The washing process was performed in three steps: ① an air washing step to remove foreign substances and mold spores, ② a water washing step to remove residual pesticides, and ③ an alcohol (Et-OH) washing step to remove microbial contamination. The raw material that completed the washing process was dried in a hot air dryer capable of mass drying, and then went through a milling process to obtain black rice bran.

[0100] Example 1-2. Obtaining fermented black rice bran (BRB-F)

[0101] Black rice bran powder, from which foreign matter and mold contamination had been removed, was treated with enzymes and heat-treated to make a liquid culture medium, after which water was added. Commercially available enzyme product, amylase series, fungamyl 800L alpha-amylase (derived from Aspergillus oryzae), a hydrolase, was used. After adding 0.02% of the enzyme, the temperature was maintained at around 50°C and pH 4-6, and the reaction was carried out for 1 hour, followed by sterilization at high temperature for 30 minutes to make a culture (fermentation) medium for black rice bran. Afterwards, shiitake mushroom mycelia, which were separately cultured as basidiomycetes, were added to the culture (fermentation) medium. The fermentation process of the bioconversion process was performed by inoculating 10% (v / v) of the basidiomycete mycelial culture and culturing for 3 to 5 days at 28 to 30 ℃ and pH 4.5 to 7 to obtain a fermented product. The basidiomycete mycelia can be selected from the mycelia of medicinal and edible basidiomycetes such as shiitake mushrooms, phellodendron mushrooms, chaga mushrooms, maitake mushrooms, oyster mushrooms, pine mushrooms, and chive mushrooms, but shiitake mushroom mycelia are preferred and have been confirmed to have the same effect. In addition, these basidiomycete mycelia can be used by receiving strains from institutions such as the Korea Center for Microorganism Conservation (KCCM), Korea Center for Biotechnology and Biotechnology (KCTC), Korea Cell Line Bank (KCLB), Korea Agricultural Microorganism Resource Center (KACC), and the American Type Culture Collection (ATCC).

BRB-F (fermented black rice bran) produced by the bioconversion process was subjected to an enzyme inactivation process and a sterilization process at 90°C for 1 hour, and then freeze-dried to obtain BRB-F (fermented black rice bran powder) (Fig. 1).

[0105] The black rice fermented product produced by the bioconversion process of fermentation and enzyme treatment was subjected to an enzyme inactivation process and a sterilization process at 90℃ for 1 hour, and then freeze-dried and powdered. In addition, the powder of the black rice fermented product having the immune-activating effect is dissolved in 20 times the amount of water, or the black rice fermented product fermentation liquid is centrifuged or passed through a filter to remove insoluble residues to obtain a solid-liquid separation fraction (BRB-F solid-liquid separation fraction, or BRB-FS), or this can be freeze-dried to obtain a powder. That is, to remove insoluble residues, centrifugation was performed at 10,000 rpm for 30 minutes or passing the product through a filter with a pore size of 200 mesh (approximately 75 μm) and 1 μm. In small-scale production, centrifugation is convenient, but in large-scale production, using a filter is preferable. The above fraction or powder refers to the bioconverted product.

[0107] Example 2. Evaluation of the anti-obesity effect of black rice biotransformation products.

[0108] To evaluate the anti-obesity effect of black rice bran bioconversion product, 6-week-old female BALB / c mice were purchased from Orient Bio (Seongnam, Korea) and maintained at an indoor temperature of 22±2℃ until use in the experiment, with sufficient water and food supplied. Mouse groups were established as shown in Table 1 below, and the mice were randomly divided into groups of 10. After a 1-week adaptation diet, they were fed a high-fat diet containing 60% fat (D12492, 60% fat) for 8 to 10 weeks to induce obesity and fatty liver formation (Fig. 2). The black rice bran bioconversion product manufactured in Example 1 was administered together with the high-fat diet, and body weight and food intake were measured once a week. The average daily intake and body weight changes of the mice during the test are shown in Figs. 3 and 4.

On the other hand, despite similar daily intakes, the high-fat diet (positive control) group showed significant weight gain after 10 weeks compared to the BRB-F solid-liquid fraction group. The high-frequency extraction was performed by treating the mixture at a frequency of 900 to 1000 kHz (970 kHz) or at a power of 200 to 400 W for 2-4 hours. Thereafter, the extract was filtered through a 1.0 μm sieve and then sterilized through a 0.2 μm sieve. The extract was used in liquid form, or lyophilized and used in powder form.
