Menin InhibitorsTECHNICAL FIELD

[0001] The present invention is directed to menin inhibitors, particularly relates to compounds represented by formula I, and further relates to a use of the compound for treating or preventing a disease or condition mediated by menin interaction.BACKGROUND OF THE INVENTION

[0002] Mixed lineage leukemia 1 (MLL1, also known as MLL or KMT2A) is a histone-H3 lysine-4 (H3K4) methyltransferase and a master regulator for transcription of important genes for embryonic development and hematopoiesis. The MLL1 gene can fuse with one of over 60 partner genes via chromosomal translocation, leading to the formation of chimeric MLL fusion proteins, which are often associated with the development of leukemia (Popovic, R., &Zeleznik-Le, N.J, 2005, J Cell Biochem, 95 (2) , 234–242) . AF4, AF9, ENL, AF6, ELL, and AF10 are the most frequent fusion partners of the MLL gene (Meyer, C. et al., 2006, Leukemia, 20 (5) , 777–784; Dou, Y., &Hess, J.L, 2008, Int J Hematol, 87 (1) , 10–18) . Transcriptional deregulation of MLL-r fusion protein target genes, including HOXA genes and MEIS1, can lead to an aggressive form of acute leukemia with limited therapeutic options.

[0003] Menin, encoded by the MEN1 gene, acts as a scaffold protein by interacting with multiple partners to regulate several important signaling pathways. While menin is ubiquitously expressed in various organs (Guru, S.C. et al., 1999, Mamm Genome, 10 (6) , 592–596; Stewart, C. et al., 1998, Oncogene, 17 (19) , 2485–2493) , its function is tissue-specific.

[0004] Menin is an essential co-factor of oncogenic MLL fusion proteins and the menin-MLL interaction is critical for development of acute leukemia in vivo. Disruption of the menin and MLL interaction has been proved to be an effective therapeutic strategy for MLL-rearranged leukemia in clinical development (Issa, Ghayas C et al. Nature. 2023; 615 (7954) : 920-924) . Additionally, a menin inhibitor has been shown to reduce the expression of downstream target genes, such as MYC and Bcl2, and results in an inhibition of the proliferation of various tumor cells, including KRAS mutated cells (Dreijerink, Koen M A et al. Endocr Relat Cancer. 2017; 24 (10) : T135-T145) . Furthermore, a menin-MLL inhibitor has been shown to enhance human β cell function and proliferation (Chamberlain et al., 2014, Journal of Clinical Investigation, 124: 4093-4101) , supporting a role for inhibitors of the menin-MLL interaction in the treatment of diabetes (Yang et al., 2010, Proc. Natl. Acad. Sci. USA, 107: 20358-20363) , which has been demonstrated in clinical trials (Rodriguez J. et al. Diabetes. 2023; 72 (Supplement_1) : 91-LB) . There are great unmet medical needs in treatment of leukemia and KRAS-driven cancers. Clinical data demonstrate that although SNDX-5613 achieves significant initial response rates, its long-term utility is limited by the development of acquired resistance mutations in menin (majorly T349M, M327I, M327V, G331R) in approximately 38.7%of treated patients, as evidenced by next-generation sequencing of progressive disease samples (Perner, F., Stein, E.M., Wenge, D.V. et al. Nature 615, 913–919 (2023) . ) . There are great unmet medical needs in treatment of leukemia and KRAS-driven cancers. Therefore, it is of great clinical significance to develop novel menin inhibitors with good safety and efficacy profiles.SUMMARY OF THE INVENTION

[0005] After in-depth research, the inventors of the present invention have designed and synthesized a series of novel structures with good pharmacodynamics and pharmacokinetics properties.

[0010] Another object of the present invention is to provide a pharmaceutical composition comprising one of the above compounds.

[0011] Another object of the present invention is to provide use of the above compounds in the manufacture of a medicament for treating or preventing a disease or condition mediated by menin-MLL interaction or at least in part mediated by menin-MLL interaction, and in the manufacture of a medicament for treating or preventing cancer including leukemia, lymphoma and myeloma.

[0483] HEK293 cell line stably expressing hERG channel (Cat. K1236) were seeded onto the coverslips in 6 cm cell culture dish and placed in bath chamber mounted on the microscope stage. After formation of a Gigaohm seal. The whole cell configuration was obtained by applying repetitive, briefly strong suction until the membrane patch has ruptured. Membrane potential was set to ‐60 mV at this point to ensure that hERG channels are not open. Holding potential was set to ‐90 mV for 500 ms; current was recorded at 50 kHz and filtered at 10k Hz. Leaking current was tested at ‐80 mV for 500 ms. The hERG current was elicited by depolarizing membrane to +30 mV for 4.8 sec and then the voltage was taken back to ‐50 mV for 5.2 sec to remove the inactivation and measure the deactivating tail current. The maximum amount of tail current size was used to determine hERG current amplitude.

[0489] The kinetic solubility of test compound in PBS (pH 7.4) , FaSSIF (pH 6.5) and FeSSIF (pH 5.0) were determined using centrifugation method. 15 μL of stock solution (10 mM) of each sample and 485 μL of buffer were added into each vial of the cap-less Solubility Sample plate and shaked at 1100 rpm for 2 hours. The supernatant was centrifuged at 4,000 rpm for 30 minutes. An aliquot of 300 μL is transferred from the supernatant, then dispensed into another 96-vial glass insert plate and centrifuged again (4,000 rpm, 30 minutes) . 5 μL filtrate, 5 μL DMSO and 490 μL of a mixture of H2O and acetonitrile containing internal standard (1: 1) (100 nM alprazolam, 200 nM caffeine and 100 nM tolbutamide) were mixed as 100 fold diluted samples. Then 20 μL of 100 fold diluted samples were further diluted by addition of 180 μL of a mixture of H2O and acetonitrile containing internal standard (1: 1) as 1000 fold diluted samples. A certain proportion of a mixture of H2O and acetonitrile containing internal standard (1: 1) was used to dilute the diluent according to the peak shape. The dilution factor was changed according to the solubility values and the LC-MS signal response. The plate was placed into the well plate auto sampler. The samples were evaluated by LC-MS / MS analysis.

[0494] The inhibition of the enzyme activity of the compound on Menin-MLL1 was tested by FP method with serial diluted concentration. Briefly, Menin (WT / M327I / M327V / T349M / G331R) and FITC-MLL13-43 were prepared in reaction buffer (50 mM HEPES (pH 7.5) , 50 mM NaCl, 0.01%BSA, 1mM DTT) . Menin was pre-incubated with the compound for 10 min before FITC-MLL1 was added, reacted for 1 h at room temperature, and the fluorescence polarization mP value (ex / em: 485 / 520) was read. The final concentrations of Menin and FITC-MLL1 in the reaction mixture were 10 nM and 5 nM, respectively.
