Abstract
Description
Technical Field
[0001] The invention belongs to the technical field of traditional Chinese medicines, and particularly relates to an anti-Parkinson's disease warming yang and relieving depression traditional Chinese medicine composition and application thereof. Background Art
[0002] With the aging of the population, the burden of Parkinson's disease (PD) is expected to increase significantly in the next few decades, becoming the second most common neurodegenerative disease, and the prevalence is on the rise. Although modern medicine has made some progress in the treatment of Parkinson's disease, the overall course of the disease has not been significantly shortened, and the survival rate within ten years of the disease is about 50%. Parkinson's disease is generally divided into four types according to the clinical symptom type: rigid type, hypokinetic type, tremor type, with or without dementia. Rigidity and hypokinetic type account for almost half of the total population, and the overall symptoms are stiff limbs, slow movements, shortness of breath, cold limbs, weak pulse, pale face, pale lips, loose stools, clear and long urine, or even depression, insomnia, etc., and it is different from other types of Parkinson's disease. It develops relatively quickly, has a relatively long course of disease, and has a relatively early onset.
[0003] The motor symptoms of Parkinson's disease are similar to the "tremor syndrome" in traditional Chinese medicine. Ancient books often use words such as "tremor, falling, shaking, trembling, and cramps" to describe its manifestations. "Suwen·Wuchang Zhengda Lun" also describes "the disease is shaking", "falling dizzy and convulsing", "trembling and trembling". Parkinson's non-motor symptoms (NMS) belong to the categories of "depression syndrome", "insomnia", and "constipation", including olfactory disorders, sleep disorders, constipation, depression, anxiety and other symptoms. Depression is a common non-motor disorder symptom of PD, which appears in all stages of the course of Parkinson's disease, and even before the appearance of motor symptoms. Patients with Parkinson's disease depression (PDD) show persistent low mood, difficulty concentrating, loss of interest in work and life, sleep disorders, indifference, pessimism, lack of humor, suicidal thoughts, anxiety, and sensitivity.
[0004] Studies have shown that traditional Chinese medicine has unique advantages in prolonging survival, improving quality of life, alleviating related adverse reactions and reducing recurrence and metastasis. Summary of the invention
[0005] The purpose of the present invention is to provide a traditional Chinese medicine composition for warming yang and relieving depression for resisting Parkinson's disease.
[0006] Another object of the present invention is to provide an application of the above-mentioned Chinese medicine composition.
[0007] According to clinical experience and theoretical research, the present invention finds that the basic pathogenesis of PDD is that the spleen and kidney have congenital and acquired yang deficiency, which is weak in warming, ascending and pushing, resulting in the lack of nourishment of the tendons, flesh and bones of the limbs, the lack of nourishment of the brain essence, the lack of yang and the stagnation of liver qi, the loss of control of the limbs and tremors, and the manifestation of various pathological manifestations such as mental emotions. Yang is closely related to the functioning of the internal organs and the driving of the spirit and body. The spleen governs the muscles of the limbs, the liver governs the tendons and veins, and the kidney governs the bones. The limbs are composed of bones, flesh and tendons. Although the fullness of the three belongs to the yin essence, the yang "fire" is needed to transport and nourish. The use of the limbs depends on the active nature of yang. The deficiency of spleen and kidney yang and the stagnation of liver qi affect each other, and together cause the motor and non-motor pathological symptoms of Parkinson's disease.
[0008] Therefore, the present invention provides a Chinese medicine composition, including raw aconite, roasted liquorice, dried ginger, bupleurum, peony root, immature bitter orange and ligusticum chuanxiong, which aims to improve the clinical symptoms of Parkinson's disease holistically from the perspective of warming and replenishing yang qi and relieving qi movement.
[0009] The present invention takes "Yang deficiency and liver depression" as the basic pathogenesis of Parkinson's disease as the "starting point", and refers to the research on Parkinson's disease pathology, and proposes to treat Parkinson's disease from the perspective of warming and tonifying the "Yang Qi of the spleen and kidney" and relieving the "stagnation of liver qi", with "warming Yang, dispersing cold, and relieving depression" as the main treatment method, and "Sini Decoction combined with Sini Powder" as the basic prescription, focusing on the "Sini Decoction" for warming Yang, treating both the symptoms and the root causes, and referring to the mechanisms for synergistic effect; at the same time, taking "commonalities with human diseases" as the "starting point", animal experiments are attached, and the feasibility, effectiveness and scientificity of the present invention are proved by in-depth basic research.
[0010] According to a specific embodiment of the present invention, the anti-Parkinson's compound Chinese medicine composition is prepared from the following raw materials in parts by weight: 5-15 parts of raw aconite, 10-120 parts of roasted licorice, 5-50 parts of dried ginger, 5-50 parts of bupleurum, 10-80 parts of peony, 5-50 parts of immature bitter orange, and 2-12 parts of ligusticum chuanxiong.
[0011] According to a specific embodiment of the present invention, a method for preparing a traditional Chinese medicine composition for warming yang and relieving depression against Parkinson's disease comprises the following steps: According to the above proportion of Chinese medicine, water is added to soak and then the first decoction is performed; after filtering, water is added to the residue and the second decoction is performed; The decoctions obtained from the two decoctions were combined.
[0012] Furthermore, according to the method for preparing the Chinese medicine composition of the specific embodiment of the present invention, the combined decoction is subjected to static precipitation, and then concentrated by decoction in a water bath to obtain a concentrated medicine.
[0013] The method for preparing the Chinese medicine composition according to a specific embodiment of the present invention comprises the following steps: According to the above-mentioned proportion of Chinese medicine, add 10 times the amount of water of the total weight of Chinese medicine for the first decoction, soak for 30 minutes, boil over high heat and then decoct for 90 minutes, filter through gauze; add 8 times the amount of water for the second decoction, boil and then decoct for 80 minutes, filter through gauze, combine the two filtrates, let stand and settle for 12 hours, take the supernatant, combine the two decoctions, decoct and concentrate in a water bath to obtain concentrated medicine.
[0014] Preferably, the traditional Chinese medicine composition is made of the following raw medicines in parts by weight: 10-15 parts of raw aconite, 20-80 parts of roasted licorice, 10-30 parts of dried ginger, 10-30 parts of bupleurum, 20-60 parts of peony, 10-30 parts of immature bitter orange, and 4-10 parts of ligusticum chuanxiong.
[0015] More preferably, the Chinese medicine composition is made of the following raw medicines in parts by weight: 15 parts of raw aconite, 30 parts of roasted licorice, 24 parts of dried ginger, 15 parts of bupleurum, 30 parts of peony, 15 parts of immature bitter orange, and 9 parts of ligusticum chuanxiong.
[0016] The Parkinson's disease treatment principle and treatment method of the present invention: In clinical practice, the treatment of Parkinson's disease should focus on the basic pathogenesis of "Yang deficiency and liver depression", and first warm and replenish the "spleen and kidney yang qi", while taking into account "soothing the liver and regulating qi", and "warming yang, dispersing cold, and relieving depression" should be carried out throughout the treatment process. At the same time, the yin essence should not be ignored, and both the symptoms and the root causes should be treated, with the mechanisms interrelated and synergistic.
[0017] Prescription composition and explanation (1) Ingredients: Aconite root, dried ginger, roasted licorice root, bupleurum root, peony root, bitter orange, and ligusticum chuanxiong.
[0018] (2) Explanation of the prescription: Aconite, dried ginger and roasted licorice are used heavily. Aconite is a ball of raging fire, which replenishes the body's yang energy, strengthens the kidney and replenishes the essence. The pungent and warming properties of dried ginger can disperse and warm the earth of the middle burner, and the sweetness of licorice can suppress fire with earth. The three herbs are used together to replenish the yang of the spleen and kidney and warm the yang energy of the whole body. Peony, Zhishi and Chuanxiong are also used. Bupleurum is used to dredge the meridians and relieve the qi mechanism, while Zhishi is pungent and opens and disperses the knots. The two are used together to lift and lower. White peony and licorice can restrain yin and relax the tendons, taking into account both the body and the function. Chuanxiong is used to use its "walking" function to regulate the qi and blood inside and outside. Although there are two prescriptions, they are actually three. Among them, Sini Tang contains aconite, dried ginger, and roasted licorice to replenish yang; Sini San contains bupleurum, peony, immature bitter orange, and roasted licorice to promote the flow of qi; the first two prescriptions contain peony and licorice, and have the meaning of Peony and Licorice Decoction, which are used to soften yin; together they have the effects of warming yang and relieving depression, softening tendons and promoting the flow of qi, and taking both physical and functional aspects into consideration.
[0019] (3) Pharmacological effects Raw aconite: enters the heart, kidney and spleen meridians. Pungent, sweet, hot and toxic. It can restore yang and rescue adverse conditions, replenish fire and boost yang, dispel cold and relieve pain. Modern pharmacological research has confirmed that aconite contains more than 100 chemical components, which are divided into alkaloids and non-alkaloids. The main biologically active components are diterpene alkaloids, flavonoids, steroidal saponins, etc., which have the effects of protecting the heart, regulating immunity, anti-tumor, protecting nerve damage, and anti-depression. Aconite plays a role in the signal transduction of the neuronal survival-related pathway MAPK1 / MAPK3. It can promote the expression of GRIN1 and MAPK1 genes and inhibit nerve damage by activating the noradrenergic system. Aconite water extract (norphine) can increase norepinephrine, affect synaptic plasticity, and improve learning and cognitive abilities. Dried ginger: It enters the spleen, stomach, kidney, heart and lung meridians. It is hot in nature and pungent in taste; it has the functions of warming the middle and dispersing cold, restoring yang and unblocking meridians, and drying dampness and eliminating phlegm. The chemical components of dried ginger are mainly volatile oils, gingerols and diphenylheptanes. It has pharmacological effects such as analgesia, anti-inflammatory, anti-tumor, anti-pathogen, liver protection and gallbladder, anti-ulcer, and improvement of cardiovascular system. Dried ginger is pungent and hot, and contains pungent ingredients such as volatile oils, which can promote local blood circulation, protect wounds and promote healing. The water extract and volatile oil of dried ginger have the effect of inhibiting platelet aggregation and preventing thrombosis. Dried ginger extract has a biphasic regulatory effect on the body's immune function. Studies have found that the enhancement effect of dried ginger extract on cytokines is time-dependent, and its anti-inflammatory effect is related to the gingerols and diphenylheptanes it contains. 6-Gingerol relieves systemic and local inflammatory responses by regulating the balance of Th17 / Treg cells, and is effective in treating DSS-induced UC in mice. 10-Gingerol can inhibit the expression of pro-inflammatory genes by blocking the activation of NF-κB, thereby leading to a decrease in the levels of nitric oxide (NO), IL-1B, IL-6, and TNF-α.
[0020] Licorice root: sweet and flat in nature, it enters the heart, lung, spleen and stomach meridians, and has the effects of clearing away heat and detoxification, removing phlegm and relieving cough, tonifying the spleen and lungs, and relieving pain. The chemical components in licorice root are diverse, mainly including triterpenoid saponins, flavonoids and polysaccharides; licorice root has pharmacological effects such as anti-tumor, anti-depression, anti-inflammatory, anti-arrhythmic, liver protection, antioxidant and immunomodulatory. The flavonoids in licorice have antioxidant, anti-tumor, anti-depression, anti-inflammatory and antispasmodic effects, among which isoliquiritigenin has the strongest antispasmodic effect. Licorice root can effectively reduce the level of proinflammatory cytokines in the lipopolysaccharide-induced depression model of mice, reduce the expression of NF-KB / p-p65 and p-IKBα, and increase the content of brain-derived neurotrophic factor (BDNF) and tyrosine kinase receptor B (TrkB) in the hippocampus.
[0021] Bupleurum: It enters the liver and gallbladder meridians. It is bitter and slightly cold in nature. It "rises but does not fall, and is the yin in the yang". It has the effects of dispersing fever, reconciling the exterior and interior, relieving depression and regulating menstruation, and raising yang. Bupleurum saponin is the main component of Bupleurum. At present, more than 100 types of Bupleurum saponin components have been confirmed. Among them, Bupleurum saponin is a popular compound studied in recent years, which has anti-inflammatory, antiviral, antipyretic, analgesic, anti-epileptic, anti-tumor, and liver protection effects. Flavonoids are mainly found in the aboveground parts of Bupleurum plants. Kaempferol, isorhamnetin and quercetin are the three main types, which have the effects of protecting nerves, anti-oxidation, anti-depression, and inhibiting the central nervous system. The active ingredients contained in Bupleurum can improve nerve function, inhibit nerve cell death and protect nerves. Bupleurum total saponin can improve learning and memory disorders by increasing the expression of brain-derived neurotrophic factor and related pathways and promoting the occurrence of neural synapses. In addition, the active ingredients of Bupleurum can protect against ischemia-reperfusion brain injury by inhibiting low-density lipoprotein, promoting mitochondrial autophagy, increasing the levels of superoxide dismutase and glutathione in the hippocampus, inhibiting oxidative stress by passing through the blood-cerebrospinal fluid barrier, and protecting vascular endothelial cells.
[0022] Citrus aurantium: enters the spleen and stomach meridians. Bitter, pungent, sour, slightly cold. It has the functions of breaking up qi and eliminating stagnation, resolving phlegm and relieving lumps. Its chemical components are mainly flavonoids, volatile oils and alkaloids, etc., which have the functions of regulating the gastrointestinal tract, anti-oxidation and anti-inflammatory. The main antioxidant effect in Citrus aurantium is flavonoids, and anti-oxidation is one of the main mechanisms of action for the treatment of AD.
[0023] Peony: Enters the liver and spleen meridians. It is sour, bitter, and slightly cold. It has the effects of nourishing blood and regulating menstruation, restraining yin and stopping sweating, and calming liver yang. It is rich in monoterpenoid glycosides, triterpenoids, flavonoids, tannins, volatile oils, and phenolic acids. It has the effects of anti-inflammatory and liver protection, antihypertensive and analgesic, anti-oxidation, anti-depression, and immunomodulation. It can be used to treat cardiovascular and cerebrovascular diseases, neurodegenerative diseases, etc. Studies have shown that paeoniflorin can treat neurodegenerative diseases such as Parkinson's disease that are related to oxidative stress by blocking the mitochondrial pathway. In PC12 cells induced by 6-hydroxydopamine (6-OHDA), paeoniflorin protects PC12 cells from apoptosis by attenuating the translocation of nuclear factor kB (NF-kB), increasing the expression of bcl-2 in anti-apoptotic cells, reducing the expression of Bax in pro-apoptotic cells, and preventing the release of cytochrome C from mitochondria to the cytoplasm.
[0024] Chuanxiong: warm in nature, pungent in taste, and enters the liver meridian, gallbladder meridian, and pericardium meridian. It has the effects of promoting blood circulation and qi, dispelling wind and relieving pain. Its chemical components are mainly volatile oils, alkaloids, polysaccharides, etc., including phthalide and its dimers, alkaloids, organic acid phenols, polysaccharides, and compounds such as cerebrosides and ceramides; it has multi-faceted pharmacological activities on multiple systems such as the cardiovascular system, liver and kidney system, nervous system, and respiratory system, mainly manifested in analgesia, anti-inflammation, antioxidant ability, anti-tumor, anti-coagulation, anti-depression, anti-aging, anti-atherosclerosis, cell protection, and improvement of cardiac function.

[0049] Example 4 The Parkinson's disease combined with depression model in mice was established by intraperitoneal injection of MPTP, a Parkinson's disease modeling drug, followed by chronic unpredictable mild stress.
[0050] At a drug concentration of 30 mg / kg, 0.2 ml of MPTP was injected intraperitoneally into mice for 7 days. According to the reference "Chronic Unpredictable Mild Stress", including fasting, water deprivation, 24-hour continuous lighting, reversed light and dark, tilted empty cages, empty cages with water at the bottom, 2-3 stimulations were applied every day for 21 days.
[0051] In the treatment phase after modeling, the Western medicine treatment group was gavaged with 0.5 mg / kg of pramipexole, and the Chinese medicine treatment group was gavaged with the composition of the present invention, a high-dose group (8.97 mg / kg) and a low-dose group (4.48 mg / kg), for a total of 21 days. After the experiment, the effects of Chinese medicine treatment on anxiety, depression and movement disorders in mice were evaluated by sugar water preference test, open field test, rotating rod test and tremor paralysis scoring test.
[0052] Sucrose preference test (SPT) After the experimental mice were housed alone for 24 h, two bottles of 1% sucrose solution were placed in the cage for 12 h of adaptive drinking, followed by 1 bottle of 1% sucrose solution and 1 bottle of distilled water. After 12 h, the positions of sucrose solution and distilled water were swapped for 12 h. After adapting to drinking, the experimental mice were deprived of water for 12 h, and then 1 bottle of 1% sucrose solution and 1 bottle of distilled water were placed in the cage for them to drink for 12 h. Before and after the experiment, the weight of sucrose solution and distilled water were measured, and the sucrose consumption ratio was calculated using the following formula: Sugar water preference index = sucrose consumption / (sucrose consumption + water consumption). Open field test (OFT) After the mice were adapted to the laboratory environment for 1 h, they were placed in a 40 cm × 40 cm × 35 cm open field box, and the 20 × 20 cm area near the center of the box was set as the central area. The total movement distance of the mice within 5 min before the experiment, the number of times they entered the central area (central area entry times), and the time they stayed in the central area (central area residence time) were recorded. The open field box was cleaned with 75% alcohol before the next batch of mice were tested in the open field. The behavioral data of the mice were analyzed using TopScanTM2.0 software from Clever Sys Inc., USA.
[0053] Rotarod Test (RT) The rotarod test requires animals to maintain balance and move continuously on the rotating shaft. It is a widely used test for detecting motor coordination. The rotarod fatigue instrument consists of a rotating shaft and four independent compartments. Each rat is placed on the rotarod instrument, and the speed is accelerated from 5r to 40r within 1min until the rat falls. Each rat is trained 4 times and trained for 3 days before the experiment. In the formal rotarod test, the rat is placed on the rotarod instrument to adapt for 10s, and the recording begins when the rat can walk more steadily and maintain balance continuously. The speed of the rotarod instrument is accelerated from 5r to 40r within 1min, and the time the rat stays on the shaft is recorded as the latency (i.e., the time from the start of the shaft to the rat falling). If the rat has not fallen, the latency is recorded as the maximum value of 300s. After each test, the rat is put back in the cage to rest for 1min. The test is repeated 7 times in a row, and the maximum and minimum values are removed, and the average of the remaining 5 results is taken.
[0054] Paralysis tremor score (PTS) The scoring of mouse tremor paralysis is based on the phenotypic characteristics of mice, including hair (hair tufts), motor performance (walking, shaking, deflection), limb activity (arched back, tremor, tail tremor, nodding, rigidity), spontaneous behavior (swallowing, salivation), etc. The grade score is divided into 5 levels, 0 points means no adverse reactions, similar to normal mice (none); 1 point means the occurrence of hair tufts, arched backs, small tremors, etc., but does not affect the activity (occasionally); 2 points means frequent swallowing, limb tremors, tail tremors, hind limbs open, etc., and the activity is affected (intermittent); 3 points means persistent limb tremors, rigidity, salivation, etc., and cannot move normally (persistent); 4 points means persistent whole body paralysis and severe death (severe). The mice were transferred to an empty cage with a quiet environment and appropriate lighting. The observation cage was cleaned with alcohol every time. The observer did not carry irritating odors and did not make abnormal sounds. The observation process lasted 10 minutes, with the first 5 minutes being the adaptation phase and the last 5 minutes being the formal observation phase. Each animal was observed once by two observers, and if the results were different, a third observer would give a score.
[0055] Results: After the experiment, the Parkinson's behavior evaluation method, the tremor paralysis score, the rotation fatigue test, and the depression behavior evaluation method, the open field test, and the sugar water preference test were adopted. The behavioral results showed that after modeling, the mice showed a significant decrease in sugar water preference and a shortened total distance of open field movement, indicating that the mice had depressive-like symptom behavior. Compared with the blank control normal mice, the tremor paralysis score was significantly increased, and the residence time in the rotating rod test was significantly reduced, indicating that the mice showed significant movement disorders. After treatment, compared with the model group, the Chinese medicine group increased the sugar water preference rate and the total distance of movement in the open field, indicating that the depression-like symptom behavior of the mice was alleviated after treatment; and the tremor paralysis score of the Chinese medicine group was lower, similar to the blank control normal mice, and the residence time in the rotating rod test was increased, indicating that the Parkinson's-like behavior of the mice was alleviated after treatment. The high and low dose experiments of Chinese medicine showed that the Chinese medicine composition of the present invention significantly improved various behavioral tests, and was dose-dependent, indicating that the composition of the present invention significantly improved depression and movement disorders in mice.
