Abstract
This invention provides CLL1-CAR-T cells, a method for preparing them, and their use. The CLL1-CAR-T cells contain a chimeric antigen receptor. The chimeric antigen receptor comprises a single-domain antibody, a hinge domain, a transmembrane domain, and an intracellular signaling domain. The amino acid sequence of the single-domain antibody is from positions 22 to 150 of SEQ ID No. 1. The CLL1-VHH-1 CAR-T cells of this invention can secrete IFN-γ, a T cell-specific effector molecule, and can specifically kill CLL1+ target cells, significantly suppress the proliferation of tumor cells in mice, and significantly extend the survival time of mice, making them suitable for immunotherapy of CLL1-target-related diseases (e.g., acute myeloid leukemia).
