[0001] EVAPORATION APPARATUS AND METHOD FOR EVAPORATION

[0002] The present patent disclosure relates to the area of solvent evaporation. The present patent disclosure also relates to the field of liquid level detection. Particular examples concern an evaporation apparatus and method for evaporation of a part of a liquid in a sample container.

[0003] Evaporation apparatuses can be found in many types of laboratories, from drug discovery to analytical chemistry laboratories, often to remove solvent from a sample by evaporation of the solvent. The sample container containing the sample, which may comprise an analyte in a solvent is often heated to increase the vapor pressure of the solvent. The heating may be done by partially submerging the sample container in a heatable liquid bath or tank, such as a heated water bath. To increase the speed of evaporation, one type of evaporation apparatus uses a nozzle to direct a stream of gas in the sample container.

[0004] The analyte may, however, comprise sensitive components, such as certain proteins or polymers, that require at least some solvent to remain stable. The evaporation should therefore be stopped before all the solvent is evaporated. Evaporation apparatuses may comprise electronic optical detectors that are positioned at a certain level of the sample container in order to detect the liquid level in the sample container. These electronic optical detectors often fail, especially when submerged in the heated liquid bath, and require replacement and may cause loss of the analyte when malfunctioning during evaporation. Patent document US 5,054,319 in accordance with its abstract states a liquid level sensor system includes a float assembly configured for placement in a liquid. The float assembly includes a float that is sufficiently buoyant to move the float assembly vertically in response to changes in the level of the liquid. The float assembly may comprise a buoyant member placed around a tube and constrained to movement only lengthwise along the tube in response to changes in the liquid level. A reflector is coupled to the float such that the position of the reflector is a function of the liquid level. A carriage may be placed inside the tube for moving with the float in response to changes in the liquid level, with the reflector being mounted to the carriage. The carriage and the float preferably are magnetically coupled together so that the carriage means moves lengthwise inside the tube in response to movement of the float assembly as the liquid level changes. A light source directs a light signal toward the reflector. Light reflected from the reflector is processed in a feedback loop to control the frequency of the feed back induced amplitude modulation of the light signal as a function of the optical path length, which depends upon the liquid level. The frequency of the amplitude modulated signal is measured to determine the liquid level.

[0008] According to a first aspect, there is provided an evaporation apparatus for evaporation of a part of a liquid in a sample container, the evaporation apparatus comprising: a sample holder for holding the sample container; a light source; a light detector configured to output an electronic signal indicative for a characteristic property of light received by the light detector; an optical fiber assembly configured to transfer light emitted by the light source along an optical path through at least a part of the sample container when placed in the sample holder, and to transfer light that travelled along the optical path to the light detector; and a controller in electrical contact with the light detector, wherein the controller is configured to obtain the electronic signal and to control the evaporation apparatus to terminate evaporation based on the electronic signal.

The reflector is preferably a retroreflector, such as a micro cube comer retroreflector. By using a reflector, the light is (at least partially) reflected back along the optical path, and either a receiving fiber end or the same fiber end from which the light originated can be placed at one side of the sample container position, thus reducing the number of fiber positions within the evaporation apparatus.

[0060] In Fig. 1, an evaporation apparatus, generally designated 1, includes a sample holder 108 for holding a sample container 100. The evaporation apparatus comprises a control system, or liquid level detection system, comprising a light source 210 and a light detector 220 configured to output an electronic signal indicative for a characteristic property of light received by the light detector 220. The control system and / or liquid level detection system comprises an optical fiber assembly 230 is configured to transfer light emitted by the light source 210 along an optical path 250 through at least a part of the sample container 100 when placed in the sample holder 108, and to transfer light that travelled along the optical path 250 to the light detector 220. The control system and / or liquid level detection system comprises a controller 240 in electrical contact with the light detector 220. The controller 240 is configured to obtain the electronic signal and to control the evaporation apparatus 1 to terminate evaporation based on the electronic signal. The light source 210, light detector 220, and controller 240 are included in electronics unit 200 in this example.

A moving average filter averaging the last 100 samples is applied to the data of Fig. 8. The light detector signal is normalized and a light detector signal value of 1 indicates that a saturation level of the light detector, and 0 indicates that no detectable amount of light is received by the light detector. Air is blown at least partially at an angle above the liquid in the sample container, so that the liquid starts to rotate relative to the sample container and a vortex is formed in the liquid. The vortex creates a deep meniscus which is wavy. The noise visible in the signal may be at least partially caused by the wavy meniscus through the optical path 250 or at least through the path the light that is detected by the light detector 220 and has gone through the sample container. In this case, since the light that passed the optical path is received by a second optical fiber, the presence of the wavy meniscus causes scattered light that disturbs the received signal, thus increasing the noise. Clearly, however, the fluid level and / or wanted endpoint is obtained using the system 400. An example threshold level 800 is indicated in the figure. If the (average) signal value goes below this level, the evaporation may be terminated. It is noted that the signal value may be inverted, depending on the type of light detector used and the way it is connected in an electrical circuit. In this case, the evaporation may in that case be terminated when the (average) signal value increases above the level 800. In general, the evaporation may be terminated when the (average, e.g. moving average) signal value has crossed the threshold level, such as the threshold level 800 or 900. For the data in Fig. 8, the evaporation was not terminated and therefore the signal level reduces also after the threshold level 800 has been crossed. If evaporation would have been terminated, then the signal level would reduce less or remain constant after the threshold level 800 has been crossed.

[0080] A plot of example data of the light detector signal versus time for the system 600 is shown in Fig. 9. The data is obtained in the same manner and is treated the same way as for Fig. 8. A retroreflector with circular shape and diameter of 6 mm was used. The distance between the retroreflector and the second optical fiber end 448 was 11 mm. The sample container diameter at which the measurement was performed was 10 mm. The offset of the tip relative to the radial axis 702 was about 2 mm. Comparing the data of Fig. 9 with that of Fig. 8, the system with the retroreflector reduces the amount of noise substantially compared to the data of Fig. 8 of the fiber-to-fiber configuration of system 400 of Fig. 2. Without being bound by any theory, the noise is reduced in the system 600 compared to the system 400 because there is less interference due to the scattering caused by the presence of waves in the vortex as the light is reflected back towards the second fiber end 448 with a much higher speed than the speed of any waves in the vortex. In other words, even though light emitted by the second fiber end 448 towards the retroreflector 260 may be scattered by the irregularities in the liquid caused by the waves in vortex, the light travels back along the same path while the irregularities have not substantially changed in that timescale and thus much of the light returns into the second fiber end 448 so that a part of that returned light is detected by the light detector 220. Close to the same noise reduction as for system 600 is achieved by the system 500.

Claims

CLAIMS1. Evaporation apparatus for evaporation of a part of a liquid in a sample container, the evaporation apparatus comprising: a sample holder for holding the sample container; a light source; a light detector configured to output an electronic signal indicative for a characteristic property of light received by the light detector; an optical fiber assembly configured to transfer light emitted by the light source along an optical path through at least a part of the sample container when placed in the sample holder, and to transfer light that travelled along the optical path to the light detector; a controller in electrical contact with the light detector, wherein the controller is configured to obtain the electronic signal and to control the evaporation apparatus to terminate evaporation based on the electronic signal; and a retroreflector, comprising a reflective side arranged at an end of and towards the optical path.

2. Evaporation apparatus according to claim 1, wherein the optical path is offset relative to a longitudinal center axis of the sample container, when placed in the sample holder.

3. Evaporation apparatus according to claim 1 or 2, wherein the retroreflector is a micro cube comer retroreflector.

4. Evaporation apparatus according to claim 1, 2 or 3, wherein the retroreflector is arranged such that, when the sample container is placed in the sample holder, light that passed through the sample container is reflected in an opposite direction along the optical path.

Evaporation apparatus according any one of the preceding claims, wherein the optical fiber assembly is configured to: receive the light emitted by the light source; emit at least a part of the received light emitted by the light source along the optical path in which at least a part of the sample container is placeable; receive the light that travelled along the optical path; and emit at least a part of the received light that travelled along the optical path towards the light detector.

Evaporation apparatus according to claim 5, wherein the optical fiber assembly comprises one or more optical fibers comprising a first optical fiber end, a second optical fiber end and a third optical fiber end, wherein the one or more optical fibers are arranged such that: the first optical fiber end is positioned to receive the light emitted by the light source; the second optical fiber end is positioned to emit the at least a part of the received light emitted by the light source along the optical path; and the third optical fiber end is positioned to emit the at least a part of the received light that travelled along the optical path towards the light detector.

7. Evaporation apparatus according to claim 6, in dependence of any one of claim 3 and 4, wherein the retroreflector is arranged opposite the second optical fiber end of the one or more optical fibers, wherein the retroreflector is arranged to reflect light emitted by the second optical fiber end back towards the second optical fiber end.

Evaporation apparatus according to claim 6 or 7, wherein the one or more optical fibers comprise a first optical fiber comprising the first optical fiber end, the second optical fiber end and the third optical fiber end, wherein the first optical fiber comprises an optical splitter, wherein a first fiber optical path through the first optical fiber goes from the first optical fiber end to the second optical fiber end via the optical splitter, and a second fiber optical path goes from the second optical fiber end to the third optical fiber end via the optical splitter.

Evaporation apparatus according to any one of the preceding claims, wherein the controller comprises processing circuitry comprising a processor and a memory, wherein the processing circuitry is in electrical contact with the light detector, wherein the processor and memory comprise: an obtaining module configured to obtain the electronic signal from the light detector, a determining module configured to determine whether to terminate evaporating based on the obtained electronic signal, and a termination module configured to terminate the evaporating if the determining module determined to terminate evaporation.

Evaporation apparatus according any one of the preceding claims, wherein the characteristic property of the light received by the light detector comprises at least one of a light intensity, a change in light intensity, a wavelength, a change in wavelength, a polarization, and a change in polarization.

Evaporation apparatus according to claim 10, wherein the characteristic property is a light intensity or a change in light intensity, wherein preferably the controller is configured to control the evaporation apparatus to terminate evaporating when the electronic signal indicates that the light intensity or the change in light intensity is above or below a threshold light intensity or change in light intensity, respectively.

Evaporation apparatus according to any one of the preceding claims, wherein the evaporation apparatus comprises at least one nozzle, wherein the sample holder is configured to hold the sample container in a defined position relative to the at least one nozzle, wherein the evaporation apparatus is configured such that a gas flow is provided through the at least one nozzle so that, when the evaporation apparatus is in use, a vortex is formed in the liquid to be evaporated in the sample container, wherein the controller is configured to control the evaporation apparatus to terminate evaporation by terminating the gas flow to the sample container.

13. Evaporation apparatus according to any one the preceding claims, comprising a heatable liquid tank, wherein the sample holder is arranged such that, when the heatable liquid tank comprises liquid, the sample container is at least partially submerged in the liquid in the heatable liquid tank when placed in the sample holder, wherein the light source, the light detector and the controller are arranged outside of the heatable liquid tank, wherein preferably the controller is configured to control the evaporation apparatus to terminate evaporation by terminating heating of the heatable liquid tank.

Evaporation apparatus according to any one of the preceding claims, wherein: the optical path correlates with a predetermined liquid level in the sample container at which the evaporation is to be terminated; and / or the light source is configured to emit light with a wavelength in a range of 650-1000 nm, preferably 700-980 nm, more preferably 750-950 nm; and / or the light detector comprises a phototransistor or photodiode.

15. Method for evaporating a part of a liquid to be evaporated in a sample container, the method comprising: performing evaporation of the liquid in the sample container; transferring, using an optical fiber assembly, light from a light source along an optical path through at least a part of the sample container; transferring light that passed along the optical path to a light detector;generating, by the light detector, an electronic signal indicative for a characteristic property of the light detected by the light detector; and terminating the evaporation based on the electronic signal.
