Enantiomers can be separated by converting the enantiomeric mixture into a diastereomeric mixture by reaction with an appropriate optically active compound (e.g., a chiral auxiliary such as a chiral alcohol or Mosher's acid chloride), separating the diastereomers and converting (e.g., hydrolyzing) each diastereomer to the corresponding pure enantiomer. Enantiomers can also be separated by using a chiral HPLC column.
[0062] By resolving the racemic mixture using methods such as the use of an optically active resolving agent to form diastereoisomers, a single stereoisomer, such as a substantially pure enantiomer, can be obtained. The racemic mixture of the chiral compounds of the invention can be separated and isolated by any suitable method, including: (1) forming ionic diastereomeric salts with chiral compounds and separating by fractional crystallization or other methods, (2) forming diastereomeric compounds with chiral derivatizing agents, separating the diastereoisomers and converting to pure stereoisomers, and (3) separating the substantially pure or enriched stereoisomers directly under chiral conditions.

[0078] Useful pharmaceutical dosage forms for administering the subject compounds disclosed herein and stereoisomers thereof and pharmaceutically acceptable salts thereof include, but are not limited to, hard and soft gelatin capsules, tablets, parenteral injections and oral suspensions.
[0079] The dosage administered will depend on factors such as the age, health and weight of the recipient, the extent of the disease, the type of concurrent treatment (if any), the frequency of treatment, and the nature of the desired effect. In general, the daily dose of the active ingredient may vary, for example, 0.1-2000 mg per day. For example, 10-500 mg once or more per day may be effective to obtain the desired result.
[0080] In certain embodiments, a large number of unit capsules can be prepared by filling standard two-piece hard gelatin capsules with, for example, 100 mg of a powdered form of a subject compound disclosed herein and stereoisomers thereof and pharmaceutically acceptable salts thereof, 150 mg of lactose, 50 mg of cellulose and 6 mg of magnesium stearate.

[0085] When the compound, its stereoisomers and pharmaceutically acceptable salts thereof are administered in steps or in combination with at least one other therapeutic agent, the same dosage form can generally be used. When the drugs are administered in physical combination, the dosage form and administration route should be selected based on the compatibility of the combined drugs. Therefore, the term co-administration should be understood to include simultaneous or sequential administration of at least two medicaments, or alternatively as a fixed dose combination of at least two active ingredients.
[0086] The compounds disclosed herein, their stereoisomers, and pharmaceutically acceptable salts thereof can be administered as the sole active ingredient or in combination with at least one second active ingredient.
[0087] The subject compound is incorporated into a pharmaceutical composition or formulation. The composition will contain a pharmaceutically acceptable diluent and / or carrier, i.e., a diluent or carrier that is physiologically compatible and substantially free of pathogenic impurities. Suitable excipients or carriers and methods for preparing compositions that can be administered are known or apparent to those skilled in the art and are described in more detail in publications such as Remington's Pharmaceutical Science, Mack Publishing Co, NJ (1991). The composition can also be in the form of controlled release or sustained release compositions known in the art. For many applications, the subject compound is administered in the morning / daytime and discontinued at night.
[0088] The subject compound can be used per se, or in the form of a pharmaceutically acceptable salt thereof such as a hydrochloride, hydrobromide, acetate, sulfate, citrate, carbonate, trifluoroacetate, etc. When the compound contains a relatively acidic functional group, a salt can be obtained by adding a desired base in pure or suitable inert solvents. Examples of pharmaceutically acceptable base addition salts include sodium salts, potassium salts, calcium salts, ammonium salts, organic amino salts, or magnesium salts, etc. When the compound contains a relatively basic functional group, a salt can be obtained by adding a desired acid in pure or suitable inert solvents. Examples of pharmaceutically acceptable acid addition salts include those derived from inorganic acids such as hydrochloric acid, hydrobromic acid, nitric acid, carbonic acid, monohydrogencarbonic acid, phosphoric acid, monohydrogenphosphoric acid, dihydrogenphosphoric acid, sulfuric acid, monohydrogensulfuric acid, hydroiodic acid or phosphorous acid, and those derived from relatively nontoxic organic acids such as acetic acid, propionic acid, isobutyric acid, maleic acid, malonic acid, benzoic acid, succinic acid, suberic acid, fumaric acid, lactic acid, mandelic acid, phthalic acid, benzenesulfonic acid, p-toluenesulfonic acid, citric acid, tartaric acid, methanesulfonic acid, etc. Also included are amino acid salts such as arginine salts, and salts of organic acids such as glucuronic acid or galacturonic acid, etc. (see, e.g., Berge et al., "Pharmaceutical Salts", Journal of Pharmaceutical Science, 1977, 66, 1-19).
[0089] By contacting the salt with a base or acid and isolating the parent compound in a conventional manner, the neutral form of the compound can be regenerated. The parent form of the compound differs from the various salt forms in certain physical properties (such as solubility in polar solvents), but for purposes of the present invention, the salt is equivalent to the parent form of the compound in other respects.
[0090] In addition to salt forms, the present invention provides compounds in the form of prodrugs. Prodrugs of compounds described herein are those compounds that are easily subjected to chemical changes under physiological conditions to provide compounds of the present invention. In addition, prodrugs can be converted into compounds of the present invention by chemical or biochemical methods in an ex vivo environment. For example, when placed in a transdermal patch reservoir together with a suitable enzyme or chemical reagent, prodrugs can be slowly converted into compounds of the present invention. Prodrugs are often useful because, in some cases, they may be easier to administer than the parent drug. For example, their bioavailability by oral administration may be higher than that of the parent drug. Prodrugs may also have improved solubility in pharmacological compositions compared to the parent drug. Various prodrug derivatives are known in the art, such as those that rely on hydrolytic cleavage or oxidative activation of prodrugs. A non-limiting example of a prodrug is a compound of the present invention that is administered as an ester ("prodrug") but then metabolically hydrolyzed into a carboxylic acid active entity.
[0091] Some compounds of the present invention may exist in unsolvated form and solvated form (including hydrated form). Generally speaking, the solvated form is equivalent to the unsolvated form, and is intended to be included within the scope of the present invention. Some compounds of the present invention may exist in multiple crystalline forms or amorphous forms. Generally speaking, all physical forms are equivalent for the purposes encompassed by the present invention, and are intended to be within the scope of the present invention.
[0092] Certain of the subject compounds possess asymmetric carbon atoms (optical centers) or double bonds; the racemates, diastereomers, geometric isomers and various isomers are intended to be encompassed within the scope of the present invention.
[0093] The compounds of the present invention may also contain unnatural proportions of atomic isotopes at one or more atoms constituting such compounds, such as deuterium (e.g., -CD3, CD2H or CDH2) in place of a methyl group. For example, the compounds may be substituted with radioactive isotopes such as tritium ( 3 H), iodine-125 ( 125 I) or carbon-14 ( 14 C) Radiolabeling. All isotopic variations of the compounds of the present invention, whether radioactive or not, are intended to be encompassed within the scope of the present invention.
[0094] The compounds are typically administered in a "therapeutically effective amount," i.e., an amount of the subject compound that will elicit the biological or medical response of a tissue, system, animal or human being that is being sought by the researcher, veterinarian, physician or other clinician. The term "therapeutically effective amount" includes an amount of the compound that, when administered, is sufficient to prevent the development of or alleviate to some extent one or more symptoms of the condition or disorder being treated. The therapeutically effective amount will vary depending on the compound, the disease and its severity, and the age, weight, etc., of the mammal to be treated.
[0095] Contact is usually achieved by administering to the subject an effective amount of one or more compounds of Formula I (supra) (including the various embodiments described above). Administration is usually adjusted to achieve a therapeutic dose of about 0.1-50, preferably 0.5-10, more preferably 1-10 mg / kg, but the optimal dose is compound-specific and is usually determined empirically for each compound.
[0096] The term "unit dosage form" refers to a physically discrete unit suitable as a unit dose for human subjects and other mammals, each unit containing a predetermined amount of active substance calculated to produce the desired therapeutic effect in combination with a suitable pharmaceutical excipient. Typical unit dosage forms include pre-filled, pre-measured ampoules or syringes of liquid compositions, or pills, tablets, capsules, lozenges, etc. in the case of solid compositions. In such compositions, the simulant is usually a minor component (about 0.1 to about 50% by weight or preferably about 1 to about 40% by weight), and the remainder is various vehicles or carriers and processing aids that help form the desired form of administration. The unit dose formulation is preferably about 5, 10, 25, 50, 100, 250, 500 or 1,000 mg per unit. In a specific embodiment, the unit dosage form is packaged in a multi-pack suitable for sequential use, such as a blister pack containing at least 6, 9 or 12 sheets of unit dosage forms.
[0097] The subject compositions may also be co-formulated and / or co-administered with different compounds to treat applicable indications, inhibit ferroptosis activity, or modulate or inhibit diseases associated with dysregulation of ferroptosis, such as neuropathy, ischemia-reperfusion injury, acute renal failure, and cancer. In embodiments, applicable indications include cancer, neuropathy and neurodegenerative diseases of the central or peripheral nervous system, muscular dystrophy, ischemia and ischemia-reperfusion injury, kidney disease and failure, degenerative arthritis, retinal necrosis, heart disease, liver, gastrointestinal or pancreatic disease, avascular necrosis, diabetes, cancer-chemo / radiotherapy-induced cell death and poisoning.
