Description

Title of the invention: Use of amitriptyline and / or one of its pharmaceutically acceptable salts as a preservative agent

[0001] The present invention relates to the use of amitriptyline and / or one of its pharmaceutically acceptable salts as a preservative in a pharmaceutical composition.

[0002] Pharmaceutical compositions and the substances they contain are subject to important standards, particularly in terms of the presence of microorganisms, such as bacteria, yeasts or fungi, in order to avoid the risks of infections for the user and the deterioration of the pharmaceutical compositions.

[0003] In Europe, Chapter 5.1.4 "Microbiological quality of pharmaceutical preparations" of the European Pharmacopoeia states that formulations administered topically do not need to be sterile. However, they must not contain more than one hundred microorganisms per gram of composition and they must not contain Staphylococcus aureus or Pseudomonas aeruginosa.

[0004] In order to verify their proper compliance with these standards before being placed on the market, the compositions must be subjected to tests such as those specified by monographs 2.6.12 and 2.6.13 requiring a count of germs and microorganisms and setting a limit threshold not to be exceeded.

[0005] To meet these standards and maintain such a low microorganism population, it is often necessary to incorporate preservatives or antimicrobial agents into pharmaceutical compositions. Chapter 5.1.3 "Effectiveness of antimicrobial preservation" of the European Pharmacopoeia clearly defines how the effectiveness of an antimicrobial preservative in a pharmaceutical product should be assessed.

[0006] Such standards and tests are also in force in the United States (USP specifications, Chapter 51).

[0007] Among the preservatives commonly used in pharmaceutical compositions, we find in particular parabens, such as methyl paraben. However, these compounds are suspected of interacting with the receptors of certain hormones and therefore of having a role as endocrine disruptors.

[0008] There is therefore a real need to provide new antimicrobial preservatives that can be used in pharmaceutical compositions, in particular topical pharmaceutical compositions.

[0009] It has been surprisingly discovered that amitriptyline and / or one of its pharmaceutically acceptable salts provides an antimicrobial effect equivalent to a paraben-type preservative in pharmaceutical compositions.

[0010] The subject of the present invention is therefore the use of ramitriptyline and / or one of its pharmaceutically acceptable salts as a preservative in a pharmaceutical composition.

[0011] The use of ramitriptyline and / or one of its salts makes it possible to formulate pharmaceutical compositions with a low or even zero amount of paraben-type preservative. Such use therefore makes it possible to limit the presence of potential endocrine disruptors or preservatives that present risks of causing allergies or irritations.

[0012] Other objects, characteristics, aspects and advantages of the invention will appear even more clearly on reading the description and the example which follows.

[0013] In this description, and unless otherwise indicated:

[0014] - the expression "at least one" is equivalent to the expression "one or more" and can be substituted for it;

[0015] - the expression "between...and..." is equivalent to the expression "ranging from...to..." and may be substituted therefor, and implies that the terminals are included;

[0016] - the expression “polyoxyalkylenated” corresponds, within the meaning of the invention, to a unit -(O-alkyl)n-, where n is an integer ranging from 2 to 200, preferably from 2 to 40, more preferably from 2 to 20;

[0017] - the expression “polyoxyethylenated” corresponds, within the meaning of the invention, to a unit -(O-CH2CH2)n-, where n is an integer ranging from 2 to 200, preferably from 2 to 40, more preferably from 2 to 20.

[0018] Amitriptyline and its salts

[0019] The use according to the present invention uses at least amitriptyline and / or one of its pharmaceutically acceptable salts in a pharmaceutical composition.

[0020] Amitriptyline has the following formula (I):

[0022] In the context of the present invention, the term “pharmaceutically acceptable amitriptyline salts” means salts compatible with a pharmaceutical composition pharmaceutical, i.e. intended for administration to humans. In particular, the term “pharmaceutically acceptable salt of amitriptyline” means the hydrates, solvates, acid salts such as hydrochlorides and clathrates of amitriptyline.

[0023] As a particularly preferred salt of amitriptyline, amitriptyline hydrochloride will be used.

[0024] Preferably, the total content of amitriptyline and / or one of its pharmaceutically acceptable salts is between 1 and 25% by weight, more preferably between 5 and 20% by weight, even more preferably between 5 and 15% by weight, relative to the total weight of the composition.

[0025] More preferably, the total content of amitriptyline hydrochloride is between 1 and 25% by weight, more preferably still between 5 and 20% by weight, even better still between 5 and 15% by weight, relative to the total weight of the composition.

[0026] The presence of amitriptyline and / or one of its pharmaceutically acceptable salts in the pharmaceutical composition according to the invention makes it possible to obtain a composition in accordance with the criteria of the European and American Pharmacopoeia, in particular with regard to the presence of microorganisms in the composition. In particular, the presence of amitriptyline and / or one of its pharmaceutically acceptable salts makes it possible to limit, or even prevent, the growth of microorganisms such as Pseudomonas aeruginosa, Staphylococcus aureus, Escherichia coli, Candida albicans, Aspergillus brasiliensis.

[0027] The composition may optionally further comprise at least one additional preservative agent, different from amitriptyline and its salts, such as methyl paraben. In this case, the content of additional preservative agent(s) is preferably less than or equal to 10% by weight, more preferably less than or equal to 5% by weight, more preferably still less than or equal to 1% by weight, better still less than or equal to 0.5% by weight, relative to the total weight of the composition.

[0115] Composition A, comprising methyl paraben but not amitriptyline, does not comply with the European and American Pharmacopoeia tests.

[0116] Composition C, comprising methyl paraben and amitriptyline, complies with the European and American Pharmacopoeia tests. The same is true for composition D, comprising amitriptyline but not methyl paraben.

[0117] Thus, the presence of amitriptyline makes it possible to reduce the populations of microorganisms such as P. aeruginosa, S. aureus, E. coli, C. albicans and A. brasiliensis, in pharmaceutical compositions, even in the absence of a preservative such as methyl paraben.
