[0055] The "oxygen permeability" of an insert or material, Dk / t, is the rate at which oxygen passes through a particular insert or material of average thickness t [in mm] over the area being measured. [0056] The term "modulus" or "elastic modulus" with respect to a contact lens or material means the tensile modulus, i.e., Young's modulus, which is a measure of the embedment of a contact lens or material. The modulus can be measured according to the procedure described in Example 1.
[0057] "Raw state" refers to an insert obtained by casting of a polymerizable composition in a mold and which has not been subjected to extraction and / or hydration post-molding processes (i.e., has not been in contact with water or any organic solvent or any liquid after molding).
[0058] In general, the present invention relates to crosslinked materials that are rigid in a dry state at room temperature (about 22° C. to about 26° C.), have relatively high oxygen permeability and a high refractive index when fully hydrated, and are capable of becoming softer at temperatures above 32° C. Such materials are useful for making inserts in implantable contact lenses for the correction of corneal astigmatism, presbyopia, and color blind lenses, and for imparting photochromic characteristics to lenses.
[0059] The present invention is based in part on the discovery that when the polymerizable composition for producing the insert comprises as two main components (i.e., in a combination that constitutes at least about 70% by weight based on the total weight of the total polymerizable material), (1) at least one aryl acrylic monomer and (2) at least one vinyl-functional polysiloxane, each of which contains at least two vinyl groups directly bonded to one silicon atom, and at least 15 mole % of the siloxane units each have at least one phenyl substituent, and at least one vinyl-based crosslinker, an insert material can be obtained that has a relatively high oxygen permeability and a high refractive index, and is hard in a dry state (raw state) at room temperature. It is believed that by incorporating at least one of the described vinyl-functional polysiloxanes into the polymerizable composition for producing the insert material (crosslinked polymer material), the resulting insert material can have a relatively high oxygen permeability and a high refractive index. However, such an insert material is softer and more tacky at room temperature, and there are manufacturing and handling problems related to softness and tackiness. It is very difficult to open the mold in the green state and remove the cast-molded insert from the mold (i.e., "dry release and lens removal"). It has been found that by incorporating aryl acrylic monomers and / or crosslinkers into the polymerizable composition for making the insert, the resulting insert material can have a higher glass transition temperature (e.g., higher than 32°C), and is thereby hard in the dry state (i.e., green state) at room temperature. Due to their hard form in the dry state at room temperature, the manufacturing and handling problems associated with the softness and stickiness of the insert material can be significantly reduced or eliminated.
[0060] The present invention is also based in part on the discovery that by adding two different initiators (e.g., one thermal initiator such as Vazo-64 and one peroxide initiator such as ter-butyl peroxide 2-ethylhexyl carbonate) in such polymerizable compositions, two types of polymerization can be used in curing the polymerizable composition in forming the insert. The first type of polymerization (curing) is a free radical polymerization initiated by a thermal initiator (Vazo-64) at a temperature below 100°C. The other type of polymerization (curing) is a peroxide-activated curing system at higher temperatures (e.g., 120°C), which involves peroxide-induced free radical coupling between one vinyl and one methyl group of the siloxane unit. Insert materials can be obtained with the desired combination of properties such as oxygen permeability, refractive index, and elastic modulus suitable for implantable contact lenses for various applications. The performance of the implantable contact lenses can be optimized for a given application.
[0061] In one aspect, the present invention provides an insert comprising a crosslinked polymeric material for embedding in a silicone hydrogel contact lens, the insert comprising: (1) repeating units of at least one vinyl-functional polysiloxane, each of which contains at least two vinyl groups directly bonded to a silicon atom, and at least 15 mole percent of the siloxane units each having at least one phenyl substituent; (2) repeating units of at least one aryl acrylic monomer; and (3) repeating units of at least one vinyl crosslinker, wherein the combined amount of components (1) and (2) is at least about 70% by weight (preferably from about 75% to about 99% by weight, more preferably from about 80% to about 98% by weight, and even more preferably from about 100% to about 150% by weight) based on the total weight of the crosslinked polymeric material. The crosslinked polymer material preferably has a glass transition temperature of about 28°C or more (preferably about 30°C or more, more preferably about 32°C or more), and the crosslinked polymer material in a dry state has a water content of less than about 5% by weight (preferably about 4% by weight or less, more preferably about 3% by weight or less, even more preferably about 2% by weight or less), an oxygen permeability of at least about 40 barrers (preferably at least about 45 Barrers, more preferably at least about 50 Barrers, even more preferably at least about 55 Barrers), and a refractive index of at least about 1.47 (preferably at least about 1.49, more preferably at least about 1.51, even more preferably at least about 1.53).
[0062] It is understood that the weight percentage of each of the components of the crosslinked polymeric material of the insert of the present invention can be obtained based on the weight percentage of its corresponding polymerizable component (material) in the polymerizable composition for manufacturing the insert. Examples of such vinyl-functional polysiloxanes include, but are not limited to, vinyl-terminated polyphenylmethylsiloxane (e.g., PMV9925 from Gelest), vinylphenylmethyl-terminated phenylmethyl-vinylphenylsiloxane copolymer (e.g., PVV-3522 from Gelest), vinyl-terminated diphenylsiloxane-dimethylsiloxane copolymer (e.g., PDV-1625 from Gelest), or combinations thereof. Preferably, the vinyl-functional polysiloxane is vinyl-terminated polyphenylmethylsiloxane (e.g., PMV9925 from Gelest), vinylphenylmethyl-terminated phenylmethyl-vinylphenylsiloxane copolymer (e.g., PVV-3522 from Gelest), or combinations thereof.

The polymerizable composition contains, based on the total amount of the polymerizable composition, (a) about 20% by weight to about 79% by weight (preferably about 20% by weight to about 75% by weight, more preferably about 25% by weight to about 70% by weight, and even more preferably about 30% by weight to about 65% by weight) of at least one silicone-containing vinyl-based monomer and / or at least one silicone-containing vinyl-based crosslinker; (b) 20% by weight to about 79% by weight (preferably about 20% by weight to about 75% by weight, more preferably about 25% by weight to about 70% by weight, and even more preferably about 30% by weight to about 65% by weight) of a hydrophilic vinyl-based monomer; and (c) 0 to about 2.5% by weight (preferably 0 to about 2.0% by weight, more preferably about 1.0% by weight to about 1.5% by weight) of a hydroxypropyl acrylate or acrylate copolymer; (d) about 0.05% by weight to about 2.0% by weight (preferably about 0.1% by weight to about 2.0% by weight, more preferably about 0.2% by weight to about 1.5% by weight, and even more preferably about 0.3% by weight to about 1.2% by weight) of a free radical initiator; (e) 0 to about 15% by weight (preferably 0 to about 14% by weight, more preferably about 2% by weight to about 13% by weight, and even more preferably about 4% by weight to about 12% by weight) of mixed vinyl monomers; and (f) 0 to about 3.0% by weight, preferably about 0.1% by weight to about 2.5% by weight, and more preferably about 0.2% by weight to about 2.0% by weight of a UV absorbing vinyl monomer and / or a UV / HEVL absorbing vinyl monomer. Preferably, the sum of the amounts of polymerizable materials (a) and (b) is at least 70% by weight (preferably at least 75% by weight, more preferably at least 80% by weight, more preferably at least 85% by weight) based on the total amount of all polymerizable materials in the polymerizable composition.
[0097] Lens molds for producing contact lenses, such as SiHy contact lenses, are well known to those skilled in the art and are utilized, for example, for cast molding or spin casting. For example, molds (for cast molding) typically include at least two mold sections (or portions) or mold halves, namely, a first and a second mold half. The first mold half defines a first molding (or optical) surface, and the second mold half defines a second molding (or optical) surface. The first and second mold halves are configured to receive each other such that a lens-forming cavity is formed between the first and second molding surfaces. The molding surfaces of the mold halves are the cavity-forming surfaces of the mold and are in direct contact with the polymerizable composition.
[0098] Methods for producing mold sections for cast molding contact lenses are generally known to those skilled in the art. The method of the present invention is not limited to a specific method for forming a mold. In fact, any method for forming a mold can be used in the present invention. The first and second mold halves can be formed by various techniques, such as injection molding or lathing. Examples of suitable processes for forming mold halves are described in U.S. Patent Nos. 4,444,711; 4,460,534; 5,843,346; and 5,894,002.
[0099] To manufacture the mold for making contact lenses, virtually all materials known in the art for manufacturing molds can be used. For example, polymer-based materials such as polyethylene, polypropylene, polystyrene, PMMA, Topas® COC grade 8007-S10 (transparent amorphous copolymer of ethylene and norbornene, Ticona GmbH, Frankfurt, Germany and Summit, New Jersey) can be used.Other materials that allow UV light to pass through can also be used, such as quartz glass and sapphire.
[0100] According to the present invention, the insert can be placed in the mold and the silicone hydrogel lens-forming composition can be introduced (dispensed) into the cavity formed by the mold by any technique known to those skilled in the art. In a preferred embodiment, the insert is placed on the molding surface of the female mold half at a specified location, then a dispensing device dispenses a specific amount of the silicone hydrogel lens-forming composition into the female mold half with the insert thereon, then the male mold half is placed and the mold is sealed. When the mold closes, any excess amount of unpolymerized lens-forming material is pressed into the overflow provided on the female mold half (or on the male mold half), and the insert is immersed in the silicone hydrogel lens-forming composition in the mold.
[0101] After the insert of the present invention is placed into the mold and the silicone-hydrogel-lens-forming composition is dispensed into the mold, the closed mold containing the silicone-hydrogel-lens-forming composition is then thermally or actinically (preferably thermally initiated) cured (i.e., polymerized) to form a green implantable silicone hydrogel contact lens.
[0102] Actinic polymerization of the silicone-hydrogel-lens forming composition in the mold can be carried out by irradiating the closed mold having the silicone-hydrogel-lens forming composition therein with UV or visible light by any technique known to those skilled in the art.
[0103] Thermal polymerization of the silicone-hydrogel-lens-forming composition in the mold can be conveniently carried out in an oven at temperatures between 25 and 120° C., preferably between 40 and 100° C., as is well known to those skilled in the art. The reaction time can vary within wide limits, but is conveniently, for example, between 1 and 24 hours, or preferably between 2 and 12 hours. The silicone-hydrogel-lens-forming composition is previously degassed and then heated under an inert atmosphere, for example N 2 Alternatively, it is advantageous to carry out the copolymerization reaction under an Ar atmosphere.
[0104] In a preferred embodiment, after the silicone-hydrogel-lens forming composition in the mold in the oven has cured to form a green implantable silicone hydrogel contact lens, the temperature of the oven is increased to a post-cure temperature of about 105° C. or higher (preferably at least about 110° C., more preferably at least about 115° C., and even more preferably at least about 120° C.), and the flow rate of the nitrogen gas through the oven is increased to a second flow rate that is at least about 1.5 times (preferably at least about 2.0 times, more preferably at least about 3.0 times, and even more preferably at least about 4.0 times) the first flow rate.
[0105] The lens mold with the green embedded silicone hydrogel contact lens therein is subjected to a post-cure processing step in an oven at the post-cure temperature under nitrogen gas flow through the oven at a second flow rate for at least about 30 minutes (preferably at least about 60 minutes, more preferably at least about 90 minutes, and even more preferably at least about 120 minutes).
[0106] After the curing and optional post-curing steps, the steps of opening the mold (i.e., separating the male mold half from the female mold half with the green implantable silicone hydrogel contact lens attached onto one of the male and female mold halves) and extracting the lens (i.e., removing the green implantable silicone hydrogel contact lens from the mold half to which it was attached) are performed.
[0107] After the raw implantable silicone hydrogel contact lens is removed, it is typically extracted with an extraction medium known to those skilled in the art. The extraction liquid medium is any solvent that can dissolve the diluent, unpolymerized polymerizable material and oligomers in the raw implantable silicone hydrogel contact lens. Water, any organic solvent known to those skilled in the art or mixtures thereof can be used in the present invention. Preferably, the organic solvent used in the extraction liquid medium is water, buffered saline, C 1 ~C 3Alkyl alcohol, 1,2-propylene glycol, polyethylene glycol having a number average molecular weight of about 400 Daltons or less, C 1 ~C 6 alkyl alcohols or combinations thereof.
[0108] The extracted implantable silicone hydrogel contact lens can then be hydrated by any method known to those of skill in the art.
[0109] The hydrated implantable silicone hydrogel contact lenses may be further subjected to additional processes such as, for example, surface treatment, packaging in a lens package using a packaging solution known to those skilled in the art, and sterilization such as autoclaving at 118-124° C. for at least about 30 minutes.
[0110] Lens packages (or containers) are well known to those skilled in the art for autoclaving and storing soft contact lenses. Any lens package can be used in the present invention. Preferably, the lens package is a blister package comprising a base and a cover, the cover being removably sealed to the base, and the base comprising a cavity for receiving a sterile packaging solution and a contact lens.
[0111] The lenses are packaged in individual packages, sealed, and sterilized (e.g., by autoclaving at or above about 120° C. under pressure for at least 30 minutes) before being distributed to users. Those skilled in the art will be familiar with methods of sealing and sterilizing lens packages.
[0112] In a further aspect, the present invention provides an implantable silicone hydrogel contact lens comprising a silicone hydrogel material and an insert of the present invention therein (as described above in this application), wherein the silicone hydrogel material is a crosslinked material having a polymer matrix and comprises (a) repeat units of at least one silicone-containing vinylic monomer and / or at least one silicone-containing vinylic crosslinker, and (b) repeat units of at least one hydrophilic vinylic monomer, wherein the implantable silicone hydrogel contact lens, when fully hydrated, has a water content of from about 15% to about 70% by weight (preferably from about 15% to about 65% by weight, more preferably from about 20% to about 65% by weight, and even more preferably from about 25% to about 60% by weight) in a fully hydrated state.

[0172] The above disclosure enables those skilled in the art to carry out the present invention.Various modifications, alterations and combinations can be made to the various embodiments described herein. In order to better enable the reader to understand specific embodiments and their advantages, reference to the following examples is proposed. It is intended that the present specification and examples are considered to be illustrative. EXAMPLES
[0173] Example 1 Oxygen permeability measurement Unless otherwise specified, oxygen transmission rate (Dk / t), intrinsic (or edge-corrected) oxygen permeability (Dk i Or Dk c) is determined according to the procedure described in ISO 18369-4.
[0174] equilibrium water content The equilibrium water content (EWC) of a contact lens is determined as follows.
[0175] The amount of water (expressed as weight percent) present in hydrated hydrogel contact lenses that have been fully equilibrated with saline is measured at room temperature. The lenses are quickly stacked and after wiping the lenses with a cloth, the stack of lenses is transferred to an aluminum pan on an analytical balance. The number of lenses in each sample pan is typically five. Record the hydrated weight of the pan + lenses. Cover the pan with aluminum foil. Place the pan in a laboratory oven at 100 ± 2 °C and dry for 16-18 hours. Remove the pan + lenses from the oven and cool in a desiccator for at least 30 minutes. Remove one pan from the desiccator and discard the aluminum foil. Weigh the pan + dry lens sample on an analytical balance. Repeat for all pans. The wet and dry weights of the lens samples can be calculated by subtracting the weight of the empty weighing pan.
[0176] Refractive index The refractive index (RI) of the inserts is determined with an Abbe transmission laboratory refractometer Reichert Abbe Mark III at 25° C. The inserts are fully equilibrated in PBS saline prior to the measurements.
[0177] Elasticity Modulus The storage modulus (Young's modulus) of the inserts is determined using a TA RSA-G2 DMA (Dynamic Mechanical Analyzer). The inserts are cut into strips 3.08 mm wide using a Precision Concept dry lens cutter. Five thickness values are measured within a gauge length of 6.5 mm. The strips are attached to the instrument using metal grips. An oscillatory temperature ramp test using a linear heating rate of 10°C to 50°C at 2°C / min is applied to the inserts to monitor the material's response to temperature increase at a constant frequency of 1 Hz, constant amplitude of 0.5% strain and a sampling rate of 10.0 pts / s. The storage modulus (E'), loss modulus (E'') and tan δ data are calculated by TRIOS software.
[0178] The elastic modulus of silicone hydrogel material or contact lenses is determined using an MTS Insight instrument. The contact lenses are first cut into 3.12 mm wide strips using a Precision Concept two-stage cutter. Five thickness values are measured within a gauge length of 6.5 mm. The strips are attached to the grips of the instrument and submerged in PBS (phosphate buffered saline) at a controlled temperature of 21±2°C. Typically, a 5N load cell is used for testing. A constant force and velocity is applied to the sample until it breaks. Force and displacement data are collected by the TestWorks software. The elastic modulus value is calculated by the TestWorks software, which is the slope or tangent of the stress versus strain curve near zero elongation in the elastic deformation region.
[0179] Glass transition temperature The glass transition temperature (Tg) of the insert is defined as the peak of tan δ from a dynamic temperature ramp test by using a TA RSA-G2 DMA (Dynamic Mechanical Analyzer).
[0180] Delamination Implantable silicone hydrogel contact lenses are tested for potential delamination using an Optimec instrument or Optical Coherence Tomography (OCT).
[0181] Regardless of the evaluation method, the contact lenses are staged for a minimum of 12 hours at room temperature after the autoclave run and before the delamination study.
[0182] After the required staging time has been met, the fully hydrated contact lens is placed into the "V" graticule assembly of the Optimec instrument (Model JCF; OPTIMEC England). After the contact lens has set under the influence of gravity, the front surface of the contact lens is carefully examined for any signs of circular patterns. Delamination will show up as a circular pattern on the Optimec image.
[0183] OCT (Thorlabs Spectral Domain Optical Coherence Tomography (Model-II)) can also be utilized to study delamination. OCT allows non-invasive imaging of contact lenses to obtain high resolution cross-sectional images. For this purpose, after meeting the minimum staging requirements, the contact lens is removed from its blister and immersed in PBS solution for a minimum of 30 minutes until equilibration. A cuvette with a "V" block feature is then filled approximately 3 / 4 with fresh PBS solution and the contact lens is transferred to the cuvette using a Q-tip. The lens is allowed to float freely in the "V" shape at the bottom of the cuvette and the entire contact lens is scanned in 10 degree increments. Delamination appears in the OCT image as air pockets at the spacing surfaces of the insert and carrier.
