Abstract
Description
Technical Field
[0001] The present invention discloses an antibacterial composition, in particular, a transparent moisturizing antibacterial composition. The transparent moisturizing antibacterial composition comprises water, alcohol, a moisturizer and a sensory oil. Background Art
[0002] Consumers are increasingly concerned about microbial and viral contamination. Therefore, hygiene has been one of the most important attributes required by consumers. Consumers expect products that will provide the desired hygiene properties. Consumers around the world use different types of antimicrobial compositions to disinfect various surfaces, including hard surfaces, such as countertops, floors, and furniture; soft and porous surfaces, such as clothing, carpets, and upholstery; and personal surfaces, such as skin and hair. Many microorganisms, such as bacteria and viruses, are found on such surfaces. It is desirable to keep these surfaces free of bacteria to minimize the risk of illness.
[0003] Many antibacterial products are commercially available. Some are provided as gels, liquids or liquid sprays containing at least 60% by weight or more ethanol. Although such products are known to produce good antibacterial benefits after application, some consumers avoid using high ethanol-based products because such products may dry out the skin, weaken the epidermal barrier and accelerate the aging process. In the current environment, consumers may use antibacterial products on their bodies throughout the day, thus increasing the likelihood of dry skin.
[0004] Therefore, there is a continuing need to develop an antibacterial composition that provides excellent antimicrobial and antiviral benefits while being safe and mild enough to be used throughout the day at any time of day. In view of consumer demand, there is increasing interest in developing an antibacterial composition that also delivers soothing and moisturizing benefits when applied topically. Summary of the invention
[0005] In various aspects, moisturizing antimicrobial compositions are disclosed.
[0006] The transparent antimicrobial composition comprises: water; alcohol; humectant; and sensory oil.
[0007] A method for preparing a transparent antimicrobial composition comprises: combining a sensory oil with a moisturizer to form a first phase; combining water and an alcohol to form a second phase; combining the first phase and the second phase to form a third phase; and adding a neutralizer to the third phase to form the antimicrobial composition.
[0008] These and other features and characteristics are described in more detail below. DETAILED DESCRIPTION
[0009] Disclosed herein is an antibacterial composition. The antibacterial composition is a transparent composition having non-sticky sensory properties. The antibacterial composition can be in any form, but is preferably in the form of a transparent liquid spray. The antibacterial composition provides moisturizing to the surface to which it is applied. For example, when the composition is applied to human skin, the antibacterial composition does not dry the user's skin, but rather moisturizes and disinfects the user's skin. The antibacterial composition can provide less than or equal to 8 hours of moisturizing to the surface to which it is applied. The antibacterial composition can also provide antimicrobial and antiviral protection against bacteria, viruses, and fungi. The antibacterial composition includes water, alcohol, a moisturizer, and a sensory oil. It was unexpectedly found that the antibacterial composition provides moisturizing without stickiness after application. This is an unexpected benefit because antibacterial compositions generally require the use of an anti-adhesive agent to reduce the stickiness typically associated with the application of the antibacterial composition.
[0010] The antimicrobial composition may be substantially free of anti-adhesives. Substantially free of anti-adhesives means that the composition contains less than 2% by weight of anti-adhesives, preferably less than 1% by weight of anti-adhesives, more preferably less than 0.5% by weight of anti-adhesives, even more preferably less than 0.1% by weight of anti-adhesives, and even more preferably 0% by weight of anti-adhesives. The weight percentages (wt%) mentioned throughout this article refer to the wt% in the total composition.
[0011] The antimicrobial composition advantageously provides moisturizing and disinfecting benefits to the surface to which it is applied. As used herein, a disinfecting or antimicrobial composition refers to a composition that can kill bacteria or inhibit bacterial growth. The antimicrobial composition may be substantially free of emulsifiers. The antimicrobial composition may be transparent or translucent, preferably the antimicrobial composition is transparent. Substantially free of emulsifiers means that the composition contains less than 2% by weight of emulsifiers, preferably less than 1% by weight of emulsifiers, more preferably less than 0.5% by weight of emulsifiers, even more preferably less than 0.1% by weight of emulsifiers, and even more preferably 0% by weight of emulsifiers.
[0012] The antimicrobial composition comprises water in an amount of 8 to 30 wt%, preferably 10 to 25 wt%, more preferably 10 to 20 wt%.
[0013] Alcohol can be used as the antimicrobial agent in the composition. When using alcohol as the antimicrobial agent, the antimicrobial composition comprises 60 to 80 wt %, preferably 62 to 75 wt %, more preferably 65 to 75 wt % of the alcohol, including all values and ranges contained therein. The alcohol can include a variety of alcohols, including but not limited to ethanol, propanol (n-propanol), isopropanol or a combination thereof. The level of alcohol present in the antimicrobial composition can be adjusted based on the purity percentage of the alcohol. For example, if 92% ethanol is used, the ethanol can be used in an amount of 70 to 80 wt %, such as 75 wt %. Optionally, the alcohol can include chloroxylenol. A preferred alcohol can be denatured SD-40 ethanol.

[0049] The antimicrobial composition may additionally contain a neutralizing agent, such as sodium hydroxide, potassium hydroxide, triethanolamine, ammonium, arginine, tromethamine, aminomethyl propanol sold by Angus as AMP 95, AMP-Ultra PC1000, AMP-Ultra PC2000, AMP-Ultra PC3000, tetrahydroxypropylethylenediamine also known as Neutrol TE from BASF, diisopropanolamine, triisopropanolamine, or a combination thereof. The neutralizing agent may be present in an amount of 0 to 1 wt %, such as 0.1 to 0.75 wt %, such as 0.2 to 0.5 wt %, including all values and ranges subsumed therein.
[0050] Regarding packaging, the antibacterial composition can be packaged in a spray bottle, a squeeze bottle, or provided as an impregnated wetting agent on a cotton swab, a wipe, a towel, a cosmetic substrate sheet (such as those described in US 6,294,182B1), etc. Because the viscosity is increased by a thickener, the antibacterial composition gels and can be provided to consumers as a gel composition in a squeeze bottle. The spray bottle can also be metal, and the antibacterial composition can be provided by conventional aerosol packaging technology, and includes those technologies that utilize air-in-bag discharge tanks, mechanisms, and actuators. It is also within the scope of the antibacterial composition to include a foaming agent (e.g., zwitterionic and / or amphoteric surfactants) so that the antibacterial composition can be discharged as a foam.
[0051] The antimicrobial composition should be provided with instructions for applying (e.g., squeezing or spraying) the composition onto a surface (e.g., skin) to kill bacteria and reduce viral activity. The antimicrobial composition may be provided in a biodegradable package, and the package used is preferably refillable or reusable, biodegradable, and / or at least 50%, preferably at least 100%, made of post-consumer recycled resin. The antimicrobial composition may be in the form of a liquid or a gel, preferably the antimicrobial composition is in the form of a liquid, such as a liquid spray antimicrobial composition.
[0052] As used herein, skin means skin including arms (including underarms), face, feet, neck, chest, hands, legs, buttocks and scalp (including hair). Disinfection as used herein refers to at least 2 bacterial logarithmic kills and at least 2 viral logarithmic inactivations (both achieved) within less than 3 minutes after topical application to the surface, preferably at least 3 bacterial logarithmic kills and at least 3 viral logarithmic inactivations. As used herein, combination means the total weight of, for example, cetrimonium chloride plus benzalkonium chloride. Skin benefit agents refer to ingredients suitable for improving skin properties. Surfaces as used herein include skin or inanimate surfaces, such as desktops, computer monitors, door handles, toilet seats, shopping cart handles or even clothing. Surfaces also mean fur including animals, such as the fur of dogs and cats. As used herein, surfaces preferably refer to human skin, and especially the skin on the face and hands.
[0053] The antimicrobial composition can be a home care composition, such as a laundry spray composition suitable for spraying clothes and interior decorations that need to be disinfected. The home care composition can also be an antimicrobial kitchen or bathroom spray composition. Preferably, the antimicrobial composition is a topical composition applied to the skin for disinfection by significantly reducing the amount of bacteria and viruses on the skin. The composition can optionally include skin-benefitting ingredients added thereto, such as emollients, vitamins and / or derivatives thereof, resorcinols, retinoic acid precursors, colorants, moisturizers or wetting agents, fragrances, sunscreens, combinations thereof, etc., as previously described herein. The skin-benefitting ingredients can be water-soluble or oil-soluble.
[0054] Thus, the antimicrobial composition is a hydroalcoholic composition having a pH of 3.0 to 8.2, and the composition is water / alcohol continuous. As used herein, viscosity is obtained as follows: using a Brookfield viscometer (using spindle 4 at 10 rpm) or using a Discovery HR-2 rheometer (using a sandblasted plate with a gap of 1000 microns and for a first viscosity V A 0.4s -1 The first shear rate S A and for the second viscosity V B 10s -1 The second shear rate S B , both at 25°C and at 20 s intervals).
[0055] It is known to use several units in the context of viscosity, but the more frequently used units are centipoise (cP), Pascal seconds (Pa*s) and milliPascal seconds (mPa*s), and these units are easily convertible to each other with the help of publicly available resources such as textbooks, encyclopedias and the Internet. In yet another embodiment, the composition is a non-therapeutic and non-pharmaceutical composition, which is a hydroalcoholic solution with a viscosity below 10,000 centipoise (cP) (10 Pa*s), preferably a viscosity below 5,000 cP (5 Pa*s), preferably a viscosity of 0.8 to 3,000 cP (0.0008 to 3 Pa*s), more preferably a viscosity of 0.8 to 2,000 cP (0.0008 to 2 Pa*s).
[0056] Typically, the viscosity of the antimicrobial composition will be less than 10,000 cP (10 Pa*s). Typically, the viscosity of the antimicrobial composition will be 0.8 to 5,000 cP (0.0008 to 5 Pa*s), preferably 0.8 to 3,000 cP (0.0008 to 3 Pa*s), more preferably 0.8 to 2,000 cP (0.0008 to 2 Pa*s), and even more preferably 0.8 to 1,000 cP (0.0008 to 1 Pa*s), including all ranges subsumed therein.
[0057] The antimicrobial composition can be prepared by any method for preparing an antimicrobial composition. In one embodiment, the method for preparing the antimicrobial composition may include the following: combining a sensory oil and a moisturizer to form a first phase. Then combining water and an alcohol to form a second phase. Then combining the first phase and the second phase to form a third phase. Adding a neutralizing agent to the third phase to form the antimicrobial composition.
[0058] Example
[0059] The following examples are merely illustrative of the antimicrobial composition disclosed herein and are not intended to limit the scope thereof.
[0060] Embodiment 1:
[0061] Samples 1 and 2 are examples demonstrating the composition of the antimicrobial composition disclosed herein.
[0062] Table 1 - All figures are shown as weight %
[0063]
[0064]
[0065] The level of alcohol present in the antimicrobial composition can be adjusted based on the percentage purity of the alcohol. For example, the ethanol purity should preferably be 92% w / w to 94% w / w, so that if 92% pure ethanol is used, the ethanol can be used in an amount of 70 to 80% by weight, such as 72% by weight.
[0066] Table 2 shows the oils that can be used in antibacterial compositions to design a transparent and fast-drying hand disinfectant spray formulation with a non-sticky feel. These examples are not meant to limit the materials that can be used, but are merely to illustrate the types and levels of oils selected based on three criteria. The oils preferably used are those with a solubility greater than 30% in at least a 75 / 25 EtOH / water blend, reflecting the good solubility in the formulation shown in Sample 1. In addition, the oil must be liquid at room temperature so that once left as a film, the composition does not impart any negative feeling. Finally, the preferred oil itself must not have a solubility for water so that during application and when drying, as ethanol and water evaporate, water is also allowed to evaporate rapidly, leaving only the oil and any skin-benefitting active substance / moisturizer (if present). Without wishing to be bound by theory, it is believed that when a hand disinfectant formulation with a preferred oil as disclosed herein is applied to the skin and as water and ethanol evaporate, if the oil has a good solubilizing ability for water, this will delay evaporation and drive towards an undesirable slower drying and stickier feel.
[0067] Table 2
[0068]
[0069] Table 2 shows examples of oils that do and do not meet three criteria, including: (1) the oil must be a liquid; (2) the oil must be soluble in at least a 75 / 25 EtOH / water mixture; and (3) the oil must have no solvent power for water and, therefore, be ideal for use in the disclosed antimicrobial compositions.
[0070] Embodiment 2:
[0071] The composition of Sample 2 was used to test moisturizing, restorative wound healing test, and melanin content.
[0072] Moisturization was measured by visual measurements (dryness and erythema) and instrumental hydration assessment with Skicon and Skin Hydration Tester.Measurements were obtained at baseline, 2 hours, 4 hours, 6 hours and 8 hours after a single controlled product application.
[0073] As can be seen from the data, the disinfectant prepared with the composition of Sample 2 showed an improvement in hydration at all time points compared to the baseline. The disinfectant prepared with the composition of Sample 2 was also superior in hydration compared to the untreated control site.
[0074] Sample 2 was tested and compared to samples 3 and 4. The formulations of samples 2 to 4 are shown in Table 3. Samples 4 and 5 were untreated skin to which no disinfecting composition was applied.
[0075] Table 3
[0076]
[0077]
[0078] Participants entered a 5-day conditioning phase, during which they were instructed to use A white soap bar was used for all general bathing and showering, including the test site (forearm). Participants were asked to avoid using any additional products on the test area during the study and to avoid scraping for two days before or during the product application and assessment phase. Participants were also instructed to bathe at least 12 hours before the study.
[0079] At the baseline visit, participants with a visual dryness score of 0 to 2.5 and an erythema score of ≤2.0 (0-6 scale) at each site on the volar forearm and a grade difference between adjacent sites of no more than ±0.5 or 1.0 units overall entered the product application phase. A total of 8 test sites (4 cm×4 cm) were randomly assigned to the participant's right or left volar forearm. Treatment sites were balanced across all sites on the volar forearm by a carry-over Latin square design.
[0080] Before the product application phase, a baseline assessment of visual assessment of dryness and erythema was obtained by an expert visual assessor and instrumental assessment of hydration (Skicon: conductance, Skin Hydration Tester: capacitance). After the baseline measurement, the investigator performed a single controlled product application. The product application procedure is described as follows:
[0081] The researcher dispensed 0.035 mL of the test product onto the test site using a repeating pipette. Using a gloved finger, the test product was thoroughly rubbed onto the designated test site in a circular motion until the site was completely wet. The site was then allowed to air dry.
[0082] Two test sites were left untreated on each subject, which formed Samples 3 and 4.
[0083] Visual assessments of dryness, dermatometrometer, and Skicon measurements were then obtained at 2, 4, 6 hours (±15 minutes), and 8 hours (±30 minutes) after application. Prior to assessment, participants acclimatized for a minimum of 20 minutes in a room maintained at 66.3°F to 70.1°F (19.0°C to 21.2°C) and 38% to 55% relative humidity. One trained assessor performed all visual assessments during the treatment phase. Each instrument used more than one trained operator to collect instrument readings.
[0084] Visual dryness analysis, such as assessing change from baseline (CFB) within a product or assessing differences in CFB for pairs of products, was done using a simple pairwise nonparametric method (Wilcoxon signed-rank test with Pratt-Lehman adjustment).
[0085] For instrumental data, within each product / untreated site, each assessment time point was compared to baseline using a two-tailed paired t-test at a 95% confidence level. Between-treatment analyses of instrumental measures applied a single linear model at each time point (with CFB as the outcome variable, BL as a covariate, and PRODUCT as the efficacy of interest), with all products compared to all others. For each product's CFB, all comparisons of interest were also performed for all product comparisons.
[0086] No visual erythema was observed during the study period for any sample or untreated site.
[0087] During the hydration assessment with the dermatothromycete, samples 2 to 4 were evaluated. As shown in Table 4, samples 3 and 4 showed no change or deterioration in dermatothromycete levels from baseline at all time points after application. Sample 2 showed improvement at all time points after one application.
[0088] Table 4: Skin hydration CFB values of the CM825 skin moisture tester on the palmar forearm
[0089]
[0090]
[0091] As can be seen in Table 2, Sample 2 showed a statistically significant increase in hydration by the Skin Hydration Meter at all time points. At the 8 hour time point, Samples 3 and 4 showed a statistically significant increase in hydration by the Skin Hydration Meter. "BL" in Table 5 refers to Baseline. The baseline is the starting point of the measurement.
[0092] Table 5: Skicon 200-EX: Skin surface hydration - CFB, based on Log10 transformed values at the volar forearm
[0093]
[0094] As shown in Table 5, Sample 2, the sample of the present invention, showed a significant increase in hydration by Skicon at all time points, and Samples 3 and 4 showed a slight increase in hydration at all time points.
[0095] Example 3
[0096] In this embodiment, the restorative wound healing and barrier repair of various combinations of beneficial agents were tested. For wound healing assessment, cells were grown to 80% confluence and treated with bioactive substances for 24 to 36 hours. After treatment, scratches were produced on the cells, and the closure of the wound was monitored after incubation for 24 hours. The closure of the scratch area was calculated as the closure % of the wound. Microscope images were captured at a magnification of 4x, and the open wound area was measured using software CellSens for analysis. Wound closure in percentage = (the average value of the readings of the open wound area at (t=24) - the average value of the readings of the open wound area at (t=0) / the average value of the readings at (t=0)) * 100. Table 6 lists the results of wound healing assays.
[0097] Table 6
[0098]
[0099] As can be seen in Table 6, a significant improvement in wound healing was observed with the combination of vitamin B3, vitamin E, aloe vera and sodium hyaluronate, a statistically significant improvement compared to either vitamin B3 or vitamin E alone.
[0100] Example 4
[0101] In this embodiment, melanin content determination is carried out to determine the inhibition or melanin reduction found with various combinations of beneficial active substances as shown in Table 7. In this embodiment, cells are grown for 24 hours and treated with bioactive substances and incubated for 72 hours. After treatment, cells are lysed and absorbance is measured at 405nm. Lysate is transferred to a fresh 384-well transparent flat-bottom plate, and the original diameter is measured at 405nm using a FlexStation 3 reader. (The reduction % of melanin content is measured by taking the average absorbance value of the control - the average absorbance value of the treated cells / the average absorbance value of the control) * 100).
[0102] Table 7
[0103]
[0104] In this melanin content assay, 10 micromoles of 4-hexylresorcinol produced about 33% inhibition (baseline). Any inhibition above ~15% is generally considered significant in a melanin content assay. Thus, from Table 7, it can be seen that the combination of vitamin B3, vitamin E, and aloe vera showed significant inhibition of melanin content at both vitamin E levels. In addition, the combination of vitamin B3, vitamin E, aloe vera, and sodium hyaluronate showed significant inhibition of melanin content.
