Abstract

Pharmaceutical compositions and medical uses of modified human arginases are described, and in particular, inter alia, a pharmaceutical composition comprising a protein comprising a human Arginase I amino acid sequence and a non-natural metal coenzyme, wherein the non-natural metal coenzyme is cobalt; a pharmaceutical composition comprising a human Arginase I protein having a metal binding site and comprising at least one amino acid substitution in the metal binding site, wherein the protein exhibits an increase in Arginine hydrolysis resulting in a Kcat / Km at least twofold greater than that of a native human Arginase I; a pharmaceutical composition comprising a fusion protein comprising any of said proteins, as well as medical uses related thereto.
