[0001] Priority is claimed of European patent application no. 19 217 100, filed on December 17, 2019.
[0002] The invention relates to an improved process for the synthesis of Fluralaner in an efficient and economical way.
[0003] Fluralaner is a compound having the chemical name 4-[(5R / S)-5-(3,5-Dichlorophenyl)-4,5-dihydro-5-trifluoromethyl-1,2-oxazol-3-yl]-N-[2-oxo-2-(2,2,2-trifluoroethylamino)ethyl]-o-toluamide or 4-[5-(3,5-dichlorophenyl)-4,5-dihydro-5-(trifluoromethyl)-3-isoxazolyl]-2-methyl-N-[2-oxo-2-[(2,2,2-trifluoroethyl)amino]ethyl]-benzamide and has the following structure:
[0004] Fluralaner, acts as an "ectoparasiticide". Fluralaner kills fleas, ticks and mites that have ingested the dog's or cat's blood by acting on their nervous system. It blocks the normal movement of charged chloride particles (ions) in and out of nerve cells, especially those associated with gamma-aminobutyric acid (GABA) and glutamate, two substances that convey messages between nerves (neurotransmitters). This results in uncontrolled activity of the nervous system and the paralysis and death of the parasites.
[0005] Fluralaner is available for oral (chewable tablets) and topical (spot-on) administration to dogs, for topical administration (spot-on) to cats, and for oral administration (additive for drinking water) to chickens. The spot-on solution contains 28 g of the active substance Fluralaner per 100 ml of solution (28% w / v). The excipients in the formulation are dimethylacetamide (DMA), glycofurol, diethyltoluamide (DEET), and acetone. Five strengths are available for dog bodyweight ranges between 2 kg and 56 kg and three strengths are available for cat bodyweight ranges between 1.2 kg and 12.5 kg; all strengths are made from identical bulk solution containing 28% w / v Fluralaner.
[0006] The chewable tablets comprise in addition to active substance the excipients sucrose, maize starch, sodium lauryl sulphate, magnesium stearate, aspartame, glycerol, soya bean oil (refined), macrogol 3350, and disodium pamoate monohydrate.

[0008] In a solution of 1.00 g of N-[4-[5-(3,5-dichlorophenyl)-5-trifluoromethyl-4,5-dihydroisoxazol-3-yl]-2-methylbenzoyl]glycine (2) in 30 ml of dichloromethane, 0.65 g of 1-[3-(diethylamino)propyl]-3-ethylcarbodiimide hydrochloride (EDC·HCl) was added, and stirred at room temperature for 15 minutes, then 0.40 g of 2,2,2-trifluoroethylamine and 0.40 g of 4-(N,N-dimethylamino)pyridine (DMAP) were added and stirred at the same temperature for further 2 hours. After the completion of the reaction, the solvent was distilled off under reduced pressure, the residue was purified with silica gel column chromatography that was eluted with ethyl acetate-hexane (1:3), and then crystallized from hexane to obtain 0.48 g of the aimed product as white crystal. Thus, the yields reported are low, only about 41%.

[0013] Although various approaches for preparing Fluralaner from various intermediates are known from the prior art, there still remains a need for improved procedures for preparing Fluralaner. There is a demand to make Fluralaner available at high yield and high purity so that synthesis is efficient and no complex and laborious work up procedures are necessary. Moreover, the procedures for preparing Fluralaner should allow for production on industrial scales.

[0016] It is an object of the invention to provide processes for the chemical synthesis of Fluralaner having advantages compared to the processes of the prior art.
[0017] This object has been achieved by the subject-matter of the patent claims.
[0018] Unexpectedly, the process of the present invention considerably improves the known approaches for preparing Fluralaner. It has been surprisingly found that the low yield of the process known from the prior art, e.g. from WO 2005 / 085216, is due to an unwanted side reaction which according to the invention can be suppressed by defined order of addition of the reagents. It has been unexpectedly found that according to prior art process up to 50% of the product material comprises a "dimeric impurity" of the following formula: which is a product of an unwanted side reaction.
[0019] It has been surprisingly found that a slight modification of the process known from the prior art provides for improved yield of Fluralaner, easy purification, and suitability of the process for industrial production. The present invention provides process which minimizes the formation of this dimeric impurity, thereby significantly improved yields and minimizes need for purification.
[0020] Without wishing to be bound to any theory, it is assumed that the order of addition of specific reagents to the reaction mixture surprisingly reduces the formation of side products, especially of the dimeric impurity, and thereby essentially contributes to the preparation of Fluralaner in high yields and with high purity (e.g. with a purity of more than 98%). The order of addition of reagents therefore provides the advantage that the formation of side product can be avoided or minimized and that purification procedures, especially complex, material-intensive, laborious and time-consuming purification procedures, such as column chromatography, may be avoided.
[0021] The aspects, advantageous features and preferred embodiments of the present invention summarized hereinafter, respectively, alone or in combination with one another, further contribute to solving the problem of the present invention.
[0022] A first aspect of the invention relates to a process for preparing Fluralaner of formula (1), said process comprising the steps of: (a) providing a composition comprising a solvent; (4-(5-(3,5-dichlorophenyl)-5-(trifluoromethyl)-4,5-dihydroisoxazol-3-yl)-2-methylbenzoyl)-glycine (2) a catalyst which is either (i) a base or (ii) a N-hydroxyl compound; and either (i) 2,2,2-trifluoroethylamine (or a salt thereof, for example hydrochloride, etc.) or (ii) a carboxylic acid activating agent, preferably a carbodiimide; (b) adding to the composition provided in step (a) either (i) a carboxylic acid activating agent, preferably a carbodiimide, or (ii) 2,2,2-trifluoroethylamine (or a salt thereof, for example hydrochloride, etc.); (c) allowing the composition obtained in step (b) to react thereby synthesizing Fluralaner (1); and (d) optionally, recovering Fluralaner (1) from the composition obtained in step (c).
[0023] Figure 1 shows a chromatogram of the crude reaction product obtained in Example 1. Figure 2 shows a chromatogram of the crude reaction product obtained in Example 2. In each case, FLRAA (retention time about 9.5 minutes) is Fluralaner; FLE82 (retention time about 3.8 minutes) is the starting compound (2); and MH913 (retention time about 8.7 minutes) is the dimeric impurity.
[0024] The 2,2,2-trifluoroethylamine may be employed in form of the free base or as a salt thereof. Suitable salts of 2,2,2-trifluoroethylamine include for instance the hydrochloride and hydrobromide.
[0025] The process according to the invention encompasses two variants, variant (i) and variant (ii). Both variants (i) and (ii) have in common that in contrast to synthetic example 214 of WO 2005 / 085216, the (4-(5-(3,5-dichlorophenyl)-5-(trifluoromethyl)-4,5-dihydroisoxazol-3-yl)-2-methylbenzoyl)glycine (2) is not allowed to react with the carboxylic acid activating agent in absence of the catalyst and the 2,2,2-trifluoroethylamine (or a salt thereof, for example hydrochloride, etc.), respectively.
[0026] The composition provided in step (a) is preferably stirred for at least 10 minutes, preferably for at least 15 minutes before addition of either (i) a carboxylic acid activating agent, preferably a carbodiimide, or (ii) 2,2,2-trifluoroethylamine (or a salt thereof, for example hydrochloride, etc.) in step (b).
[0027] When the carboxylic acid activating agent is the last reagent that is added to the composition (variant i), it is preferably added to the composition provided in step (a) after the composition is stirred for at least 5 minutes, preferably for at least 10 minutes, more preferably for at least15 minutes. In this regard, the reagents comprised in the composition provided in step (a) are the compound of formula (2), the 2,2,2-trifluoroethylamine (or a salt thereof, for example hydrochloride, etc.) and the base as a catalyst.

In this regard, the reagents comprised in the composition provided in step (a) are the compound of formula (2), the carboxylic acid activating agent, and the N-hydroxyl compound as a catalyst.

Then 4.8 g (25 mmol) 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride (EDC·HCl) were added and the mixture was agitated until reaction was finished according to HPLC analysis.
[0075] Then 300 mL of 5 % aqueous solution of hydrochloric acid were added, agitated for at least 15 minutes and the two phases were separated. After some time (approximately 75 min) to the mixture 12.5 g (126.2 mmol) 2,2,2-trifluoroethan-1-amine were added and the mixture was agitated until reaction was finished according to HPLC analysis.

Claims
1. A process for preparing Fluralaner of formula (1), said process comprising the steps of: (a) providing a composition comprising - a solvent which is selected from the group consisting of acetonitrile, acetone, N,N-dimethylformamide, tetrahydrofuran, ethyl acetate, dichloromethane, and mixtures thereof; - (4-(5-(3,5-dichlorophenyl)-5-(trifluoromethyl)-4,5-dihydroisoxazol-3-yl)-2-methylbenzoyl)glycine (2) - a catalyst which is either (i) a base or (ii) a N-hydroxyl compound; and - either (i) 2,2,2-trifluoroethylamine or a salt thereof, or (ii) a carboxylic acid activating agent which is selected from carbodiimides, phosphonium-type reagents, and aminium / uronium-imonium-type reagents; (b) adding to the composition provided in step (a) either (i) a carboxylic acid activating agent which is selected from carbodiimides, phosphonium-type reagents, and aminium / uroniumimonium-type reagents, or (ii) 2,2,2-trifluoroethylamine or a salt thereof; wherein the relative molar ratio of the 2,2,2-trifluoroethan-1-amine or a salt thereof and the compound of formula (2) is less than 15; (c) allowing the composition obtained in step (b) to react thereby synthesizing Fluralaner (1); and (d) optionally, recovering Fluralaner (1) from the composition obtained in step (c).
2. The process according to claim 1, wherein (i) the composition provided in step (a) comprises 2,2,2-trifluoroethylamine or a salt thereof, and wherein in step (b) the carboxylic acid activating agent is added; preferably the entire amount of 2,2,2-trifluoroethylamine or a salt thereof that is employed in the process is contained in the composition provided in step (a), and wherein the entire amount of the carboxylic acid activating agent that is employed in the process is added in step (b).
3. The process according to claim 1 or 2, wherein the composition provided in step (a) comprises the solvent, the (4-(5-(3,5-dichlorophenyl)-5-(trifluoromethyl)-4,5-dihydroisoxazol-3-yl)-2-methylbenzoyl)glycine (2), the catalyst, and the 2,2,2-trifluoroethylamine or a salt thereof, but not yet the carboxylic acid activating agent; and wherein the carboxylic acid activating agent is subsequently added in step (b) to the composition provided in step (a); and step (a) comprises the sub-steps of: (a-1) providing a composition comprising the solvent and the (4-(5-(3,5-dichlorophenyl)-5-(trifluoromethyl)-4,5-dihydroisoxazol-3-yl)-2-methylbenzoyl)glycine (2); and (a-2) adding to the composition provided in sub-step (a-1) the 2,2,2-trifluoroethylamine or a salt thereof and the catalyst.
4. The process according to claim 1, wherein (ii) the composition provided in step (a) comprises the carboxylic acid activating agent, and wherein in step (b) 2,2,2-trifluoroethylamine or a salt thereof is added; preferably the entire amount of 2,2,2-trifluoroethylamine or a salt thereof that is employed in the process is added in step (b), and wherein the entire amount of the carboxylic acid activating agent that is employed in the process is contained in the composition provided in step (a).
5. The process according to claim 1 or 4, wherein the composition provided in step (a) comprises the solvent, the (4-(5-(3,5-dichlorophenyl)-5-(trifluoromethyl)-4,5-dihydroisoxazol-3-yl)-2-methylbenzoyl)glycine (2), the catalyst, and the carboxylic acid activating agent, but not yet the 2,2,2-trifluoroethylamine or a salt thereof; and wherein the 2,2,2-trifluoroethylamine or a salt thereof is subsequently added in step (b) to the composition provided in step (a); and step (a) comprises the sub-steps of: (a-1) providing a composition comprising the solvent and the (4-(5-(3,5-dichlorophenyl)-5-(trifluoromethyl)-4,5-dihydroisoxazol-3-yl)-2-methylbenzoyl)glycine (2); and (a-2) adding to the composition provided in sub-step (a-1) the carboxylic acid activating agent and the catalyst.
6. The process according to any of the preceding claims, wherein the catalyst is a base which is selected from the group consisting of 4-dimethylaminopyridine, triethanolamine, N,N-diisopropylethylamine, N-methyl-morpholine, 2-dimethylaminopyridine, 2,4,6-trimethylpyridine, piperidine and 4-(2-piperidinoethyl)pyridine; preferably 4-dimethylaminopyridine.
7. The process according to any of claims 1 to 6, wherein the catalyst is a N-hydroxyl compound which is selected from the group consisting of N-hydroxysuccinimide, N-hydroxy-benzotriazoles, N-hydroxy-benzotriazines, and 2-hydroxyimines; preferably N-hydroxysuccinimide.
8. The process according to any of the preceding claims, wherein the solvent is dichloromethane.
9. The process according to any of the preceding claims, wherein the carboxylic acid activating agent is a carbodiimide selected from 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride, dicyclohexylcarbodiimide, N-cyclohexyl-N'-(β-[N-methylmorpholino]ethyl)carbodiimide, and diisopropylcarbodiimide; preferably 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride.
10. The process according to any of the preceding claims, wherein the relative molar ratio of the 2,2,2-trifluoroethan-1-amine or a salt thereof and the compound of formula (2) is less than 14.
11. The process according to any of the preceding claims, wherein the relative molar ratio of the compound of formula (2) and the 2,2,2-trifluoroethan-1-amine or a salt thereof is within the range of from 1:1 to 1:10.
12. The process according to any of the preceding claims, wherein the relative molar ratio of the compound of formula (2) and the carboxylic acid activating agent is within the range of from 1:0.5 to 1:5.
13. The process according to any of the preceding claims, wherein step (c) is carried out at a temperature within the range of from 10 to 50 °C.
14. The process according to any of the preceding claims, wherein - the catalyst is 4-(N,N-dimethylamino)pyridine (DMAP) or N-hydroxysuccinimide (NHS); - the solvent is dichloromethane; and - the carboxylic acid activating agent is 1-[3-(diethylamino)propyl]-3-ethylcarbodiimide hydrochloride (EDC·HCl).
15. The process according to any of the preceding claims, which provides an overall yield of Fluralaner (1) of at least 45%, preferably at least 50%.
