Description

[0001] PREPARATION OF BRIDGED PENTACYCLIC IMIDAZOLE DERIVATIVES AS MODULATORS OF TNF ACTIVITY, INTERMEDITATES AND THEIR PREPARATION

[0002] Disclosed herein is a method of preparing a compound of Formula (I):

[0003] Formula (I) i.e. (7R,14R)-1 l-[2-(l-aminocyclobutyl)pyrimidin-5-yl]-l-(difluoromethoxy)-6-methyl-6,7- dihydro-7,14-methanobenzimidazo[l,2-b][2,5]benzodiazocin-5(14H)-one. Also disclosed herein are intermediates, and methods of manufacturing intermediates.

[0004] Background

[0005] (7R,14R)-1 l-[2-(l-aminocyclobutyl)pyrimidin-5-yl]-l-(difluoromethoxy)-6-methyl- 6,7-dihydro-7,14-methanobenzimidazo[l,2-b][2,5]benzodiazocin-5(14H)-one (the compound of Formula (I)) is a pharmacologically active substituted fused pentacyclic benzimidazole derivative which shows pharmacological activity on TNFa signalling. The compound of Formula (I) is described in patent publication WO 2018 / 197503.

[0006] TNFa is the prototypical member of the Tumour Necrosis Factor (TNF) superfamily of proteins that share a primary function of regulating cell survival and cell death. One structural feature common to all known members of the TNF superfamily is the formation of trimeric complexes that bind to, and activate, specific TNF superfamily receptors. By way of example, TNFa exists in soluble and transmembrane forms and signals through two receptors, known as TNFR1 and TNFR2, with distinct functional endpoints.

[0007] Various products capable of modulating TNFa activity are already commercially available. All are approved for the treatment of inflammatory and autoimmune disorders such as rheumatoid arthritis and Crohn’s disease. All currently approved products are macromolecular and act by inhibiting the binding of human TNFa to its receptor. Typical macromolecular TNFa inhibitors include anti-TNFa antibodies and soluble TNFa receptor fusion proteins. Examples of commercially available anti-TNFa antibodies include fully human antibodies such as adalimumab (Humira®) and golimumab (Simponi®), chimeric antibodies such as infliximab (Remicade®), and pegylated Fab' fragments such as certolizumab pegol (Cimzia®). An example of a commercially available soluble TNFa receptor fusion protein is etanercept (Enbrel®).

[0008] The compound of Formula (I) is a potent modulator of human TNFa activity and is therefore beneficial in the treatment and / or prevention of various human ailments. These include autoimmune and inflammatory disorders, neurological and neurodegenerative disorders, pain and nociceptive disorders, cardiovascular disorders, metabolic disorders, ocular disorders, and oncological disorders.

[0009] The compound of Formula (I) is described in Example 6 of WO 2018 / 197503 Al. The synthetic method used to produce the compound involves a step in which the pentacyclic moiety (7R, 14R)-11 -chloro- l-(difluoromethoxy)-6-methyl-6,7-dihydro-7, 14-methano- benzimidazo[l,2-Z>][2,5]benzodiazocin-5(14 / / )-one is reacted with bis(pinacolato)diboron in the presence of a palladium catalyst. / c / V-Butyl A-[l-(5-bromopyrimidin-2- yl)cyclobutyl]carbamate is then added to the reaction mixture. The pyrimidine moiety is therefore added to the pentacyclic core by a Suzuki coupling in which a boronated derivative of the pentacyclic core is reacted with a brominated pyrimidine.

[0010] There is a desire to provide improved methods of manufacturing the compound of Formula (I), for example methods which are suitable for the large-scale manufacture of the compound of Formula (I). There is a particular desire to provide methods which do not involve the use of flash chromatography during purification steps.

[0203] Overall, by performing Steps 1 to 3, Intermediate 4 may be synthesized at an overall yield of 65%, with excellent purity control.

[0204] The above combination of Steps 1 to 3 can be compared to the synthetic route described in Example 11 of WO 2016 / 050975 Al which describes a reaction in which (lR,3R)-l-[2-bomo-6-(difluoromethoxyphenyl)-7-chl oro-2, 3-dihydro-lH-pyrrolo[ 1,2- a]benzodiazol-3-amine is converted to Intermediate 4 using potassium carbonate and dichloro[9,9-dimethyl-4,5-bis(diphenylphosphino)xanthene]palladium (II). The reaction in WO 2016 / 050975 Al takes place in one step and has a yield of 33.1%. In contrast, the combined yield of Steps 1 to 3 is up to 65% showing that such a synthetic route is associated with a significant improvement in yield. The improved yield of the present invention as compared to WO 2016 / 050975 Al is also evident when taking into account the preliminary step of obtaining the protected Starting Material la (starting material in Step 1), which is described in WO 2016 / 050975 Al. In particular, Starting Material la is obtained with a 70% yield from Intermediate 40 in WO 2016 / 050975 Al. Thus, overall, the preliminary protection step followed by Steps 1 to 3 described herein provides Intermediate 4 in a yield of up to 46%.

[0205] In addition, Steps 1 to 3 herein provide Intermediate 4 with excellent purity without using flash chromatography as described in WO 2016 / 050975, which cannot be used at an industrial scale.

A 5% solution of sulfate sodium in water (212 kg; 6.6 vol) and 10.5 vol of 2-methyltetrahydrofuran were added and the mixture was stirred for 0.5-2 h. The aqueous layer was washed with 5.25 vol 2-methyltetrahydrofuran and the combined organic layers were washed with a 5% aqueous solution of sulfate sodium (100 kg; 3.1 vol) and with a lithium hydroxide solution. The mixture was stirred for 0.5-1 h and the organic layer was concentrated below 50 °C under vacuo to 3-4 vol. The solvent was replaced with methanol by distillation below 50 °C under vacuo. The volume was adjusted to 8-10 vol and 4.5 vol of water was added at 45-55 °C. The mixture was stirred for 1-2 h at 45- 55 °C, then at 5-15 °C for 3-5 h and finally filtered at 5-15 °C. The aqueous layer obtained was washed at 20-30 °C three times with 3.8 vol of di chloromethane (DCM) and finally with 4.0 vol of 2-methyltetrahydrofuran. To the aqueous layer, 19 vol of 2-methyl tetrahydrofuran and a potassium phosphate solution in water (27%; 4.5 eq; 278.4 mol) were added at 20-30 °C. The mixture was stirred for 6-10 h and the organic layer was washed three times with 3.9 vol of water and with a lithium hydroxide solution. [0229] Conclusion on the obtention of the compound of Formula (I):

[0230] Overall, Steps 1 to 9 described herein allow the compound of Formula (I) to be obtained in a yield of up to 35%. Including the preliminary step of producing Starting Material la from Intermediate 40 of patent application WO 2016 / 050975, the overall yield of Formula (I) obtained by the present invention is approximately 25%. This is improved as compared to the yield of compound of Formula (I) described in WO 2018 / 197503 (21%, starting from Intermediate 40 of WO 2016 / 050975). Further advantageously, the process described herein is scalable at an industrial production level, as it produces the end-product with excellent purity without performing purification steps involving flash chromatography, which is carried out at three points in the process described in WO 2018 / 197503. Comparison of the Formation of Intermediate 6 by Different Methods

[0231] Experiments were performed to investigate the efficacy of different methods for the formation of Intermediate 6:

[0232] Intermediate 6

[0233] The experiments involved the coupling of Intermediate Ba and Intermediate 5:

[0234] Intermediate Ba Intermediate 5

[0235] In a first experiment, the pentacycle was reacted with bis(pinacolato)diboron to form a boronated complex. This was then reacted with Intermediate Ba to produce Intermediate 6.

[0236] In a second experiment, Intermediate Ba was reacted with bis(pinacolato)diboron to form a boronated complex. This was then reacted with Intermediate 5 to produce Intermediate 6.

[0237] A number of differences were observed between the two experiments: in the first experiment it was found that the boronated pentacycle was unstable and led to impurities identified as the H-pentacycle (due to protodeboronation) and a pentacycle dimer (due to homocoupling of boronated pentacycle), see formulae below:

[0238] H-pentacycle Dimer

[0239] The H-pentacycle was formed in an amount of 39% and the pentacycle dimer in an amount of 7%. The overall yield of the reaction for the desired end-product (Intermediate 6) was 38%.

[0240] In the second experiment, no such impurities were observed (no pyrimidine homocoupling by-product and below 0.50% of deboronated pyrimidine reactant detected), and the target product was obtained with a yield of 89%.

[0241] It can therefore be concluded that the pentacycle Intermediate 5 and the pyrimidine Intermediate B can be coupled by using a Suzuki reaction to produce Intermediate 6. However, performing the Suzuki reaction in such a manner that the pyrimidine is boronated is a more efficient reaction, leading to a higher yield compared to the reaction in which the pentacycle Intermediate 5 is boronated.

[0242] In addition, this high reaction yield for the Suzuki coupling reaction is afforded herein without performing a purification by flash chromatography, which is not scalable for an industrial production but was needed in the coupling step described in the previously published patent application WO 2018 / 197503.
