Abstract
Description
Technical Field
[0001] The invention relates to the field of a decoction and extraction method of a Mongolian medicine Xanthoceras sorbifolia-4-flavor soup, and in particular to a decoction and extraction method of a Xanthoceras sorbifolia-4-flavor soup. Background Art
[0002] The Mongolian medicine Singuanmu-4 Decoction is recorded in the ancient Mongolian medicine book "Four Medical Classics" (subsequent volume), "the decoction of Singuanmu, Terminalia chebula, Gentiana macrophylla, and Phellodendron chinense can treat diseases caused by Xie Ri Wu Su fever." It is also recorded in the works "Four Ganlu" and "Gao Shige Meilin Fang", and is mainly used to treat various Xie Ri Wu Su diseases, rheumatoid diseases, rheumatism, and has the proportion and method of taking prescription drugs.
[0003] Decoctions in Chinese and Mongolian medicine are the most widely used traditional dosage forms in clinical practice. Improper operation of the decoction and extraction process will lead to unstable clinical efficacy. Mongolian medicine decoctions are crushed into coarse powder, then sieved, and all the crushed coarse powder is evenly mixed through an 80-mesh sieve to form a powder; generally, the dosage of Mongolian medicine decoctions is small each time the medicine is taken, and the patients generally take 10 ml-100 ml of water decoctions; Mongolian medicine decoctions are generally decocted once. There are two common decoctions for Mongolian patent medicine: decoction soup and soaking soup. Decoction soup is to crush 3g-5g of medicine into coarse powder according to the decoction preparation requirements, put it into 250mL of water or the required liquid, decocted on a slow fire, and evaporate 2 / 3 of the liquid, such as decoction of seven treasures soup. Soaking soup is to put 3g-5g of medicine into 200L of warm boiled water or the required liquid, soak for 8h-10h, remove the residue and take the juice, such as making three-flavor black alum soup.
[0004] At present, no relevant research on this prescription has been found through literature review, and there is no relevant report on how to determine the content of each component. In order to study the quality standard in the future, a method that can solve the ambiguity in the content determination of Mongolian medicine Sendeng-4-flavor soup is urgently needed. Summary of the invention
[0005] The invention aims to provide a method for decocting and extracting Xanthoceras sorbifolia-4-flavor soup.
[0006] The present invention is achieved through the following technical solutions:
[0007] The present invention relates to a decoction and extraction method of Xanthoceras sorbifolia-4-flavor decoction, comprising the following steps: taking a single factor investigation, selecting decoction time, water addition amount and soaking time as factors; taking dry extract yield and gallic acid, gentiopicroside, epicatechin, dihydromyricetin, ellagic acid and berberine contents as indicators, establishing an L9 (34) orthogonal experiment, determining and preparing the content and dry extract rate of a test solution, and obtaining the optimal process conditions as follows: decoction time of 35 minutes, water addition amount of 40 times, and soaking time of 30 minutes; the specific steps are as follows:
[0008] Step 1, preparation of reference solution
[0009] Accurately weigh the reference substance: gentiopicroside 0.79 mg, placed in a 10 mL brown volumetric bottle, gallic acid 0.98 mg, epicatechin 0.75 mg, dihydromyricetin 0.95 mg, ellagic acid 0.66 mg, berberine 0.61 mg, respectively placed in a 5 mL brown volumetric bottle, add methanol to dissolve and dilute to the scale, shake well, and prepare a stock solution;

[0030] Weigh 5g of the powder of Crownwood-4 Decoction, put it in a decoction pot, decoct once, soak for 20min, decoct for 35min, accurately add water at 40, 60, 80, and 100 times, filter with a 200-mesh sieve, collect the filtrate, and determine the indicators and dry paste rate according to the above method; the results show that the increase in the amount of water added is accompanied by a decrease in the content of each indicator and the dry paste rate, and there is no obvious trend when the amount is 80 times, so the highest amount of water is abandoned, and 40 times, 60 times, and 80 times are selected for subsequent experiments;
[0031] Step 12, choose the soaking time
[0032] Weigh 5g of the powder of Crownwood-4 Decoction, put it in a decoction pot, accurately add 60 times the amount of water, decoct once, the decoction time is 20min, the soaking time is 0min, 10min, 20min, 30min respectively, filter with a 200-mesh sieve, collect the filtrate, and determine the various indicators and dry paste rate according to the above method; the results show that the increase of soaking time increases the content of various indicators and the dry paste rate, so the minimum decoction time is abandoned, and 10min, 20min, and 30min are selected for subsequent experiments;
[0033] Step 13, orthogonal experiment
[0034] A single factor study was conducted, and decoction time (A), water addition (B), and soaking time (C) were selected as factors; the dry extract yield and the contents of gallic acid, gentiopicroside, epicatechin, dihydromyricetin, ellagic acid, and berberine were used as indicators to establish an L9 (34) orthogonal experiment. The content and dry extract yield of the test solution were determined and prepared according to the above method; the results showed that the order of influence of each factor on the comprehensive index component was A>B>C, that is, decoction time>water addition>soaking time; the order of influence on the dry extract yield was B>A>C, that is, water addition>decoction time>soaking time; each factor had no significant effect (P>0.05); considering the cost saving and extraction effectiveness, the optimal process conditions were a decoction time of 35 minutes, a water addition of 40 times, and a soaking time of 30 minutes.
[0035] Step 14: Verification test
[0036] Weigh 3 portions of medicinal materials, select the best process optimization conditions obtained in step 13 above for decoction, and measure various indicators and dry paste rate according to the above method. The results show that the content of various indicator components and dry paste rate are relatively stable, and the optimization process is reasonable and feasible.
[0037] The present invention has the following advantages:
[0038] (1) This study used high performance liquid chromatography (HPLC) and L9 (34) orthogonal experiments to determine the contents of gallic acid, epicatechin, dihydromyricetin, gentiopicroside, ellagic acid and berberine in the Mongolian medicine Sendeng-4-flavor soup, and studied the decoction and extraction process. This method is simple and rapid, and solves the scientific problem of unclear content determination of Mongolian medicine Sendeng-4-flavor soup, laying a foundation for its further content determination and quality control.
[0039] (2) The present invention uses high performance liquid chromatography to obtain six index components from the four-flavor decoction, and then classifies them with the four single herbs in the prescription. The identification results show that gallic acid and ellagic acid are derived from Terminalia chebula; epicatechin and dihydromyricetin are derived from the clary sibiricum; gentiopicroside is derived from Gentiana macrophylla; and berberine is derived from Phellodendron amurense. The above six active ingredients may become markers when determining the content of the four-flavor decoction, which will provide new development concepts for the formulation of the quality standards of the prescription or the development of innovative drugs. [0081] (5) Determination of dry paste yield
[0082] 25 mL of the decoction extract is accurately measured and placed in an evaporating dish (M1) of constant mass, then dried in a drying oven at 105°C to constant weight, placed in a desiccator to cool, and accurately weighed (M2). Dry paste yield = (M2-M1) / m, where m is the total mass of the medicinal material in the sample to be tested.
[0083] (6) Single factor study
[0084] A single factor investigation was conducted using the decoction extraction method (decoction time, amount of water added, soaking time), dry paste yield, and the contents of gallic acid, gentiopicroside, epicatechin, dihydromyricetin, ellagic acid, and berberine as indicators.
[0085] (6.1) Cooking time
[0086] Weigh 5g of the powder of Crownwood-4 Decoction, put it in a decoction pot, add 60 times the amount of water, decoct once, soak for 20 minutes, and the decoction time is 5 minutes, 15 minutes, 25 minutes, and 35 minutes respectively. Filter with a 200-mesh sieve, collect the filtrate, and determine the various indicators and dry paste rate according to the above method. The results show that the increase in decoction time increases the content of various indicators and the dry paste rate, so the minimum decoction time is abandoned, and 15 minutes, 25 minutes, and 35 minutes are selected for subsequent experiments.
[0087] (6.2) Amount of water added
[0088] Weigh 5g of the powder of Crownwood-4 flavor soup, put it in a decoction pot, decoct once, soak for 20min, decoct for 35min, accurately add water at 40, 60, 80, and 100 times, filter with a 200-mesh sieve, collect the filtrate, and determine the indicators and dry paste rate according to the above method. The results show that the increase in the amount of water added reduces the content of each indicator and the dry paste rate, and there is no obvious trend when it reaches 80 times, so the highest amount of water is abandoned, and 40 times, 60 times, and 80 times are selected for subsequent experiments.
[0089] (6.3) Soaking time
[0090] Weigh 5g of the powder of the 4-flavor soup of the crown wood, put it in a decoction pot, add 60 times the amount of water, decoct once, decoction time 20min, soak time 0min, 10min, 20min, 30min, filter with a 200-mesh sieve, collect the filtrate, and determine the various indicators and dry paste rate according to the above method. The results show that the increase in soaking time increases the content of various indicators and the dry paste rate, so the minimum decoction time is abandoned, and 10min, 20min, and 30min are selected for subsequent experiments.
[0091] (7) Orthogonal experiment
[0092] The results of single factor investigation were adopted, and decoction time (A), water addition (B), and soaking time (C) were selected as the investigation factors. The dry paste yield and the contents of gallic acid, gentiopicroside, epicatechin, dihydromyricetin, ellagic acid, and berberine were used as indicators to establish the L9 (34) orthogonal experiment, as shown in Table 5. The content and dry paste rate of the test solution were determined and prepared according to the above method, as shown in Table 6 and Table 7. From the results in Table 7, it can be concluded that the order of influence of each factor on the comprehensive index component is A>B>C, that is, decoction time>water addition>soaking time; the order of influence of dry paste rate is B>A>C, that is, water addition>decoction time>soaking time; each factor has no significant effect (P>0.05). Considering the cost saving and extraction effectiveness, the optimal process conditions are selected as decoction time of 35min, water addition of 40 times, and soaking time of 30min.

[0105] The method involved in the present invention uses the extract rate and the content of each effective ingredient as indicators for content determination. Through single factor investigation and orthogonal test results, it is concluded that the optimal process conditions of the 4-flavor decoction of Senna sorbifolia are 35 minutes of decoction time, 40 times of water addition, and 30 minutes of soaking time. The decoction uses gallic acid, gentiopicroside, epicatechin, dihydromyricetin, ellagic acid and berberine as indicators; in addition, the present invention also uses high performance liquid chromatography to obtain 6 index components from the 4-flavor decoction of Senna sorbifolia respectively and attributes them to the 4 single herbs of the decoction. The identification results show that gallic acid and ellagic acid are derived from Terminalia chebula; epicatechin and dihydromyricetin are derived from Senna sorbifolia; gentiopicroside is derived from Gentiana macrophylla; and berberine is derived from Phellodendron chinense. The above 6 effective ingredients may become markers when determining the content of the Senna sorbifolia sorbifolia 4-flavor decoction, which will provide new development concepts when studying the formulation of the quality standards of the decoction or developing innovative drugs.
