Plasmodium berghei genetically modified parasite strains and their use in screening transmission blocking agents

By knocking out and labeling the Plasmodium berghei gene PBANKA_1112700, its expression and function during the sexual stage of the parasite were clarified. This solved the problem of the limited number of candidate antigens in existing vaccines, provided a screening method for candidate targets to block transmission, and achieved effective blocking of malaria transmission.

Abstract

The application belongs to the technical field of genetic engineering, and particularly relates to a Plasmodium berghei gene modified strain and application thereof in screening of a transmission blocking agent. The application discloses, for the first time, a nucleotide sequence of a sexual stage gene PBANKA_1112700 of Plasmodium berghei and an amino acid sequence of a protein coded by the gene. By constructing an HA-tagged strain, it is determined that the gene is highly expressed in a ookinete stage and located on a membrane surface. By constructing a gene knockout strain, it is proved that the gene is inhibited after being knocked out, and ookinete formation and mosquito stomach oocyst formation are inhibited. The application provides application of PBANKA_1112700 as a malaria transmission blocking candidate gene, and provides a theoretical and experimental basis for research and development of a novel malaria transmission blocking.
