Description

USE OF COMPOUND 1 IN TREATING OR PREVENTING DISEASE ASSOCIATED WITH C-KIT MUTATION OR C-KIT FUSION MUTATIONTECHNICAL FIELD

[0001] The present invention relates to use of N- (3-chloro-5- (trifluoromethyl) phenyl) -3- ( (6- (4-hydroxypiperidin-1-yl) imidazo [1, 2-b] pyridazin-3-yl) ethynyl) -2-methylbenzamide (also referred to herein as Compound 1) or a pharmaceutically acceptable salt thereof in treating or preventing a disease associated with a c-Kit mutation or a c-Kit fusion mutation.BACKGROUND

[0002] c-Kit is a transmembrane receptor tyrosine kinase protein, also known as tyrosine protein kinase KIT, CD117 (cluster of differentiation 117) or mast / stem cell growth factor receptor (SCFR) . The c-Kit protein increases intracellular signaling by activating three pathways of PI3K, MAPK, and STAT, playing a crucial role in cell proliferation, differentiation, survival and migration. c-Kit, together with a platelet-derived growth factor receptor (PDGFR) , a macrophage colony stimulating factor-1 receptor (CSF-1R) , and an Fms-like tyrosine kinase receptor 3 (FLT3) , forms the type III receptor tyrosine kinase superfamily, which is significantly involved in tumor occurrence, development, invasion, migration, and recurrence.

[0003] Abnormalities in c-Kit function, such as gain of function, loss of function, overexpression, and point mutations, can lead to continuous activation of intracellular signaling pathways, promoting the occurrence and development of tumor cells. High expression of c-Kit is found in various malignant tumors, including gastrointestinal stromal tumor, mantle cell lymphoma, acute myeloid leukemia, prostate cancer, germ cell tumor, breast cancer, small cell lung cancer, etc. c-Kit mutations are common and have been found in exons 2, 8, 9, 10, 11, 13, 14, 15, 16, 17, etc., with higher mutation rates in exons 11, 9, 13 and 17. Inhibiting the c-Kit-mediated signaling pathway for the purpose of tumor therapy has become a hot spot of tumor therapy in recent years.

[0004] Currently, the main targeted drugs related to c-Kit gene mutation diseases include imatinib, sunitinib, regorafenib, pazopanib, sorafenib, dasatanib, ponatinib, and ripretinib. Sunitinib, regorafenib, pazopanib, sorafenib, dasatanib, ponatinib, and ripretinib are all multi-target drugs with poor kinase selectivity, leading to significant side effects for patients during use. In addition to susceptibility, imatinib causes or aggravates psoriasis, resulting in dermatitis, rashes, edema and the like during use. Existing kinase inhibitors, including imatinib and the like, have high inhibitory effects on wild-type c-Kit, thus highlighting the clinical need to develop a high-selectivity c-kit mutant kinase inhibitor.

[0005] The present invention provides a novel highly selective c-Kit mutant kinase inhibitor, Compound 1, and salts thereof. The inhibitory effect of Compound 1 on a wild-type c-Kit, mutant proteins and related cells is determined through inhibition experiments. In the present invention, it is identified through these characterizations that Compound 1 has an inhibitory effect on c-Kit gene  mutations, while it has no significant impact on the wild-type c-Kit.

[0006] CN113831344B discloses a series of TRK inhibitors, particularly N- (3-chloro-5- (trifluoromethyl) phenyl) -3- ( (6- (4-hydroxypiperidin-1-yl) imidazo [1, 2-b] pyridazin-3-yl) ethynyl) -2-methylbenzamide.SUMMARY OF THE INVENTION

[0043] EXAMPLE

[0044] Example 1: In Vitro Inhibitory Activity of Compound 1 Against c-Kit and Mutant Protein Kinases Thereof

[0045] A stock solution of 10 mM Compound 1 was prepared and then Compound 1 was diluted to 200× of the test concentration (amaximum concentration of 2000 μM) with DMSO. 25 nL of stock solution was transferred to a 384-well reaction plate (784075, Greiner) using Echo 655 ( 655 SYSTEM, Beckman) . 2× Kinase solutions (containing c-KIT at 0.6 ng / μL, c-KIT-V560G at 1.25 ng / μL, and c-KIT-T670I at 1.0 ng / μL, respectively) were prepared with a 1×kinase reaction buffer (1× Buffer, 5 mM MgCl2, 1 mM DTT, H2O) . Then, 2.5 μL of the kinase solution was transferred to the 384-well reaction plate. The plate was centrifuged at 1000 rpm for 60 s using a centrifuge and incubated at 25 ℃ for 10 min. A mixed solution of 2× substrate biotin (2 μM) and ATP (16 μM) was prepared using the kinase reaction buffer. To the reaction plate, 2.5 μL of the mixed solution of the substrate biotin and ATP was added to start the reaction, and the plate was centrifuged at 1000 rpm for 60 s using the centrifuge. The reaction plate was sealed and incubated at 25 ℃ for 40 min. To each well of the reaction plate, 5 μL of kinase detection reagent was added. The plate was centrifuged at 1000 rpm for 1 min and incubated at 25 ℃ for 60 min. The fluorescence signals at 615 nm (Cryptate) and 665 nm (XL665) were read using a BMG microplate reader. The ratio of the fluorescence signals, i.e., 665 nm / 615 nm, in each well was calculated. Inhibition rate %= 1 - (ratio of Compound 1 -ratio of positive control)  /  (ratio of negative control -ratio of positive control) × 100%. Data analysis was performed using Graphpad 7.0 software, and the IC50 (half maximal inhibitory concentration) values of Compound 1 were obtained using the following non-linear fitting formula: Y = Bottom + (Top -Bottom)  /  (1 + 10^ ( (LogIC50 -X) × Hill Slope) ) , wherein X denotes the log value of the concentration of Compound 1, and Y denotes the inhibition rate (%inhibition) .

[0046] Table 1. Inhibition of c-Kit and mutant kinases protein thereof by Compound 1

[0047] The test results showed that the IC50 values of Compound 1 for c-KIT-V560G and c-KIT-T670I mutations were 4.389 nM and 85.89 nM, respectively, indicating that Compound 1 had excellent inhibitory activity against c-Kit mutant protein kinases. Meanwhile, the activity of imatinib against the c-Kit protein kinase was 0.1 μM, whereas Compound 1 did not affect the activity of c-Kit protein, indicating that Compound 1 had a highly selective inhibitory effect on mutant proteins.

[0048] Example 2: Evaluation of Activity of Compound 1 Against Ba / F3 Cell Line with c-Kit Fusion or Fusion Mutant Thereof

[0049] Ba / F3 cell lines with c-Kit fusion or mutant gene were cultured at 37 ℃ with 5%CO2. The cells growing in the logarithmic growth phase were taken for plating. Compound 1 was prepared as a 20 mM solution in DMSO and serially 4-fold diluted from the highest concentration to the lowest concentration with DMSO for a total of 9 concentration gradients.

[0050] The cell culture medium was added to a 96-well plate. The Compound 1 solution was taken from the plate containing 1000× Compound 1 (10 mM) , and added to the cell culture medium in the 96-well plate. The mixture was pipetted and mixed well to prepare a 20× Compound 1 working solution. DMSO was added to the control wells.

[0051] The cell concentration was adjusted to an appropriate concentration (final concentration of 2000 cells / well) . To each well of the Compound 1 test cell plate, 95 μL of cell suspension was added. The cell-free culture medium was added to the Min control well. 5 μL of 20× Compound 1 working solution was added to the cell culture plate. To the Max control well, 5 μL of a mixed solution of DMSO and cell culture medium (final DMSO concentration of 0.1%) was added. The culture plate was incubated in an incubator at 37 ℃ with 5%CO2 for 72 h.

[0052] A CellTiter-Glo buffer was added to a flask of CellTiter-Glo substrate to dissolve the substrate, thereby preparing a CellTiter-Glo working solution. 50 μL (equal to half the volume of cell culture medium in each well) of CellTiter-Glo working solution was added to each well. The culture plate was shaken on an orbital shaker for 2 min to induce cell lysis. The culture plate was left to stand at room temperature for 10 min to stabilize the luminescent signals. The luminescent signals were detected on a SpectraMax Paradigm plate reader.

[0053] The cell proliferation inhibition rate (Inhibition Rate) data was processed using the following formula: Inhibition Rate (Inh%) = 100 - (RLUDrug -RLUMin)  /  (RLUMax -RLUMin) × 100%. The inhibition rates corresponding to different concentrations of Compound 1 were calculated in EXCEL, followed by plotting the inhibition rate curves and calculating relevant parameters, including maximum cell inhibition rate, minimum cell inhibition rate, and IC50 values, using Graphpad 7.0 software.

[0054] Table 2. Activity of Compound 1 against Ba / F3 cell lines containing c-Kit fursion or mutants thereof

[0055] The experimental results described above showed that Compound 1 of the example of the present invention exhibited a low inhibitory effect on wild type Ba / F3-TEL-c-KIT (IC50 value of 3383 nM) , significantly lower than that of imatinib (IC50 value of 516.2 nM) . However, Compound 1 can effectively inhibit Ba / F3-TEL-c-KIT-C674S, Ba / F3-TEL-c-KIT-V559D-T670I, and Ba / F3-TEL-c-KIT-D820E, and highly effectively inhibit various c-KIT mutant cell lines such as Ba / F3-TEL-c-KIT-V559A, Ba / F3-TEL-c-KIT-V559D, Ba / F3-TEL-c-KIT-V560D, Ba / F3-TEL-c-KIT-V560G, Ba / F3-TEL-c-KIT-L576P, Ba / F3-TEL-c-KIT-S709F, Ba / F3-TEL-c-KIT-T670I, Ba / F3-TEL-c-KIT-557 / 558 Del, Ba / F3-TEL-c-KIT-559 / 560 Del, Ba / F3-TEL-c-KIT-557 / 558 Del-V654A, and Ba / F3-TEL-c-KIT-557 / 558 Del-T670I.

[0056] Example 3: Evaluation of In Vivo Efficacy of Compound 1 Against c-Kit Mutant

[0057] An appropriate amount of Compound 1 was weighed, and dissolved and mixed well in a dissolution medium containing 5%DMSO + 10%Solutol + 85%Saline. Compound 1 was administered at low, medium, and high doses once a day. High doses of imatinib mesilate were dissolved in the same dissolution medium as the positive control, and the negative control was a pure solvent. Ba / F3 cell lines were cultured using RPMI1640 medium + 10%fetal bovine serum + 1%bispecific antibody. Approximately 0.2 mL of cells (1 × 106 cells) with corresponding mutations were mixed with matrigel at a 1: 1 volume ratio, and subcutaneously inoculated into the right dorsal side of Balb / c nude mice (female, 6-8 weeks old, weighing 18-22 g) . The mice were divided into groups for intragastric administration when the mean tumor volume reached approximately 100-120 mm3 (as close to 100 mm3 as possible) .

[0058] Tumor growth inhibition rate (TGI (%) ) = [ (1 – (mean tumor volume at the end of administration  of a treatment group –mean tumor volume at the start of administration of the treatment group) )  /  (mean tumor volume at the end of treatment of the solvent control group –mean tumor volume at the start of treatment of the solvent control group) ] × 100%.

[0059] Results of in vivo efficacy study

[0060] The body weight of mice in each group was within the normal range.

[0061] Compound 1 was found to have excellent anti-tumor effects in various mouse xenograft tumor models.

[0062] For example, for the BaF3-TEL-KIT-V559A mouse xenograft tumor model (FIG. 1) , the tumor volume in the Compound 1 treatment group was significantly lower than that in the control group on days 1-13 of administration, and the mice were sacrificed on day 13. The results showed that the TGIs of 30, 60, and 120 mg / kg Compound 1 were 55%, 84%, and 101%, respectively, while the TGI of 120 mg / kg positive drug imatinib mesilate was 89%, indicating that Compound 1 had a better inhibitory effect on the BaF3-TEL-KIT-V559A mouse xenograft tumor, and was better than the positive compound.

[0063] For the BaF3-TEL-KIT-V559D mouse xenograft tumor model (FIG. 2) , the tumor volume in the Compound 1 treatment group was significantly lower than that in the control group on days 1-13 of administration, and the mice were sacrificed on day 13. The results showed that the TGIs of 30, 60, and 120 mg / kg Compound 1 were 61%, 63%, and 79%, respectively, while the TGI of 120 mg / kg positive drug imatinib mesilate was 72%, indicating that Compound 1 had a better inhibitory effect on the BaF3-TEL-KIT-V559D mouse xenograft tumor, and was better than the positive compound.

[0064] For the BaF3-TEL-KIT-V560D mouse xenograft tumor model (FIG. 3) , the tumor volume in the Compound 1 treatment group was significantly lower than that in the control group on days 1-13 of administration, and the mice were sacrificed on day 13. The results showed that the TGIs of 25, 50, and 100 mg / kg Compound 1 were 43%, 79%, and 98%, respectively, while the TGI of 100 mg / kg positive drug imatinib mesilate was 79%, indicating that Compound 1 had a better inhibitory effect on the BaF3-TEL-KIT-V560D mouse xenograft tumor, and was better than the positive compound.

[0065] For the BaF3-TEL-KIT-V560G mouse xenograft tumor model (FIG. 4) , the tumor volume in the Compound 1 treatment group was significantly lower than that in the control group on days 1-13 of administration, and the mice were sacrificed on day 13. The results showed that the TGIs of 10, 30, and 100 mg / kg Compound 1 were 33%, 77%, and 106%, respectively, while the TGI of 100 mg / kg positive drug imatinib mesilate was 99%, indicating that Compound 1 had a better inhibitory effect on the BaF3-TEL-KIT-V560G mouse xenograft tumor, and was better than the positive compound.
