g. liver failure caused by antibiotics, nonsteroidal anti-inflammatory drugs and anticonvulsants, toxic liver failure such as acute liver failure caused by herbal supplements (e.g. kava, ephedra, skullcap, pennyroyal), liver disease and liver failure due to vascular diseases such as Budd-Chiari syndrome, acute liver failure of unknown cause, chronic liver disease due to right heart failure;
[0062] Galactosemia, cystic fibrosis, porphyria, hepatic ischemia / perfusion injury, small-for-size graft syndrome after liver transplantation, primary sclerosing cholangitis or hepatic encephalopathy.
[0063] To promote liver regeneration or inhibit or prevent liver cell death, the compounds of the present invention are administered to patients in need thereof in therapeutically effective amounts. To detect the presence of liver disease, various diagnostic methods are available. Blood levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) that are above the clinically accepted normal range are known to indicate ongoing liver damage. Blood bilirubin levels or other liver enzymes are used as detection or diagnostic criteria.
[0064] Routine monitoring of liver disease patients for blood levels of ALT and AST is used to measure the progression of liver disease during treatment. The reduction of elevated ALT and AST levels to acceptable limits is considered clinical evidence reflecting a decrease in the severity of the patient's liver damage. Commercial tests such as FibroTest / FibroSURE, HepaScore™, FibroMeter or Cirrhometer evaluate the combined results of five or more biochemical parameters for the detection of fatty liver, fibrosis and cirrhosis. In addition, non-invasive and innovative physical imaging methods such as magnetic resonance imaging, sonography and especially elastography are available to detect and monitor the status and progression of liver disease.
[0065] Furthermore, shRNA-mediated inhibition of MKK4 has been shown to attenuate TNF-α-driven cartilage matrix degradation in osteoarthritis (Cell Death and Disease (2017)8,e3140). Thus, inhibition of MKK4 activity using the compounds of the present invention is further useful for the treatment of osteoarthritis and rheumatoid arthritis.
[0066] The compounds of the present invention are conventionally administered in the form of a pharmaceutical composition containing at least one compound according to the invention, optionally together with an inert carrier (e.g., a pharma- ceutically acceptable excipient), and, where appropriate, other drugs. These compositions can be administered, for example, orally, rectally, transdermally, subcutaneously, intraperitoneally, intravenously, intramuscularly, or intranasally.

Any and all treatment regimens in which a patient receives separate but simultaneous or overlapping therapeutic administration of a compound of the present invention and at least one additional therapeutic agent are within the scope of the present invention. In one embodiment of adjunctive therapeutic administration described herein, a patient is typically stabilized for a period of time on a therapeutic administration of one or more components, and then receives administration of another component.
[0072] The combination therapy of the present invention can also be administered simultaneously. By simultaneous administration, it is meant that each component is administered together in the form of a single pharmaceutical composition or device that has or contains both components, or as separate compositions or devices, each of which contains one of the components that is administered simultaneously. This combination of each separate component for simultaneous combination can be provided in the form of a kit of parts.

[0130] Example 15: Biological activity The kinase activity of the compounds of the present invention was measured using the 33PanQinase™ assay service provided by ProQinase GmbH (Freiburg, Germany). Detailed assay conditions are disclosed on the ProQinase website (https: / / www.proqinase.com / products-services-biochemical-assay-services / kinase-assays). Briefly, all kinase assays were performed in 50 μl reaction volumes using 96-well FlashPlates™ from PerkinElmer (Boston, MA, USA). The reaction cocktail was pipetted in four steps in the following order: 20 μl Assay Buffer (standard buffer) 5 μl of ATP solution (in H2O) 5 μl of test compound (10% DMSO solution) 20 μl enzyme / substrate mix
[0131] All protein kinase assays contained 70 mM HEPES-NaOH pH 7.5, 3 mM MgCl2, 3 mM MnCl2, 3 μM Na-orthovanadate, 1.2 mM DTT, 50 μg / ml PEG20000, ATP (variable concentrations, corresponding to the apparent ATP-Km of each kinase), [γ-33P]-ATP (approximately 8 × 1005 cpm per well), protein kinase and substrate. The reaction cocktail was incubated for 60 min at 30° C. The reaction was stopped with 50 μl of 2% (v / v) H3PO4, and the plates were aspirated and washed twice with 200 μl of 0.9% (w / v) NaCl. 33 Pi incorporation was determined using a microplate scintillation counter (Microbeta, Wallac). All assays were performed on a BeckmanCoulter / SAGIAN™ core system.
[0132] For single concentration assays, efficacy of test compounds is expressed as % remaining activity. To determine IC50 values, serial dilutions were tested with a final concentration range of 100 μM to 3 nM (10 concentrations). The fitting model for the determination of IC50 values was "sigmoidal response (variable slope)" with parameters "top" fixed at 100% and "bottom" fixed at 0%. The fitting method used was least squares.
[0133] MKK4 Efficacy:
[0134] [Table 4]
[0135] The selectivity of test compounds against BRaf, JNK1, and MKK7, all referred to as off-targets, was calculated by the ratio IC50(off-target) / IC50(MKK4) and classified as follows:
