[0004] CD3+ bispecific T-cell redirection molecules engage T cells and tumor cells simultaneously to form an artificial immunologic synapse, which then results in the lysis of the target tumor cells. Since the approval of the first bispecific CD3+ T cell engager (BiTE) -Blinatumomab (anti-CD3 × anti-CD19) to treat patients with Philadelphia chromosome-negative B-cell acute lymphoblastic leukemia (ALL) by the United States Food and Drug Administration (FDA) in 2014 (D. Przepiorka et al., 2015) , there are tremendous efforts to develop next-generation CD3+ bispecific antibodies for both hematological and solid tumors with more than a hundred clinical trials (S. Wang et al., 2021) . In order to improve the clinical benefit, there are still obstacles to be overcome (A. Singh et al., 2021) . One of the major mechanisms for resistance to Blinatumomab is the upregulation of immune checkpoints after Blinatumomab therapy (J. Feucht et al., 2016; J. Sam et al., 2020) . The addition of checkpoint inhibition antibodies to CD3+ bispecific T cell redirection antibodies could reverse the resistance and improve the BiTE-mediated cytotoxicity (J. Feucht et al., 2016; J. Sam et al., 2020) . CD3 engagement and PD-L1 blockade was also integrated into one molecule to increase efficacy (M. Passariello et al., 2022) .

[0005] Many of the current generation of CD3+ bispecific antibodies lack the presence of a costimulatory signaling, which is called “signal 2” . The ligation of costimulatory molecule in T cells with its ligands is required to form an immune synapse for durable T cell activation, proliferation, and cytokine release. In the absence of a second signal, T cell stimulation through CD3 alone typically leads to activation-induced cell death (N. Borthwick et al., 2000) . The value of co-stimulation has also been validated by the successful development of 2nd-generation CAR-Ts, which include at least one costimulatory moiety (A. J. Harrison et al., 2021) . Therefore, the unsustained T cell activity may be one of the reasons that treated patients with large tumor burden had lower complete regression (CR) rate when compared to treated patients with small tumors (H. Liu et al., 2023) . There are descriptions of the combination of “costimulatory bispecifics” that cross-link a tumor-associated antigen (TAA) (P. G. Groothuis et al. ) to CD28 on T cells (TAA x CD28 bispecifics) . The TAA x CD3 bispecifics had an improved antitumor activity with the addition of a TAA x CD28 when compared to TAA x CD3 alone (D. Skokos et al., 2020) . [0006] In one aspect, the present disclosure provides novel anti-CD3, anti-CD28, and anti-PD-L1 antibodies and multispecific (e.g., bispecific, trispecific, or tetra-specific) antibodies comprising one or more of these antibodies. In some embodiments, domains with a range of affinities are integrated into a single molecule with multiple specificities, including architectures such as (1) anti-CD3 x anti-TAA, (2) anti-CD3 x anti-CD28 x anti-TAA, (3) anti-CD3 x anti-PD-L1 x anti-TAA, and (4) anti-CD3 x anti-CD28 x anti-PD-L1 x anti-TAA. These molecules are designed to vary binding affinity and molecular format to enable improved T-cell redirection for tumor cytotoxicity with decreased cytokine release for patients with unmet medical needs, such as in hematological and solid tumors.

[0279] Bispecific antibodies comprising a single anti-ROR1 arm (JNJ134) paired with a variety of anti-CD3 arms, respectively, were mixed in a series of increasing concentrations of the bispecific antibodies with PBMC and a target cell line. The target cell line was the Jeko-1 human ROR1 positive mantle cell lymphoma cell line. The results shown in Figure 6 are representative curves, demonstrating concentration dependent T cell killing as a consequence of the binding of the bispecific antibodies to both CD3 and ROR1 proteins. The data further supported the conclusion that the binding mediated the desired T cell directed target cancer cell killing. Due to the limit of the experimental setting, for example the limited coculture time, some of the variants showed minimal killing effect due to the low affinity binding of CD3 arm, which will be useful in some specific diseases to reduce the cytokine release syndrome while maintain the killing effect of diseased cells.

[0283] In order to verify the benefit of addition of an anti-CD28 arm to CD3 engagers, anti-ROR1 x CD3 x CD28, and anti-ROR1 x CD3 x inert trispecific antibodies were designed and generated. Schematic structures of the tri-specific antibodies were shown in Figure 8C. Trispecific antibodies were mixed in a series of increasing concentrations with PBMCs and a target cell line. The target cell lines were ROR1 positive MAVER-1 and MDA-MB-231, respectively. The results shown in Figure 8A (MAVER-1) and 8B (MDA-MB-231) demonstrated concentration dependent T cell killing of anti-ROR1 x CD3 x inert trispecific antibody although the killing effect is minimal, while the molecule with anti-CD28 incorporated enhanced both the potency and efficacy of T cell killing compared to the anti-ROR1 x CD3 x inert. The data further supported the conclusion that anti-CD28 arm potentiates PBMCs cytotoxicity induced by CD3 engagers.

[0285] In order to verify the enhancement of PBMCs killing by an anti-PD-L1 arm, anti-ROR1 x CD3 x PD-L1 x inert and anti-ROR1 x CD3 x inert trispecific antibodies were designed and generated. Schematic structures of the trispecific antibodies are shown in Figure 9C. The trispecific antibodies were mixed in a series of increasing concentrations with PBMCs and a target cell line. The target cell lines were ROR1 positive MAVER-1 and MDA-MB-231, respectively. The results shown in Figure 9A (MAVER-1) and 9B (MDA-MB-231) showed minimal killing effect of anti-ROR1 x CD3 x inert trispecific antibody, while the antibody with anti-PD-L1 incorporated enhanced both the potency and efficacy of T cell killing compared to the anti-ROR1 x CD3 x inert, which is especially obvious in PD-L1 positive MDA-MB-231 cell line. The data further supported the conclusion that the anti-PD-L1 arm potentiates PBMCs cytotoxicity induced by CD3 engagers.

[0287] To further verify the PBMC killing effect of anti-CD3 x CD28 x PD-L1 x EGFR, Anti-CD3 x CD28 x PD-L1 x EGFR, anti-CD3 x inert arm x EGFR x PD-L1, anti-inert arm x CD28 x PD-L1 x EGFR, and anti-CD3 x CD28 x inert x inert arm tetra-specific antibodies were designed and generated. Schematic structures of these tetra-specific antibodies were shown in Figure 10C. Tetra-specific antibodies were mixed in a series of increasing concentrations with PBMCs and a target cell line. The target cell lines were EGFR and PD-L1 positive H1975 human non-small cell lung cancer cell line and BT-20 human breast cancer cell line with low EGFR and high PD-L1 expression. The results shown in Figure 10A (H1975) and 10B (BT20) demonstrated concentration dependent T cell killing of anti-CD3 x CD28 x PD-L1 x EGFR tetra-specific antibody. The killing effects were CD3 and TAA dependent, as the comparator molecule either without TAA (EGFR / PD-L1) binding arms or without a CD3 binding arm did not produce any killing effect for both cell lines. The antibody without CD28 binding domain slightly compromised the killing potencies (increased EC50 values) as observed in both cell lines.

[0288] Example 10: Anti-tumor efficacy of anti-CD3 x 5T4 bispecific antibodies in PBMC humanized mouse model

[0289] The efficacy of anti-CD3 x 5T4 bispecific antibodies in tumor cell killing was evaluated in a PBMC humanized mouse tumor model. The novel anti-CD3 (huSP34_v1) was paired in a bispecific antibody with an anti-5T4 arm, denoted as 5T4-2, known to be capable of binding to 5T4, on tumor cells to assess the ability of huSP34_v1 to direct T cell killing of a 5T4 expressing cell. The positive control anti-CD3 arm was designated as GEN_CD3 with well-defined capability to bind to CD3 on T cells, and was paired with 5T4-2 as well to make a bispecific antibody as benchmark. These bispecific antibodies were evaluated in two 5T4 positive tumor models-BxPC3 human pancreatic cancer and MDA-MB-231 human breast cancer models, respectively. PBMCs containing CD3 positive effector T cells were engrafted in immunocompromised mice 3 days before BxPC3 tumor cell inoculation and 6 days after MDA-MB-231 cell inoculation depending on the tumor growth rate. The humanized tumor-bearing mice were randomized into treatment groups when the tumors were established and the first day of dose administration was denoted as day 0. The test molecules were intraperitoneally administered to the mice and the antibody-mediated tumor shrinkage was assessed (Figure 11A and 11B) . The results of the experiment shown in Figure 11 demonstrated that the 5T4-2 / huSP34_v1 were highly active in inhibiting tumor growth in both models, with low and medium dose levels (0.5 and 1 mg / kg) showing even better tumor inhibition effect than high dose level (3 mg / kg) , which could be explained by the bell‐shaped concentration response relationship observed for CD3 bispecific antibodies (A. Betts et al., 2020) . The benchmark molecule, 5T4-2 x GEN_CD3 at high dose level (3 mg / kg) , only showed minimum tumor inhibition effects in both models, and the anti-tumor activity was either comparable or inferior to 5T4-2 / huSP34_v1 in BxPC3 (Figure 11 A) and in MDA-MB-231 (Figure 11 B) xenograft models, respectively. A primary conclusion is that huSP34_v1 of the bispecific tested in this experiment is highly efficacious in redirecting CD3+ T cells for killing against the two solid tumor models in vivo.

[0291] The efficacy of anti-CD3 x CD28 x PD-L1 x EGFR multispecific antibodies in tumor cell killing was evaluated in a PBMC humanized mouse tumor model. The tetra specific antibody - anti-CD3 x CD28 x PD-L1 x EGFR, along with comparator molecule lacking specificities to both CD28 and PD-L1 were evaluated in a BT20 TNBC model, which has low EGFR and high PD-L1 expression. PBMCs containing CD3+ effector T cells were engrafted in immunocompromised mice 10 days before BT20 tumor cell inoculation. The humanized tumor-bearing mice were randomized into treatment groups when the tumors were established and the first day of dose administration was denoted as day 0. The tumors from some of the treatment groups were collected at the end of the study (day 25) to assess the infiltrated immune cells by flow cytometry. The test molecules were intraperitoneally administered to the mice and the antibody-mediated tumor shrinkage was assessed (Figure 12A) . The results of the experiment shown in Figure 12A demonstrated that the tetra-specific antibody - anti-CD3 x CD28 x PD-L1 x EGFR, was active in inhibiting well established BT20 tumors at both dose levels tested. With both PD-L1 and CD28 targeting arms silenced, the bispecific CD3 x EGFR even showed a trend of tumor growth promoting effect, highlighting the critical role of CD28 and PD-L1 targeting arms. Consistent with the observed tumor growth inhibition effect, the infiltration of CD45+human lymphocytes were increased in the tumor treated with the tetra-specific antibody (Figure 12B) . A primary conclusion is that the anti-CD3 arm-huSP34_v1, anti-CD28 arm-CD28-8 and anti-PD-L1 arm -PD-L1_56tav1 of the tetra-specific antibody tested in this experiment are highly efficacious either as T cell redirector and stimulators or as homing TAA in the BT20 TNBC model in vivo.
