Description

Technical Field

[0001] The invention belongs to the technical field of microorganisms and relates to an excellent strain of Coprinus comatus and an application method thereof. Background Art

[0002] Coprinus comatus (OFMüll.) Pers., scientifically known as Coprinus comatus, commonly known as Coprinus comatus or thorny mushroom, belongs to the phylum Basidiomycetes, class Agaricales, order Agaricales, family Agaricaceae, genus Coprinus, and is the model species of the genus Coprinus. Coprinus comatus has high nutritional and medicinal value. It is a rare edible fungus that can be used as both medicine and food. It not only contains rich nutrients such as protein, amino acids, sugars and mineral elements, but also has the effects of enhancing the body's immunity, lowering blood sugar, lowering blood pressure and anti-tumor. In addition, Coprinus comatus has the effects of aiding digestion, benefiting the spleen and stomach, and treating hemorrhoids. It also has potential value in treating alcoholic liver disease, Alzheimer's disease and leukemia.

[0003] Coprinus comatus has a high bioconversion rate, short growth cycle and is easy to cultivate, making it a new choice for industrial poverty alleviation in poor areas. However, the fruiting body of Coprinus comatus is very prone to browning and autolysis after opening, and will continue to autolyze after harvesting, making it inedible. In addition, serious diseases have been found in the cultivation of Coprinus comatus, which seriously affects its product quality and shelf life.

[0004] Although Coprinus comatus is easy to cultivate, its autolysis and browning problems have not been properly solved, and there are still serious diseases in the cultivation. It is urgent to improve the strains and promote the development of the Coprinus comatus industry. Patented wild Coprinus comatus strain and cultivation method thereof (CN201710142321.9) discloses a wild strain Coprinus comatus and its cultivation method, which adopts straw secondary fermentation method and layer-frame cultivation in the shed. Patented Coprinus comatus strain and preparation method (CN201210503847.2) provides a strain that uses Coprinus comatus fruiting bodies from Yunnan origin to screen out excellent strains, and uses biotechnology to identify strains, and prepares excellent strains of Coprinus comatus liquid and solid. However, there is currently a lack of reports on excellent hybrid strains of Coprinus comatus, especially hybrid strains with high yield, slow autolysis rate and strong disease resistance. Summary of the invention

[0005] One object of the present invention is to provide a Coprinus comatus strain with high yield, slow autolysis rate and strong disease resistance.

[0006] The invention provides a coprinus comatus, which is characterized in that the strain of the coprinus comatus is Coprinus comatus DCH1507, and its registration number in the General Microbiological Center of China Culture Collection of Microorganisms is CGMCC No. 22447.

[0007] Wherein, the Coprinus comatus has at least one of the following characteristics:

[0008] 1) The autolysis time of the Coprinus comatus is later than that of Coprinus comatus cc900;

[0009] 2) The harvesting period of the Coprinus comatus is shorter than that of Coprinus comatus cc900;

[0010] 3) The bioconversion rate of the Coprinus comatus is higher than that of Coprinus comatus cc900.

[0011] The fruiting body, mycelium and / or spores of the Coprinus comatus should also be within the protection scope of the present invention.

[0012] The protoplasts of Coprinus comatus should also be within the protection scope of the present invention.

[0013] The Coprinus comatus spawn sticks of the Coprinus comatus should also be within the protection scope of the present invention.

[0014] The use of Coprinus comatus in the preparation of Coprinus comatus fruiting bodies should also be within the protection scope of the present invention.

[0015] The application of Coprinus comatus in the preparation of Coprinus comatus mycelium should also be within the protection scope of the present invention.

[0016] The use of Coprinus comatus in the preparation of Coprinus comatus spores should also be within the protection scope of the present invention.

[0017] The application of Coprinus comatus in breeding of Coprinus comatus should also be within the protection scope of the present invention.

[0018] The Coprinus comatus strain is obtained by screening a hybrid culture of a strain isolated and purified from a wild Coprinus comatus fruiting body and a homokaryotic monosporic strain of a cc900 strain purchased from a strain enterprise.

[0019] The strain selection method comprises the following steps: parent screening, collecting basidiospores after parent cultivation, sequentially performing basidiospore suspension coating culture, picking monospore colonies, homokaryotic monospore strain verification, homokaryotic monospore strain hybridization, hybrid strain verification and hybrid strain variety comparison experiment.

[0020] Specifically, the method may include the following steps:

[0021] (1) Parent screening: The wild isolated strain and the cc900 strain were screened through confrontation experiments, comparison of mushroom production and comparison of fruiting body autolysis rates. The cc900 strain had the slowest autolysis rate, followed by the wild isolated strain. In addition, the wild isolated strain produced mushrooms in an even pattern with basically the same maturity. Therefore, these two strains were selected as parents for hybrid breeding.

[0022] (2) Parent cultivation: The above-mentioned wild isolated strains and cc900 strains were cultivated, with PDA medium used for mother strain cultivation, wheat grain medium used for original strain cultivation, and rice straw medium used for cultivated strain cultivation. After the mycelium has grown all over the rice straw medium, the bags are opened and covered with soil for fruiting, with a soil thickness of about 3 cm.

[0023] (3) Collection of basidiospores: When the fruiting bodies in step (2) have matured and the edge of the cap has slightly changed color but has not yet opened, take a portion of the gills and place them in a 5 mL centrifuge tube in a clean bench and wait for them to naturally dissolve into liquid at room temperature. Cut filter paper into small strips and sterilize them. Then use a sterile filter paper strip to dip the spore liquid into a 5 mL centrifuge tube. After drying in a clean bench, seal the centrifuge tube with a sealing film and store it in a dry place for later use.

[0024] (4) Preparation of basidiospore suspension and smear culture: Take the filter paper strip stained with basidiospores in step (3) and place it in a 2 mL centrifuge tube. Add 1 mL of Tween solution (0.05%) and shake to elute the spore suspension. Pipette an appropriate amount of spore liquid and drop it on a hemocytometer to count under an ordinary optical microscope. Then dilute the spore liquid to different concentrations, spread it on a PDA plate, and place it in a 25° C. biochemical incubator in the dark for culturing until the spores germinate.

[0025] (5) Picking single spore colonies: Use a sterile toothpick to pick a single germinated colony from step (4) onto a new PDA plate, and continue to culture in a biochemical incubator at 25°C in the dark until mycelium germinates.

[0026] (6) Verification of homokaryotic monosporic strains: After the mycelium germinates in step (5), a small amount of mycelium is picked up and observed under an ordinary optical microscope to see whether it has a lock-like joint structure. The strain without a lock-like joint structure is a homokaryotic monosporic strain, and the strain is further cultured to obtain a homokaryotic monosporic strain.

[0027] (7) Hybridization of homokaryotic monosporic strains: Use a sterile toothpick to pick up two homokaryotic monosporic strains and place them on the same PDA plate in an ultra-clean workbench. Place the plate in a 25°C biochemical incubator and culture it in the dark until the hyphae of the two colonies come into contact. Observe the contact phenomenon and pick up a small amount of hyphae from the contact area for observation under an ordinary optical microscope. If a lock-like joint structure is observed, the hybridization is successful.

[0028] (8) Hybrid strain verification: Use a sterile hole puncher to punch holes at the edge of the Coprinus comatus colony. Then use a sterile toothpick to pick up the colonies of the hybrid strain and the colonies of the two parent strains and place them on the same PDA plate. Distribute them in a triangular shape, with the colonies about 2 cm apart. Place the plate in a dark incubator at 25°C and observe the antagonism after the mycelium of the two colonies come into contact. If the hybrid has antagonism with the parent, it is preliminarily proved that a new hybrid strain of Coprinus comatus has been obtained by hybridization.

[0029] (9) Hybrid strain comparison experiment: The obtained hybrid strains are cultivated for mushroom production, and the earliest appearance of primordium, fruiting body harvesting cycle, first wave mushroom yield, fruiting body height, cap size, stipe thickness, fruiting body unit weight, autolysis rate and antagonism to pathogens of each strain are compared to select hybrid strains with excellent traits.

[0030] The culture medium formula and preparation method in step (2) are:

[0031] The PDA culture medium is composed of 200 g peeled potatoes, 20 g glucose, 18 g agar powder, and 1000 mL tap water.

[0032] The wheat grain culture medium comprises 93% wheat grains, 5% cow dung, 1% gypsum, 1% calcium carbonate and 0.1% lime.

[0033] The wheat grains in the wheat grain culture medium should be soaked overnight before pre-cooking, and the cow dung should be pre-wetted in advance. After the wheat grains are rinsed, they are fished out and drained, and then immersed in water and boiled for about 20 minutes, so that the wheat grains change from beige to light brown, and attention should be paid to prevent them from swelling and cracking. The cooked wheat grains are fished out and spread in a clean large basin to evaporate the moisture on the surface of the wheat grains until they are not hot to the touch, and other auxiliary materials are mixed in while they are hot. The moisture content is preferably 42% to 45%, and the pH is 7 to 8.

[0034] The rice straw compost comprises 30% of cut rice straw, 15% of miscellaneous wood chips, 20% of cottonseed hulls, 15% of dry cow dung powder, 18% of wheat bran and 2% of lime.

[0035] Before the rice straw compost is mixed, the rice straw needs to be soaked overnight, and the miscellaneous wood chips, cottonseed hulls and dry cow dung powder need to be pre-wetted in advance, with a water content of about 65%.

[0036] The DCH1507 strain of Coprinus comatus provided by the present invention is a hybrid strain obtained by hybridizing the cc900 strain purchased from the Lianyi Edible Fungi Promotion Center in Yangzhou, Jiangsu Province with a wild specimen isolated from Yanqi Lake in Huairou District, Beijing. The strain can obtain fruiting bodies after cultivation. The fruiting bodies obtained by cultivating the strain are 8.5 to 20 cm high and 7 to 60 g in weight; the cap is elliptical, bell-shaped or umbrella-shaped; the outer surface of the cap has brown scales, which are fleshy and often curled; the cap is initially white and then turns black and self-dissolves; the stipe is mesogenic, fibrous, 0.5 to 1.5 cm in diameter, hollow, and has a cotton-like string in the center; the gills are separated from the stipe, the flesh is white, has a ring structure, is membranous, and is connected to the cap to tightly wrap the stipe when the cap is not opened, and the ring is easy to loosen and slide off after the cap matures and opens.

[0037] The invention also provides a cultivation method of the Coprinus comatus, comprising the step of carrying out mushroom cultivation on the Coprinus comatus.

[0038] Beneficial effects of the present invention:

[0039] The invention is a new variety of Coprinus comatus with excellent properties. Compared with other hybrid strains and parent strains, the invention has a high bioconversion rate of 117.5%, a slow autolysis rate, which is slower than other hybrid strains, 2 days slower than the cc900 parent, and 8 days slower than the 935 parent. The invention has strong anti-pathogenic bacteria ability, and has strong antagonism to Lecanicillium fungicola, Mucor racemosus and Trichoderma virens.

[0040] The invention provides a hybrid strain of Coprinus comatus with high yield, slow autolysis speed and strong disease resistance, develops a new variety of rare edible fungi and enriches edible fungi variety resources; at the same time, the fungus is a typical grass-rotting fungus, can utilize crop straw, and has ecological significance.

Morphological observation and nuclear rDNA-ITS sequencing proved that the strain was a Coprinus comatus strain. The wild Coprinus comatus fruiting bodies were separated, purified and cultured to obtain Coprinus comatus 935 strain.

[0065] Example 1, identification and hybridization of parent strains

[0066] The hybrid culture of the homokaryotic monospore strain of Coprinus comatus 935 and Coprinus comatus cc900 was cultivated for comparative screening. The specific breeding method has the following steps:

[0067] (1) Parent screening: The wild isolated strain and the cc900 strain were screened through confrontation experiments, comparison of mushroom production and comparison of autolysis rates. The cc900 strain had the slowest autolysis rate, followed by the wild isolated strain. In addition, the wild isolated strain produced mushrooms in an even pattern with basically the same maturity. Therefore, these two strains were selected as parents for hybrid breeding.

[0068] (2) Parent cultivation: The wild isolated strains and cc900 strains were cultivated, with PDA medium used for mother strain cultivation, wheat grain medium used for original strain cultivation, and rice straw medium used for cultivated strain cultivation. When the mycelium is full of rice straw medium, the bags are opened and covered with soil for fruiting.

[0069] (3) Collection of basidiospores: When the fruiting bodies in step (2) have matured and the edge of the cap has slightly changed color but has not yet opened, take a portion of the gills and place them in a 5 mL centrifuge tube in a clean bench and wait for them to naturally dissolve into liquid at room temperature. Cut filter paper into small strips and sterilize them. Then use a sterile filter paper strip to dip the spore liquid into a 5 mL centrifuge tube. After drying in a clean bench, seal the centrifuge tube with a sealing film and store it in a dry place for later use.

[0070] (4) Preparation of basidiospore suspension and smear culture: Take the filter paper strip stained with basidiospores in step (3) and place it in a 2 mL centrifuge tube.

[0071] (5) Picking single spore colonies: Use a sterile toothpick to pick a single germinated colony from step (4) onto a new PDA plate, and continue to culture in a biochemical incubator at 25°C in the dark until mycelium germinates.

[0072] (6) Verification of homokaryotic monosporic strains: After the mycelium germinates in step (5), a small amount of mycelium is picked up and observed under an ordinary optical microscope to see whether it has a lock-like joint structure. The strain without a lock-like joint structure is a homokaryotic monosporic strain, and the strain is further cultured to obtain a homokaryotic monosporic strain.

[0073] (7) Hybridization of homokaryotic monosporic strains: Use a sterile toothpick to pick up two homokaryotic monosporic strains and place them on the same PDA plate in an ultra-clean workbench.

[0074] Example 2: Hybrid strain verification and comparison experiment

[0075] (1) Hybrid strain verification:

[0076] Use a sterile hole puncher to punch holes at the edge of the Coprinus comatus colony, and then use a sterile toothpick to pick up the bacterial blocks of the hybrid strain and the bacterial blocks of the two parent strains and put them into the same PDA plate, distributing them in a triangular shape, with the bacterial blocks about 2 cm apart. The plate is placed in a dark incubator at 25°C for culture, and the antagonism phenomenon is observed after the mycelium of the two colonies comes into contact. If the hybrid has antagonism with the parent, it preliminarily proves that the hybridization has obtained a new hybrid strain of Coprinus comatus.

[0077] (2) Hybrid strain comparison experiment:

[0078] The hybrid strains were cultivated for mushroom production and cultured in confrontation with pathogenic fungi to compare their mushroom production, fruiting body autolysis time and their antagonism to pathogenic fungi in confrontation culture.

[0079] The method for comparing the autolysis of the fruiting bodies is as follows: the hybrid strains are selected to have basically the same maturity, that is, the middle part of the cap has swelled and become loose, but the fungus ring has not loosened, still tightly wrapping the stipe, and the fruiting bodies whose cap edges have not changed color are placed in a 4°C refrigerator, and the fruiting bodies of the cc900 and 935 parent strains are used as controls, and the color change of the gills at the edge of the cap is used as the autolysis standard, and the time required for all the fruiting bodies of each strain to reach the autolysis standard is observed and compared. The results are shown in Table 1, indicating that the patented variety has a shorter harvesting period than the parent control, from covering the soil to harvesting, only 28±1d; the bioconversion rate is high, and the bioconversion rate of the first wave mushroom is 117.5±0.9%; the autolysis is slow, and it starts to autolyze after being stored at 4°C for 18d after harvesting.

[0080] The method for the confrontation culture of the hybrid strain and the pathogenic fungus is as follows: the hybrid strain block is picked to one side of the PDA plate, placed in a 25°C incubator for 3 days, then taken out, and Trichoderma viride, Mucor racemosus and Verticillium truncatum are respectively transferred to the other side of the PDA plate, and then placed in a 25°C incubator for further culture, and the antagonistic ability of the hybrid strain to the pathogenic fungus is observed. The results are as follows: Figure 8 As shown. Figure 8 It can be seen that the hybrid strain of the present invention has strong disease resistance and has strong antagonism against Verticillium, Mucor racemosus and Trichoderma virens ( Figure 8 ).

[0081] Table 1 shows the results of the comparison between the patented strain and the parent strain

[0082]

[0083] Note: The same letters in the same row indicate no significant difference in multiple tests (P=0.05).

[0084] (3) Fruiting body characteristics of the invented strain

[0085] The fruiting bodies of Coprinus comatus strain DCH1507 were successfully cultivated. The fruiting bodies are 8.5 to 20 cm high and weigh 7 to 60 g each. The cap is elliptical, bell-shaped or umbrella-shaped. The outer surface of the cap has brown scales, which are fleshy and often curled. The cap is initially white and then turns black and self-dissolves. The stipe is mesogenous, fibrous, 0.5 to 1.5 cm in diameter, hollow, and has a cotton-like string in the center. The gills are separated from the stipe, the flesh is white, has a ring structure, is membranous, and is connected to the cap and tightly wraps the stipe when it is not opened. The ring is easy to loosen and slide off after the cap matures and opens.

[0086] Example 3 Screening of parent strains:

[0087] (1) Comparison of cultivation characteristics and fruiting body autolysis speed of strains from different sources: The collected strains of Coprinus comatus from different sources were cultivated, and the fruiting body morphology was evaluated and compared after the mushrooms were produced. The fruiting body autolysis speed of different strains was also compared. First, fruiting bodies with basically the same maturity, that is, the middle part of the cap had swelled and loosened, but the annulus had not loosened and still tightly wrapped the stipe, and the edge of the cap had not changed color, were selected and placed in a 4°C refrigerator. They were observed and compared every 12 hours and photographed. The color change of the gills at the edge of the cap was used as the autolysis standard. The fruiting body autolysis rate was calculated according to the following formula (fruiting body autolysis rate = number of fruiting bodies autolyzed during storage time / storage time). Parents with excellent traits were selected for hybrid breeding based on the strain characteristics and autolysis speed.

[0088] (2) Different strains confrontation experiment: Use a sterile hole puncher to punch holes at the edge of the Coprinus comatus colony, then pick out the fungus blocks of different strains and put them on the same PDA plate. Place the two fungus blocks opposite to each other and culture the plate in a dark incubator at 25°C. After the hyphae of the two colonies come into contact, observe the antagonism phenomenon. The strains with antagonism are different strains.

[0089] Embodiment 4 Research on fungal diseases of Coprinus comatus:

[0090] (1) The diseased fruiting bodies of Coprinus comatus brought back from the soil cave cultivation base of Coprinus comatus in Pingyin, Shandong and Guanghao Edible Fungi Company in Xuzhou, Jiangsu were gently wiped clean with alcohol cotton balls, and the diseased tissues were picked out with a sterile inoculation needle and transferred to a PDA plate. The plate was then cultured in an incubator at 20°C. After the mycelium germinated, part of the mycelium was picked out and placed under an optical microscope to observe the microstructure.

[0091] (2) Pick part of the hyphae into a centrifuge tube, shake and grind, and extract DNA using the CTAB method. PCR amplification was performed using ITS4 / ITS5 primers. The amplified products were sent to Beijing Sangon Biotechnology (Shanghai) Co., Ltd. for sequencing. The obtained sequences were compared to NCBI. The species were identified based on sequence similarity and morphological characteristics. Results Six fungi were isolated and purified from the infected fruiting bodies of Coprinus comatus, namely Lecanicillium fungicola, Mucor racemosus, Trichoderma virens, Clonostachys rosea, Trichoderma rossicum, and Fusarium sp., among which Verticillium has been reported to be the pathogen of blackhead disease of Coprinus comatus.

[0092] Example 5 Comparison experiment between DCH1507 strain and other hybrid strains:

[0093] (1) Comparison of fruiting of DCH1507 and other hybrid strains: The colony edges of DCH1507 and other hybrid strains were punctured and the fungus blocks were transferred to new PDA plates for culture. Their growth rates and colony morphologies were compared. The plates were then transferred to wheat grain culture medium for culture. After full growth, they were transferred to straw culture medium. After the mycelium had fully grown on the straw culture medium, they were cultured for 5 to 10 days before being opened and covered with soil for fruiting. The earliest appearance of primordia, harvesting cycle, bioconversion rate, and fruiting body morphology of each strain were compared.

[0094] (2) Comparison of autolysis time between DCH1507 and other hybrid strains: The fruiting bodies of the strains were selected to be basically the same in maturity, that is, the middle part of the cap had swelled and loosened, but the annulus had not loosened and still tightly wrapped around the stipe, and the edge of the cap had not changed color. The fruiting bodies of the parent strains cc900 and 935 were used as controls, and the color change of the gills at the edge of the cap was used as the autolysis standard. The time required for all the fruiting bodies of the strains to reach the autolysis standard was observed and compared.

[0095] Table 2 shows the comparison results of the patented strains with other hybrid strains

[0096]

[0097] Note: The same letters in the same row indicate no significant difference in multiple tests (P=0.05); “-” indicates missing experimental results.

[0098] (3) Comparison of disease resistance of strains: DCH1507 strain and other hybrid strains were picked up on one side of the PDA plate, placed in a 25°C incubator for 3 days, and then taken out. Trichoderma, Mucor racemosus and Verticillium were transferred to the other side of the PDA plate respectively, and then placed in a 25°C incubator for further culturing to observe the antagonistic ability of the hybrid strains to common diseases. According to the antagonistic phenomenon, the antibacterial ability of Coprinus comatus was divided into 4 levels: Level I (****): strong antibacterial ability, the diameter of the Coprinus comatus colony was greater than 1 / 2 of the plate diameter; Level II (***): relatively strong antibacterial ability, the diameter of the Coprinus comatus colony was approximately equal to 1 / 2 of the plate diameter; Level III (**): weak antibacterial ability, the diameter of the Coprinus comatus colony was less than 1 / 2 of the plate diameter; Level IV (*): extremely weak antibacterial ability, the diameter of the Coprinus comatus colony was less than 1 / 3 of the plate diameter (such as Figure 9-11 shown).

[0099] Table 3 is a comparison of the antibacterial ability of each hybrid strain and parent strain
