[0004] The survival way of tumor cells is different from that of normal cells, so does the energy intake and utilization. The common metabolic pathway in aerobic organisms is tricarboxylic acid cycle, in which isocitrate dehydrogenase (IDH) catalyzing the conversion of isocitrate to α-ketoglutaric acid (α-KG) is a rate-limiting step. The known IDH family comprises three isozymes, IDH1, IDH2 and IDH3, which are located in different organelles and perform the same biological functions, i.e., catalyzing the formation of α-KG. Recent studies have shown that heterozygous IDH1 / 2 mutations were present in a certain proportion of a variety of tumors, such as glioma (60-80%) , chondrosarcoma (55%) , acute myeloid leukemia (15-25 %) , etc. The mutant IDH1 or IDH2 loses the capability of catalyzing the conversion of isocitrate to α-KG, whereas has the ability of catalyzing the reaction of α-KG to α-hydroxyglutaric acid (2HG) , which then interferes with normal cell differentiation, and results in the proliferation of immature cells. Many evidences have shown that, the mutant IDH inhibitors can effectively inhibit the generation of 2HG mediated by mutant IDH in cells and can induce differentiation of abnormally proliferating cancer cells. Furthermore, the mutant IDH1 / IDH2 inhibitors have also shown significant effects on diseases with IDH gene mutations in clinical trials. Therefore, inhibition of mutant IDH enzyme activity is useful for treating diseases induced by IDH mutations, such as cancer.

[0005] PCT patent application WO2019047909A1 disclosed the compound (S) -3- (4, 6-bis ( ( (R) -1, 1, 1-trifluoropropan-2-yl) amino) -1, 3, 5-triazin-2-yl) -2, 6, 6-trifluorocyclohex-2-e n-1-ol for the first time (i.e. Compound 197 in WO2019047909A1) as well as the preparation thereof. The chemical structure of the compound is shown as Formula A:

[0006] Studies have shown that, the compound (S) -3- (4, 6-bis ( ( (R) -1, 1, 1-trifluoropropan-2-yl) amino) -1, 3, 5-triazin-2-yl) -2, 6, 6-trifluorocyclohex-2-e n-1-ol can effectively inhibit mutant IDH enzyme activity. Thus, it is useful for preventing and treating diseases induced by IDH mutations, such as cancer.

[0007] The phenomenon that a compound could exist in two or more crystal structures is known as polymorphism. Many compounds may exist as various polymorph crystals and also in a solid amorphous form. However, until polymorphism of a compound is discovered, it is highly unpredictable (1) whether a particular compound will exhibit polymorphism, (2) how to prepare any such unknown polymorphs, and (3) how are the properties, such as stability, solubility, hygroscopicity, etc., of any such unknown polymorphs. See, e.g., J. Bernstein "Polymorphism in Molecular Crystals" , Oxford University Press, (2002) .

[0008] Since the properties of a solid material depend on the structure as well as on the nature of the compound itself, different solid forms of a compound can and often do exhibit different physical and chemical properties as well as different biopharmaceutical properties. Differences in physicochemical properties can be determined, analyzed and compared through a variety of analytical techniques. Those differences may ultimately be used to differentiate among different solid forms. For example, differences in physical properties, such as solubility, stability, and biopharmaceutical properties, such as bioavailability, are also of importance when describing the solid state of a pharmaceutical compound.

[0009] Contents of the InventionSummary of the Invention

[0010] Upon extensive exploration and research, the inventors have found that the compound of Formula A, i.e. (S) -3- (4, 6-bis ( ( (R) -1, 1, 1-trifluoropropan-2-yl) amino) -1, 3, 5-triazin-2-yl) -2, 6, 6-trifluorocyclohex-2-e n-1-ol can exist in different crystalline forms (i.e. polymorphs) , and can form solvates with certain solvents. The inventors have made extensive studies on the polymorphs of the compound of Formula A and have finally prepared and determined the crystalline forms which meet the requirement of pharmaceutical use. Based on these studies, the present invention provides the various crystalline forms of the compound of Formula A and the solvates and the crystalline forms thereof, which are designated as Form I, Form III, Form IV, Form V, Form VI, Form VII, Form VIII, and Form XI respectively.

[0011] In one aspect, the present invention provides the polymorphs of the compound of Formula A or the solvates thereof, which having the characteristics of good crystallinity, reproducible preparation, high solubility, non-hygroscopicity and good stability.

[0012] Firstly, the present invention provides Form I, Form IV, Form VII, Form VIII, and Form XI of the compound of Formula A.

[0013] Secondly, the present invention provides the solvates of the compound of Formula A, which are tert-butanol solvate, and hydrate of the compound of Formula A.

[0014] Further, the present invention provides mono-tert-butanol solvate, hydrate containing one molecule of water, and hydrate containing 5.25 molecules of water of the compound of Formula A. Even further, the present invention provides mono-tert-butanol solvate of the compound of Formula A, which is Form III of the compound of Formula A; hydrate (containing one molecule of water) of the compound of Formula A, which is Form V of the compound of Formula A; hydrate (containing 5.25 molecules of water) of the compound of Formula A, which is Form VI of the compound of Formula A.
