The first and second ends puncture the sealed primary and secondary containers in order to provide fluid communication between the first and second containers. The low-density high-viscosity liquid of the primary container blocks flow through the cannula upon entering therein.

A PRP separator-concentrator comprising a housing, a separation assembly, and a concentration assembly. The concentration assembly has a concentration sump. An axially concentric rigid stationary outlet tube is secured to the housing and extends through the separation assembly to the sump. The separation assembly is attached to and positioned above the concentration assembly to form a combined separator-concentrator assemblage that is rotatable about the outlet tube. The separation assembly includes a separation chamber lined with a depth filter having pores and passageways that are sized to receive and entrap erythrocytes during centrifuging. The concentration chamber has a floor for supporting desiccated beads and a wall with at least one opening closed with a screen. The concentrator can have a distribution of upright screen supports, the upright screen supports having an inner surface and an outer surface, the cylindrical screen being supported on the outer surface of the upright screen supports. A stationary bead rake can be secured to the stationary tube and extend outward therefrom, the rake having distal ends that are spaced at a distance from the upright screen supports. The rake can comprise a longitudinal body, the center of which is secured to the rigid outlet tube. The separator-concentrator includes a valve assembly connecting the separation chamber and the concentration chamber. PRP concentrate is produced by contacting PRP with desiccated beads while the beads are stirred with a stationary rake, and rotating the concentration chamber at centrifugal speeds to separate PRP concentrate from the beads.

A tube and float system for use in separation and axial expansion of the buffy coat is provided. The system includes a transparent, or semi-transparent, flexible sample tube and a rigid separator float having a specific gravity intermediate that of red blood cells and plasma. The sample tube has an elongated sidewall having a first cross-sectional inner diameter. The float consists of a main body portion and one or more support members protruding from the main body portion to engage and support the sidewall of the sample tube. The main body portion and the support members of the float have a cross-sectional diameter less than that of the first cross-sectional inner diameter of the tube when the sample tube is expanded, such as by centrifugation. The main body portion of the float together with an axially aligned portion of the sidewall define an annular volume therebetween. The support members protruding from the main body portion of the float traverse said annular volume to produce one or more analysis areas. During centrifugation, the centrifugal force enlarges the diameter of the tube to permit density-based axial movement of the float in the tube. Thereafter, the centrifugal force is reduced to cause the tube sidewall to return to its first diameter, thereby capturing the float and trapping the buffy coat constituents in the analysis area. The buffy coat constituents can then be evaluated or measured.

A flow path is provided through which oil, separated from gases generated by the internal combustion engine and collecting on the partition wall, can travel from the separation chamber to the drainage chamber. A pump wheel is coupled to a shaft portion defined by the rotor and extends radially outwardly therefrom. During operation the oil, upon exiting the separation chamber, contacts the pump wheel. Centrifugal forces generated due to the rotation of the pump wheel cause the oil to be thrown into the drainage chamber. The pump wheel also minimizes the potential for oil resident in the drainage chamber to re-enter the separation chamber.
