Abstract

A system for nucleic acid synthesis comprising: a nucleic acid synthesis device; at least one array of surface-bound oligonucleotides; at least one partially complementary oligonucleotide such that it will at least partially hybridise to one of the array of surface-bound oligonucleotides to generate an overlapping oligonucleotide having at least one overhanging region; means for extending the overlapping oligonucleotide; means for cleaving the extended oligonucleotide; means for assembling the at least one cleaved oligonucleotide to provide a synthesised nucleic acid product. The system further comprises means for extending the synthesised nucleic acid product and means for cleaving the extended synthesised nucleic acid product to generate a plurality of products. Optionally the plurality of surface-bound oligonucleotides comprise at least one repeating sequence of 8 base pairs in length. Preferably the overhang region is a 5’ overhang and the synthesised nucleic acid is a gene or gene fragment. Also disclosed is a method for synthesizing nucleic acids and a nucleic acid synthesis device comprising an input; an array of surface-bound oligonucleotides; a means for extending the surface-bound oligonucleotides; a means for cleaving the extended oligonucleotides; an assembly chamber and a means for assembling and an output.
