Abstract
Description
Field of the Invention
[0001] The present invention relates to a novel indoor space spray composition comprising a pyrethroid insecticide as an active ingredient. Specifically, the present invention relates to a novel and effective insecticidal composition comprising cyfluthrin (Renofluthrin) alone or in combination with at least one other insecticide, for delivery by aerosol, trigger spray, or single-shot delivery, wherein the composition exhibits significant activity in controlling flying and crawling insects. Background of the Invention
[0003] Pest control inside and outside the home is a never-ending problem. Mosquitoes and other crawling and flying insects need to be controlled to prevent the spread of vector-borne diseases. Flying insects like mosquitoes and flies, and crawling insects like cockroaches and red ants, are known to carry disease-causing bacteria. Flying insects cause significant human suffering and are a global problem. Diseases such as malaria, filariasis, chikungunya, dengue fever, yellow fever, Japanese encephalitis, Ross River virus, Burmese forest fever, and Murray Valley encephalitis are all transmitted by mosquitoes. Crawling insects like ants and cockroaches can also cause health problems. Cockroaches can be carriers of intestinal diseases such as diarrhea, dysentery, typhoid fever, and cholera.
[0004] Furthermore, dust mites should not be neglected, as they are the most common indoor allergen. They are tiny organisms, almost invisible to the naked eye, that feed on indoor dust and moisture in the air. They can trigger asthma in humans.
[0005] Aerosol cans, oil sprays, mosquito coils, and slow-release fogging systems (including mosquito mats and liquid atomizers) are the most commonly used technologies for controlling household insect infestations. Neem oil, pyrethrins, malathion, and carbaryl are all common household insecticides. However, long-term exposure to the odor or toxic chemical emissions of common commercial insecticides may pose health risks. Excessive exposure to these commonly used insecticides has been linked to negative health effects and the development of insecticide resistance in vectors, which is a concern.
[0006] Pyrethrum is a natural insecticide with high insecticidal activity and low toxicity to mammals. However, natural pyrethrum is easily oxidized by air (photooxidation) and degraded under light, making it unsuitable for insecticide use.
[0007] In order to retain insecticidal activity and reduce toxicity to mammals, many compounds with slightly modified natural pyrethrum structures have been synthesized.
[0008] While some of these compounds (synthetic pyrethroids) possess these properties, conventional aerosol compositions can be highly flammable and toxic due to inhalation, and can be harmful to the environment due to the need to continuously formulate high active concentrations. Aerosols are designed to be effective even in enclosed spaces, so exposure to such high doses of insecticide compositions could be considered a health risk to the user. Therefore, it is necessary to formulate a suitable composition that not only has higher efficacy but also protects the molecules from photooxidation as much as possible, ensuring a high degree of persistence of the insecticide action at lower active concentrations.
[0009] Recently, the inventors of IN4121 / MUM / 2013 developed an active ingredient, "Renofluthrin," which has been demonstrated to be an effective household insecticide. Renofluthrin has been shown to be effective and practical in existing formulations and also has the potential to develop novel formulations and delivery systems. The active ingredient, "Renofluthrin," is a composition of isomers of 2,3,5,6-tetrafluoro-4-(methoxymethyl)benzyl-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropanecarboxylate, with the total trans isomers (naturally occurring 1R, 3S, and 1S, 3R in nearly equal amounts) comprising 90% to 99.90% and the total cis isomers (naturally occurring 1R, 3R, and 1S, 3S in nearly equal amounts) comprising 0.1% to 10%.
[0010] There is a need in the art to formulate an indoor spray composition that is safer and more economical to use. These goals can be achieved by providing an insecticide composition that can achieve a higher knockdown effect at a lower active dose than conventional insecticides. Furthermore, there is a need in the art for a pyrethroid insecticide composition that has both insecticidal and acaricidal properties.
[0011] Therefore, the present invention aims to overcome the problems in the prior art and provide a stable and effective indoor spray composition comprising cyfluthrin alone or in combination with other insecticides / miticides known in the art. Summary of the Invention

[0118] Toxicity profile of cyfluthrin
[0119] Toxicity and efficacy play crucial roles in insecticide / pesticide compounds. New insecticide compounds are developed based on their efficacy against target pests and their toxicological profiles.
[0120] Acute inhalation toxicity:
[0121] Inhalation toxicity is of paramount importance in mosquito and flying insect repellent products. Most flying insect repellent products rely on the evaporation and vaporization of the active ingredient. Household insecticide product forms such as coils, mats, aerosol sprays, trigger sprays, liquid vaporizers, and other combustion and vaporization formulations all rely on the evaporation of the active ingredient. The insecticide's airborne concentration and its evaporation are the most important aspects of an insect repellent product.
[0122] Inhalation toxicity is measured using the "lethal concentration" (LC50) value. The LC50 (lethal concentration) is expressed as the weight of the test substance per standard volume of air (i.e., mg / L). The higher the LC50 (mg / L) value, the lower the inhalation toxicity and the safer the product.
[0123] Known insecticide compounds with a similar mode of action have the following reported LC50 values:
[0124] The insecticide "Prallethrin" LC50 is 0.658 mg / L (the unit of measurement reported in the literature is 658
[0125] mg / m3, converted to mg / L is 0.658)
[0126] The LC50 of the insecticide "Transfluthrin" is 0.513 mg / L (the unit of measurement reported in the literature is
[0127] 513mg / m3, converted to mg / L is 0.513)
[0128] The insecticide "Metofluthrin" LC50 is reported to be >1.08 and <1.96 mg / L
[0129] These results have been published and reported for the respective pesticides.
[0130] An acute inhalation toxicity study of renofluthrin was conducted on rats according to OECD Guideline No. 403. In the Globally Harmonized System of Classification and Labelling of Chemicals (GHS), the insecticide "renofluthrin" of the present invention belongs to Category 4 for dust and mist (mg / L), i.e. >1.0 and <5 mg / L.
[0131] The LC50 of the insecticide "Renofluthrin" of the present invention is greater than 3.81 mg / L, which indicates that among all compounds commonly used as active ingredients of household insecticides, Renofluthrin has the lowest inhalation toxicity. Contains Renofluthrin and its components The biological efficacy and activity of the combined insecticide aerosol composition.
[0132] The bioavailability of a cyfluthrin aerosol composition against target pests was determined by comparing it to the bioavailability of a commercial aerosol composition containing d-trans allethrin and transfluthrin. Aedes aegypti is the primary vector of dengue, chikungunya, yellow fever, and other viral diseases. Peet Grandy ) test chamber to test the effectiveness and efficacy of the aerosol composition of the present invention.
[0133] The effectiveness / efficacy of mosquito repellent formulations is often measured as "knockdown time" (KT), which is the time, measured in minutes, required to knock down a certain percentage of a test mosquito population. For example, the KT50 value refers to the time required to knock down 50% of the test insect population. The shorter the KT50 time, the more effective the insecticide. Results may vary depending on study conditions, mosquito species, and protocol.
[0134] A comparative study on the bio-effectiveness evaluation of the aerosol composition of the present invention and other aerosol compositions known in the art against Aedes aegypti and German cockroaches was conducted; the results are shown in the following table:
[0135] 1) In Pete Grady Peet Grandy test Indoor, d-TransAllethrin 0.25% aerosol and Comparison of the bioefficacy of renofluthrin 0.03% aerosol against Aedes aegypti
[0136] Methods: Two replicates of each aerosol were tested against Aedes aegypti. Before pre-treatment testing with the aerosol in the smoke, the aerosol was placed in and out of the test area 2-3 times. After filling, the weight of the aerosol can was recorded. A single spray of 0.65 ± 0.10 g of the formulation was applied to the center of the test chamber through a nozzle located on the side wall. Knockdown was recorded every 1 minute for up to 10 minutes, then every 2 minutes for up to 20 minutes, and then every 5 minutes for up to 60 minutes.
[0137] Table 1: Comparison of the bioefficacy of d-trans-allethrin 0.25% aerosol and Renofluthrin 0.03% aerosol against Aedes aegypti in a Peet Grandy test chamber
[0138]
[0139] result:
[0140] As can be seen from the table above, the Renofluthrin aerosol formulation showed better efficacy than the d-trans allethrin aerosol formulation. At an active concentration of 0.03% (relative to 0.25%), the knockdown time of Renofluthrin on 50% and 90% of mosquitoes was shorter than that of d-trans allethrin.
[0141] Thus, Renofluthrin 0.03% aerosol exhibits superior efficacy at lower active concentrations.
[0142] 2) In Peet Grandy test Initial and residual biomass of aerosols tested indoors against free-flying Aedes aegypti Efficacy testing.
[0143] Methods: Mosquitoes were released in a Peet Grady chamber and, after an acclimation period, sprayed with aerosol product at a dose of 0.65 g ± 0.1 g. Knockdown readings were recorded for up to 60 minutes and the data were plotted to calculate KT50 and KT90 values using SPSS software. The product was sprayed once, and readings were recorded at 0 (fresh), 4 hours, 8 hours, and 10 hours after the start of spraying, and residual efficacy was assessed.
[0144] Table 2: RNF 0.05% aerosol, RNF 0.03% + Cypermethrin 0.1% Aerosol and Transfluthrin (TFT)
[0145] Initial and Residual Bioefficacy Testing of 0.08% Aerosol
[0146]
[0147] result:
[0148] TFT 0.08% aerosol showed residual protection for up to 4 hours.
[0149] RNF 0.03% + CYPERMETHRIN 0.1% aerosol showed residual protection for up to 8 hours.
[0150] The RNF 0.05% aerosol formulation had the highest residual efficacy, lasting 8 to 10 hours.
[0151] Under the test conditions, 100% mortality was observed within 10 hours for RNF 0.05% aerosol, however, this was up to 8 hours for RNF 0.03% + Cypermethrin 0.1% aerosol, and only up to 4 hours for the TFT 0.08% aerosol formulation.
[0152] Thus, RNF 0.05% aerosol showed enhanced residual protection against mosquitoes in the test.
[0153] 3) RNF0.05%, RNF0.03%+Cypermethrin0.1% and Deltamethrin0.02%+Allethrin0.13% Biological efficacy of aerosol samples against German cockroaches on cement surfaces
[0154] Method: The product was sprayed on the cement board. After drying, 10 cockroaches were exposed to the panel twice. The mortality was checked after 30 minutes of exposure and after 4 and 24 hours.
[0155] Table 3: Biological efficacy of aerosol samples against German cockroaches on cement surfaces
[0156]
[0157] result:
[0158] The RNF 0.05% aerosol formulation had a 30% mortality rate within 24 hours.
[0159] However, the RNF 0.03% + cypermethrin 0.1% aerosol formulation showed higher efficacy with a mortality rate of 80% compared to the art known deltamethrin 0.02% + allethrin 0.13% aerosol (24 hour mortality rate of 55%).
[0160] Therefore, even at low doses of the active ingredients, the Renofluthrin 0.03% + Cypermethrin 0.1% aerosol formula provides excellent control of crawling insects.
[0161] 4) The direct spray knockdown method was used to treat RNF0.05%, RNF0.03% + cypermethrin
[0162] Bioefficacy of 0.1% and deltamethrin 0.02% + allethrin 0.13% aerosol samples against German cockroaches
[0163] Method: Five cockroaches were placed in a 6-inch stainless steel mesh cartridge (with stainless steel mesh on both sides) and placed at a predetermined distance in the room, and then the aerosol product was sprayed on the cartridge.
[0164] Observe and record 100% mortality in seconds; if available, record 24-hour resuscitation data.
[0165]
[0166] Results: All three treatments achieved the same 100% mortality. However, RNF 0.03% + Cypermethrin 0.1% achieved 100% KD in 161 seconds. Therefore, RNF 0.03% + Cypermethrin 0.1% demonstrated superior control in the shortest time.
