[0007] [Non-Patent Document 1] Remington: The Science and Practice of Pharmacy, 20th ed.; Gennaro, AR, Ed.; Lippincott Williams & Wilkins: Philadelphia, PA, 2000 [Non-Patent Document 2] Ashworth, S., et al., Evaluation of 11C-GSK189254 as a novel radioligand for the H3 receptor in humans using PET. J Nucl Med, 2010. 51(7):1021~9 Summary of the Invention [Problem to be solved by the invention]
[0008] Generally speaking, it is desirable to identify drug candidates that are as potent as possible: greater efficacy generally comes with greater selectivity for the target, lower risk of off-target binding, and therefore greater clinical safety. It is therefore desirable to identify drug candidates that are as potent as possible and have the lowest possible effective dose. [Means for solving the problem]
[0009] Despite the disclosed doses of H3-ligands disclosed in WO2006 / 117609, the present inventors have now discovered that H3-ligands of the formula:

[0047] The following examples are included as illustrative, non-limiting embodiments of the present invention. EXAMPLES
[0048] Example 1 PK results in humans C of compound (A) given orally once daily at doses of either 60 or 90 μg max were reached approximately 2.5-3.5 h after dosing on days 1 and 10, averaging 121 and 171 pg / mL, and 172 and 344 pg / mL after a single dose and after repeated doses of 60 and 90 μg, respectively. The anti-insomnia activity of compound (A) at doses as low as 30 μg daily was evidenced by a delay in falling asleep when going to bed at night, whereas repeated treatment with 90 μg once daily for 10 days led to some insomnia. Mean t 1 / 2 was determined on the 10th day after multiple dosing and was approximately 33 h. Total body clearance (Clss / F) was reduced by 65% and 74% after repeated dosing of 60 and 90 μg of Compound (A), respectively. Serum levels of Compound (A) after the 60 μg dose were 42% higher at C max and 73% higher AUC 0~24h As reflected in the % accumulation after repeated dosing (day 10) was higher than after a single dose (day 1). The % accumulation after repeated dosing was more pronounced at the 90 μg dose, and C max is 101% higher, and AUC 0~24h was 113% higher. Average C max and AUC 0~24h Based on dose-normalized ratios, Compound (A) serum exposure in human volunteers tended to increase greater than dose proportionally from od 60 to 90 μg on days 1-10. Steady state appears to be achieved after 7-8 days of treatment at the 60 μg dose.
[0049] Example 2 Determination of the active dose of compound (A) The therapeutic activity of a histamine H3-receptor antagonist / inverse agonist drug can be predicted by assessing the degree of occupancy of its target in the brain, namely the H3 receptor. The degree of occupancy by this agent can be reliably measured in humans by their displacement of the binding of selective radioactive probes labeled with short half-life radioisotopes such as 11C. Thus, PET imaging allows the non-invasive measurement of receptor occupancy. The imaging method requires the injection of a positron-emitting radiotracer that binds to the receptor and the subsequent measurement of this binding using a PET scanner. Radioligands selective for the H3 receptor have been developed, among which [ 11 C]GSK189254 is a high affinity histamine H3 receptor antagonist with good brain penetration and a terminal half-life of 1.6 + 0.4 hours. This tracer has recently been used to characterize the dose-response occupancy of novel histamine H3 antagonists, and [ 11 This provides validation of the ability of 11C-GSK189254 to measure H3 receptors using PET in humans (Ashworth, S., et al., Evaluation of 11C-GSK189254 as a novel radioligand for the H3 receptor in humans using PET. J Nucl Med, 2010. 51(7):1021-9). This method allows predicting the therapeutic dose of new H3 receptor inverse agonists / antagonists in two steps: 1 / determine the degree of receptor occupancy associated with a dose that ensures a therapeutic effect of a known drug A and 2 / evaluate the dose of a new drug B that ensures the same receptor occupancy as the therapeutic dose of A. This strategy was applied by comparing H3R occupancy in healthy volunteers receiving the approved and marketed H3R inverse agonist pitolisant at its maximum oral therapeutic dose of 40 mg with occupancy by compound (A) given at several dosages. H3R occupancy after 40 mg of pitolisant in six healthy volunteers was found to be 82.33±8.71%. As shown below, very similar H3R occupancy was found at a dose of 60 μg of Compound (A), and at a dose of 30 μg of Compound (A), a very high degree of occupancy was still present after repeated treatment once daily for 10 days, especially when steady state was reached.
[0050] [Table 3]
[0051] Since dose-response studies of pitolisant show therapeutic activity in various pathologies (narcolepsy, Parkinson's disease or excessive daytime sleepiness associated with obstructive sleep apnea) at oral doses of 20-40 mg, the above results lead to the conclusion that the therapeutic dose of compound (A) is in the region of 30-60 μg. The latter compound is therefore deemed to be 1,000 times more potent than the marketed drug pitolisant.
[0052] Melting points are determined on a Buchi capillary melting point apparatus. Proton NMR spectra were recorded on a Varian 400 MHz NMR instrument. Chemical shifts δ are expressed in ppm. The following abbreviations are used to describe the signal patterns: s=singlet, d=doublet, t=triplet, q=quartet, m=multiplet, ms=massif. The recorded spectra are consistent with the proposed structures.
[0053] Infrared spectra (range 4000-450 cm-1) were recorded on a THERMO Electron Corporation Nicolet 380 FT-IR equipped with an attenuated total reflection system. Wavelengths are in cm-1.
[0054] Example 3 (3S)-4-{4-[3-(3-methylpiperidin-1-yl)propoxy]phenyl}pyridine 1-oxide dihydrochloride tetrahydrate Charge (3S)-4-{4-[3-(3-methylpiperidin-1-yl)propoxy]phenyl}pyridine 1-oxide dihydrochloride (210 g) and water (200 g) into an evaporation vessel. Set the temperature of the bath in the evaporation system to about 40° C. and stir the mixture until all precipitate has dissolved. Evaporate the water until the product separates as a solid form. The evaporation vessel containing the product is transferred to a vacuum tray dryer and the product is dried at 30° C. The product is transferred from the evaporation vessel to a practical drying vessel and drying is continued at 30° C. until the water content of the product is 15.3±0.5% by mass.
[0055] Example 4 (3S)-4-{4-[3-(3-methylpiperidin-1-yl)propoxy]phenyl}pyridine 1-oxide dihydrochloride tetrahydrate Charge acetone (10 kg) and (3S)-4-{4-[3-(3-methylpiperidin-1-yl)propoxy]phenyl}pyridine 1-oxide (2.5 kg). Set the temperature to 30-40°C. Stir the mixture until the (3S)-4-{4-[3-(3-methylpiperidin-1-yl)propoxy]phenyl}pyridine 1-oxide is dissolved, filter through a pressure filter and concentrate. Add a solution of hydrochloric acid (32%, 1.84 kg, 2.1 equiv.) in water (2 kg). Set the temperature to 22-30°C and add acetone (6-7 L). After seeding, add acetone to a total volume of 20 kg at 22-30°C. Stir the suspension at 18-24°C for 1-2 hours and filter. Rinse the cake with a mixture of acetone (3.5 kg) and water (240 g). The product is dried in a vacuum tray dryer under nitrogen flow (Tmax=30° C.) until the water content is 15.0-16.5% by mass.
[0056] Example 5 (3S)-4-{4-[3-(3-methylpiperidin-1-yl)propoxy]phenyl}pyridine 1-oxide base (3S)-4-{4-[3-(3-methylpiperidin-1-yl)propoxy]phenyl}pyridine 1-oxide dihydrochloride is treated with ethyl acetate, water, concentrated sodium hydroxide, and potassium carbonate to liberate the base. The phases are filtered through a pad of Celite and decanted. The organic phase is washed with saturated aqueous sodium chloride, dried over magnesium sulfate, filtered, and concentrated under reduced pressure to give (3S)-4-{4-[3-(3-methylpiperidin-1-yl)propoxy]phenyl}pyridine 1-oxide base. IR (main bands): 3258, 3102, 2930, 2356, 1603, 1470, 1411, 1287, 1232, 1177, 1059, 947, 819, 696, 568, 509, 456.
[0073] Example 22 Comparative pharmacokinetic data in mice Compounds were administered to male Swiss mice at doses approximating 1 mg base / kg. Plasma and brain samples were taken 24 hours later. Results, normalized to 1 mg base / kg, are shown in Table 1.
[0074] [Table 4]
[0075] To some extent, the exemplified compounds show similar absorption rates. The ratios of AUC of the examples to the AUC of the dihydrochloride tetrahydrate are shown in Table 2. The ratios all fit within the interval of 0.6 to 1.2, which is consistent with the similarity of experimental error associated with the tests.
[0076] [Table 5]
[0077] The striking results are for the pamoate salt (Example 8). Pamoate (embonic acid) salts are widely used to make sustained release drugs (see, for example, Zypadhera / Zyprexa relprevv, the pamoate salt of olanzapine given only every 2-4 weeks). Example 8 shows a similar AUC and similar time to Cmax compared to the dihydrochloride tetrahydrate (Example 4). Therefore, low doses of the claimed compound A are possible, regardless of its salts and / or solvates therefrom.
