Their presence may be associated with a specific disease, often directly or indirectly linked with an autoimmune response. In other cases, a disease may be associated with the absence of one or more autoantibodies. Similarly, rheumatoid arthritis requires accurate recognition of early synovial inflammation to ensure timely initiation of disease-modifying therapies; however, classical criteria frequently fail to diagnose early disease, necessitating updated, biomarkerintegrating classification systems to correctly distinguish rheumatoid arthritis from other inflammatory arthritides. Psoriatic arthritis presents additional diagnostic challenges due to its heterogeneous clinical domains and the absence of definitive diagnostic biomarkers, making differentiation from rheumatoid arthritis, osteoarthritis, crystal-induced arthritis, and axial spondyloarthritis essential to avoid misclassification and delayed intervention. In inflammatory bowel diseases, including Crohn’s disease and ulcerative colitis, accurate diagnosis demands a combination of endoscopic evaluation, radiologic imaging, and biomarker analyses to distinguish between these two closely related yet pathologically distinct disorders, as no single blood test can confirm either condition and symptoms frequently overlap with non-inflammatory gastrointestinal diseases. Across all these entities, misdiagnosis may lead to inappropriate therapies, delayed intervention, progression of irreversible tissue or organ damage, and reduced treatment efficacy. Therefore, there is a substantial unmet medical need for diagnostic approaches capable of reliably discriminating among these conditions and identifying diseasespecific molecular signatures.

[0145] IgG was isolated from patient serum using affinity chromatography. IgG purification was carried out at room temperature, and serum samples as well as the eluted IgG were kept on blue ice. Serum samples from patients were diluted four-fold in binding buffer (20 mM Na2HPO4 and 20 mM NaH2PC>4 at pH 7) and centrifuged for 10 min at 4 °C and 10000 x g. Using a syringe and needle, the serum was aspirated from underneath the lipid layer and transferred to a new tube on ice.
