[0042] The system is configured to determine the effect of a magnesium complex on mitochondrial membrane potential using flow cytometry. To determine the effects, 1 × 10 6CAL 27 cells are seeded into a 6-well plate and incubated overnight. The monolayer is treated with 1.7, 2, and 2.3 µM of the magnesium complex and incubated for 48 hours. Untreated control cells remain untreated under the same conditions. After the incubation period, the cells are harvested and collected in centrifuge tubes. A positive control is created by treating the cells with 2 µl of CCCP and incubating them for 10 minutes at 37°C. All samples are stained with 2 µM JC-1 dye for 30 minutes. The cells are then pelleted by centrifugation and resuspended in 500 µl of PBS. The system includes a flow cytometer configured for sample analysis using FITC and PI channels. Data processing is performed using the system's data processing unit.

[0043] The system also evaluates the cell cycle modulating effects of the magnesium complex. For this purpose, 2 × 10 6CAL 27 cells were seeded into a 6-well plate and incubated overnight. The monolayer was treated with 1.7, 2, and 2.3 µM of the magnesium complex and incubated for 48 hours. Control cells remained untreated. After treatment, the cells were harvested and collected in centrifuge tubes. The samples were washed with PBS and fixed overnight at 4°C with chilled 70% ethanol. The ethanol solution was added dropwise while vortexing to prevent clumping. After fixation, the cells were pelleted at 300 x g for 30 minutes and washed three times with PBS. The system processed the samples by treating them with 0.5 µg / ml DNase-free RNase solution and then incubating them for 2 hours at 37°C. Subsequently, 50 µg / ml PI was added, and the samples were incubated for 1 hour at room temperature. The flow cytometer is used for cell cycle analysis on the PI channel and the data processing unit processes the data.

[0044] The system also includes a gene expression analysis module by 1 × 10 6 CAL 27 cells per well in a 6-well plate were treated with 1.7 and 2 µM of the magnesium complex for 48 hours. Control cells remained untreated. Total RNA was extracted using a reagent according to the manufacturer's protocol. This procedure involved treating the cells with the reagent and chloroform to separate the aqueous phase, followed by RNA purification with isopropyl alcohol and 75% ethanol. The RNA samples were then transcribed into cDNA using a First Strand cDNA Synthesis Kit. The synthesized cDNA served as a template for gene expression analysis using Maxima SYBR Green / ROX qPCR Master Mix along with forward and reverse primers of the target gene.

[0045] The system is configured for immunocytochemistry. CAL 27 cells are seeded in a 48-well plate at a density of 40,000 cells per well and incubated overnight. The cells are then treated with 1.7 and 2 µM of the magnesium complex for 48 hours, while the control cells remain untreated. Following treatment, the cells are washed three times with PBS, fixed with 4% paraformaldehyde for 15 minutes, and permeabilized with 0.1% Triton X-100 for 10 minutes. [0046] The system includes a module for assessing acute toxicity using Balb / c mice of both sexes, randomly divided into four groups. The magnesium complex is dissolved in drinking water containing 4% DMSO. The control group receives only the vehicle, while the treatment groups receive a single oral dose of 10, 50, and 250 mg / kg body weight. The system continuously monitors behavioral changes for three hours, followed by half-hourly observations for the next four hours. Mortality is monitored until the fourth day after administration. Following the study, organs such as the kidney and liver are surgically removed, fixed in 4% formaldehyde, and gradually dehydrated with 70%, 90%, and 100% ethanol. The system processes tissue samples by incubating them in a 1:1 ratio of xylene and paraffin for one hour, followed by incubation in paraffin alone overnight at 65°C.Tissue sections of 5-6 micrometers are prepared on slides, rehydrated stepwise with decreasing ethanol concentrations (100%, 90%, and 70%), and stained with hematoxylin and eosin Y dyes. The slides are mounted with DPX and visualized under a bright-field microscope at 10x magnification.

[0047] The system's data processing unit is also configured to perform statistical analyses of the data obtained and then present the results in a graphical representation.

[0048] The results showed that the synthesized magnesium complex exhibited potent anticancer activity against OSCC cells (CAL 27) while exhibiting significantly lower toxicity to normal human cells (BJ cells). It effectively induced apoptosis by disrupting the mitochondrial membrane potential, causing DNA condensation, and driving the cells into the late apoptotic stage. The complex arrested the cell cycle in the G1 phase, reduced clonogenic potential, and significantly inhibited migration and invasion. At the molecular level, the treatment downregulated key oncogenic genes and proteins, including WNT, β-catenin, c-MYC, COX-2, RAS, MMP-2, EGFR, ERK, and VEGF-A, while upregulating pro-apoptotic markers such as BAX, caspase-3, and E-cadherin. This modulation of gene and protein expression suppressed the proliferation, migration and angiogenesis of cancer cells. In vivo toxicity studies showed no significant adverse effects on liver and kidney tissue, confirming its safety at higher doses. These results suggest that the magnesium complex is a promising anticancer agent, with selective cytotoxicity against OSCC cells, minimal toxicity to normal cells, and potent pro-apoptotic and antimetastatic properties.

[0049] The results suggest that the magnesium complex is a novel metal-based anticancer compound that demonstrated potent cytotoxic and antimigratory effects against oral squamous cell carcinoma (OSCC) in vitro, while remaining non-toxic to normal human fibroblast cells. The complex significantly induced apoptosis and reduced the clonogenicity, migratory, and invasive potential of cancer cells. The treatment also modulated the expression of apoptotic and metastatic genes and proteins. The complex showed no major signs of toxicity in the acute in vivo toxicity analysis in mice. Taken together, the data suggest that the magnesium complex may be a potential drug candidate for OSCC therapy.
