[0002] The present disclosure generally relates to inhibitors of proteases, in particular, to inhibitors of Cathepsin L (CatL) and their pharmaceutical use for the treatment or prevention of CatL-related diseases.BACKGROUND

[0003] Human cathepsins are found to be involved in multiple physiological processes and are highly related to most human diseases, including osteoimmunology, osteoarthritis, oral diseases, cancer, liver diseases, pulmonary fibrosis, kidney diseases, atherosclerosis, Alzheimer’s disease, Parkinson’s disease and neuroinflammation. Cathepsin is a lysosomal protease with serine, cysteine, or aspartic acid residues as the nucleophiles, which is vital for digestion, coagulation, immune response, peptide synthesis, etc. There are generally three families of cathepsin: serine proteases (cathepsins A and G) , aspartic proteases (cathepsin D and E) , and eleven cysteine proteases (cathepsins B, C, F, H, K, L, O, S, V, X, and W) .

[0004] As a member of the cysteine protease family of cathepsin, CatL plays a key role in various biological and disease processes through intracellular and extracellular protein degradation. CatL is produced as preprocathepsin L, transported via the Golgi apparatus as procathepsin L in secretory vesicles and then stored as mature cathepsin L in lysosomes. In contrast to most of the other cysteine cathepsins, CatL is nearly ubiquitously expressed in all tissues and it is involved in multiple physiological processes, including apoptosis, and extracellular matrix modeling. CatL regulated intracellular protein turnover is involved in several important processes. Cell cycle was affected by CatL as it is able to degrade nuclear transcription factors. CatL also plays a role in the immune system, in particular by degrading the invariant chain in MHC class II processing which is a critical step in antigen presentation. The expression of CatL in the thymus was shown to be essential for development of natural killer cells. In central nervous system and neurodegenerative diseases, CatL is involved in recycling processes during axon outgrowth and synapse formation. Increased extracellular release of CatL from α-Syn-activated microglia through exosomes amplifies and aggravates of the neurotoxic effect of microglia, implying that CTSL is involved in a fresh mechanism of Parkinson’s disease (PD) pathogenesis. Recently, CatL is known to be involved in some oral diseases, uncluding oral lichen planus and dental caries. It is also implicated in pathological conditions, such as tumor invasion and metastasis. CatL expression is dysregulated in a variety of cancers, including glioma, melanoma, pancreatic, breast, oral squamous cell carcinoma, and prostate carcinoma. Extensive empirical evidence indicates their direct participation in cancer growth, angiogenic processes, metastatic dissemination, and the development of treatment resistance. Recently, CaL has been reported to play a major proangiogenic role and plays a critical role in endothelial progenitor cell mediated neovascularization. In addition, large amounts of CatL could be activated in inflammatory cells under inflammatory conditions and CatL has a significant involvement in chronic inflammation, diabetes, cardiovascular disease, renal disease, bone disease, as well as viral infection. Besides its above roles, CatL is also significantly involved in the development of proteinuria and deterioration of renal function contribute to glomerular fltration barrier dysfunction through enhanced production of reactive oxygen species and the cleavage of the large GTPase, dynamin as well as synaptopodin in podocytes. Long-term increased tubular protein load further increases CatL activity in the proximal tubule and degrades tubular protein in the progression of renal disease.

[0005] Thus, it is desirable to develop novel CatL inhibitors for therapeutic uses in treating or preventing CatL-related diseases.SUMMARY

[0006] According to an aspect of the present disclosure, a compound is provided. The compound may be represented by formula (I) : wherein X is an alkyl group, an alkoxy group, or a 4-6 membered heterocyclic ring; and Y is H,  halogen, an alkyl group, an amino group, a hydroxyl group, a haloalkyl group, an alkoxy group, a cyclopropyl, or a 4-6 membered heterocyclic ring, wherein the alkyl group and the alkoxy group is optionally substituted with a 4-6 membered heterocyclic ring.
