Abstract
Description
OSMANTHUS FRAGRANS EXTRACT FOR A COSMETIC TREATMENT
[0001] The present invention relates to the use of an extract ofOsmanthus fragransor a composition comprising said extract for non-therapeutic cosmetic treatment, in particular a topical treatment.
[0002] « Cosmetic or cosmeceutical treatment » means a treatment which treats healthy skin and / or skin appendages, said treatment being intended to improve or beautify their appearance and condition. Such treatment has no therapeutic purpose. The skin and appendages for which the treatment is intended are healthy and normal, i.e. not diseased.
[0003] The present invention relates particularly to the cosmetic industries, which manufacture products for the treatment of the skin, including the scalp, mucous membranes and the appendages (such as body hair, eyelashes, eyebrows, nails, hair) of mammals, animals or humans.
[0004] These industries are in increasing demand for new products, particularly new active ingredients that are derived from plants, because they offer a combination of efficacy, reduced risks of irritation and allergy, reduced side effects and biodegradability, with the possibilities of labelling / certification and compliance with a logic of sustainable development and / or fair trade.
[0005] At any age, a ‘beautiful’ skin will mean first a radiant and even complexion, i.e. with few spots and no unsightly redness.
[0006] Redness of the skin and scalp correspond to a cutaneous reaction that can have multiple origins.
[0007] The cause may be external, for example a contact with a skin care or cleansing product or a hair product (perm or colour) that is too aggressive, or mechanical action on the skin such as shaving, scrubbing, depilation, rubbing against clothing or jewellery. The effects of temperature, climate, ultraviolet rays (UVs), infrared rays (IR), atmospheric pollution, xenobiotics such as undesirable micro-organisms and allergens can also cause unsightly redness.
[0008] The cause may also be internal, such as psychological or hormonal stress, or linked to perspiration.
[0009] A skin that reddens is often a skin that feels uncomfortable, overheated, tight and tingling, and therefore a skin that needs to be treated gently to restore not only its evenness and radiance, but also its comfort.
[0010] The aim of the present invention is to provide a topical non-therapeutic cosmetic treatment that meets this cosmetic need as well as the need for an active ingredient derived from a plant.
[0011] To this end, the present invention proposes the use of an extract ofOsmanthus fragrans, or of a composition including it and a physiologically acceptable medium, for a topical non-therapeutic cosmetic treatment to prevent and / or reduce unsightly redness of the skin.
[0012] Osmanthus fragrans(Fragrant Osmanthus) is also known as the fragrant olive tree. It is an evergreen shrub native to eastern Asia. It is used in Indian pharmacopoeia and traditional Chinese medicine for its many properties, including antitussive, antioxidant, anti-rheumatic, antimicrobial, antifungal and longevity properties.
[0013] Osmanthus flowers are very fragrant and, like jasmine flowers, are used to flavour tea, wine and some liqueurs. They are also commonly used in Asian cuisine as a condiment or flavouring.
[0014] An extract ofOsmanthus fragranshas already been proposed as an anti-glycation agent (JP5969738 patent), or likely to promote the viability of epidermal skin cells (CN107456411 patent).
[0015] According to the invention, theOsmanthus fragransextract can be made from the whole plant or from one or more parts of the plant such as the leaf, stem, flower, fruit, seed, root, tuber and / or pericarp. Preferably, theOsmanthus fragransextract according to the invention is made from the flowers.
[0016] TheOsmanthus fragransextract according to the invention may be obtained by the usual solid / liquid extraction techniques including, for example, maceration, simple decoction, infusion, lixiviation, leaching, extraction under reflux, extraction by subcritical or supercritical fluid, extraction by means of ultrasound or microwaves, or any other physical and / or chemical method, such as distillation, percolation, digestion, cryoextraction, enzymatic digestion, etc.
[0017] « Maceration » means a process consisting of soaking a plant or part of a plant in an extraction solvent at room temperature.
[0018] « Infusion » means a process consisting of bringing the extraction solvent to the boil before pouring it onto the plant or part of the plant.
[0019] « Decoction » means a process consisting of blending the plant or the part of plant with the extraction solvent, then heating the mixture at the boiling point.
[0020] « Digestion » means a process consisting of blending the plant or the part of plant with the extraction solvent, then heating the mixture below boiling point.
[0021] Preferentially, theOsmanthus fragransextract is obtained by extraction by using ultrasound or microwaves.
[0022] According to the invention, the extraction is carried out in at least one aqueous, hydroalcoholic, glycolic or lipidic solvent or a mixture thereof, or a polyol chosen from pentanediol, sorbitol, butylene glycol, pentylene glycol, propylene glycol, hexanol, caprylic / capric triglyceride, a vegetable oil, a vegetable milk such as almond, hazelnut, coconut, oat, spelt, soya or rice, or a mixture thereof, or a basic or acidic aqueous solvent for hydrolysis.
[0023] The duration of extraction using ultrasound can vary between 30 minutes and 3 hours, preferably between 1 hour and 2 hours. The temperature is between 10°C and 60°C, preferably at room temperature. The power is between 10 and 40 Watts per kilogram of mixture of plant material and solvent.
[0024] According to another aspect of the invention, the extraction with ultrasound is carried out in at least one solvent chosen from: water, glycerin, hydroalcoholic, propylene glycol, butylene glycol, polyethylene glycol of any molecular weight, diethylene glycol monoethyl ether, hexylene glycol, but also ethanol, propanol, butanol and other polyols (propanediol, pentanediol, hexanol, sorbitol and other liquid sugars), vegetable milk, as well as all mixtures of these solvents.
[0025] Preferably, the extraction solvent is aqueous, comprising water or a mixture of at least one glycolic solvent and at least one aqueous solvent, more preferably a glycerin / water mixture.
[0026] Preferably, according to the invention the extraction is carried out by means of ultrasound in water or in a glycerin / water mixture.
[0027] According to another aspect of the invention, the extraction with microwaves is carried out in at least one solvent chosen from: water, glycerin, hydroalcoholic, propylene glycol, butylene glycol, polyethylene glycol of any molecular weight, diethylene glycol monoethyl ether, hexylene glycol, but also ethanol, propanol, butanol and other polyols (propanediol, pentanediol, hexanol, sorbitol and other liquid sugars), vegetable milk, as well as all mixtures of these solvents.
[0028] The extraction duration can vary between 10 and 300 seconds, preferably between 30 and 100 seconds. The power of the microwave generators can vary between 100 and 2000 Watts, the microwave processing frequency being limited to between 2000 and 3000 Mhz.
[0029] The advantage lies in the fact that theOsmanthus fragransextract according to the invention thus obtained is highly soluble in water and can therefore be formulated in various formulations for topical application.
[0030] In-vivotests are given below in the description showing the efficacy of treatment with an extract ofOsmanthus fragransaccording to the invention or of a composition containing it in preventing and / or reducing skin redness. As the treatment according to the invention is a purely cosmetic non-therapeutic treatment, it does not include the treatment of redness due to a burn, in particular due to prolonged sun exposure, or redness due to invasive cutaneous action with a wound.
[0031] Thus, the treatment according to the invention is more particularly suitable for treating unsightly redness linked to:
[0032] - exposure to radiation, in particular solar radiation, UVAs, UVBs, IR, visible light such as red, blue or green visible light, which can be used in photobiomodulation, also known as low-energy lasers, or high-energy radiation used in radiotherapy;
[0033] - climatic aggression or linked to exposure to abnormally high or low temperatures, for example the heat of a bath or wearing a mask, which increases skin temperature;
[0034] - friction between two parts of the skin or with another surface in contact with the skin, such as clothing, jewellery, etc.
[0035] - following undertaking of a tattoo; skin redness may be linked to the repeated passage of the needle inserting the pigments into the dermis; and / or
[0036] - an emotional reaction, called a ‘flush’, linked to stress, anxiety, etc.
[0037] The invention is therefore particularly suitable for treating the so-called reactive skin, which is susceptible to redness following the stimuli mentioned above.
[0038] According to another aspect, the treatment according to the invention is particularly suitable for increasing the production of adenosine triphosphate (ATP). ATP is one of the main sources of energy responsible for cell vitality. A reduction in ATP, caused for example by age or exposure to UVs, can lead to cell death, resulting in a slowdown in cell renewal and non-functional cell differentiation, leading to a reduction in the thickness of the epidermis, which is responsible for a duller, inhomogeneous complexion and less effective defense against UV damage. An increase in ATP production can counteract these effects by activating cellular respiration and increasing energy production in the cells. As a result, the cells are more functional and the harmful effects responsible for redness are better absorbed by the skin and appear later.
[0039] Advantageously, the treatment according to the invention is also suitable for treating the sensations of tingling and tightness which often accompany redness. In fact, as shown by thein vitrotests given below, theOsmanthus fragransextract according to the invention is also particularly effective in treating skin dryness thanks to an increase in hyaluronic acid synthesis. Improved skin hydration will restore suppleness to the skin, thereby calming it.
[0040] The cosmetic effects according to the invention, of an aesthetic nature, i.e. influencing the colour of the skin, and of a sensory nature, i.e. influencing the comfort felt, can be considered separately or in combination. These effects are beneficial for the skin, including the scalp.
[0041] The treatment according to the invention can be obtained by using the extract directly or by using a composition including it in a physiologically acceptable medium, and optionally other active ingredients.
[0042] "Physiologically acceptable medium" means according to the present invention, without limitation, an aqueous or hydro-alcoholic solution, a water-in-oil emulsion, an oil-in-water emulsion, a micro-emulsion, an aqueous gel, an anhydrous gel, a serum, a dispersion of vesicles, or a powder.
[0043] "Physiologically acceptable" means that the compositions are suitable for topical or transdermal use, in contact with mucous membranes, appendages (nails, hair and body hair), scalp and skin of mammals, particularly human, compositions which may be ingested, or injected into the skin, without risk of toxicity, incompatibility, instability, allergic response, and others. This "physiologically acceptable medium" forms what is commonly called the excipient of the composition.
[0044] According to the invention, the physiologically acceptable medium can be an aqueous, hydroglycolic or hydroalcoholic solution, or a water-in-oil emulsion, an oil-in-water emulsion, or a microemulsion. Preferably again, it is hydroglycolic. Preferably again, the physiologically acceptable medium is a blend of glycerin and water.
[0045] The present invention thus covers a non-therapeutic topical cosmetic treatment suitable for beautifying or improving the appearance and general condition of the skin and / or dander, more particularly for preventing and / or reducing unsightly redness of the skin and / or sensations of tightness and discomfort, including the topical application to the skin of a subject in need of an effective amount of the extract ofOsmanthus fragransaccording to the invention or of a composition including it in a physiologically acceptable medium.
[0046] The "effective" amount ofOsmanthus fragransextract according to the invention, i.e. its dosage in the composition, depends on the intended use of the composition. It depends on various factors, such as the age and condition of the patient and the severity of the disorder. An effective amount means a non-toxic amount sufficient to obtain the desired effect.
[0047] In a cosmetic composition comprisingOsmanthus fragransextract according to the invention, to be present in an effective amount, it is generally present in an amount ranging from 0.000001% and 15% based on the total weight of the composition, preferably ranging from 0.00001% and 10%, depending on the destination of the composition and the more or less pronounced desired effect. More preferably, the effective amount is between 0.0001% and 5% based on the total weight of the composition.
[0048] All percentages and ratios used herein are by weight of the total composition and all measurements are made at 25°C unless it is otherwise specified.
[0049] For indication, for a cosmetic face treatment, the European standard dosage of a cream is 2.72 mg / cm² / day / person and for a cosmetic body treatment the European standard dosage of a lotion is 0.5 mg / cm² / day / person.
[0050] According to other features, the cosmetic treatment method according to the invention may be combined with one or more other treatment methods targeting the skin such as luminotherapy, heat or aromatherapy treatments.
[0051] According to the invention, devices with several compartments or kits may be proposed to apply the method described above which may include for example and non-restrictively, a first compartment containing a composition comprisingOsmanthus fragansextract according to the invention, and in a second compartment an additional active ingredient, the compositions contained in the said first and second compartments in this case being considered to be a combination composition for simultaneous, separate or stepwise use in time, particularly in one of the treatment methods described above.
[0052] According to other advantageous features, theOsmanthus fragransextract according to the invention may be associated with one or more other active ingredients at effective concentrations that can act synergistically or additionally for reinforcing and achieving the desired effects.

[0075] 3.1. Effect on skin dryness - maintaining hydrationPrinciple
[0076] Hyaluronic acid is a major constituent of the epidermis contributing to the barrier function as well as to the maintaining of a satisfactory hydration of the skin. It is capable of absorbing 1,000 times its own weight in water. It is in the form of an aqueous and nourishing gel which fills the spaces between the keratinocytes. It prevents from dryness of the skin, which is known to alter the texture of the skin, giving it a rough touch.Protocol
[0077] Human epidermal keratinocytes were cultured at sub-confluence and then contacted with the active ingredient according to the invention for 3 days. After this contact, the synthesis of hyaluronic acid is assessed using ELISA methods. The quantity of cells is estimated using the Hoechst method and used to standardize the results. Results
[0078] Variation of hyaluronic acid production by keratinocytes. Effect of the active ingredient according to the invention at 3% compared with the control (n=5):

[0080] The results show that the active ingredient according to the invention significantly increases the synthesis of hyaluronic acid by keratinocytes.3.2. ATP synthesisProtocol
[0081] Human keratinocytes (HaCaT) were cultured at confluence in their culture medium. Then, the cells were brought into contact with the active ingredient according to the invention for 2 hours. Luciferin is converted with ATP into a form capable of being catalytically oxidised (oxyluciferin) by luciferase. The produced oxyluciferin (in an excited state) returns to a stable state with the emission of a yellow-green light that is measured (λ max = 565 nm), so the obtained signal is proportional to the amount of ATP in the cells. Results
[0082] Variation of ATP production by human keratinocytes. Effect of the active ingredient according to the invention at 3% compared with the control (n=5):

[0086] The study was carried out on a panel of 19 healthy volunteers, 9 men and 10 women with an average age of 39 (between 23 and 58).
[0087] The inclusion criteria that had to be met were:Being male or female,Be aged between 18 and 65, andHave a skin phototype between I and III on the Fitzpatrick scale and not be tanned.
[0088] The exclusion criteria that had to be met:Not to have skin disease or any other medical condition interfering with the objectives of the study,For women, do not be pregnant or breastfeeding, andDo not have uneven skin tone, sunburn, scars or lesions in the test area.Type of study, duration, applications
[0089] For 7 days prior to the test, volunteers were asked not to use any topical preparations on the inside of the forearms (test area).
[0090] Firstly, the test product was applied to the inside of the forearms by the volunteers approximately 15 minutes before UV irradiation. A second application was made immediately after irradiation and a final application was made 24 hours after irradiation.
[0091] For the duration of the study, only water or the mild soap provided was allowed for cleaning. In addition, volunteers were advised to avoid excessive exposure to the sun and sunbathing was prohibited.Statistics
[0092] For redness values, the statistical calculation is performed using a one-tailed paired Student's t-test for parametric values.Principle
[0093] The aim of this study is to evaluate the effect of the cream according to the invention on skin redness linked to exposure to UV radiation.
[0094] UV Source: 300W Xenon arc lamp solar simulator (Model 601 V2.5 Multisport, Solar Light Co. Inc, USA) equipped with a homogenizer. The entire UVA+UVB spectrum complies with the requirements of ISO 24444:2019. The uniformity of the solar simulator is measured prior to each test and the spectral output controlled annually.
[0095] An irradiation equivalent to 1.25 MED (mild but defined) is performed.Measurement conditions: Acclimatisation time: 30min.
[0096] Room temperature: 21 ± 2°C
[0097] Humidity: 45 ± 10%Measurement protocol
[0098] Colorimetric skin measurements were taken on the treated areas of the forearms using a MINOLTATM CM400 Spectrophotometer before the first application of the test product (D0), 24 hours after irradiation and before the second application (T=24 hours) and 48 hours after irradiation (T=48 hours). Each measurement was an average of six acquisitions over the same area of skin. The mean value of the measurements was calculated for each subject.
[0099] The spectrophotometer converts the colours perceived by humans into a numerical code. The parameter a* is used to study the spectrum from green to red. A decrease in the a* parameter over time indicates a decrease in skin redness. Results
[0100] The values measured at T0 are subtracted from all the results to obtain results relating to the induction of redness while eliminating the initial conditions, i.e. the skin's natural red hue.
[0101] Variation in redness (parameter a*, arbitrary units) (N=19).
