Abstract
Description
PROCESS FOR OBTAINING STEM CELL ENRICHED SUTURE THREAD, SUTURE KIT AND ITS USES Certificate of addition to the invention BR 10 2013 012895 3, filed on 05 / 24 / 2013.
[001] The first descriptive report for deposit refers to a process for obtaining suture threads enriched with stem cells. The threads obtained and their uses are an additional object.
[002] This Addition Certificate refers to an improvement to the process of obtaining enriched suture thread and a suture kit comprising suture thread, thrombin, calcium chloride, fibrinogen and culture medium. The use of the enriched suture thread and the suture kit is an additional object. Detailed description
[003] This Addition Certificate refers to an improvement to the process of obtaining stem cell-enriched suture thread and a suture kit comprising suture thread, thrombin, calcium chloride, fibrinogen and culture medium. An additional object is the use of the enriched thread obtained and the suture kit for sutures of any kind.
[004] The process of obtaining enriched suture thread comprises the following steps: a) Dripping of thrombin and calcium chloride solution along the entire length of the suture thread; b) Stem cell drip; c) Fibrinogen drip; d) Addition of sufficient culture medium to coat the suture thread; (e) Addition of 10% serum relative to the amount of culture medium added, and 2 / 3 f) Maintain for a period varying between 2 and 24 hours in a CO2 incubator for cell culture.
[005] The ratio between thrombin, calcium chloride solution and fibrinogen varies between 4-6:0.5-1.5:1-3, preferably 3:1:2.
[006] The concentration of the calcium chloride solution is preferably 40 µmol / mL.
[007] The culture medium can be selected from group 199 medium, MEM medium, RPMI medium, Dulbecco MEM medium, HAM F-12 nutrient, Leibowitz L-15 medium, HAM F-10 nutrient, SOL. BAI. De Hanks, MEM Alfa medium, Glasgow medium, DMEM / HAM F-12 medium, RPMI 1640 medium with Hepes, DMEM medium, Schneider's medium, TC-100 medium, CTS, MesenCultSF Culture Kite MesenCultXF Medium.
[008] Step (e) is omitted if the selected culture medium is CTS, MesenCult SF Culture Kit and MesenCult XF Medium.
[009] Serum may be selected from the group of inactivated fetal bovine serum (10-bio1 OOi) (FBS), fetal bovine serum (10-bio100) (FBS), sterile equine serum, calf serum (NCS) and human serum.
[0010] The suture kit comprises the following components: - suture thread; - thrombin and calcium chloride; - fibrinogen; - means of culture, and - serum.
[0011] The ratio between thrombin, calcium chloride solution and fibrinogen varies between 4-6:0.5-1.5:1-3, preferably 3:1:2.
[0012] The concentration of the calcium chloride solution is preferably 40 µmol / mL.
[0013] The culture medium can be selected from group 199 medium, MEM medium, RPMI medium, Dulbecco MEM medium, HAM F-12 nutrient, Leibowitz L-15 medium, HAM F-10 nutrient, Hanks BAI solution, MEM Alpha medium, Glasgow medium, DMEM / HAM F-12 medium, RPMI 1640 medium with Hepes, DMEM medium, 3 / 3 Schneider's medium, TC-100 medium, CTS, MesenCultSF Culture Kite MesenCultXF Medium.
[0014] Serum may be selected from the group of inactivated fetal bovine serum (10-bio1 OOi) (FBS), fetal bovine serum (10-bio100) (FBS), sterile equine serum, calf serum (NCS) and human serum.
[0015] It is an optional component of the stem cell kit.
