Abstract
Description
Biofilm disinfectant composition
[0001] The present invention relates to a biofilm disinfectant composition and a method for disinfecting a biofilm.
[0002] BACKGROUND ART Once bacteria attach to an object, they multiply and form a biofilm. Biofilms, also known as biofilms or slimes, generally refer to three-dimensional structures formed by microorganisms such as bacteria, enveloping them with polymeric substances such as polysaccharides, proteins, and nucleic acids produced by the microorganisms as they attach to and multiply on the surface of a material. Biofilms are a problem for individuals in a variety of situations. For example, they cause unpleasant slimy stains and foul odors in household wet environments such as kitchens, sinks, and bathroom drains. Biofilms are also prone to forming and causing problems not only in these direct water environments, but also in situations where water, moisture, and dirt coexist, such as scratches on cutting boards, the inside of dishwashing sponges, washing machine drums, and fabric products such as clothing, dish towels, dustcloths, and foot mats. Furthermore, detergents containing various surfactants are widely used to clean textile products such as clothing. In recent years, with increasing hygiene awareness among consumers, interest in bacteria and viruses adhering to the surfaces of textile products has also increased. As a result, consumers take the extra effort and expense of washing items such as socks and sweaty clothing, which are prone to bacterial growth, by soaking them in cleaning solution for a while, or by using disinfectants or bleach in combination with textile cleaners.
[0003] US Patent No. 5,728,667 discloses germicidal light-duty dishwashing detergent compositions, either gel or liquid, containing dioctyldimethylammonium salts and alkyl ether carboxylate salts.
[0004] Biofilms attached to target articles, particularly textile products, are extremely difficult to remove because the polymers that make up the biofilm function as a protective barrier that protects bacteria from various bases, and it is also extremely difficult to sterilize the bacteria present within the biofilm. The present invention provides a biofilm disinfectant composition that has excellent disinfecting properties against bacteria present within biofilms attached to target articles, particularly textile products, and a method for sterilizing biofilms using the same.

[0012] According to the present invention, there are provided a biofilm disinfectant composition that has excellent disinfecting properties against bacteria present in biofilms attached to target articles, particularly textile products, and a method for disinfecting biofilms using the same.
[0013] Modes for Carrying Out the Invention The reason why the biofilm disinfectant composition of the present invention and the method for disinfecting biofilms using the same have excellent disinfecting properties against bacteria present in biofilms attached to target objects, particularly textile products, is not entirely clear, but is presumed to be as follows. In the present invention, it is believed that the combined use of a specific cationic surfactant (component (a)) and a specific anionic surfactant (component (b)) allows the base to penetrate into the biofilm and act thereon, thereby enabling biofilm disinfection. Specifically, the specific cationic surfactant (component (a)) and the specific anionic surfactant (component (b)) form a loose ion complex, preventing the bactericidal component (a) from nonspecifically adsorbing to the biofilm surface, and allowing the ion complex of components (a) and (b) to penetrate into the biofilm. In addition, the penetrated ion complex has a moderate cationicity, which is thought to act on bacteria and enable disinfection. It should be noted that the present invention is not limited to the above-mentioned mechanism of action.

The number of carbon atoms in the internal olefin sulfonate in component (b2) refers to the number of carbon atoms in the internal olefin to which the sulfonate is covalently bonded.
[0029] The internal olefin sulfonate (b2) is a sulfonate obtained by sulfonating, neutralizing, and hydrolyzing a raw material internal olefin (an olefin having a double bond within the olefin chain) preferably having 10 or more carbon atoms, more preferably 14 or more carbon atoms, and preferably 20 or less, more preferably 18 or less carbon atoms. Such internal olefins also include those containing trace amounts of so-called alpha olefins (hereinafter also referred to as α-olefins), in which the double bond is located at the first position of the carbon chain. Furthermore, sulfonation of internal olefins quantitatively produces β-sultone, and a portion of the β-sultone is converted to γ-sultone and olefin sulfonic acid. These are further converted to hydroxyalkanesulfonate and olefin sulfonate in the neutralization and hydrolysis steps (e.g., J. Am. Oil Chem. Soc. 69, 39 (1992)). The hydroxy group of the resulting hydroxyalkanesulfonate is located within the alkane chain, while the double bond of the olefin sulfonate is located within the olefin chain. The resulting product is primarily a mixture of these compounds, and may contain trace amounts of hydroxyalkanesulfonates having a hydroxy group at the end of the carbon chain or olefinsulfonates having a double bond at the end of the carbon chain. In this specification, these products and their mixtures are collectively referred to as internal olefinsulfonates (component (b2)). Hydroxyalkanesulfonates are also referred to as hydroxy forms of internal olefinsulfonates (hereinafter also referred to as HAS forms), and olefinsulfonates are also referred to as olefin forms of internal olefinsulfonates (hereinafter also referred to as IOS forms). The mass ratio of the HAS form to the IOS form of the compounds in component (b2) can be measured using a high-performance liquid chromatography mass spectrometer (hereinafter abbreviated as HPLC-MS). Specifically, the mass ratio can be determined from the HPLC-MS peak area of component (b2).

The contents of compounds in component (b2) in which the sulfonic acid group is located at different positions can be measured by HPLC-MS. In this specification, the content of each compound having a sulfonic acid group at a different position is determined as a mass ratio based on the HPLC-MS peak area of the compound having the sulfonic acid group at each position in the total HAS form of component (b2).
[0034] The content of olefin sulfonate salts in which the sulfonic acid group is at position 1 in component (b2) is preferably 10% by mass or less, more preferably 7% by mass or less, even more preferably 5% by mass or less, and even more preferably 3% by mass or less in component (b2) from the viewpoint of biofilm sterilization, and is preferably 0.01% by mass or more from the viewpoint of reducing production costs and improving productivity. [0107] [Preparation of biofilm disinfectant composition] The preparation method of the biofilm disinfectant composition shown in Tables 1 to 7 is as follows. A 3 cm long Teflon stirrer piece was placed in a 200 mL glass beaker, followed by 10 g of ion-exchanged water, and the components (a) or (a'), (b) or (b') were added in the formulations shown in Tables 1 to 7. The mixture was stirred at 100 r / min for 10 minutes, and 95% by mass of the amount of ion-exchanged water needed to be added to make the total amount 100 g was added. After stirring at 100 r / min for 5 minutes, the pH (25 ° C.) was adjusted to 8 with monoethanolamine or hydrochloric acid (Examples 35 and 36 in Table 7 were adjusted to the pH listed in the table). The total amount was 100 g. The ion-exchanged water was added, and the mixture was stirred at 100 r / min for 15 minutes. The biofilm disinfectant composition shown in Tables 1 to 7 was prepared. After preparation, the pH was checked again, and the pH of each biofilm disinfectant composition in Tables 1 to 7 was found to be unchanged from the pH at the time of preparation. The amounts of each component in each biofilm disinfectant composition shown in Tables 1 to 7 are all active components. For Example 29 in Table 4, the amounts of each component in the diluted test treatment solution are shown below.
[0108] [Biofilm disinfection evaluation] (1-1) Preparation of biofilm-formed fabrics Glycerol stock solutions of Rhodotorula mucilaginosa (clothing isolate) and Methylobacterium variable (clothing isolate) were smeared on potato dextrose agar medium (manufactured by Difco Laboratories) and statically cultured for 2 days at 30 ° C. A glycerol stock solution of Moraxella osloensis (clothing isolate) was smeared on SCD agar medium (Shiotani MS Co., Ltd. SCD agar medium "Daigo", for general bacterial testing) and statically cultured for 1 day at 37 ° C. After culturing, the bacterial cells were scraped off with a disposable loop (As One Corporation, Disposable Loop 10 Type) and suspended in R2A medium (Shiotani MS Corporation, R2A medium "Daigo"). The turbidity (OD600) of each bacterial suspension was measured using a spectrophotometer (Apel Corporation, Model No. PD-303S). The bacterial solution was diluted to an OD600 of 0.2 for R. mucilaginosa and 0.01 for M. variable and M. osloensis. Hiraori cotton cloth (Yagashira Shoten, Cotton 2003) cut into 3 x 3 cm pieces was autoclaved in an autoclave (PHC Corporation, High-Pressure Steam Sterilizer, Lab Autoclave, Model No. MLS-3781). The sterilized Hiraori cotton cloth was then placed in the No. The samples were placed in a No. 6 screw tube (Maruem Corporation, screw tube bottle No. 6), and 1 mL of each of the three bacterial strains and 7 mL of R2A medium were added. The tubes were then sealed with a lid and cultured for two days at 30°C and 200 rpm in a small constant-temperature shaking incubator, Bioshaker (TAITEC Corporation, model number: BR-23FP). After culturing, the Hiraori cotton fabric was transferred to a new No. 6 screw tube, and 10 mL of autoclave-sterilized ion-exchanged water was added. The fabric was washed by shaking for 3 minutes at 25°C and 150 rpm in a small constant-temperature shaking incubator, Bioshaker. After washing, the Hiraori cotton fabric was air-dried for 1 hour in a safety cabinet to prepare a biofilm-forming fabric. Biofilm formation on the fabric was confirmed by loosening the fabric into threads with tweezers, staining the threads, and observing them with a confocal laser scanning microscope (LSM880 ZEN, manufactured by Carl Zeiss KK).The threads to be observed were stained with a dye for staining live cells (C375-Cellstain™-CFSE, manufactured by Dojindo Laboratories, Inc.), a dye for staining dead cells (P378-Cellstain™-PI solution, manufactured by Dojindo Laboratories, Inc.), a dye for staining cellulose (Calcofluor White staining solution, manufactured by Sigma-Aldrich), a dye for staining sugars (Lectin PNA From Arachis hypogaea (peanut), Alexa Fluor™ 647 Conjugate, manufactured by Thermo Fisher Scientific Co., Ltd.), a dye for staining sugars (Concanavalin Alexa Fluor™ 647, manufactured by Thermo Fisher Scientific Co., Ltd.), and a dye for staining sugars (Lectin PNA From Arachis hypogaea (peanut), Alexa Fluor™ 647 Conjugate, manufactured by Thermo Fisher Scientific Co., Ltd.). The cells were stained with HCl (Conjugate) for 30 minutes.
[0109] (1-2) Preparation of biofilm-forming hard surfaces For Example 31 in Table 5, 0.3 mL of each of the three bacterial species and 2.1 mL of R2A medium were added per well of a polystyrene 6-well plate (Corning Falcon Cell Culture 6-well multiwell plate with flat bottom lid), and the plate was sealed with a lid and allowed to stand at 30 °C for 2 days. After incubation, the upper layer of the 6-well plate was absorbed and air-dried for 1 hour in a safety cabinet to form a biofilm-forming hard surface. Biofilm formation on the hard surface was confirmed by observation with a digital microscope (Keyence Corporation VHX-7000).
[0110] (2) Biofilm Sterilization Test (Biofilm Sterilization Evaluation) The prepared biofilm-formed cloth was cut into 1.5 × 1.5 cm pieces, and the sterilized Hiraori cotton cloth was cut into 1.5 × 3 cm pieces. A total of three pieces, one cut biofilm-formed cloth and two cut sterilized Hiraori cotton cloths, were stacked and placed in a 5 mL tube (Eppendorf Tube 5.0 mL, model number: 0030 119.401, manufactured by Eppendorf Co., Ltd.). To this was added 3 mL of a test treatment solution prepared by diluting each of the biofilm disinfectant compositions in Tables 1 to 7 3000 times with model tap water (4 ° dH) (in Table 4, Example 29 used the test treatment solution diluted to the concentration listed in the table, and Example 30 was used as is at the concentration listed in the table without dilution. For Examples 32, 33, and 34 in Table 6, the hardness of the model tap water for dilution 3000 times was changed to that listed in the table to prepare a test treatment solution.) The solution was then washed by shaking in a small thermostatic shaking incubator bioshaker at 25 ° C. and 150 rpm for 10 minutes, which was the washing step. The washed cloth was transferred to a 5 mL tube containing 3 mL of model tap water and washed by shaking for 3 minutes under the same conditions, which was the rinsing step. This washing and rinsing step was repeated four times, after which only the biofilm-formed cloth was removed and air-dried in a safety cabinet for 30 minutes. For biofilm-forming hard surfaces, 3 mL of the test treatment solution prepared by diluting the biofilm disinfecting composition of Example 31 in Table 5 30,000 times with model tap water (4 ° dH) was added per well, and the well was shaken and washed for 10 minutes at 25 ° C and 150 rpm in a small constant temperature shaking incubator bioshaker. This was the washing step. The test treatment solution in the bioshaker after washing was aspirated with a micropipette (Eppendorf Research (R) plus, manufactured by Eppendorf Co., Ltd.), and 3 mL of model tap water was added. The well was shaken and washed for 3 minutes under the same conditions. This rinsing step was repeated four times, and the model tap water in the bioshaker after rinsing was then aspirated with a micropipette. The rinsed biofilm-forming hard surface was then air-dried for 30 minutes in a safety cabinet.
[0111] (Measurement of the number of remaining viable bacteria) The biofilm-formed fabric after air drying and the biofilm-formed fabric before contact with the test treatment solution were each placed in a 1.5 mL tube (Eppendorf Co., Ltd., Eppendorf tube 1.5 mL, model number: 0030 125.150) containing 1 mL of LP dilution solution (Shiotani MS Co., Ltd., LP dilution solution "Daigo"). With the lid on, the tube was placed in an ultrasonic cleaner (Emerson Japan Co., Ltd., tabletop ultrasonic cleaner, model number: 2510JDTH) for 15 minutes, and the bacteria remaining on the fabric were dispersed in the LP dilution solution. After stirring the solution for 3 seconds using a vortex mixer (Vortex Mixer, Model No. VTX-3000L, manufactured by AS ONE Corporation), the bacterial solution was serially diluted 10-fold with the LP dilution solution and 100 μL of each solution was dispensed into a petri dish (Aznol Petri dish, 90 mm diameter, 15 mm height, manufactured by AS ONE Corporation). Approximately 15 mL of SCD agar medium was added, the dish was covered, gently mixed, and then left to stand at room temperature until the agar solidified. After static culture at 37 °C for 1 day, the number of remaining viable M. variable bacteria was counted using a colony counter pen (Colony Counter Pen Type, manufactured by AS ONE Corporation, Model No. 2-5764-01). For the biofilm-forming hard surfaces, 3 mL of the LP dilution solution was added per well to the air-dried biofilm-forming hard surface and to the biofilm-forming hard surface before contact with the test treatment solution. The biofilm was detached from the wells by pipetting 20 times with a micropipette and dispersed in the LP dilution solution. This bacterial solution was serially diluted 10-fold with the LP dilution solution, and the number of remaining viable bacteria of M. variable was counted in the same manner as above. (Method for calculating biofilm bactericidal activity value) The biofilm bactericidal activity value was calculated using the following formula. The results are shown in 1 to 7. The higher the bactericidal activity value, the better the biofilm bactericidal ability. Biofilm bactericidal activity value = Log A - Log B A: average number of remaining viable bacteria before contact with the test treatment solution B: average number of remaining viable bacteria after contact with the test treatment solution
