Description

Imidazolopyridopyrimidone compound and its preparation method and application Technical Field

[0001] The present invention relates to the technical field of medicinal chemistry, and in particular to an imidazopyridopyrimidone compound, a preparation method and an application thereof. Background Art

[0002] Mitochondria play an important role in the occurrence, development, and metastasis of various tumors. Human caseinolytic enzyme P (HsClpP) is an unfolding peptidase in the mitochondrial matrix that controls protein quality, regulates mitochondrial metabolism, and maintains the integrity and enzyme activity of the mitochondrial respiratory chain. Studies have shown that abnormalities in HsClpP can lead to mitochondrial dysfunction and a variety of human diseases, including cancer, neurological diseases, diabetes, gastrointestinal diseases, cardiovascular diseases, skin diseases, and aging. ClpP is overexpressed in multiple myeloma, various lymphomas, chronic myeloid leukemia (CML), and some solid tumors. Further activation of ClpP is expected to provide new interventions for the treatment of these tumors.

[0003] The potential mechanisms by which HsClpP may participate in cancer are related to mitochondrial bioenergetics and proteostasis, reactive oxygen species (ROS) production, and autophagy activation. HsClpP is associated with members of the electron transport chain (ETC), particularly SDHA and other enzymes involved in metabolism. Inhibition of HsClpP activity results in the inability of the HsClpXP complex to effectively degrade damaged or malfunctioning proteins involved in oxidative phosphorylation, thereby affecting mitochondrial oxidative phosphorylation (OXPHOS) and inducing cell death. Furthermore, HsClpP inhibition reduces the enzymatic activity of SDHA in respiratory chain (RC) complex II, increasing mitochondrial ROS production and initiating non-apoptotic tumor cell death. Furthermore, overactivation of HsClpP can inhibit cancer cell growth. Overactivation of HsClpP can degrade ClpP substrates, including RC complex proteins (SDHB and SDHB), impair OXPHOS, disrupt mitochondrial structure and function, and selectively induce cancer cell death. Therefore, HsClpP is a unique tumor target, and both inhibition and activation of HsClpP can inhibit tumor growth.

[0004] HsClpP is also highly expressed in acute myeloid leukemia (AML). In primary AML, approximately 45% of patients have elevated HsClpP expression compared to normal hematopoietic progenitor cells. Blocking HsClpP expression in vitro and in vivo significantly reduced the growth and viability of various AML cell lines and significantly inhibited tumor development in vivo. Furthermore, overactivation of HsClpP has been proposed as an effective therapeutic strategy for AML. When HsClpP is activated, the rates of cell death and apoptosis in primary AML cells increase compared to normal cells, and HsClpP activators also reduce leukemia burden in mice. HsClpP expression is also elevated in many other hematological malignancies, such as CML, multiple myeloma, and various lymphomas. Therefore, modulating HsClpP activity is considered a therapeutic strategy for AML and other hematological malignancies.

[0005] HsClpP is also overregulated in many solid tumors, including gliomas, breast cancer, ovarian cancer, prostate cancer, liver cancer, lung cancer, thyroid cancer, and colon cancer. Overexpression of HsClpP is associated with shorter survival in patients with breast cancer and uveal melanoma. Inhibition of HsClpP can deregulate mitochondrial RC and oxidative stress, inhibiting tumor cell proliferation, motility, and metastasis. In prostate cancer, inhibition of HsClpP activity reduces the expression of cyclins A, B1, and D1, thereby inducing cell cycle arrest in PC3 prostate cancer cells and inhibiting PC3 cell proliferation. In a PC3 cell wound wound assay, interference with HsClpP expression inhibited tumor cell invasion and migration. In tumor-bearing mice, inhibition of HsClpP slowed tumor progression and the extent of liver metastasis.

[0006] Therefore, developing anti-tumor drugs with new mechanisms targeting HsClpP will help improve the therapeutic effect of anti-tumor drugs and effectively prolong the survival of patients.

[0118] S1. Compound 1-1 (5.0 g, 33.0 mmol), compound 1-2 (3.2 mL, 36 mmol), DBU (0.5 mL, 3.3 mmol) and triethylamine (4.5 mL, 33.0 mmol) were dissolved in dichloromethane (60 mL) and stirred at 40°C for 12 hours. Triethylamine (9.0 mL, 66.0 mmol) and Boc2O (9.0 mL, 39.0 mmol) were then added and the reaction continued at 40°C for 24 hours. After the reaction was completed, the mixture was cooled to room temperature and the solvent was removed by distillation under reduced pressure. Methyl tert-butyl ether was added and stirred for 30 minutes, then filtered. The filter cake was washed twice with methyl tert-butyl ether, the organic phases were combined and dried over anhydrous sodium sulfate, and concentrated to give a colorless oil 34-1 (7.3 g, 75% yield), which was used directly in the next step.

[0119] S2. Dissolve compound 13-1 (10.6 g, 34.9 mmol) in tetrahydrofuran (175 mL). Add 1.0 M potassium tert-butoxide / tetrahydrofuran solution (42 mL, 42.0 mmol) under ice-cooling. React at 0°C for 4 hours. After completion of the reaction, dilute with water and extract with ethyl acetate. The organic layer is washed with saturated brine, dried over anhydrous sodium sulfate, concentrated, and purified by column chromatography (petroleum ether:ethyl acetate = 4:1) to obtain a colorless oil 13-2 (mixture, 9.6 g, total yield 85%).

[0120] S3. Compound 13-2 (2.7 g, 10.0 mmol) and compound 13-3 (4.5 g, 12.0 mmol) were dissolved in ultra-dry methanol (20 mL). Sodium methoxide (2.7 g, 50.0 mmol) was added and stirred at 100°C for 16 hours. After the reaction was completed, the mixture was diluted with water and extracted with ethyl acetate. The organic layer was dried over anhydrous sodium sulfate, concentrated, and purified by column chromatography (4% MeOH in DCM) to obtain 13-4 as a yellow solid.

[0121] S4. Compound 13-4 (2.3 g, 5.0 mmol) was dissolved in dichloromethane (10 mL), trifluoroacetic acid (5 mL) was added, and the mixture was stirred at room temperature. After completion of the reaction, the solvent was removed by distillation under reduced pressure, and the reaction was quenched by addition of saturated aqueous NaHCO₃. The mixture was then extracted with dichloromethane. The organic layer was dried over anhydrous sodium sulfate, concentrated, and purified by column chromatography (8% MeOH in DCM) to afford 13-5 as an off-white solid.

[0122] S5. Compound 13-5 (73 mg, 0.2 mmol) was dissolved in acetonitrile (2 mL), and 13-6 (46 mg, 0.24 mmol) and cesium carbonate (130 mg, 0.4 mmol) were added. The mixture was stirred at 80°C overnight. After the reaction, the solvent was removed by distillation under reduced pressure. The reaction mixture was diluted with water and extracted with dichloromethane. The organic layer was dried over anhydrous sodium sulfate, concentrated, and purified by column chromatography (4% MeOH in DCM) to obtain 13 as a yellow solid.
