Description

Technical Field

[0001] The present invention relates to a crystalline form of 1-(1-oxo-1,2-dihydroisoquinolin-5-yl)-5-(trifluoromethyl)-N-[2-(trifluoromethyl)pyridin-4-yl]-1H-pyrazole-4-carboxamide (Compound A) monohydrate. The crystalline form can be used to treat diseases, syndromes, conditions or disorders, in particular MALT1-related diseases, syndromes, conditions or disorders, including but not limited to cancer and immune diseases. The present invention also relates to a pharmaceutical composition comprising such a crystalline form, a method for preparing such a crystalline form, and the use of such a crystalline form or a pharmaceutical composition for treating cancer and autoimmune diseases, syndromes, disorders or conditions associated with MALT1 inhibitors. Background Art

[0002] MALT1 (Mucosa-associated lymphoid tissue lymphoma translocation 1) is a key mediator of the classical NFKB signaling pathway. MALT1 is the only human paracaspase and transduces signals from the B cell receptor (BCR) and the T cell receptor (TCR). MALT1 is the active subunit of the CBM complex formed after receptor activation. The CBM complex consists of multiple subunits of three proteins: CARD11 (caspase recruitment domain family member 11), BCL10 (B cell CLL / lymphoma 10) and MALT1. MALT1 affects NFKB signaling through two mechanisms: first, MALT1 acts as a scaffold protein and recruits NFKB signaling proteins, such as TRAF6, TAB-TAK1 or NEMO-ΙKKα / β; second, MALT1, as a cysteine protease, cuts and thereby inactivates negative regulators of NFKB signaling, such as RelB, A20 or CYLD. The ultimate endpoint of MALT1 activity is nuclear translocation of the FKB transcription factor complex and activation of FKB signaling (Jaworski et al., Cell Mol Life Science 2016. 73, 459-473).

[0003] The constitutive activation of FKB signaling is a hallmark of ABC-DLBCL (diffuse large B-cell lymphoma of activated B-cell-like subtype, which is a more aggressive form of DLBCL). DLBCL is the most common form of non-Hodgkin's lymphoma (NHL), accounting for approximately 25% of lymphoma cases, while ABC-DLBCL accounts for approximately 40% of DLBCL. In ABC-DLBCL patients, NFKB pathway activation is driven by mutations in signaling components (e.g., CD79A / B, CARD11, MYD88 or A20) (Staudt, Cold Spring Harb Perspect Biol [Cold Spring Harb Biology Outlook] 2010, 2; Lim et al., Immunol Rev [Immunology Review] 2012, 246, 359-378).

[0004] The use of BTK inhibitors (e.g., ibrutinib) provides clinical proof of concept that inhibition of NFKB signaling is effective in ABC-DLBCL. MALT1 is downstream of BTK in the NFKB signaling pathway, and MALT1 inhibitors can target ABC-DLBCL patients who do not respond to ibrutinib (primarily those with CARD11 mutations), as well as treat patients who have acquired resistance to ibrutinib.

[0005] Small molecule tool compound inhibitors of the MALT1 protease have shown efficacy in preclinical models of ABC-DLBCL (Fontan et al., Cancer Cell 2012, 22, 812-824; Nagel et al., Cancer Cell 2012, 22, 825-837). Interestingly, covalent catalytic sites and allosteric inhibitors of MALT1 protease function have been described, suggesting that inhibitors of this protease may be useful as pharmaceutical agents (Demeyer et al., Trends Mol Med 2016, 22, 135-150).

[0006] Chromosomal translocations that produce API2-MALT1 fusion oncoproteins are the most common mutations identified in MALT (mucosa-associated lymphoid tissue) lymphomas. API2-MALT1 is an effective activator of the NFKB pathway (Rosebeck et al., World J Biol Chem [World Journal of Biochemistry] 2016, 7, 128-137). API2-MALT1 mimics ligand-bound TNF receptors, promoting TRAF2-dependent ubiquitination of RIP1 (which serves as a scaffold for activating classical NFKB signaling). In addition, it has been shown that API2-MALT1 cuts and generates stable constitutively active fragments of NFKB-induced kinases (NIK), thereby activating non-classical FKB pathways (Rosebeck et al., Science [Science], 2011, 331, 468-472).

[0007] In addition to lymphoma, MALT1 has been shown to play a key role in innate and adaptive immunity (Jaworski M et al., Cell Mol Life Sci. 2016). MALT1 protease inhibitors can reduce disease onset and progression in experimental allergic encephalomyelitis in mice, a mouse model of multiple sclerosis (Mc Guire et al., J. Neuroinflammation 2014, 11, 124). Mice expressing catalytically inactive MALT1 mutants show loss of marginal zone B cells and B1 B cells, as well as general immune defects characterized by reduced T and B cell activation and proliferation. However, those mice also develop spontaneous multi-organ autoimmune inflammation at 9 to 10 weeks of age. It is still poorly understood why the knock-in mice with disabled MALT1 protease show a breakdown of tolerance, while the conventional MALT1 KO mice do not. One hypothesis suggests that the imbalance in immune homeostasis in MALT1 protease knock-in mice may be caused by an incomplete lack of T cells and B cells but a severe lack of immunoregulatory cells (Jaworski et al., EMBO J. [Journal of the European Molecular Biology Association] 2014; Gewies et al., Cell Reports [Cell Reports] 2014; Bornancin et al., J. Immunology [Journal of Immunology] 2015; Yu et al., PLOS One [Public Library of Science Journal] 2015). Similarly, human MALT deficiency is associated with combined immunodeficiency disorder (McKinnon et al., J. Allergy Clin. Immunol. 2014, 133, 1458-1462; Jabara et al., J. Allergy Clin. Immunol. 2013, 132, 151-158; Punwani et al., J. Clin. Immunol. 2015, 35, 135-146). Given the differences between genetic mutations and pharmacological inhibition, the phenotype of knock-in mice with ineffective MALT1 protease may not be similar to the phenotype of patients treated with MALT1 protease inhibitors. Reducing immunosuppressive T cells by MALT1 protease inhibition may benefit cancer patients by potentially improving anti-tumor immunity.

[0008] Therefore, MALT1 inhibitors may provide therapeutic benefit to patients suffering from cancer and / or immune diseases.

[0009] WO 2018 / 119036 (which is incorporated herein by reference) discloses compounds that inhibit MALT1, including 1-(1-oxo-1,2-dihydroisoquinolin-5-yl)-5-(trifluoromethyl)-N-[2-(trifluoromethyl)pyridin-4-yl]-1H-pyrazole-4-carboxamide (referenced in WO 2018 / 119036 as Compound 158).

[0010] There is a need to provide alternative forms of 1-(1-oxo-1,2-dihydroisoquinolin-5-yl)-5-(trifluoromethyl)-N-[2-(trifluoromethyl)pyridin-4-yl]-1H-pyrazole-4-carboxamide. Summary of the invention

[0069] Crystalline Form I can be characterized by an X-ray powder diffraction pattern. The X-ray powder diffraction pattern can be obtained using copper K-alpha X-rays having a wavelength of 1.5406.

[0070] The X-ray powder diffraction pattern comprises peaks at 8.4, 12.7, 13.3 and 16.7 ° 2θ ± 0.2 ° 2θ. The X-ray powder diffraction pattern may further comprise at least one peak selected from 6.7, 10.0, 10.7, 12.0, 12.3, 13.5, 14.1, 14.6, 15.4, 15.6, 16.0, 18.1, 18.4, 19.2, 20.0, 20.3, 21.1, 22.0 and 24.9 ° 2θ ± 0.2 ° 2θ.

[0077] Crystalline Form III of Compound A Monohydrate

[0078] Crystalline form III exhibits non-hygroscopic behavior (water absorption is less than 0.2% at 80% relative humidity). Unlike hydrate crystalline form I, crystalline form III avoids the loss of water at low relative humidity (e.g., relative humidity is 20% or less). These are desirable properties in active pharmaceutical ingredients (particularly those intended for solid dosage forms). When variations in water content are avoided, accurate amounts of compound A can be dispensed, and weighing errors can be avoided (e.g., when preparing formulations or dosage forms).

[0079] Crystalline Form III can be characterized by an X-ray powder diffraction pattern. The X-ray powder diffraction pattern can be obtained using copper K-alpha X-rays having a wavelength of 1.5406.

[0090] The crystalline form III of compound A monohydrate can be prepared by the following method, which comprises the following steps: a) adding compound A or its hydrate or solvate to a mixture of ethyl acetate and ethanol and heating to a temperature ranging from about 30°C to the reflux temperature of the solvent; b) adding water to the mixture and filtering out any precipitate, maintaining the temperature; c) adding n-heptane to the mixture, seeding with crystalline form III, and maintaining the temperature; and d) cooling to room temperature to produce a precipitate of the crystalline form; wherein the amount of water is from about 0.1w / w% to about 3w / w% relative to the total weight of the solvent; specifically, wherein the amount of water is from about 1.0w / w% to about 3w / w% relative to the total weight of the solvent.

[0091] In one embodiment, the amount of water is at least 0.3w / w%, specifically 0.4w / w%, more specifically at least 0.8w / w%, even more specifically at least 1.0w / w%, relative to the total weight of the solvent. In one embodiment, the amount of water is at least 0.3w / w%, specifically 0.4w / w%, specifically at least 0.8w / w%, even more specifically at least 1.0w / w%, relative to the total mass of the reaction mixture. In one embodiment, the reaction mixture should contain an amount of water exceeding 0.038 times the amount of compound A.

[0092] In one embodiment, the amount of water is at most 3.0 w / w%, 2.5 w / w%, 2.0 w / w%, 1.5 w / w% or 1.0 w / w% relative to the total weight of the solvent.

[0093] In one embodiment, the amount of water is 1.0 w / w% relative to the total weight of the solvent. In one embodiment, the amount of water is from about 0.3 w / w% to about 1.5 w / w% relative to the total weight of the solvent. In one embodiment, the amount of water is from 0.3 w / w% to 1.5 w / w% relative to the total weight of the solvent.

[0094] Alternatively, the crystalline form III of compound A monohydrate can be prepared by the following method, which comprises the following steps: a) adding compound A or a hydrate or solvate thereof to a mixture of water and isopropanol and heating to a temperature ranging from about 30°C to the reflux temperature of the solvent; b) adding n-heptane to the mixture, seeding with crystalline form III, and maintaining the temperature; and c) cooling to room temperature to produce a precipitate of the crystalline form; wherein the amount of water is from about 1.0 w / w% to about 6.0 w / w% relative to the total weight of the solvent.

[0095] In one embodiment, the amount of water is at least 2.0 w / w%, specifically 2.5 w / w%, more specifically at least 2.8 w / w%, even more specifically at least 3.0 w / w%, relative to the total weight of the solvent. Compound A or a hydrate or solvate thereof may comprise hydrate crystalline form I.

[0104] The temperature to which the mixture is heated can be from about 40° C. to about the solvent reflux temperature or from about 50° C. to about the solvent reflux temperature. The temperature to which the mixture is heated can be about 60° C. The suspension can be stirred for at least 1 day, at least 2 days, at least 3 days, at least 4 days, or at least 5 days.

[0105] The method may further comprise the step c) of filtering out the precipitate. The method may further comprise the step d) of drying the precipitate under vacuum, optionally at a temperature of from about 45°C to about 55°C.

[0106] Alternatively, the crystalline form III of the monohydrate of compound A can be prepared by a process comprising the steps of: a) adding compound A or a hydrate or solvate thereof to a mixture of water and isopropanol; and b) stirring the resulting suspension at a temperature ranging from about 30° C. to the reflux temperature of the solvent to produce a precipitate of the crystalline monohydrate of crystalline form III of compound A. [0117] For buccal or sublingual administration, the pharmaceutical compositions of this invention may be administered in the form of tablets or lozenges which can be formulated in conventional manner.

[0118] As another example, according to conventional pharmaceutical compounding techniques, a pharmaceutical composition containing the crystalline form as an active pharmaceutical ingredient can be prepared by mixing the crystalline form with a pharmaceutically acceptable carrier, a pharmaceutically acceptable diluent, and / or a pharmaceutically acceptable excipient. Carriers, excipients, and diluents can take a variety of forms, depending on the desired route of administration (e.g., oral, parenteral, etc.). Therefore, for liquid oral preparations (e.g., suspensions, syrups, elixirs, and solutions), suitable carriers, excipients, and diluents include water, glycerol, oils, alcohols, flavoring agents, preservatives, stabilizers, colorants, etc.; for solid oral preparations (e.g., powders, capsules, and tablets), suitable carriers, excipients, and diluents include starch, sugar, diluents, granulating agents, lubricants, adhesives, disintegrants, etc. Solid oral preparations can also optionally be coated with substances such as sugars, or enteric coatings to regulate the main sites of absorption and disintegration. For parenteral administration, the carrier, excipient and diluent will usually include sterile water, and other ingredients may be added to increase the solubility and preservation of the composition. Injectable suspensions or solutions may also be prepared using an aqueous carrier and appropriate additives (such as solubilizers and preservatives).

[0181]

[0182]

[0183] DSC curve of crystalline form I ( Figure 8 ) shows a broad endothermic signal at 98.7°C (142 J / g) with an onset temperature of 66.3°C, which is due to the dehydration of the hydrate. A second endothermic signal is observed at 157.0°C (17 J / g) with an onset temperature of 145.2°C, corresponding to the melting of the dehydrated form.

[0184] TGA curve of crystalline form I ( Fig. 9) shows a weight loss of 4.7% from RT up to 82°C due to evaporation of the solvent (water) present. A weight loss of 0.4% was recorded between 82°C and 122°C and this weight loss corresponds to further evaporation of the solvent.

[0185] DVS mass change diagram of crystalline form I ( Fig.11 ) showed a weight loss of approximately 2.7% during the initial drying step. The compound rapidly adsorbs and desorbs water in the humidity range of 0-20% RH (relative humidity) ( Fig.10 and Fig.11 ). Depending on the atmospheric humidity, it can absorb up to 4.1%-4.2% of water (RH between 0%-20%) and completely dry during the desorption cycle. In the humidity range of 20%-95% RH, the compound slowly and reversibly adsorbs and desorbs with 1.8% of water adsorbed and desorbed. The XRPD pattern and IR spectrum of the fractions obtained after DVS testing were comparable to the starting material. No indication of a change in the solid state form was observed.

[0186] The data indicate that the water of hydration in crystalline Form I is loosely bound and is lost at RH of 20% and below.
