Description

[0001] Organometallic compounds, their production and use

[0002] Introduction

[0003] Numerous organometallic compounds, especially palladium compounds, are known as catalysts in chemical synthesis. Numerous palladium-catalyzed reactions, such as the Heck and Stille reaction, the Hartwig-Buchwald reaction, the Negishi coupling, the Suzuki coupling, and the Sonogashira coupling, are established in preparative organic chemistry.

[0004] Nevertheless, there is a constant need for new catalysts that meet specific requirements or have new properties in order to expand and complement the range of preparative chemistry.

[0005] WO 2017 / 093427, for example, shows a palladium-catalyzed, selective arylation process.

[0071] Table 5 shows the 35 compounds 3.01 to 3.35 of formula I and the 35 compounds 3.36 to 3.70 of formula II, where R1 and R2 have the meanings defined in Table 1, R3 is cyclohexyl and X is Br.

[0072] Table 6 shows the 35 compounds 6.01 to 6.35 of formula I and the 35 compounds 6.36 to 6.70 of formula II, where R1 and R2 have the meanings defined in Table 1, R3 is phenyl and XI.

[0073] Table 7 shows the 35 compounds 7.01 to 7.35 of formula I and the 35 compounds 7.36 to 7.70 of formula II, where R1 and R2 have the meanings defined in Table 1, R3 is cyclohexyl and XI.

[0074] The compounds of formula I can be obtained by a process wherein a ligand of the type R1-C-(P(R2)2)(P(R3)3), where R1, R2 and R3 are as defined above, is reacted with a palladium compound of the type PdX2 or L n (PdX2), where X is halogen as defined above, L is a neutral electron donor ligand and n = 1 or 2.

[0075] The ligand used of the type R1-C-(P(R2)2)(P(R3)3) has the structure of formula III.

[0076] Formula III

[0077] The substituents R1, R2 and R3 are as defined above. In particular, the

[0078] Substituents R1, R2 and R3 are as defined in Table 8 below.

[0079] The palladium compounds of the type PdX2 are the well-known palladium halides PdCl2, PdBr2 and Pdl2, in particular palladium chloride PdCl2.

[0080] Palladium compounds of type L have also proven to be suitable as starting materials for the preparation of compounds of formulas I and II. n (PdX2), where L is a neutral electron donor ligand and n = 1 or 2. If the neutral electron donor ligand is a monodentate ligand, n = 2; for a bidentate ligand, n = 1.

[0081] In specific embodiments, L is selected from the group consisting of acetonitrile (CH3CN), dimethyl sulfoxide (DMSO), dibenzylideneacetone (DBA) and 1,5-cyclooctadiene (COD), norbornadiene (NBD)

[0082] Compounds such as (CH3CN)2PdCl2, (COD)PdCl2 or (DBA)PdCl2 are well suited.

[0083] To prepare the compound of formula II, a ligand of the type R.1-CH- (P(R2)2)(P(R3)3)(X) of formula IV can be used:

[0084] Formula IV

[0085] This can be obtained by acid treatment of a ligand of the type R.1-C-(P(R2)2)(P(R3)3) of formula III. For this purpose, the ligand is suspended in a suitable solvent, such as dichloromethane, and treated with a suitable acid, such as concentrated hydrochloric acid or HBF4.

[0086] The reaction is then carried out with a palladium compound. Palladium compounds of the PdX2 type are the well-known palladium halides PdCl2, PdBr2, and Pd12, in particular palladium chloride PdCl2.

[0087] In strongly acidic, aqueous solutions, these compounds usually occur in the form of the corresponding acid H2PdX4, i.e., in the case of palladium chloride, in the form of the compound H2PdCl4. These acidic solutions, usually produced industrially, contain palladium halides, especially palladium chloride or H2PdX4 or H2PdCl4, and are well-suited as starting materials for the preparation of the compounds of formula II. The order of addition of the starting materials is uncritical in this procedure and leads to the same product in comparable purities and yields.

[0088] When using such an acidic solution of PdX2, in particular PdCl and / or PdBr2, the ligand according to formula III can also be used; a ligand according to formula IV is then not required.

[0089] In a further embodiment, a compound of formula II can be obtained by first preparing a compound of formula I and then treating it with acid. For this purpose, the ligand is suspended in a suitable solvent, such as dichloromethane, and mixed with, for example, concentrated hydrochloric acid. Conversely, a compound of formula II can also be converted into a compound of formula I by treatment with a base. Various alkali metal carbonates, alkali metal alcoholates and HMDS bases (hexamethylenedisilazides) such as sodium or potassium ethanolate, sodium or potassium carbonate and sodium or potassium hexamethyldisilazide (Na-HMDS or K-HMDS) are generally suitable for this purpose; in practice, potassium tert-butoxide and sodium carbonate have proven suitable. The reaction can be carried out in a solvent or mechanochemically.For example, alcohols such as ethanol or isopropanol, ethers such as tetrahydrofuran or aromatic solvents such as toluene can be used as solvents.

[0090] Palladium compounds of type L have also proven to be suitable as starting materials for the preparation of compounds of formulas I and II. n (PdX2), where L is a neutral electron donor ligand and n = 1 or 2. If the neutral electron donor ligand is a monodentate ligand, n = 2; for a bidentate ligand, n = 1.

[0091] In specific embodiments, L is selected from the group consisting of acetonitrile (CH3CN), dimethyl sulfoxide (DMSO), dibenzylideneacetone (DBA) and 1,5-cyclooctadiene (COD).

[0092] Compounds such as (CH3CN)2PdCl2, (COD)PdCl2 or (DBA)PdCl2 are well suited.

[0093] The reaction of the ligand of formula III or formula IV with a palladium compound to form the compounds of formula I or II can be carried out at temperatures from 0°C to 100°C, in particular from 10°C to 50°C, advantageously from 15°C to 30°C, or the respective room temperature. The reaction times are from 2 hours to 72 hours, in particular from 2 hours to 12 hours, or from 2 hours to 8 hours, or from 2 to 4 hours.

[0094] The reaction can be carried out in a solvent. Polar solvents, usually polar, aprotic solvents, are particularly suitable, although in some cases good results have been achieved with ethanol. It may be advantageous to select water-miscible solvents. When using an acidic palladium halide solution, especially acidic palladium chloride solution, a water-soluble solvent is required.

[0095] Tetrahydrofuran, dichloromethane, acetone, ethanol, ethyl acetate and acetonitrile have proven particularly suitable solvents.

[0096] The palladium complexes of formula I and formula II described above can be used in homogeneous catalysis, in particular in coupling reactions, where the coupling reaction can be selected from the group consisting of

[0097] (i) catalytic hydrofunctionalization reactions of alkynes and alkenes;

[0098] (ii) catalytic hydroamination reactions of alkynes and alkenes;
