Description

Title of the invention: NEW COSMETIC COMPOSITIONS FOR INJURED SKIN

[0001] The present invention falls within the field of dermo-cosmetic or dermatological compositions for the treatment of damaged skin, in particular cracked and / or fissured skin.

[0002] More particularly, the present invention relates to a dermatological or dermo-cosmetic composition in the form of an emulsion, in particular oil-in-water, of the type applicable topically, particularly suitable for the repair of damaged skin. The invention also relates to the use of such a composition for the repair of damaged skin, in particular for the repair of skin lesions such as cracks, fissures and crevices.

[0003] The skin is made up of different tissues that form a vital barrier for the body against the external environment. This barrier protects the body against external attacks, particularly chemical or mechanical.

[0004] The skin is made up of three main parts, the superficial one, the epidermis, the internal part, the dermis and a deeper layer, the hypodermis, which interact with each other.

[0005] The human epidermis is composed of four to five distinct layers (depending on the anatomical site) and four types of cells which are keratinocytes, which are the vast majority, melanocytes, Langerhans cells and Merkel cells. Each of these cell types contributes through its own functions to the essential role played in the body by the skin, in particular the role of protecting the body against external aggressions. The epidermal cells proliferate at the level of the deepest layer, the basal layer, and differentiate during their migration towards the upper layers to successively form the spinous layer, the granular layer and finally the horny layer (or stratum corneum) which is the most superficial layer of the epidermis.

[0006] The skin thus forms a vital barrier for the body against external attacks, particularly chemical, mechanical or infectious.

[0007] It is clear that the quality of the skin barrier and mucous membranes is dependent on complex endogenous biological mechanisms involving numerous growth and differentiation factors, adhesion molecules, hormones and lipid metabolism enzymes.

[0008] Thus, an alteration of the skin barrier and / or a break in the continuity of the skin surface can occur in the presence of external aggressions (cold, humidity...), mechanical stress (friction, shocks, abrasion, tearing of the surface, shaving or hair removal...). Thus when the skin barrier is damaged, the skin loses its shield, water loss accelerates and, inevitably, the skin dries out, or even becomes dehydrated. While transepidermal water loss is a normal phenomenon, dehydration is not. When the hydrolipidic film is damaged, it no longer plays its role as a protective barrier and the water contained in the skin then evaporates more.

[0009] Dehydrated skin becomes more fragile, it can no longer withstand overuse, it ends up tearing, creating cracks, fissures, and even crevices.

[0010] Thus, these lesions of the skin and / or mucous membranes, which are cracks, fissures and crevices, result from the combination of intense drying, dehydration and thickening of the epidermis.

[0011] Throughout the remainder of this description, the term skin includes both the skin itself and the mucous membranes. The term cutaneous includes both that which relates to the skin and that which relates to the mucous membranes.

[0012] The term "damaged skin" within the meaning of the present description, in a conventional manner in itself, means an area of skin presenting cutaneous lesions, in particular cracks, fissures, and / or crevices.

[0013] For the purposes of the present description, the term "crack" means an area of damaged skin, more or less extensive, having a rough appearance, a red color, this area of skin is dry and flaky, with a loss of material due to small superficial cracks.

[0014] For the purposes of this description, the term "crack" means a skin lesion presenting a more or less deep furrow, with a loss of material. The cracks can reach a few millimeters in width, a depth of more than one centimeter and sometimes even bleed.

[0015] For the purposes of the present description, the term "crevasse" means a lesion of the skin having a deeper furrow in the skin than a crack.

[0016] These skin lesions generally appear on certain parts of the skin, most often on areas without hair and exposed to the air, such as the face, especially the lips, the feet, especially the heels, the hands, but also the elbows and knees, the nipples and external intimate areas.

[0017] These skin lesions are more severe and frequent in women, people suffering from obesity and people with diabetes.

[0018] These skin lesions are not only unsightly, they can also cause discomfort such as itching or burning sensations. In addition, deep cracks or crevices are an open door to pathogens such as bacteria or fungal spores. If these lesions become infected, they can degenerate and lead to serious skin ulcers.

[0019] Several solutions for dermo-cosmetic compositions adapted to this type of skin lesions exist. One of the traditional solutions consists of applying petroleum jelly in the form of a thick layer on the damaged skin area. This ingredient promotes skin self-repair by forming an occlusive film on the surface of the skin.

[0020] Solid petroleum jelly or petrolatum gel is thus widely used as a therapeutic agent for local applications. This therapeutic agent indeed has well-known characteristics of lubricating, softening and treating the skin, and many dermatologists prescribe it as an effective skin moisturizing and protecting agent for patients with dry or flaky skin. Among the many characteristics of petroleum jelly that make it useful in topical applications, mention may be made of a moisturizing property, a barrier activity with respect to water, its hydrophobicity, its resistance to washing with water and its chemical stability.

[0021] However, there are disadvantages to adding petroleum jelly to compositions for topical application. Such disadvantages include its greasy consistency, its lack of ability to produce a thin film over a large area of skin, and its occlusive nature. It is also unfortunately comedogenic. Finally, its petrochemical origin and its composition (mixture of hydrocarbons including mineral oil and microcrystalline hydrocarbon waxes) make it an increasingly criticized ingredient. It is also flammable and is regularly suspected of causing skin burns.

[0022] For all these reasons, the inventors have developed a new composition useful for repairing skin lesions, such as skin cracks and fissures, overcoming the drawbacks of the compositions of the prior art containing petroleum jelly, that is to say a new composition which, while being free of petroleum jelly, has sensorial properties at least as good as these compositions of the prior art containing petroleum jelly, and has an effectiveness as good, or even better.

[0023] An additional objective of the invention is that this composition does not contain any, or as little as possible, ingredients of petroleum-based origin.

[0024] It has now been discovered that these objectives are achieved by a dermo-cosmetic or dermatological composition combining specific ingredients.

[0025] Thus, according to the present invention, there is provided a dermo-cosmetic or dermatological composition applicable topically, in the form of an emulsion comprising an aqueous phase and an oily phase, and comprising, by weight relative to the total weight of the composition: - 5 to 10% water, - 10 to 30% polyol(s), - 45 to 70% oily emollient(s), - 4 to 15% of waxy consistency factor(s), - 1 to 2.5% of emulsifying agent(s) chosen from sucrose and fatty acid esters, this composition being free of petrolatum and paraffin.

[0026] The composition according to the invention is in the form of an emulsion, containing an aqueous phase and an oily phase, one of these phases, called the dispersed phase, being dispersed homogeneously in the other of these phases, called the continuous phase, in the form of fine droplets. This emulsion may be of the water-in-oil type. Preferably, the composition according to the invention is in the form of an emulsion of the oil-in-water type, that is to say in which the oily phase is kept homogeneously dispersed in the aqueous phase.

[0027] The composition according to the invention has a particularly good skin repair effect. This effect is felt in particular after only two days of topical application of the composition to the damaged skin, according to an administration dosage of twice a day, and it is exerted in a particularly strong manner after seven days of such treatment. Without prejudging the mechanisms underlying such skin repair effectiveness of the composition according to the invention, it can be thought that it is contributed, on the one hand, by its strong moisturizing power, which is exerted as soon as the composition is absorbed by the skin, which is maximum two hours after application, and which is maintained over a long period, for at least 24 hours after application; and, on the other hand, by its semi-occlusive nature only, allowing the skin to breathe, as opposed to petroleum jelly or paraffin-based compositions, which are highly occlusive. This performance of the composition according to the invention is not obtained to the detriment of its sensory properties, which are advantageously very close to those of petroleum jelly.

[0028] The composition according to the invention comprises from 5 to 10% by weight, preferably 6 to 10% by weight, and preferentially from 6 to 9% by weight, of water relative to the total weight of the composition.

[0096] For example, as such a hyaluronic acid, mention may be made of the hyaluronic acid supplied by the company Givaudan under the trade name CRISTALHYAL®, having a molecular weight of 1 to 1.5 MDa.

[0097] Thus, in particular embodiments of the invention, the composition further comprises hyaluronic acid with a molecular weight greater than or equal to 100 kDa, preferably greater than or equal to 1000 kDa, in particular between 1000 and 2000 kDa, or between 1000 and 1500 kDa, or one of its salts.

[0098] The main property of hyaluronic acid is its hydrophilicity. Hyaluronic acid is an exceptional water trapper since each molecule can absorb 500 to 1000 times its volume of water, which explains its strong volumizing properties. Thus, high molecular weight hyaluronic acid significantly hydrates and contributes to skin healing by stimulating regeneration and maintaining a moisture level essential for this healing.

[0099] In preferred embodiments of the invention, the composition further comprises, as active agent, an oat extract, in particular from aerial parts of oats. Preferably, the oat extract is obtained from oat seedlings, and preferably as described in application WO 2010 / 054879, i.e. an extract of aerial part(s) of oats excluding the seeds, characterized by 2 to 15% by weight of flavonoids and 0.2 to 2% by weight of avenacosides A and B. Such an oat extract has anti-inflammatory activity and helps with skin healing.

[0100] Healing agents may also be present in the composition according to the invention, alone or in combination, such as for example madecassoside, panthenol, allantoin, zinc sulfate, Aloe vera extracts or honey, or the healing complex described in application WO 2014 / 167039, comprising L- alanyl-L-glutamine, hyaluronic acid or one of its salts and an oat extract.

[0101] The composition according to the invention may be in any conventional form in itself, this form being suitable for topical application to the skin (including mucous membranes), in particular human or animal skin. It may in particular, but not limited to, be in the form of a fluid emulsion, a rich emulsion, a cream, a milk, a serum, an ointment or a balm.

[0102] The composition according to the invention is intended more particularly for topical application, on any area of damaged skin, in particular on the skin of the face, on the lips, on the skin of the feet, in particular on the heel, of the hands, but also of the elbows and knees, on the nipples and on the external intimate areas.

[0103] Another aspect of the invention relates to the use of a composition according to the invention for the repair of damaged skin, this use comprising the topical application of the composition to the area of damaged skin. The skin may be that of a mammal, in particular a human.

[0104] By repair of damaged skin, it is meant, within the meaning of the present invention, the attenuation of at least one of the physical signs of the damaged skin such as redness, roughness, dryness, edema, desquamation.

[0105] The composition according to the invention can in particular be used for the repair of skin lesions such as cracks, fissures and / or skin crevices.

[0106] The composition according to the invention can for example be applied topically, to an area of damaged skin of the subject concerned, such as the skin of the face, feet, in particular the heel, hands, elbows, knees, lips, nipples and / or external intimate areas, in an effective amount, once or twice a day, for example in the morning and evening, and for example for a period of between 1 week and 1 month or more. As explained above, the repair of the damaged skin is visible after two days of treatment, and is significant after four days of treatment.

[0107] The application of the composition according to the invention may in particular consist of spreading the composition over the entire surface of the injured area, for example at a rate of 2 g / cm2 of skin.

[0108] The invention is also expressed in terms of a method for the therapeutic treatment of an area of damaged skin of a subject, in particular a human, for the repair of this area of damaged skin, this method comprising the topical application, to this area of damaged skin, of an effective amount of a composition according to the invention.

[0109] This method may meet one or more of the characteristics described above with reference to the use of the composition according to the invention for the repair of a damaged skin.

[0110] The present invention also relates to a method for non-therapeutic cosmetic treatment of an area of damaged skin of a subject, in particular a human, for the repair of this area of damaged skin, by topical application, to this area of damaged skin, of an effective amount of a composition according to the invention.

[0111] Here again, this method can meet one or more of the characteristics described above with reference to the use of the composition according to the invention for the repair of damaged skin.

[0112] Another subject of the invention relates to the non-therapeutic cosmetic use of a composition according to the invention for the repair of an injured area of skin of a subject, in particular a human. This use comprises the topical application of this composition to this injured area of skin. It may meet one or more of the characteristics described above with reference to the use of the composition according to the invention for the repair of injured skin.

[0113] The characteristics and advantages of the invention will appear more clearly in light of the examples of implementation below, provided for purely illustrative purposes and in no way limiting the invention.

[0114] 1 / Example 1 - Occlusion test

[0115] The study consists of measuring the water loss in a glass container closed by a filter paper held by Parafilm® plastic film and on which the product to be tested is placed. In more detail, the protocol is as follows: - 80 ± 0.05 mL of water is weighed into a Kola glass bottle using a balance, - a filter paper is placed on the bottle, smooth side visible at the top of the bottle, - the filter paper is folded and surrounded using Parafilm® film, - 0.5 g of the product to be tested is placed on the filter paper using a pipette, taking care to spread the product over the entire filter paper, and the bottle is weighed: the value obtained is called T0, - all samples are placed in a study at 30°C, - after 72 hours, the bottles are weighed again: the value obtained is called T72product.

[0116] A negative control without product is also carried out. The final mass of the bottle equipped with only filter paper is then called T72blanc.

[0117] For each product tested, the occlusion factor F is calculated by the mathematical equation (1):

[0118]

[0119]

[0120]

[0121]

[0122]

[0123] [Math.l] (1) This occlusion factor F is therefore between 0 and 100, the value of 100 corresponding to a total occlusive effect. Several products are thus tested, with 3 repetitions for each: - pure vaseline (Vaseline Gifrer), - a composition in accordance with the invention (Composition 1), as described in Table 1 below, - a composition containing paraffin, not in accordance with the invention (Composition 2), which is a commercial skin repair balm, containing the following ingredients (INCI names): Paraffinum Liquidum, Cera Microcristallina, Ceresin, Lanolin Alcohol, (Eucerit), Panthenol, Glycerin, Bisabolol. [Tables 1] Ingredients % (w / w) Water 6-9 Caprylic / Capric Triglycerides 45-50 Hydrogenated Vegetable Oil 8- 12 Jojoba Oil 4-8 Shea Butter 2-5 Cupuaçu Butter 0.5-2 Glycerin 20-30 Sucrose Laurate 1-2 Sodium Hyaluronate 0.01 - 1 Oat Aerial Parts Extract 0.01 - 1 Sodium Citrate 0-0.5 Table 1 - Composition 1 in accordance with the invention The values of the occlusion factor F obtained for each of the products tested, and for the negative control, are indicated in Table 2. Product Value of F Vaseline Gifrer 64+1 Composition 1 39 + 2 Composition 2 (with paraffin) 57 + 7 Negative control (without product) 1

[0125] Table 2 - Occlusion factor F

[0126] Vaseline and Composition 2 (without vaseline but with paraffin) both have a very high occlusion factor F, revealing total occlusion, thus forming an almost impermeable layer. On the contrary, Composition 1 according to the invention has an intermediate occlusion factor F, i.e. a very interesting semi-occlusive character, because the composition according to the invention forms a protective but not completely impermeable film, which allows the skin to breathe and promotes its repair.

[0127] 2 / Example 2 - Sensory profiles of Composition 1 and petroleum jelly

[0128] The purpose of this test is to compare the sensory characteristics of Composition 1 according to the invention to those of Gifrer Vaseline.

[0129] The evaluation is carried out monadic by 15 volunteers trained in the sensory technique for topical compositions. Three evaluation phases are carried out: upon application of 0.5 mL of Composition 1 or Gifrer Vaseline to the skin of each forearm of the volunteer, and 5 minutes after then 15 minutes after application.

[0130] The tests are carried out in a room where the lighting, temperature and humidity are monitored.

[0131] Different criteria are evaluated and are detailed in Table 3 (for phase (1): upon application) and Table 4 (for phases (2) and (3): respectively 5 min and 15 min after application). Criteria Description Opacity - Thickness - Penetration speed Speed of the product to penetrate the skin Wet Sensation of humidity of the product between the fingers Oily Like cooking oil between the fingers Sticky Adherence of the finger or hand on the area of application of the product Squeaky Discontinuous adhesion of the product between the fingers

[0135] Table 4 - Descriptive criteria for phases (2) and (3): 5 min and 15 min after application

[0136] A computer questionnaire presents the unstructured analog measurement scales from 0 to 100, for each of the descriptive criteria of the different phases on the computer screen.

[0137] The results of the evaluation carried out are summarized in Table 5 below. Descriptive criterion Composition 1 Vaseline Opacity - application 63 44 Thickness - application 76 75 Penetration - application 34 31 Tension - 5 min 1 8 Tension - 15 min 2 4 Gloss - 5 min 59 60 Gloss - 15 min 50 46 Squeaky - application 4 15 Wet - application 18 20 Sticky - application 45 65 Sticky - 5 min 52 51 Sticky - 15 min 38 39 Oily - application 75 74 Oily - 5 min 72 75 Oily - 15 min 59 57 Film-forming - 5 min 54 55 Film-forming - 15 min 49 53 Resistant - 5 min 38 43 Resistant - 15 min 42 41 Powdery feel - 5 min 9 8 Powdery touch - 15 min 8 7 Suppleness - 5 min 68 63 Suppleness - 15 min 67 59 Plump skin - 5 min 36 43 Plump skin - 15 min 39 44 Fluffy - 5 min 0 0 Fluffy - 15 min 0 0

[0139] Table 5 - Sensory analysis results for petroleum jelly and Composition 1 according to the invention

[0140] It should be noted that the sensory analysis results of these two products are similar.

[0141] For the majority of the descriptive criteria, the composition according to the invention has the characteristics of petroleum jelly, such as thickness, an oily, film-forming feel, and a shiny appearance. The residual sensations after spreading are not significantly different.

[0142] However, compared to petroleum jelly, the composition according to the invention has significant and advantageous differences on some descriptor criteria. In particular, the composition according to the invention is more opaque and has a less sticky feel.

[0143] 3 / Example 3 - Sensory profiles of Composition 1 and Composition 2

[0144] The purpose of this test is to compare the sensory characteristics of Composition 1 according to the invention to those of the control composition, Composition 2 (without petroleum jelly but with paraffin).

[0145] The test is carried out in a similar manner to that described in Example 2. The same descriptor criteria are evaluated, and an additional criterion is added: spreading (ease of the product to spread without resistance on the skin).

[0146] The results obtained are summarized in Table 6 below. Descriptive criterion Composition 1 Composition 2 Opacity - application 72 55 Thickness - application 72 83 Spreading - application 61 66 Penetration - application 30 34 Tension - 5 min 8 7 Tension - 15 min 7 7 Gloss - 5 min 57 70 Gloss - 15 min 52 63 Crunchy - application 11 23 Wet - application 29 23 Sticky - application 43 61 Sticky - 5 min 39 56 Sticky - 15 min 34 42 Oily - application 74 74 Oily - 5 min 74 69 Oily - 15 min 59 53 Film-forming - 5 min 52 55 Film-forming - 15 min 37 50 Resistant - 5 min 32 43 Resistant - 15 min 30 35 Powdery touch - 5 min 12 10 Powdery touch - 15 min 20 12 Suppleness - 5 min 74 64 Suppleness - 15 min 71 65 Plump skin - 5 min 46 45 Plump skin - 15 min 43 43 Fluffy - 5 min 1 1 Fluffy - 15 min 0 0

[0148] Table 6 - Sensory analysis results for Composition 1 in accordance with the invention and the composition with paraffin (Composition 2)

[0149] It turns out that upon application, Composition 1 according to the invention presents numerous significant and advantageous differences with Composition 2: it is more opaque, less thick and less crunchy between the fingers, less shiny and it presents a truly less sticky feel.

[0150] The residual sensations after spreading are also significantly different: Composition 1 has a less sticky, less resistant feel, it is less film-forming, leaving the skin more supple.

[0151] The inventors thus show that the composition according to the invention has sensory characteristics closer to pure petroleum jelly than the comparative composition comprising paraffin (without petroleum jelly), in particular in terms of residual sensations after spreading.

[0152] The inventors have therefore succeeded in developing a composition without petroleum jelly and without paraffin, which nevertheless has a sensoriality very close to petroleum jelly.

[0153] 4 / Example 4 - Clinical evaluation of a moisturizing effect of Composition 1 according to the invention

[0154] The skin hydration capacity of Composition 1 in accordance with the invention is determined. To do this, a study measuring a skin hydration index is carried out on 31 subjects.

[0155] An application of 2 g / cm2 of Composition 1 is made on an area of skin of the forearm of approximately 3.5 x 4.3 cm of each subject. The composition is applied using a latex finger cot by massaging the area until completely absorbed.

[0156] Skin capacitance is measured by corneometry. The corneometer is a simple device which, when applied to the skin, measures its dielectric capacity using a capacitor. This is directly linked to its hydration state. These measurements are carried out at different times (before application of the composition: T0; immediately after application, i.e. approximately 15 min after: Timm; then at 1 h (+ 1 h), 2 h (+ 2 h), 4 h (+ 4 h), 6 h (+ 6 h) and 24 h (+ 24 h) after application) on the skin of the test subjects. Measurements are also carried out at the same time on these same subjects but on an area which has not received any application of product (control data), allowing statistical comparisons to be made.

[0157] The results presented in Table 7 below show the percentage increase in the hydration index compared to T0 (before application of the composition). A statistical analysis of covariance on the repeated measurements is carried out, the results are also presented in this Table 7. Time % increase in hydration index Analysis Inter-group Timm + 55.9 P<0.0001 + 1 h + 66.4 P<0.0001 + 2h + 70.7 P<0.0001 + 4h + 66.1 P<0.0001 + 6h + 68.9 P<0.0001 + 24 h + 31.4 P<0.0001

[0159] Table 7 - Increase in the skin hydration index after topical application of Composition 1 in accordance with the invention

[0160] It therefore appears that the composition according to the invention is highly moisturizing, from its complete absorption and for at least 24 hours, with a peak moisturizing effect of +70% at 2 hours after application.

[0161] It should be noted that no adverse effects were noted, thus revealing the excellent tolerance of the composition according to the invention.

[0162] 5 / Example 5 - Clinical evaluation of the skin repairing effect of Composition 1 according to the invention

[0163] This study is carried out on 25 adult subjects aged 18 to 65 years. All these subjects present skin lesions, such as cracks, fissures or crevices.

[0164] The study lasts 22 days. 5 visits are scheduled during this period. The skin lesions are examined by a dermatologist who evaluates the lesions in the form of a clinical score. The exact course of the study is as follows: - 1st day (Dl): inclusion of subjects in the study, - Composition 1 is applied from the 2nd day (D2), for a period of 21 days, twice a day (application of approximately 2 g / cm2 of the composition to the damaged skin area), - the other visits are carried out on the 3rd day (D3), 5th day (D5), 8th day (D8) and 22nd day (D22, end of study).

[0165] Patients also evaluate these lesions themselves and can thus judge the effectiveness of the tested composition.

[0166] The results obtained, concerning the effectiveness of the composition as a skin repairer, are summarized in Table 8 below, showing the percentage of patients satisfied or not, during the study, on this criterion of effectiveness.

[0167] [Tables8] D3 D5 D8 D22 No efficacy (%) 0 0 0 0 Moderate efficacy (%) 16 12 4 8 Good efficacy (%) 56 50 48 24 Very good efficacy (%) 28 38 48 68

[0168] Table 8 - Patient evaluation of the efficacy of Composition 1 according to the invention on the repair of skin lesions, expressed as a % of patients for each degree of efficacy

[0169] This table clearly shows that patients with skin cracks or fissures find the composition according to the invention effective in skin repair. They also find it increasingly effective during treatment. After only 4 days of treatment, more than one in three patients consider the composition according to the invention to be very effective.

[0170] The clinical score of physical cutaneous signs (redness, edema, desquamation, etc.) is assessed by the dermatologist in patients presenting cracks or fissures during visits. The improvement of these criteria is summarized in Table 9 below.

[0171] [Tables9] Sum of physical signs D3 D5 D8 D22 % improvement compared to D1 48 66 67 84

[0172] Table 9 - Evaluation by the dermatologist of the effectiveness of Composition 1 according to the invention on the repair of skin lesions

[0173] These results are very revealing about the improvement of physical signs of patients presenting skin cracks or fissures.

[0174] After only 4 days of treatment with the composition according to the invention, a 66% improvement in physical signs is thus demonstrated.

[0175] All of these results demonstrate that the inventors have developed a new composition without petroleum jelly or paraffin, which is very moisturizing and has a beneficial effect on skin cracks and / or fissures, while maintaining a sensoriality close to petroleum jelly. The composition according to the invention therefore has all the advantages of petroleum jelly and without its disadvantages.
