Composition for improving cognitive function containing a complex of natural extracts and the use thereof
[0001] The present invention relates to a composition for improving cognitive function comprising a natural extract complex and the use thereof.
[0002] This invention is a study conducted with the support of the Gyeonggi Regional Cooperation Research Center (Project No. GRRCAjou2023-B01).
[0003] Cognitive impairment refers to an intermediate state where cognitive function has declined compared to normal elderly individuals, but independent daily living is still possible, making it impossible to diagnose as dementia; it signifies a condition with a very high risk of developing dementia. This decline in cognitive function is distinct from the natural phenomena that occur during the normal aging process and can be severe enough to interfere with daily life.
[0004] Mild cognitive impairment (MCI) is an early stage at risk of progressing to dementia, referring to a state where cognitive decline occurs but does not significantly affect daily life. Generally, MCI begins five years before the onset of dementia, and after six years, about 80% of these individuals reach the state of dementia.
[0005] While the causes of cognitive impairment are diverse, apoptosis, which leads to the damage and loss of nerve cells, is known to be the ultimate cause. Reactive oxygen species, which are highly oxidative oxygen molecules that damage cells, are identified as a major cause of neuronal cell death, the leading cause of degenerative neurological diseases. Oxidative stress caused by reactive oxygen species inflicts irreversible damage on brain cells and leads to cognitive impairments such as Parkinson's disease and Alzheimer's disease by causing changes in the structure and function of mitochondria.
[0006] As we enter a super-aged society, the incidence of neuropsychiatric disorders and Alzheimer's disease is increasing, and recently, 43.9% of the 2030 generation are suffering from 'Young-Alzheimer's,' which refers to severe forgetfulness in young adults.
[0007] To date, many patents related to compositions having efficacy in treating cognitive impairment, improving cognitive ability, or enhancing memory have been disclosed and registered in Korea. Publication No. 2014-0144785 relates to a composition for enhancing memory and learning ability containing yuzu extract as an active ingredient, and Registration No. 10-1823892 relates to an impatiens extract exhibiting effects of improving memory, improving cognitive ability, and preventing, delaying, or treating dementia.
[0008] In addition, representative drugs currently on the market for the treatment of cognitive impairment include cholinesterase inhibitors (donepezil, rivastigmine, galantamine) and NMDA receptor antagonists (memantine). Donepezil increases the concentration of neurotransmitters by inhibiting the breakdown of acetylcholine in the brain, while memantine is effective in preventing cell damage by inhibiting excessive stimulation of nerve cells. However, side effects such as nausea, diarrhea, muscle spasms, and sleep disorders have been reported with donepezil, and side effects such as headache, drowsiness, dizziness, and vomiting may also occur with memantine.
[0009] Therefore, the development of therapeutic agents using biocompatible natural materials is advancing, and there is also a need to develop new materials with fewer side effects.
[0010] In order to solve the above-mentioned problems, the inventors completed the present invention by confirming the efficacy of a natural extract complex mixed with natural substances to improve cognitive ability and memory, based on the fact that natural substances such as Perilla frutescens var. acutakudo, Moringa oleifera, and amla (Phyllanthus emblica) are easily accessible, economical, and effective in improving cognitive ability.

[0070] Example 2. Construction of a Sleep Deprivation (SD) Stress Model
[0071] To induce a decline in cognitive ability in mice, a sleep deprivation stress model was constructed. First, eight platforms were installed in the rat cages and four in the mouse cages, and water was filled up to 1 cm below the platforms. Subsequently, utilizing the principle that placing experimental animals on the platforms prevents them from reaching a sleep state because their bodies come into contact with the water when they fall asleep or doze off, rats or mice were placed in each cage and sleep was continuously deprived for approximately 72 hours while changing the water every 24 hours.

[0074] To determine whether the natural extract complex was effective on mice with induced cognitive decline, a Y-maze test was conducted. The Y-maze test is an experiment designed to evaluate the voluntary alternation behavior ability of mice and was performed on an acrylic plate measuring 3 cm in width, 36 cm in length, and 15 cm in height. After designating zones A, B, and C, mice were carefully placed in one zone, and their movement paths within the zones they entered were observed and recorded for 8 minutes. Subsequently, spontaneous alteration was evaluated by measuring the number of times and the order in which the mice entered each maze. One point was awarded for entering three different zones sequentially, while no points were awarded for non-consecutive entry. Alternation behavior (%) was calculated using the following formula.

[0078] Example 4. Passive avoidance task
[0079] To determine whether the natural extract complex was effective on mice with induced cognitive decline, a passive avoidance experiment was conducted. The passive avoidance experiment, designed to evaluate the short-term memory of mice regarding unpleasant stimuli, was performed in a shuttle box consisting of two chambers—a light room and a dark room—made of black polyvinyl plastic with a 20 × 20 × 20 cm floor covered with a 2 mm grid. The chambers were separated by a guillotine door. The light room was equipped with a 50W light bulb, while the dark room was connected to an electrical stimulator to allow electricity to flow along the grid. Mice were placed in the illuminated section and given a 20-second exploration period; afterward, the guillotine door was opened, allowing them to enter the dark section. Once it was confirmed that all four paws had entered, the door was closed, and a 0.5 mA electrical stimulus was applied for 3 seconds to initiate fear learning (acquisition trial).
[0080] After 24 hours, the mice were placed back in the brightly lit compartment, and after a 20-second search period, the guillotine door was opened to allow entry into the dark compartment. The time it took for all four paws to enter the dark compartment (latency time) was measured (retention trial). [0082] Example 5. Measurement of cell viability
[0083] To confirm the neuroprotective efficacy of the natural extract complex according to the present invention, cell viability was evaluated using an MTT assay.
[0084] First, place 5 × 10 HT22 cells per well in a 96-well plate.3 After seeding the cells, they were cultured for 24 hours. Subsequently, the cells were divided and pretreated for 2 hours with a single natural extract and a natural extract complex dissolved in DPBS (Dulbecco's Phosphate-Buffered Saline). After pretreatment, glutamate (final concentration 10 mM) was added to each well, and the cells were cultured for an additional 24 hours.
