Description

Technical Field

[0001] The invention belongs to the technical field of cosmetics, and specifically relates to a whitening and anti-aging composition, a whitening and anti-aging cream and a preparation method thereof. Background Art

[0002] With the continuous development and progress of society, people's quality of life has gradually improved, and skin care products have increasingly entered thousands of households. Among them, the demand for skin care products with whitening and anti-aging functions has become increasingly prominent.

[0003] Melanin is synthesized in melanocytes, and its production process requires several basic substances: tyrosine, tyrosinase, oxygen, and DOPA. Tyrosine is the main raw material for melanin production, tyrosinase plays a key role in promoting the conversion of tyrosine into melanin, and oxygen is essential in the oxidation process of DOPA. The production of melanin in the skin involves a series of complex physiological and biochemical processes, including the migration, division and maturation of melanocytes, the transport of melanin particles, and the final excretion of melanin.

[0004] Aging is an inevitable natural process in everyone's life. It involves complex physiological mechanisms and is usually manifested as reduced skin moisture leading to dryness, gradual loss of collagen, increased facial expression lines, and gradual sagging of the skin.

[0005] Most products on the market currently only focus on a single whitening or anti-aging effect, and their mechanism of action is single, which cannot achieve comprehensive skin care. At the same time, products that can take into account both whitening and anti-aging are very scarce. Therefore, developing a product that can work in all directions and integrate whitening and anti-aging effects is of great significance to meet consumer needs. Summary of the invention

[0006] The purpose of the present invention is to overcome the shortcomings and deficiencies of the prior art and to provide a whitening and anti-aging composition, a whitening and anti-aging cream and a preparation method thereof.

[0007] To achieve the above objectives, the technical solutions adopted by the present invention include:

[0008] In a first aspect, the present invention provides a whitening and anti-aging composition, which comprises the following components in percentage by weight: 0.1-1% glabridin, 0.1-10% rhamnosylsilanol, 0.1-10% yarrow extract, 0.1-40% peony root extract, 0.1-10% acetylated sodium hyaluronate, 20-24% moisturizer and the remainder water.

[0009] The whitening and anti-aging composition of the present invention is prepared by scientifically combining specific whitening and anti-aging ingredients and plant extracts. Experimental research has found that the whitening and anti-aging effects of each component raw material are not obvious when used alone, but after compounding with the above-mentioned specific raw materials, the components cooperate with each other and give full play to their functions, so that the whitening and anti-aging effects of the composition are significant, and the whitening component and the anti-aging component promote each other's whitening and anti-aging effects, and have a synergistic effect.

[0010] Among them, glabridin, as a whitening ingredient, acts on the epidermis. It can inhibit the activity of tyrosinase, dopachrome interconvertase (TRP-2), DHICA oxidase, and α-MSH melanocyte-stimulating hormone, thereby inhibiting melanin production and melanosome transfer. It can also help reduce the total amount of melanin by inhibiting the proliferation of melanocytes and B16 cells. It also has the ability to scavenge oxygen free radicals similar to superoxide dismutase (SOD), thereby inhibiting the generation of reactive oxygen species.

[0011] As a whitening ingredient, yarrow extract acts on the epidermis, epidermal junction layer, and dermis. It can promote the expression of small molecule heat shock proteins sHSPs and HSP27 that protect skin function on the skin, thereby assisting the correct folding of proteins and binding to misfolded proteins to prevent their irreversible aggregation. It can also inhibit the production of melanin by inhibiting the activity of tyrosinase.

[0012] Rhamnosylsilanol, as an anti-aging ingredient, acts on the epidermal junction layer. It can promote the production of laminin and improve the binding of keratinocytes and DEJ. It can also promote the production of type VII collagen and improve the binding of the extracellular matrix of the dermis and DEJ.

[0013] As an anti-aging ingredient, peony root extract acts on the skin barrier and epidermis. It can inhibit a variety of bacteria and molds that act on the skin and promote the balance of skin microecology. At the same time, it can also inhibit the release of histamine and tumor necrosis factor (TNF-α), inhibit the production of antibodies to activate cells CD 40 monoclonal antibodies, recombinant interleukin-4 (RIL-4) and recombinant histamine releasing factor (RHRF), and down-regulate the expression of IL-4 in the reverse transcription polymerase chain reaction of cell activation, thereby helping the skin to better fight inflammation and allergies and keep the skin in better condition.

[0014] As an anti-aging ingredient, acetylated sodium hyaluronate acts on the epidermis, epidermal junction and dermis. It can penetrate deeper into the skin than standard non-acetylated hyaluronic acid and is 13 times more resistant to hyaluronidase than standard hyaluronic acid. At the same time, it protects 9 types of collagen and elastin by inhibiting the expression of proteolytic enzymes such as MMP-1, MMP-3, and MMP-9.

[0015] Preferably, the whitening and anti-aging composition comprises the following components in percentage by mass: 0.5-1% glabridin, 1-10% rhamnosylsilanol, 1-10% yarrow extract, 10-40% peony root extract, 1-10% acetylated sodium hyaluronate, 20-24% moisturizer and the balance water.

[0016] Preferably, the whitening and anti-aging composition comprises the following components in percentage by mass: 1% glabridin, 10% rhamnosylsilanol, 10% yarrow extract, 40% peony root extract, 10% acetylated sodium hyaluronate, 22% moisturizer and the balance water.

[0017] Through experimental research, it was found that when the components in the composition are used in the above-mentioned optimal amount, the whitening and anti-aging effects of the obtained whitening and anti-aging composition are the best, and when the amount of the components exceeds the above-mentioned limited range, the whitening and anti-aging effects of the obtained composition decrease, and the cost-effectiveness is significantly reduced.

[0018] Preferably, the moisturizing agent comprises butanediol and 1,2-hexanediol, the mass percentage content of the butanediol is 19-21%, and the mass percentage content of the 1,2-hexanediol is 1-3%.

[0019] In a second aspect, the present invention provides a method for preparing the whitening and anti-aging composition, comprising the following steps:

[0020] (1) mixing butanediol and glabridin, heating to 60-65° C., and stirring to mix uniformly to obtain a mixture;

[0022] (3) adding 1,2-hexanediol, stirring evenly, and filtering the material to obtain the whitening and anti-aging composition.

[0023] In a third aspect, the present invention provides a whitening and anti-aging product, wherein the whitening and anti-aging product contains the above-mentioned whitening and anti-aging composition.

[0024] By adopting the above technical scheme, the whitening and anti-aging composition can be directly added to the skin care products to make the skin care products have whitening and anti-aging effects. The use is simple and convenient and the application range is wide, which is beneficial to improve the economic benefits of the whitening and anti-aging composition and make the whitening and anti-aging composition more suitable for market demand.

[0025] In a fourth aspect, the present invention provides a whitening and anti-aging cream, comprising the above-mentioned whitening and anti-aging composition.

[0059] Effect Example 1

[0060] 1. Skin irritation test

[0061] Test method: 200 voluntary testers were selected, each with 50 testers as a group, divided into 4 groups, and the whitening and anti-aging compositions of Examples 1-4 were tested respectively. Each tester used the inner side of the left and right arms as a control test, and 0.3 mL of the composition prepared in Examples 1-4 of the present invention was applied to the inner side of the left hand each time, and the right hand was not treated as a control, twice a day, and the skin reaction phenomenon after 24 hours and 72 hours was recorded.

[0062] The results show that after applying the product of the present invention for 24 hours and 72 hours, no skin redness or allergic phenomena such as erythema are observed, indicating that the whitening and anti-aging composition of the present invention is safe to be used in cosmetics without any adverse phenomena.

[0063] 2. Whitening test

[0064] Test samples: The samples prepared in Examples 1-4 and Comparative Examples 1-9 were diluted 50 times with deionized water.

[0065] 20 testers were selected, aged 30-40 years old, with an average age of 34 years old, and the whitening efficacy evaluation method of the human skin melanization model induced by ultraviolet light was used. A xenon arc lamp sunlight simulator (290-400nm) with continuous spectral radiation and capable of generating UVA+UVB wavelength ultraviolet rays was used to mark 13 areas of skin on the back (same area and area) for irradiation experiments. After the irradiation experiment, the diluted samples of Examples 1-4 and Comparative Examples 1-9 were applied to the test areas for 28 consecutive days. The skin color instrument was used clinically to detect and record the percentage of melanin reduction on the 7th, 14th, 21st, and 28th days. The test results are shown in Table 4.

[0066] Table 4 Melanin reduction percentage

[0067] Sample source Day 7 Day 14 Day 21 Day 28 Example 1 3 5 10 15 Example 2 5 9 15 20 Example 3 7 12 20 28 Example 4 12 20 27 35 Comparative Example 1 2 3 5 8 Comparative Example 2 4 6 9 11 Comparative Example 3 6 8 13 18 Comparative Example 4 6 11 15 18 Comparative Example 5 8 12 16 19 Comparative Example 6 3 5 10 14 Comparative Example 7 6 8 14 18 Comparative Example 8 9 15 19 23 Comparative Example 9 1 2 3 3

[0068] From the results in Table 1, it can be seen that the whitening and anti-aging compositions prepared in Examples 1-4 of the present invention decreased melanin after 7 days, 14 days, 21 days and 28 days of use test, among which the percentage of melanin decrease in Example 4 was the most significant and the whitening effect was the best.

[0069] When any of the whitening components, glycyrrhizin or yarrow extract, is lacking in comparative examples 1-2, the whitening effect of the obtained composition is significantly reduced (the percentage of melanin reduction is not obvious), indicating that the specific whitening components of glycyrrhizin and yarrow extract used in the present invention have a synergistic enhancement of whitening efficacy; when there is no lack of whitening components in comparative examples 3-5, but only any of the anti-aging components, rhamnosylsilanol, peony root extract and acetylated sodium hyaluronate, are lacking, the percentage of melanin reduction in the obtained composition is significantly reduced, which is significantly lower than that in Example 4, indicating that the specific anti-aging component used in the present invention, in addition to its own anti-aging effect, can also promote the whitening effect of the whitening component, that is, the whitening component selected in the present invention has a synergistic effect with the anti-aging component, which significantly enhances the whitening and anti-aging effect of the composition.

[0070] In Comparative Example 6, glabridin and yarrow extract in the whitening component are replaced with niacinamide which also has a whitening effect, and in Comparative Example 7, rhamnosylsilanol, peony root extract and acetylated sodium hyaluronate in the anti-aging component are replaced with a compound of glycerol, water, tetradecylaminobutyryl valinamidobutyric acid urea trifluoroacetate and magnesium chloride which also have an anti-aging effect. According to the test results, the melanin reduction percentages of the compositions prepared in Comparative Examples 6-7 are all lower than those in Example 4, indicating that the composition with whitening and anti-aging effects provided by the present invention has a significant whitening effect through the synergistic effect between the components, and any one component cannot achieve the same effect by replacing it with an existing whitening component or anti-aging component that is recognized to have a better effect.

[0071] At the same time, when the dosage ratio of glycyrrhizin, rhamnosilanol, yarrow extract, peony root extract and acetylated sodium hyaluronate components in the compositions of Comparative Examples 8-9 exceeds the specified range of the present invention, the melanin removal effect of the prepared composition is significantly reduced, indicating that the components in the composition of the present invention can better exert a synergistic effect when they are within a specific dosage range, thereby maximizing the whitening and anti-aging effect of the composition.

[0072] 3. Evaluation of free radical scavenging effect

[0073] The DPPH method was used for detection, as follows:

[0074] (1) Weigh a certain amount of DPPH and prepare a 0.04 mg / mL DPPH solution with anhydrous ethanol;

[0075] (2) Take 0.2 mL of the samples of Examples 1-4 and Comparative Examples 1-9, 3.8 mL of pH 6.8 phosphate buffer, add them to 4 mL of DPPH solution, mix well, and place at 25° C. for 30 minutes. Then take the same amount of sample and measure the absorbance at 517 nm (each sample is measured in parallel 3 times and the average value is taken). According to the formula, DPPH free radical scavenging rate (%) = (absorbance of DPPH at 517 nm without adding sample - absorbance of DPPH at 517 nm with added sample) / absorbance of DPPH at 517 nm without adding sample × 100%. The test results are shown in Table 5 below.

[0076] Table 5 DPPH free radical scavenging rate

[0077]

[0078]

[0079] 4. Zebrafish anti-wrinkle efficacy test

[0080] The zebrafish anti-wrinkle efficacy test was performed on the compositions prepared in Examples 1-4 and Comparative Examples 1-9.

[0081] Wild-type AB strain zebrafish was used as the test system. The relative expression levels of three type I collagen genes (collala, collalb and colla2 encoding α1(I), α2(I) and α3(I) chains, respectively) present in zebrafish were detected to indicate whether the sample had anti-wrinkle effects. The specific method is as follows.

[0082] Zebrafish 4 days after fertilization were randomly selected in 6-well plates, with 30 fish in each well; water-soluble samples were given, and a blank control group was set up at the same time, with a volume of 3 mL per well, and three biological replicates; incubated at 28°C in the dark for 24 h; total RNA of zebrafish in each experimental group was extracted, cDNA was synthesized, and q-PCR was used to detect the gene expression of β-actin and the target gene; β-actin was used as the internal reference for gene expression, and the relative RNA expression of the target gene was calculated (Table 6), and the calculation formula is as follows:

[0083] Relative RNA expression = 2 ΔΔC(t) , where ΔΔC(t)=ΔC(t) 正常对照组 -ΔC(t) 样品组 , ΔC(t)=C(t) 目的基因 -C(t) β-actin

[0084] Table 6 Relative expression of Collala gene

[0085]

[0086]

[0087] From the results in Table 6, it can be seen that the compositions prepared in Examples 1-4 all have good anti-wrinkle effects at a test concentration of 2%, and the relative expression levels of the Collala gene are all increased, with the increase showing an increasing trend. The composition prepared in Example 4 has the largest relative expression level of the Collala gene and the best anti-aging and anti-wrinkle effect.

[0088] While in Comparative Examples 1-2, there is no lack of anti-aging components, and only any one of the whitening components, glycyrrhizin or yarrow extract, is lacking. The anti-aging effect of the composition obtained in Comparative Examples 1-2 is significantly reduced compared with that in Example 4. Combined with the whitening test results, it can be obtained that the whitening component used in the present invention can not only exert its own whitening effect, but also promote the anti-aging effect of the anti-aging component, that is, there is a synergistic effect between the whitening component and the anti-aging component, which significantly enhances the whitening and anti-aging effect of the composition; in Comparative Examples 3-5, when any one of the anti-aging components, rhamnosylsilanol, peony root extract and acetylated sodium hyaluronate, is lacking, the effect of the obtained composition on improving the relative expression of the Collala gene is significantly inferior to that in Example 4, indicating that there is a synergistic effect between the three anti-aging components of the present invention, which significantly enhances the anti-aging and anti-wrinkle effect of the composition.

[0089] In Comparative Examples 6-7, when conventional whitening components and conventional anti-aging components were used to replace the specific whitening and anti-aging components described in the present invention, the obtained compositions had lower effects on improving the relative expression of the Collala gene than those in Example 4. It can be concluded that the composition with whitening and anti-aging effects provided by the present invention has significant anti-aging effects through the synergistic effect between the components, and the same effect cannot be achieved by replacing any one component with an existing whitening component or anti-aging component that is recognized to have a better effect.

[0090] Similarly, when the dosage ratio of the components in the compositions of Comparative Examples 8-9 exceeds the specified range of the present invention, the effect of the prepared compositions on improving the relative expression of the Collala gene is significantly reduced, that is, the anti-wrinkle and anti-aging effects are reduced, indicating that the components in the composition of the present invention can better exert a synergistic effect when they are within a specific dosage range, thereby maximally enhancing the whitening and anti-aging effects of the composition.

[0091] Effect Example 2

[0092] 1. Appearance and stability test

[0093] The whitening and anti-aging cream prepared in Examples 5-7 of the present invention has a white cream-like appearance, and the pH value is tested to be 5.0-7.0. The heat stability and cold stability are investigated and it is found that the whitening and anti-aging cream samples are placed at 45°C for 24 hours and -15°C for 24 hours, respectively, and the appearance and pH value of the samples do not change, and the samples have good stability, and the samples are qualified in the inspection.

[0094] 2. Evaluation of long-term anti-aging effects

[0095] The whitening and anti-aging creams prepared in Examples 5-7, Comparative Examples 10-12 and Blank Examples were used as samples. The anti-aging efficacy of the test samples was evaluated by continuous use of the test samples by adult female volunteers under normal circumstances. 350 female subjects with dull facial complexion, expression lines and fine lines, aged 35-60 years old, were selected, and each group of 50 was divided into 7 groups. The whitening and anti-aging creams prepared in Examples 5-7, Comparative Examples 10-12 and Blank Examples were tried respectively, and applied evenly after cleansing, twice a day, once in the morning and evening, and the products were used for four weeks and eight weeks. A questionnaire survey was conducted to understand the evaluation of the subjects on the improvement of wrinkles after using the product 4 weeks and 8 weeks after using the product. The evaluation indicators are mainly divided into: light lines and brightening effect. The results are divided into: the effect is not obvious, the effect is average, more obvious, and very obvious. The evaluation results are shown in Tables 7-8.

[0096] Table 7 Evaluation results after four weeks of use

[0097]

[0098] The results in Table 7 show that when 1% of the whitening and anti-aging composition prepared in Example 4 is used in Example 5, after the volunteers use the prepared whitening and anti-aging cream for 4 weeks, 52% of the volunteers feel that the whitening and anti-aging cream has a very obvious effect of lightening and brightening. When 5% of the whitening and anti-aging composition is used to prepare the whitening and anti-aging cream, 70% of the volunteers feel that the whitening and anti-aging cream has a very obvious effect of lightening and brightening. When 8% of the whitening and anti-aging composition is used to prepare the whitening and anti-aging cream, 72% of the volunteers feel that the whitening and anti-aging cream has a very obvious effect of lightening and brightening. This shows that the whitening and anti-aging composition of the present invention has excellent effects on lightening and brightening, anti-aging and whitening; and compared with the cream prepared with commercially available anti-aging actives or commercially available whitening actives, the effect of lightening and brightening is more significant.

[0099] Table 8 Evaluation results after eight weeks of use

[0100]

[0101] The results in Table 8 show that after using the whitening and anti-aging cream prepared by the present invention for eight consecutive weeks, 70% or even 84% of the volunteers felt a very obvious wrinkle lightening and brightening effect, and the effect was significantly better than that of the cream prepared with commercially available anti-aging actives or commercially available whitening actives, indicating that the whitening and anti-aging cream prepared with the whitening and anti-aging composition of the present invention has excellent long-lasting anti-aging, anti-wrinkle, wrinkle lightening and brightening effects.

[0102] 3. Zebrafish anti-wrinkle efficacy test

[0103] The zebrafish anti-wrinkle efficacy test was performed on the whitening and anti-aging creams prepared in Examples 5-7 and Comparative Examples 10-12.

[0104] Wild-type AB strain zebrafish was used as the test system. The relative expression levels of three type I collagen genes (collala, collalb and colla2 encoding α1(I), α2(I) and α3(I) chains, respectively) present in zebrafish were detected to indicate whether the sample had anti-wrinkle effects. The specific method is as follows.

[0105] Zebrafish 4 days after fertilization were randomly selected in 6-well plates, with 30 fish in each well; water-soluble samples were given, and a blank control group was set up at the same time, with a volume of 3 mL per well, and three biological replicates; incubated at 28°C in the dark for 24 hours; total RNA of zebrafish in each experimental group was extracted, cDNA was synthesized, and q-PCR was used to detect the gene expression of β-actin and the target gene; β-actin was used as the internal reference for gene expression, and the relative RNA expression of the target gene was calculated (Table 9), and the calculation formula is as follows:

[0106] Relative RNA expression = 2 ΔΔC(t) , where ΔΔC(t)=ΔC(t) 正常对照组 -ΔC(t) 样品组 , ΔC(t)=C(t)目的基因 -C(t) β-actin

[0109] 4. Whitening test

[0110] Test samples: The whitening and anti-aging creams prepared in Examples 5-8 and Comparative Examples 10-12 were used as samples.

[0111] 20 testers were selected, aged 30-40 years old, with an average age of 34 years old, and the whitening efficacy evaluation method of the human skin melanization model induced by ultraviolet light was used. A xenon arc lamp sunlight simulator (290-400nm) with continuous spectral radiation and capable of generating UVA+UVB wavelength ultraviolet rays was used to mark 6 areas of skin on the back (same area and area) for irradiation experiments. After the irradiation experiment, the samples of Examples 5-7 and Comparative Examples 10-12 were applied to the test areas for 28 consecutive days. The skin color instrument was used clinically to detect and record the percentage of melanin reduction on the 7th, 14th, 21st, and 28th days. The test results are shown in Table 10.

[0112] Table 10 Melanin reduction percentage

[0113]

[0114]

[0115] From the results in Table 10, it can be seen that the whitening and anti-aging creams prepared in Examples 5-7 of the present invention have significantly decreased melanin after 7 days, 14 days, 21 days and 28 days of use test, and as the content of the whitening and anti-aging composition increases, the decrease in melanin increases, and the percentage of melanin decrease in Example 7 is the largest.

[0116] The effect is the best, but the amount of the whitening and anti-aging composition added in Example 6 is less than that in Example 7. The melanin reduction percentage in Example 7 is not significantly improved compared with that in Example 6. The addition ratio of the whitening and anti-aging composition in Example 6 is more practical and has a higher cost-effectiveness.

[0117] In Comparative Example 10, due to the lack of whitening ingredients, the whitening effect of the obtained cream was significantly reduced (the percentage of melanin reduction was not obvious); in Comparative Example 11, the conventional niacinamide whitening ingredient was replaced, but the anti-aging ingredient was lacking, and the percentage of melanin reduction in the obtained cream was significantly lower than that in Example 6, indicating that the anti-aging component used in the present invention can not only exert its own anti-aging effect, but also promote the whitening effect of the specific whitening component, that is, there is a synergistic effect between the whitening component and the anti-aging component; in Comparative Example 12, the commercially available whitening and anti-aging ingredients were replaced, but the percentage of melanin reduction was still significantly lower than that in Example 6, and the same effect could not be achieved by replacing them with existing whitening ingredients or anti-aging ingredients that are recognized to have better effects, indicating that there is an obvious synergistic effect between the specific whitening and anti-aging components used in the composition of the present invention, which can exert their own whitening or anti-aging effects while synergistically promoting the anti-aging or whitening effects of other components, thereby maximizing the whitening and anti-aging effects of the composition, and each component is indispensable.

[0118] Finally, it should be noted that the above embodiments are only used to illustrate the technical solution of the present invention rather than to limit the scope of protection of the present invention. Although the present invention has been described in detail with reference to the preferred embodiments, those skilled in the art should understand that the technical solution of the present invention can be modified or replaced by equivalents without departing from the essence and scope of the technical solution of the present invention.
