Description

TECHNICAL FIELD

[0001] The present application belongs to the technical field of medicine, and particularly relates to a pyrazine carboxamide compound, a preparation method, a pharmaceutical composition and application thereof. BACKGROUND

[0002] Immunotherapy has completely changed the way cancer is treated. In particular, immune checkpoint inhibitors targeting the PD-1 and CTLA-4 pathways have shown remarkable clinical effects, but they are only effective for specific patient populations and specific types of tumors, have a low tumor response rate, and most patients do not show sustained symptom relief, and drug resistance occurs after a period of use. Therefore, it is urgent to discover new tumor immunotherapy targets to improve the clinical effects of immune checkpoints.

[0003] Hematopoietic progenitor kinase 1 (HPK1, MAP4K1) is a member of the MAP4K family, which is a Ste20 serine / threonine protein kinase. HPK1 is mainly expressed in hematopoietic cells such as T cells, B cells, neutrophils, dendritic cells (DCs), natural killer (NK) cells and macrophages. HPK1 is a negative regulator of T cell receptor and B cell receptor. After TCR activation, HPK1 in the cytoplasm is recruited to the plasma membrane, and its amino acid residues Y381, S171 and T165 are phosphorylated to obtain fully activated HPK1 kinase. Activated HPK1 phosphorylates the amino acid residue S376 of its linker protein SLP76 to provide a binding site for the negative regulator 14-3-3, promotes the proteasomal degradation of SLP76, and ultimately disrupts the stability of the TCR signaling complex, thereby hindering the downstream kinase signaling pathway that promotes T cell activation and proliferation. In addition to TCR signals, HPK1 also negatively regulates T cell signals through the prostaglandin E2 (PGE2) receptor. Under the stimulation of growth factors, stress, inflammatory factors and differentiation factors, HPK1 can also transmit immunosuppressive signals in B cells, NK cells and dendritic cells. The kinase activity of HPK1 inhibits the immune function of various cells, including CD4 + T cells, CD8 +T cells, NK cells and dendritic cells, and showed that inhibition of HPK1 kinase activity was sufficient to induce anti-tumor immune responses. In addition, inhibition of HPK1 kinase activity can further promote T cell effector function, significantly increasing the therapeutic effect of PD-L1 mAb. HPK1 gene deletion, HPK1 inhibitor or HPK1 protac degrader, can enhance CAR-T cell-based immunotherapy, and show better anti-tumor activity in various preclinical hematological and solid tumor mouse models. Notably, HPK1 knockout and inactive kinase knock-in mice do not show lethal inflammatory responses, while these are significant in the absence of some other immune negative regulators, such as CTLA-4 and Cbl-b, indicating that HPK1 inhibitors have high safety. In summary, HPK1 is a potential new target for effective tumor immunotherapy, and the research and development of HPK1 inhibitors is of great significance to improve the key problems currently faced by tumor immunotherapy.

[0004] There are currently four HPK1 inhibitors in early clinical research, including CFI-402411 by Treadwell Therapeutics, BGB-15025 by BeiGene, PRJ1-3024 by Zhuhai Yufan and NDI-101150 by Nimbus Therapeutics. Although various HPK1 inhibitors have been reported, there is currently no related drug on the market. The main challenge faced by the development of HPK1 inhibitors is that the functions of HPK1 family members are different, but the structural homology is very high, and it is difficult to design highly selective inhibitors.

[0005] Compared with traditional drugs, PROTAC has significant advantages: wider range of action, higher activity, can target "undruggable" targets; improve selectivity, activity and safety; overcome tumor drug resistance, etc. PROTAC can achieve selectivity on some targets that small molecules cannot achieve, so developing HPK1 PROTAC molecules is expected to solve the problems of low selectivity and dose-dependent efficacy of HPK1 inhibitors, and there are few reports of HPK1 PROTAC molecules at present. SUMMARY

[0006] Invention purposes: In view of the low selectivity of HPK1 small molecule inhibitors, the first purpose of the present application is to provide a pyrazine carboxamide HPK1 PROTAC degrader, which can realize the selectivity of small molecules by using PROTAC, and develop a HPK1-targeted PROTAC molecule. The PROTAC of the present application can efficiently and selectively degrade HPK1 protein, and has no degradation activity on other proteins in the same family. Compared with HPK1 small molecule inhibitors, it can more efficiently stimulate T cell antitumor immune response, release effector cytokines, and has stronger antitumor activity. The second purpose of the present application is to provide a preparation method of the compound, the third purpose is to provide a pharmaceutical composition containing the compound, and the fourth purpose is to provide a pharmaceutical application of the compound and the pharmaceutical composition thereof.

[0165] The "pharmaceutically acceptable salts" of the present application can be synthesized from the parent compound that contains an acidic or basic moiety by conventional chemical methods. Generally, such salts are prepared either by contacting the free acid or base forms of these compounds with a sufficient amount of the appropriate base or acid in water or in an organic solvent or in a mixture of both to produce the desired salt. Generally, non-aqueous media like ether, ethyl acetate, ethanol, isopropanol or acetonitrile are preferred.

[0166] The term "metabolite" means a pharmacologically active product produced through metabolism in the body of a compound of formula I or salt thereof. Accordingly, the present application includes metabolites of compounds of the present application, including those produced by contact of a compound of the present application with a mammal for a period of time sufficient to yield a metabolic product thereof.

[0167] Identification of metabolites is typically performed by preparing a radiolabelled isotope of a compound of the application, administering it parenterally to an animal such as rat, mouse, guinea pig, monkey, or to man, at a dose that will result in a detectable amount of the compound in the blood or other biological fluid, waiting sufficient time for metabolism to occur (typically about 30 seconds to 30 hours) and isolating the transformation products from the urine, blood or other biological sample. These products are readily isolated because of the label (or otherwise by utilizing antibodies capable of binding to the epitope present on the metabolite). The structure of the metabolite is determined in a conventional manner, e.g., by MS, LC / MS or NMR analysis. In general, the analysis of metabolites is performed in the same way as a conventional drug metabolism study as is known to those skilled in the art. As long as the metabolite products are not otherwise found in the body, they are useful in the determination of the therapeutic dosage range of a compound of the application. The compounds of the application can contain unnatural proportions of atomic isotopes at one or more of the atoms in a compound, for example, at any position of the compounds. For example, the compounds can be radiolabeled with radioactive isotopes, such as for example tritium ( 3 H), iodine-125 ( 125 I) or carbon-14 ( 14 C). All isotopic variations of the compounds of the present application, whether radioactive or not, are encompassed within the scope of the present application.

[0168] In addition to salt forms, the compounds of this application can exist in a variety of
