Description

[0001]COSMETIC COMPOSITIONS AND USES THEREOF FIELD The invention relates generally to cosmetic compositions and specifically topically applied cosmetic formulations comprising natural active ingredients. In particular, the invention relates to cosmetic compositions, and more particularly, “dermocosmetics”, for example, in highly functioning or science-based topical applications to address cosmetic skin concerns such as dry skin, inflammation, hyperpigmentation, acneic production, prevent and reverse photo-ageing and other aesthetic features such as radiance. BACKGROUND The skin is the largest organ of our body and acts as our physical shield against the external environment, retaining complex structure, function, metabolic action and regulatory processes. There are four predominant layers of dermal tissue including the stratum corneum, epidermis, dermis and adipose tissue. There is a basal layer between the epidermis and dermis which gives rise to cellular communication, signalling and maintains a repository of extracellular proteins. The pH of healthy skin ranges from 4.8-5.5, and this is maintained precisely. This is due to an acid mantle which is present over the skin’s barrier. Pollution, temperature, dermal pathogens and UV exposure disrupts skin function, barrier function, pH value and a subsequent cascade of events may ensue. Dermal protein signalling and the production of growth factors cannot occur optimally in alkaline skin states. Patients with dermatological skin disorders such as eczema or dermatitis have higher pH values which are closer to neutral or slightly alkaline. As a consequence, these subjects and their skin are unable to form lipids, ceramides or other integral structural features of the dermal tissue. Moreover, acidity is required for appropriate function and regulation of the skin’s microbiome. The stratum corneum is the uppermost layer, commonly referred to as the barrier, where the skin’s microbiomic community reside in unison with corneocytes. Generally, keratinocytes are structured in a manner which prevents external aggressors from entering the skin and again, which is why the term ‘barrier’ is often used. This is a matrix of intercrossed involucrin and keratohyalin which form the basis of a cornified involucre. Intercellular spaces exist within these matrices or crossed structure where lipids accumulate; simply put, these lamellar lipids ‘fill in the gaps’. However, this lipid bi- layer contains polar heads and intracellular channels which allow the passing and binding of hydrophilic substances. However, this layer usually presents problems for modern skincare formulations due to this structure and tortuous pathway as described by dermatologists for substances to pass through. The epidermis is responsible for pigment creation, holding melanocytes, as well as generalised presence of keratinocytes and other antigen-presenting cells. The dermis is a reservoir of vasculature which innervates the epidermis and is responsible for the production of growth factors and other messenger proteins or enzymes as fibroblasts are abundant within this layer. The adipose tissue resides in the ‘bottom’ layer which lines the connective tissue network between the dermis and the subcutaneous layer beneath the skin. The skin is a complex organ and is affected materially by extrinsic and intrinsic factors which lead to depletion, inflammation, irradiation, degradation or reconstruction, regeneration, rejuvenation and endogenous healing processes. Extrinsic factors are the major cause of ageing within skin, most notably from UV-A induced irradiation and further lipid peroxidation. The generation of reactive oxygen species and free radicals pose various issues including induction of matrix metalloproteinase activity, activation of inflammatory pathways such as the NF-kB or TNF-alpha sequences, tyrosinase based action or induction of disordered melanosome function. Another more insidious culprit which leads to skin damage and photo-aging is blue light. Blue light is emitted visible light between the wavelengths of 380 to 500 nm, also referred to as high-energy visible (HEV) light. The main source of blue light is sunlight, but digital computer screens, light- emitting diodes (LEDs), and fluorescent lighting serve as additional sources. Concerns about the negative effects of blue light on the skin have rapidly increased over the past 15 years, and consequently, the urge to learn more about this topic is increasing as well together with cosmetic remedies to minimise or reduce the damaged caused by blue light exposure. The aforementioned effects gives rise to issues including barrier dysfunction, pathogenic material generation via a disordered microbiome which may cause acne, alongside inflammatory pathways which cause acneic production, disordered sebum regulation, degradation of proteins such as collagen and elastin and suppress fibroblast proliferation. Moreover, intrinsic factors such as DNA and telomerase dysfunction shorten these caps or telomeres and remains the biological nature of ageing with loosened DNA coiling and structure. However, most of these features are gradual, despite their nefarious nature. Dysregulated pH of the skin will result in disorders and dermal complications from very early ages. Modern skincare and evolving advancements have revealed high functioning, scientific and dermatologically based dermocosmetics to address these factors. Genetics play a part in how skin is expressed within an individual, however at least 80% of all skin damage and deterioration is a result of UV-A exposure, however as we spend more time in from of computer screens the additional aging effects of blue light become an increasing concern. The present invention seeks to address one or more of the above current problems in the art. SUMMARY The present inventors have identified a need to develop an improved cosmetic composition with blue light reflective, absorption, or filtering capabilities. The chemical compositions of the present invention, which are presented as water based liquid compositions act as improved topically applied compositions with natural reflective, absorption, and filtering agents to treat various cosmetic skin conditions (referred herein as “dermal conditioning”), or mixed with additional ingredients, for instance, via emulsion and other techniques (such as sonication) to produce other dermatologically beneficial formulations. Without wanting to be bound by any particular theory, the present inventors have identified and developed naturally sourced actives to provide novel cosmetics, which are beneficial to the dermal layers. The compositions may be complemented by additional components / ingredients may encourage endogeny, inhibit inflammation and enzyme processes which cause ageing, acneic production, hyperpigmentation and dermal dehydration. The effects of UV and HEV light on skin over time can be profound. Exposure to sunlight and tanning machines is the main source of UV light (both UV-A and UV-B). Over exposure to UV-B is the main cause of skin burn with UV-B only managing to penetrate the epidermis layer. This can lead to increased melanin and result in photo-aging, hyperpigmentation, sun allergies, freckles and melisma. UV-A light can penetrate pass the epidermis and into the dermis layer. This can lead to hyaluronic acid decrease (which causes dryness, inflammation, and skin texture changes), collagen fibre damage (causing fine lines and wrinkles and loss of skin tone), DNA Damage (ROS and oxidative stress to skin lipids, proteins and cell regeneration slowed), and degradation of elastin production (ie, loss of elasticity and skin resilience). High-energy visible (HEV) light (blue light) is also a production of sunlight, but digital computer screens, light-emitting diodes (LEDs), and fluorescent lighting serve as additional sources. In a modern world people are nowadays all the more exposed to blue light by spending large amounts of time in front of computers (and the like). Blue light can penetrate even deeper than UV-A and UV-B and damage the hypodermis layer. This leads to subcutaneous fat changes and ultimaelty impairs the support base of skin. In certain embodiments the compositions disclosed herein are able to, in part, penetrate and reside in the epidermis and / or dermis layer protecting the hypodermis layer from the damaging effects of blue light. In certain embodiments the compositions disclosed herein are able to, in part, penetrate and reside in the epidermis layer protecting the dermis and hypodermis layer from the damaging effects of blue light and UV light. The compositions disclosed herein may be developed further to offer clinical grade formulations within Dermatology or Cosmetic Procedure Practices. Such expansions may include cosmetic procedural formulations such as a novel ‘filler’ preparation. The present invention also contemplates the development of novel cosmetic and dermocosmetics compositions with an acidic buffer capacity to ensure optimal dermal outcomes. Thus, the present invention is predicted, in part, on the understanding that pH control plays a crucial role in healthy skin function conducive towards dermal conditioning and the inventors have developed a series of cosmetic aqueous based solutions which provide effective dermal conditioning properties at a pH of from 3 to about 6 which are capable of minimising / reducing or preventing photo-aging. In certain embodiments the present cosmetic compositions of the present invention are specially designed to reduce or prevent photo-ageing in a subject and contain as active ingredients plant extracts that comprise mixtures of verbascoside and echinacoside. Verbascoside (structure below) is a caffeoyl phenylethanoid glycoside in which the phenylpropanoid caffeic acid and the phenylethanoid hydroxytyrosol form an ester and an ether bond respectively, to the rhamnose part of a disaccharide, namely β- (3′,4′-dihydroxyphenyl)ethyl-O-α-L-rhamnopyranosyl(1→3)-β-D-(4-O-caffeoyl)- glucopyranoside. Verbascoside can be found in species in all the plant families of the order Lamiales (syn. Scrophulariales). Only two examples are known from outside the order, in the class Asterids. In the family Lamiaceae, it can be found in the medicinal plants in the genus Phlomis, in the Scrophulariaceae, in Verbascum phlomoides, Verbascum mallophorum, or, in the family Buddlejaceae, in Buddleja globosa or Buddleja cordata, in the family Bignoniaceae, in Pithecoctenium sp and Tynanthus panurensis, in the family Orobanchaceae, in Cistanche sp and Orobanche rapum-genistae, in the Plantaginaceae, in Plantago lanceolata, in Verbenaceae, in Verbena officinalis (common vervain), Aloysia citrodora (lemon verbena) and Lantana camara, in the Oleaceae, in Olea europaea (olive), in the Lentibulariaceae, in the carnivorous plant Pinguicula lusitanica, and, in the Byblidaceae, in Byblis liniflora. Echinacoside which is a natural plant derived phenol, is a caffeic acid glycoside from the phenylpropanoid class. It is constituted from a trisaccharide consisting of two glucose and one rhamnose moieties glycosidically linked to one caffeic acid and one dihydroxyphenylethanol (hydroxytyrosol) residue at the centrally situated rhamnose. This water-soluble glycoside is a distinctive secondary metabolite of Echinacea angustifolia and Echinacea pallida (to about 1%) but only occurs in trace amounts in Echinacea purpurea. It is also isolated from Cistanche spp. Echinacoside was first isolated Echinacea angustifolia and has been shown to have weak antibiotic activity in vitro. As such the present compositions are thought to also provide support to acneic skin. Accordingly, in one aspect the invention provides a water based cosmetic composition comprising: (i) about 0.03-6% wt / wt of a plant extract which comprises both verbascoside and echinacoside; and (ii) about 1-4% wt / wt of 1,3-propandiol. In certain embodiments the present cosmetic compositions of the present invention are specially designed to reduce or prevent photo-ageing in a subject and contain as active ingredients plant extracts that comprise mixtures of verbascoside and echinacoside. In certain embodiments the present cosmetic compositions of the present invention are specially designed to reduce or prevent photo-ageing in a subject and contain as active ingredients plant extracts that comprise bisabolol or synthetic bisabolol (structure shown below): Bisabolol, or more formally α-(−)-bisabolol or also known as levomenol, is a natural monocyclic sesquiterpene alcohol (a terpenoid). It is a colourless viscous oil that is the primary constituent of the essential oil from German chamomile (Matricaria recutita) and Myoporum crassifolium. High concentrations of bisabolol can also be found in certain medicinal cannabis cultivars. It is poorly soluble in water and glycerine, but soluble in ethanol (alcohol) which is also used in the formulations herein. The enantiomer, α-(+)-bisabolol, is also found naturally but is rare. Synthetic bisabolol is usually a racemic mixture of the two, α-(±)-bisabolol. In respect to the present invention it is considered that in embodiments which contain this ingredient there is a triple light action effect which protects against UVA and UVB and HEV. In certain embodiments the present cosmetic compositions of the present invention are specially designed to reduce or prevent photo-ageing in a subject and contain as active ingredients plant extracts that comprise bisabolol (or synthetic bisabolo) and a Polygonum aviculare extract. This Polygonum aviculare extract is a member of the Polygonaceae family of plants for instance in the knotweed family or smartweed or buckwheat family, which has been known for its antioxidant and anti-obesity effects. Polygonum aviculare L. and its active compounds, quercitrin hydrate, caffeic acid, and rutin, activate the Wnt / β-catenin pathway are also known to and induce cutaneous wound healing. The present inventors use the extract to aid the reduction or prevention of photo-ageing an in particular photo- ageing caused by exposure to blue light. DETAILED DESCRIPTION All wt / wt% values mentioned herein refer to wt / wt% amounts relative to the total weight of all ingredients in aqueous compositions of the present invention (including the water content). In certain embodiments the water based (or aqueous based) compositions of the present invention comprise from 85-96% wt / wt water content, such as about 86, 87, 88, 89, 90, 91, 92, 93, 94, 95 or about 96% wt / wt water. In certain other embodiments where the compositions are in the form of a balm the water content may be as low as 0.2-5% wt / wt such as about 0.3-1% wt / wt of the total composition. Accordingly, the skilled person will understand The skilled person will understand that based on a high water content the compositions of the present invention will normally be presented in the form of lightweight non-viscous serums or mists. The term "subject" or "subjects" encompasses as used herein refers to human subjects. As used herein the terms "compositions" and "formulations" are used interchangeably. The term “dermal conditioning” refers to the normalisation of the skin tissue after having a cosmetic condition requiring treatment or for cosmetic purposes. Such conditions includes birthmarks, botox injections or filler preparation due to extrinsic ageing concerns, acne scarring, stretch marks, dry skin, inflammation, hyperpigmentation, acneic production, ageing and other aesthetic features such as radiance. The present invention is particularly concerned with dermal conditioning associated with photo-aging. Dermal conditioning is a complex process which is characterised by intercalating degradation and re-assembly of connective tissue and epidermal layer and will largely depend on the condition which has caused the skin to be preforming non-optimally. The pH value influences indirectly and directly all biochemical reactions taking place within and on the skin. In the modern world and with hour upon hour of exposure to blue light through computer use, collagen is broken down through oxidative stress by ROS (reactive oxidative species). Primarily this is caused by the compound Flavin, naturally in the skin, absorbing blue light (380 to 500 nm- also known as high-energy visible (HEV)). Studies have shown that exposure (even for as little as 60 minutes) from blue light emitting electronic devices (e.g., computer screens) triggers changes in skin cells such as cell shrinkage and death. Some of the advantages of the present inventive compositions include: ^ Protection against blue light exposure damage. ^ Protection against blue / UVA / UVB light exposure damage. ^ Encouragement of dermal healing and restoration of integrity via regulation of micro-circulation between dermal cells and connective tissue during metabolic action and promotion of epidermal cell growth. ^ Regeneration of elasticity via epidermal re-uptake. ^ Regulates sebum and oil production, anti-acneic and anti-congestive properties and suppression of comedone formation via downregulation of 5alpha reductase enzyme. ^ Further acneic regulation via stagnation of lipid peroxidation, anti-inflammatory activity and antimicrobial / antibacterial properties. ^ Stimulation of endogenous glycosaminoglycans, peptides and hyaluronic acid. Prevents denaturation and depletion of macromolecules. ^ Hydration and moisturisation throughout all levels of dermal matrix. ^ Protects against blue light damage of endo-peptides such as various types of collagen. ^ Mitigates post-inflammatory hyperpigmentation via transfer cycle of melanosomes from melanocytes into keratinocytes. ^ Anti-aging qualities through promotion of growth factors and other complementary molecules including elastin, flavin, filaggrin, involucrin, fibrillin etc. and denaturation or suppression of MMP activity. ^ Potent anti-inflammatory action, suppressing the TNF-alpha, NF-kb and other pathways, and reducing the release of inflammatory factors such as ILs and PEGs. ^ Calming and soothing action against irritated skin due to various aggressors or compromised skin condition ^ Reversal of irradiation in dermal cells via exposure to UV light – restores and reverses prior photodamage at the cellular level whilst offering photoprotective properties ^ Significant antioxidation ability and suppresses ROS, lipid peroxidation and other free radicals. Multiple scavenger agents of free radicals. ^ Humectant properties which draw water to the upper levels of the skin from the environment, emollience as well as occlusive properties of the moisturiser to seal in dermal conditioning treatments via product use. ^ Prevention of barrier damage and exposure to external aggressors due to an invisible film forming over the barrier, which in turn, significantly decreased trans- epidermal water loss. ^ Increases skin cell turnover rate without irritation / skin sensitivity or increased photosensitivity associated, maintaining anti-aging effect. Many of these factors occur at a microscopic level and therefore can be referred to as the microenvironment. Inside this microenvironment, factors are at play that influence whether the dermal condition can be ameliorated or becomes chronically stalled. It has been proven that the surface pH of a skin plays an important role in skin conditioning as it helps control infection and increase antimicrobial activity, oxygen release, angiogenesis, protease activity, and bacterial toxicity. Therefore, the pH value of the skin affects the regular cellular events. Information regarding benefits of certain ingredients used in embodiments of compositions of the present invention: 1,3-Propanediol - Propanediol is a natural alternative and used to replace the bad mouth feel of propylene glycol. It is produced sustainably from corn sugar and it's Ecocert approved. It can be used to improve skin moisturization, as a solvent, to boost preservative efficacy or to influence the sensory properties of the end formula. Butylene glycol – a colourless, syrupy liquid with nice feel. Its main job is to be a solvent for the other ingredients. Other tasks include helping the product to absorb faster and deeper into the skin (penetration enhancer), making the product spread nicely over the skin (slip agent), and attracting water (humectant) into the skin. Hydrolyzed rhodophycea - a type of red algae that helps improve skin’s hydration and ability to look healthy while also being a good source of antioxidants. Research has shown the antioxidants in many types of algae, including red algae, help offset the oxidative damage UV-B light exposure triggers. Red algae is a rich source of carrageenan, which in compositons serves as a natural thickening and gelling ingredient. The structure of carrageenan closely resembles glycosaminoglycans, which are a key part of skin’s barrier. In this capacity, the carrageenan in red algae can help deliver other ingredients throughout skin’s uppermost layers. Along with carrageenan, red algae is a source of plant cholesterol, whose fatty acid compounds benefit skin. Panax Ginseng Root Extract - A traditional Korean medicine used for more than 2000 years. Regarding skin care, it aims to enhance skin nutrition and metabolism as a result of improving blood circulation. It also contains biologically active components referred to as ginseng saponins (ginsenosides) that have potent antioxidant properties. Glycerin - A natural moisturizer that’s also in our skin. A common, safe, effective and cheap molecule used for more than 50 years. Not only a moisturizer but also keeps the skin lipids between our skin cells in a healthy (liquid crystal) state, protects against irritation, helps to restore barrier. Effective from as low as 3% with even more benefits for dry skin at higher concentrations up to 20-40%. High-glycerin moisturizers are beneficially used for treating severely dry skin. Xanthan Gum – A thickener and emulsion stabilizer. If the product is too runny (e.g., serum), a little xanthan gum will make it more gel-like (e.g., cream). Can be used with other thickeners and so-called rheology modifiers (i.e., helper ingredients that adjust the flow and thus the feel of the formula). The typical use level of Xanthan Gum is below 1%wt / wt, it is usually in the 0.1-0.5%wt / wt range. Xanthan gum is all natural, a chain of sugar molecules (polysaccharide) produced from individual sugar molecules (glucose and sucrose) via fermentation. It’s approved by Ecocert and also used in the food industry (E415). Lecithin- An ingredient that can be found in all cell membranes. In cosmetics it has a few different roles: it's an emollient and water-binding ingredient but it's also an emulsifier and can be used for stabilization purposes. It's also often used to create liposomes. Pullulan - Versatile skin care ingredient known for it's hydration and antioxidant properties. Functions well as a “carrier” to deliver active ingredients where they need to go. Has potential to help strengthen skin’s microbiome and can provide temporary mechanical support for skin’s underlying structures. Continually being studied for its many therapeutic, film-forming benefits for skin. Sclerotium gum - A naturally derived, sugar-based ingredient. Produced by the fermentation of the Sclerotium rolfsii fungus. Functions as a thickener and stabilizer in cosmetics compositions. Silica - Natural ingredient found in clays as well as in parts of plants and animals Used as an absorbent powder and thickener in cosmetics and can enhance the absorption of ingredients and serve as a drug delivery system. Is also FDA-approved as a food additive. Sodium stearoyl glutamate- is a synthetic or animal-derived ingredient that is added to cosmetics to condition skin or work as an emulsifier, keeping oil-and-water formulas blended. Sodium benzoate - Salt of benzoic acid, which is found naturally in foods like cranberries. Used as a preservative in both cosmetics and food products Deemed safe as used in cosmetics and is a white, crystalline solid in its raw form. Sodium Hyaluronate - Salt form of hyaluronic acid and good skin-replenishing ingredient. One gram can hold up to six litres of water. More readily absorbed into skin than hyaluronic acid. Available in different molecular weights for controlled penetration. Lactic acid - Can be extracted from milk or synthetically made. Exfoliates the surface of skin and has hydrating properties and strengthens skin’s barrier. Helps positively influence skin’s microbiome. Benzyl alcohol - An organic alcohol that can be found naturally in some fruits. Serves chiefly as a preservative in cosmetics products and is considered one of the least sensitising preservatives. Deemed safe for use in concentrations up to 5%wt / wt of the total composition. Dehydroacetic acid - is a synthetic preservative used in amounts of 0.6%wt / wt or less where it’s proven to be safe and effective. Dehydroacetic acid is characterized as a white to cream crystalline powder. Sometimes its salt form (known as sodium dehydroacetate) is used, as it is more soluble in certain types of cosmetic formulas. Dehydroacetic acid is most effective as an anti-fungi ingredient.
