Abstract
Description
Technical Field
[0001] The invention relates to the field of chemical medicine, and in particular to a method for controlling the particle size of a drug. Background Art
[0002] Oral administration is currently the most convenient and commonly used route of administration. Oral bioavailability depends on factors such as water solubility, permeability, dissolution rate and first-pass metabolism, among which the most common reasons are poor solubility and low permeability. Among the newly developed chemical drugs, about 40% have poor water solubility, resulting in slow drug absorption and extremely poor bioavailability. Low water solubility is the main problem encountered in the development of new chemical entity formulations and generic drug development. The solubility of a drug is usually intrinsically linked to the particle size of the drug. As the particle size decreases, the specific surface area increases, resulting in greater interaction with the solvent to achieve the purpose of increasing solubility.
[0003] In addition, the particle size distribution of APIs not only affects its own transportation, packaging and storage, but also has a great impact on the entire preparation process, such as premixing / mixing, granulation, drying, whole grain, coating, packaging and tableting. Its impact on preparations is mainly in the following aspects: a) Impact on product performance (such as solubility, bioavailability, content uniformity, stability, etc.); b) Impact on manufacturability (such as fluidity, mixing uniformity, compressibility, etc.), ultimately affecting the safety, efficacy and quality of the drug. When the particle size distribution between product batches is unstable or the particle size distribution is bimodal or multimodal, it will bring some adverse effects to the preparation process, such as stratification and poor mixing uniformity during direct tableting.
[0004] At present, the control of the particle size distribution and size of the API can generally be achieved through two ways. The first way is to control it during the crystallization process, for example, by adjusting the process parameters in the crystallization process (such as cooling rate, stirring rate, anti-solvent drop rate, etc.) to control the particle size distribution of the API. The second way is to micronize the API after the API has been prepared and before the formulation process to reduce the particle size of the API and (or) make the particle size of the API uniform.
[0005] The method of controlling the particle size distribution of APIs through route 1 is not universal, and it is necessary to develop a corresponding crystallization process, i.e., a particle size control process, according to the characteristics of the drug itself. In the crystallization process, adding seed crystals to control the particle size and distribution of the finished product is a common means. Crystallization is a complex process, and different physical and chemical environments will affect the crystallization process, and the influence of the amplification effect needs to be considered during the amplification process. Therefore, controlling the particle size of the finished product by adding seed crystals has high requirements for the size of the seed crystals themselves and the uniformity of the particles. Especially for poorly soluble drugs, the particle size of the API needs to be controlled at a relatively small level, and the API obtained only by conventional crystallization may not meet this requirement. Therefore, it is necessary to add finer seed crystals to obtain API particles with a sufficiently small particle size, uniform distribution, and meeting the requirements of the formulation process.
[0006] When the subsequent preparation process has strict requirements on the particle size of the API, it is necessary to strictly control the crystallization parameters, which brings inconvenience to the actual production and reduces the operability. At this time, controlling the particle size of the API through the second approach is a more direct method, and this method is also a commonly used method in the current pharmaceutical industry. Traditional micronization methods, such as airflow crushing and grinding, rely on mechanical force to crush drug particles. However, the mechanical force during crushing usually exerts a large amount of physical stress on the drug product, thereby destroying its crystal structure, resulting in risks such as product crystal form transformation, decreased crystallinity, or degradation impurities; when dealing with heat-sensitive or unstable active compounds, the temperature increase during the crushing process is also a problem worthy of attention; airflow crushing and grinding can also cause static electricity in the material, which is not conducive to the packaging, transportation and storage of the product. Unconventional particle size control methods such as ultrasound and high shear also have certain limitations. The ultrasonic crushing method is difficult to industrialize and has low production capacity; although high shear can reduce the particle size of the crystal to a certain extent, it is often uneven, resulting in a wide particle size distribution, and the amplification effect is obvious. In actual industrial production, it is likely that the shear crushing efficiency of the small test cannot be achieved.
[0007] Therefore, there is an urgent need to develop a new, stable, and industrializable powder control method for application in the pharmaceutical field, which can take into account the particle size, physicochemical stability, thermal sensitivity, and yield of the raw materials and can be industrialized. [0102] After long-term and in-depth research, the inventor unexpectedly discovered a stable drug particle size control method with uniform product particle size distribution, simple and easy operation, and suitable for industrial production. The raw material drug prepared by the method of the present invention has a narrow particle size distribution, good fluidity and compressibility, and is suitable for different preparation processes. The conventional method for controlling the particle size of raw materials solves the problems of uneven particle size distribution, insufficient particle size, static electricity, caking, and poor physical and chemical stability of the product. On this basis, the inventor completed the present invention.
[0103] Terminology
[0104] Unless otherwise specified herein, each abbreviation has a common meaning understood by those skilled in the art.
[0105] As used herein, unless otherwise specified, the terms "API A" and "API B" refer to the amorphous form (form) and / or various crystalline forms (including the various crystalline forms and amorphous forms mentioned herein, and the crystalline forms or amorphous forms (forms) mentioned in various published or unpublished documents or patents) of API A and API B.
[0106] High-pressure homogenization method for producing raw materials
[0107] As used herein, "high pressure homogenization" or "high pressure homogenization method" are used interchangeably and refer to a method of treating a liquid dispersion containing a drug substance with a high pressure homogenizer to obtain a drug substance with a desired particle size.
[0108] High-pressure homogenizers are mainly used in the biological, pharmaceutical, food, chemical and other industries for cell disruption, beverage homogenization, fine chemicals, and the preparation of liposomes, fat emulsions, nanosuspensions, microemulsions, lipid microspheres, emulsions, dairy products, large infusions, dyes, solar panel coatings, and conductive coatings. High-pressure homogenizers can make materials in a suspended state flow through a cavity with a special internal structure (high-pressure homogenization cavity) at high speed under ultra-high pressure, causing the material to undergo structural property changes and ultimately achieve a homogenization effect.
[0109] The inventors unexpectedly discovered that after preliminary crystallization, high-pressure homogenization treatment of the feed liquid can change the morphology of the material, and further crystallization of the treated material can obtain a raw material drug of the desired particle size. Since the method changes the morphology of the material through physical treatment, it is not affected by the type of raw material drug and can be used for the production of any type of raw material drug.
