Abstract
Description
Title of the invention: Composition and skin care product for inhibiting the TRPV1 pathway and reducing skin irritation Technical field
[0001] The present invention belongs to the field of daily cosmetic technology, and particularly relates to a composition for inhibiting the TRPV1 pathway and reducing skin irritation and a skin care product for inhibiting the TRPV 1 pathway and reducing skin irritation. TECHNICAL CONTEXT
[0002] The skin is the organ that covers the surface of the human body and is one of the most important organs, which plays a role in protecting the human body from the external environment and external irritations and maintaining the water balance of the human body. Strong irritation on the skin is likely to cause damage to the skin's surface barrier and cause a series of immune responses, and at the same time, the nerve endings on the skin surface receive irritation signals and send them to neurons, causing a series of sensations such as tingling, itching, burning, etc. There is an irritation threshold for nerve endings to receive irritation signals, when irritation is below the threshold, no signaling will occur and no response will occur.Over-response of nerves may cause hypersensitive reactions and mild irritation may cause reactions such as tingling, itching, burning, etc. Sensitive skin may react more strongly to minor disturbances due to barrier damage, exposure of nerve endings or excessive activity of nerve endings.
[0003] Transient receptor potential vanilloid subfamily 1 (TRPV1) channel is a non-selective cation channel that, once activated, causes an inward flow of extracellular calcium ions, resulting in an increase in intracellular calcium concentration, which in turn mediates a series of physiological or pathological responses. In recent years, the TRPV1 channel has been shown to have a significant effect on stinging, itching, and burning. Exogenous administration of TRPV1 antagonists can effectively reduce the skin stress response and improve the skin barrier.
[0004] Camellia (scientific name: Camellia sp.), also known as Japanese camellia, contains over 700 compounds that have been found to promote health human, including flavonoids and polyphenols, camellia has been shown to have a variety of physiological activities. Camellia extract can inhibit the production of tyrosinase, thereby evening out skin tone, resisting UVB-induced photoaging, and reducing skin damage caused by environmental pollution.
[0005] Purslane, also known as Portulaca oleracea, is a commonly used traditional Chinese medicine, and its young stems and leaves can also be used in dishes. It is rich in amino acids, proteins, polysaccharides and other components. Studies have shown that its extracts and chemical components exhibit biological activities such as anti-inflammatory, antidiabetic, antioxidant, anticancer and antibacterial activities, and exhibit effects of reducing skin redness and helping to improve the skin barrier.
[0006] Based on this, the present invention provides a composition for inhibiting the TRPV1 pathway and reducing skin irritation, capable of inhibiting stinging, itching and burning of the skin due to activation of the TRPV1 channel. DISCLOSURE OF THE INVENTION

[0075] 1. Test of the TRPV1 receptor protein on keratinocytes
[0076] Keratinocytes are the most important cells found on the superficial layer of the skin, constituting 90% of the epidermal cells of the skin. By inhibiting the expression of epidermal cells, the activation of the TRPV1 pathway induced by skin irritation is reduced. For the verification of the inhibition of the TRPV1 pathway, the method of detecting the content of TRPV1 receptor (protein) in keratinocytes was used, which allows detecting the inhibition on the TRPV1 pathway by testing the inhibition of the expression of the TRPV1 receptor protein.
[0077] For this, the cytotoxic concentrations of Camellia Redsnow extract and Purslane Protulaca extract were detected first, and the specific results were shown in Tables 1 and 2.
[0078] Table 1 Cytotoxic concentrations of Redsnow Camellia extract Concentration gradient 1 2 3 4 5 6 7 8 PC Control 0.078 1 0.156 3 0.312 5 0.625 1.25 2.5 5 10 10%D MSO / Vi ab ilit é cel lui air e Moye nne 1 2 .78 .70 40% 108.8 1% 108.1 9% 104.6 5% 84.9 4% 79.6 7% 35.3 9% 1.8% 100 % SD 3.58 % 2.40 % 3.58 % 1.56 % 8.82 % 8.27 % 1.27 % % 0.12% 7.52 % Value rp 1.000 0.127 0.141 0.138 0.525 0.07 9 0.01 0 0.00 0 0.000 / Table 2 Cytotoxic concentrations of purpier protulaca extract Ion concentration gradient 1 2 3 4 5 6 7 8 PC Control 0.078 1 0.156 3 0.312 5 0.625 1.25 2.5 5 10 10%D MSO / Vi ab ilit é cel lui air e Average 100.0 10.6% 99.58 % 96.39 % 88.62 % 84.66 % 73.14 % 37.47 % 1.8% 100 % SD 3.18 % 3.27 % 5.23 % 3.86 % 6.38 % 4.23 % H 5.48 % 0.12% 7.52 % P -Value 1.000 0.773 0.941 0.501 0.116 0.036 0.007 0.000 0.000 /

[0082] According to the test protocol in the table above, when the cell plating rate in the 6-well plate reaches 40% to 50%, perform administration in groups at an administration dose of 2 ml / well, 3 wells for each group. After administration, incubate the 6-well plate in an incubator (37°C, 5% CO2) for 24 h.
[0083] TRPV1 detection: treat with 4% paraformaldehyde to fix for 30 minutes, At the end of fixation, perform immunofluorescence detection for the capsaicin receptor (TRPV1), observe under a microscope and take pictures, acquire images and analyze them.
[0084] Plot the graphs using GraphPad Prism, and express the results as Mean+SD. Compare the different groups using t-test statistical analysis. The statistical analysis was two-fold. P<0.05 represents a significant difference and P<0.01 represents a highly significant difference.
[0085] Based on the test protocol, TRPV 1 detection was performed according to the steps of immunofluorescence operation, and the results were shown in the view of the Fluorescence detection of TRPV1 protein in Figures 1 to 8, and the evolution trend was shown in [Fig.9]. The figures (# represents that the NC group has a significant difference compared with the BC group, * represents p<0.05, ** represents p<0.01, **** represents p<0.001) show that the use of single camellia extract and the use of single purslane extract has an inhibitory effect on TRPV1. When the two are combined at a certain proportion, they exhibit a synergistic effect and the inhibition effect is more significant, Example 2 has the best effect. This shows that the composition of these two plant extracts can effectively inhibit TRPV1 channels, thus playing a role in reducing skin irritation.
[0086] 2. Capsaicin test on the human body
[0087] Capsaicin is a classic irritant agent that activates TRPV1, which causes stinging, burning, and redness when it comes into contact with the human body. In this test, 0.01% capsaicin was used as an irritant to irritate both sides of the nasolabial fold, and then the test samples were administered on one side, and the improvement was compared with the white area. The test samples are the skin care products prepared in Examples 1 to 3 and Comparative Examples 1 and 2, respectively.
[0088] During the test, apply a filter paper (diameter of about 0.8 cm) soaked with 50 μL of 0.01% capsaicin aqueous solution to each of the two sides of the subject's nasolabial fold, and when the subject judged that the sensory score of both sides of the nasolabial fold > 3 points (moderate sensation and above), remove the filter paper. Apply 50 μL of test sample by the operator to one side of the test area, no treatment on the other side (blank control). Before sample administration (after capsaicin irritation, before sample administration) and 0.5 min, 5 min, 10 min, 15 min, 20 min and 30 min after sample administration, subjects subjectively evaluated the burning pain in the nasolabial fold and rated it in 5 points, the rating standard is: 0 points corresponds to no sensation, 1 point corresponds to a just perceptible sensation, 2 points corresponds to a slightly perceptible sensation, 3 points corresponds to a moderately perceptible sensation, 4 points corresponds to a severe perceptible sensation and 5 points corresponds to pain. The specific results are shown in Figures 10 to 14. .
[0089] Figures 10 to 14 show that the burning sensation caused by capsaicin was improved in the comparative examples and the embodiment examples, the effect of relieving irritation in the embodiment examples was better than that in the comparative examples, among which Example 2 exhibits the best effect, with the burning pain significantly reduced compared to the blank control at 5min.
[0090] The processes for implementing the present invention are described through the above embodiments, but the embodiments of the present invention are not limited thereto, the person skilled in the art can achieve the purpose of the present invention by complying with the contents disclosed in the present invention, any modifications and variations proposed based on the idea of the present invention shall be included within the scope of protection of the present invention, and the scope of protection shall be defined by the appended claims.
