Abstract
Description
FIELD OF THEINVENTION
[0001] The present disclosure relates to aerosolizable formulations, methods of making aerosolizable formulations, containers containing aerosolizable formulations, devices containing aerosolizable formulations, and methods and uses of aerosolizable formulations.
[0002] Electronic aerosol delivery systems, such as e-cigarettes, generally contain a reservoir of liquid to be vaporized, typically containing nicotine. When a user inhales on the device, a heater is activated to vaporize a small amount of the liquid, which is then inhaled by the user.
[0003] Use of e-cigarettes in the UK is growing rapidly and it is now estimated that there are over one million e-cigarette users in the UK.
[0004] One challenge faced in providing such a system is to deliver an inhaled aerosol from the aerosol delivery device that provides an acceptable experience to the consumer. Some consumers may prefer e-cigarettes that generate an aerosol that "resembles" the smoke inhaled from tobacco products such as cigarettes. The aerosol from e-cigarettes and the smoke from tobacco products such as cigarettes provide the user with a complex chain of flavors in the mouth, nicotine absorption in the mouth and throat, and subsequently nicotine absorption in the lungs. These various aspects are described by users in terms of flavor, strength / quality, impact, spiciness / smoothness, and nicotine reward. Nicotine contributes to several of these factors and is strongly associated with factors such as spiciness, spiciness, and smoothness, which are easily perceived by consumers, and e-cigarettes may give consumers too much or too little of these parameters, depending on personal preferences. Nicotine reward is particularly complex because it is a result of both the amount and rate at which nicotine is absorbed through the oral mucosa (which is typically nicotine in the vapor phase) and the amount and rate at which nicotine is absorbed through the lungs (which is typically nicotine in the particle phase of the inhaled aerosol). Each of these factors, and the balance of factors, can strongly contribute to consumer acceptability of an e-cigarette. Thus, delivery measures that optimize the overall vaping experience are desirable for e-cigarette manufacturers.
[0005] A further challenge facing such systems is the ongoing demand for harm reduction: the harm caused by cigarettes and e-cigarette devices comes primarily from toxins, and therefore it is desirable to reduce or eliminate components that may produce toxins. Summary of the Invention
[0006] In one embodiment, (i) water in an amount of at least 50 wt.% based on the aerosolizable formulation; (ii) Nicotine wherein nicotine is present in an amount of 1 wt.% or less based on the aerosolizable formulation.
[0007] In one embodiment, a method for generating an aerosol comprises: (i) water in an amount of at least 50 wt. % based on the aerosolizable formulation; and (ii) Nicotine wherein nicotine is present in an amount of 1 wt.% or less based on the aerosolizable formulation.
[0008] In one embodiment, (a) a container; (b) (i) water in an amount of at least 50 wt. % based on the aerosolizable formulation; and (ii) Nicotine and an aerosolizable formulation comprising: wherein nicotine is present in an amount of 1 wt.% or less based on the aerosolizable formulation.
[0009] In one embodiment, an electronic aerosol delivery system is provided, comprising: (a) an aerosolization device for aerosolizing the formulation for inhalation by a user of the electronic aerosol delivery system; (b) a power source comprising a cell or battery for powering the aerosolization device; and (c) (i) water in an amount of at least 50 wt. % based on the aerosolizable formulation; and (ii) Nicotine and an aerosolizable formulation comprising: and wherein the nicotine is present in an amount of 1 wt.% or less based on the aerosolizable formulation.
[0010] As discussed herein, an aerosolizable formulation is provided, comprising (i) water in an amount of at least 50 wt.% based on the aerosolizable formulation, and (ii) nicotine, wherein the nicotine is present in an amount of 1 wt.% or less based on the aerosolizable formulation. The inventors have found that it is possible to provide an advantageous system in which the aerosolizable formulation comprises water in an amount of at least 50 wt.% based on the aerosolizable formulation, and comprises nicotine in an amount of 1 wt.% or less based on the aerosolizable formulation. The inventors have surprisingly found that in such a high water system, providing nicotine in a maximum amount of 1 wt.% or less provides optimal sensory perception to the user. In particular, controlling the nicotine to this maximum amount provides the nicotine "hit" and / or sensory perception preferred by the user. This desirable nicotine "hit" and / or flavor release is in contrast to "traditional" e-liquids based on glycerol and propylene glycol, which users sometimes avoid due to a perceived difference in the nicotine "hit" and / or flavor release provided by tobacco-based cigarettes.
[0011] The inventors have also discovered that it is possible to provide an aqueous system in which an aerosolized formulation is generated at low temperature from an aerosolizable formulation. This is in contrast to "conventional" e-cigarettes that use heaters that are typically applied to glycerol and propylene glycol based liquids to generate an aerosolized formulation. The provision of such a system and avoidance of heating can address the prior art problems of toxicant generation. More specifically, the inventors have discovered an aqueous system that provides the desired nicotine "hit" and / or flavor release by selecting a very high water content, but avoids the need to use heaters associated with conventional glycerol and propylene glycol based liquids. In the inventive system, the number of components present can be reduced, leading to a reduced chance of decomposition product / toxicant generation. In particular, the use of water allows for the replacement of some or all of the glycerol, propylene glycol, 1,3-propanediol and mixtures thereof that are typically used in e-cigarettes.

[0026] The relevant amount of nicotine present in the formulation in protonated form is specified herein. These amounts can be easily calculated by those skilled in the art. Nicotine (3-(1-methylpyrrolidin-2-yl)pyridine) is a dibasic acid with a pKa of 3.12 for the pyridine ring and 8.02 for the pyrrolidine ring. Nicotine can exist in protonated (mono- and di-) and non-protonated (free base) forms with different bioavailability depending on pH. [ka]
[0027] The distribution of protonated and unprotonated nicotine will change at various pH increments. [ka]
[0028] The proportion of unprotonated nicotine dominates at high pH levels, while lowering the pH will see an increase in the proportion of protonated nicotine (mono- or di-, depending on the pH). If the relative proportion of protonated nicotine and the total amount of nicotine in a sample are known, the absolute amount of protonated nicotine can be calculated.
[0029] The relative proportion of protonated nicotine in a formulation can be calculated by using the Henderson-Hasselbalch equation, which expresses pH as a derivation of the acid dissociation constant equation, and which is widely used in chemical and biological systems. Consider the following equilibrium: [ka]
[0030] The Henderson-Hasselbalch equation for this equilibrium is:
number
[0031] where [B] is the amount of unprotonated nicotine (i.e., the free base), [BH+] is the amount of protonated nicotine (i.e., the conjugate acid), and pKa is the reference pKa value of the pyrrolidine ring nitrogen of nicotine (pKa=8.02). The relative proportion of protonated nicotine can be derived from the alpha value of unprotonated nicotine calculated from the Henderson-Hasselbalch equation as follows:

[0068] The invention will now be described with reference to the following non-limiting examples.
[0069] Example 1 A series of tests were conducted using a commercially available vibrating mesh nebulizer device. A formulation containing 88.0% (w / w) water, 6.0% propylene glycol with vanilla flavor, and 6.0% glycerol was loaded into the "nicotine free" device.
[0070] Similar devices were prepared in which 1.2% w / w nicotine was added to the formulation and the water content was correspondingly reduced to 86.8% (w / w).
[0071] Similar devices were prepared in which 0.6% w / w nicotine was added to the formulation and the water content was adjusted accordingly to 87.4% (w / w).
[0072] Similar devices were prepared in which 0.3% w / w nicotine was added to the formulation and the water content was adjusted accordingly to 87.7% (w / w).
[0073] Similar devices were prepared in which 0.1% w / w nicotine was added to the formulation and the water content was adjusted accordingly to 87.9% (w / w).
[0074] Each of these devices was given to eight subjects, including e-cigarette users, who were asked to take five puffs on each device in a sequential monadic manner and to identify their preferred nicotine strength from the four provided to them.
[0075] Eight panelists preferred a nicotine strength below 1.2% (w / w), seven preferred a nicotine strength below 0.6% (w / w), and five preferred a nicotine strength below 0.3% (w / w).
[0076] Example 2 Three flavoured formulations with different water ratios and nicotine contents were evaluated by up to ten experienced vapers with different nicotine tolerances.
[0077] A series of tests were conducted using a commercially available vibrating mesh nebulizer device. The device was filled with a formulation containing 97.2% (w / w) water, 2.0% (w / w) propylene glycol with amaretto flavor, and 0.8% (w / w) nicotine.
[0078] Similar devices were prepared in which 0.9% (w / w) nicotine was added to the formulation and the water content was correspondingly reduced to 97.1% (w / w).
[0079] Similar devices were prepared in which 1.1% (w / w) nicotine was added to the formulation and the water content was correspondingly reduced to 96.9% (w / w).
[0080] A summary of the tested formulations and their properties is given in the table below. Panelists were asked to rate the formulations in a randomized fashion. During the sensory evaluation, panelists ranked several sensory attributes, such as throat irritation and chest impact, from low (1) to high (9). Panelists were also asked to indicate which formulation they liked best among those tested.
[0081] [Table 1]
[0082] Four out of ten monitors preferred Amaretto with a nicotine strength of 0.8% (w / w), three out of ten monitors preferred Amaretto with a nicotine strength of 0.9% (w / w), two out of ten monitors who were high nicotine users preferred Amaretto with a nicotine strength of 1.1% (w / w), and one out of ten monitor did not prefer any of the formulations. Overall, seven out of ten preferred Amaretto formulations containing less than 1% (w / w) nicotine.
[0083] An additional formulation also containing (β)-cyclodextrin was prepared and tested by six subjects in the same manner as the Amaretto formulation.
[0084] The devices were filled with a formulation containing 73.4% (w / w) water, 10.0% (w / w) 2-hydroxypropyl-β-cyclodextrin, 13% (w / w) glycerol, 2.0% (w / w) propylene glycol with mojito flavoring, 0.8% (w / w) nicotine, and 0.8% (w / w) benzoic acid.
[0085] Similar devices were prepared in which 0.9% (w / w) nicotine was added to the formulation and the water content was correspondingly reduced to 73.2% (w / w).
[0086] Similar devices were prepared in which 1.1% (w / w) nicotine was added to the formulation and the water content was correspondingly reduced to 72.8% (w / w).
[0087] A summary of the formulations tested and their properties is given in the table below.
[0088] [Table 2]
[0089] Four out of six monitors preferred the Mojito with a nicotine strength of 0.8% (w / w), one out of six monitors preferred the Mojito with a nicotine strength of 1.1% (w / w) and one out of six monitors did not prefer any of the formulations. Overall, four out of six preferred the Mojito formulation containing less than 1% nicotine. Nicotine hit, kick and impact were evaluated. These are shown in Figure 2. As can be seen from Figure 2, the Mojito containing 1.1% (w / w) nicotine formulation was rated the highest in terms of nicotine hit, kick and impact.
