Abstract
Description
Detailed Description of the Invention
[0001] [Technical field] The present invention relates to water-dispersible, edible particles containing vitamins. Such particles can be used to prepare foods or beverages.
[0002] [Background of the invention] Folic acid has been known for some time. Folic acid is also called vitamin B9.
[0003] Folic acid is commercially available. EP 1026166 B1 discloses a process for preparing folic acid.
[0004] Folic acid is water soluble. Example 1 of EP 0954986B1 discloses a beverage containing folic acid.
[0005] Fortified beverages can be made by dispersing instant powders in water, fruit juice, or the like. Prior to use, such instant powders are stored in bags. To be commercially viable, the sealed bag must have a long shelf life along with its contents.
[0006] Various methods are known for producing instant powders. The preferred method is extrusion. Unfortunately, vitamin extrudates containing folic acid have a short or very short shelf life. In the presence of riboflavin and / or thiamine, the shelf life of extrudates containing folic acid is even shorter.
[0007] Shelf life can be improved by choosing different manufacturing methods such as spray drying, etc. However, these alternative manufacturing methods trigger high capital expenditures (CAPEX) and have other disadvantages such as the need for organic solvents and / or low output per operating hour.
[0008] There is a need for water-dispersible, edible powders that contain vitamin B9 and are storage stable even in the presence of riboflavin and / or thiamine. The manufacture of such powders should be low cost, simple, and / or solvent-free.
[0009] [Summary of the Invention] The problem underlying the present invention is solved by extruding a composition comprising vitamin B9, at least one pH buffer, at least one excipient, optionally at least one lubricant and water.
[0010] The addition of a pH buffer significantly reduces the degradation of vitamin B9 during storage of the extrudates. Surprisingly, the degradation of vitamin B9 is reduced even in the presence of other members of the B vitamin group. Normally, folic acid is rapidly degraded in the presence of riboflavin and / or thiamine.
[0011] A solid pH buffer can be used to reduce the degradation of vitamin B9 in the extrudate. This is surprising since very little water is used during the extrusion process. If the composition being extruded contains a lot of water, the resulting extrudate would need to be thoroughly dried. Extensive drying increases energy costs and reduces output per operating hour.
[0012] Solid state reactions are notoriously slow, so it is surprising that a solid pH buffer can be used in a relatively fast process using only a short extruder and little water.
[0013] [Detailed Description of the Invention] The present invention relates to extruded strands. After extrusion, the strands obtained are cut into small pieces that are subsequently dried. The multiple extrudates thus obtained are water-soluble or water-dispersible coarse powders. To increase the flowability of the powder, the extrudates can be spheronized before drying.
[0014] [Extrusion strand] The extruded strands of the present invention are obtained by extruding a composition comprising vitamin B9, at least one pH buffer, at least one excipient, optionally at least one lubricant, and water.
[0015] In the context of the present invention, vitamin B9 is a synonym for folic acid. In one embodiment of the present invention, the term "folic acid" includes congeners of folic acid, such as tetrahydrofolic acid, methyltetrahydrofolic acid, methenyltetrahydrofolic acid, folinic acid, folates, and folacin.
[0016] The extruded strands of the composition preferably contain at least one further micronutrient. Preferably, said at least one further micronutrient is selected from the group consisting of minerals, fat-soluble vitamins, water-soluble vitamins, amino acids, polyunsaturated fatty acids and carotenoids. The water-soluble vitamins according to the invention are preferably vitamin B1 (thiamine), vitamin B2 (riboflavin), vitamin B3 (niacin, niacinamide), vitamin B5 (pantothenic acid), vitamin B6 (pyridoxine, pyridoxamine, pyridoxal), vitamin B7 (biotin), vitamin B9 and vitamin B12 (cyanocobalamin, hydroxycobalamin, methylcobalamin).
[0017] In a preferred embodiment of the invention, the extruded strands of the invention are obtained by extruding a composition comprising vitamin B9, vitamin B1, vitamin B2, at least one pH buffer, at least one excipient, optionally at least one lubricant and water.
[0018] In the context of the present invention, a "pH buffer" is a mixture of a weak acid and a salt of its conjugate base or a mixture of a weak base and a salt of its conjugate acid. In a preferred embodiment of the present invention, the pH buffer is a mixture of a weak acid and a salt of its conjugate base, both of which are solid at room temperature and standard atmosphere. By "buffer" is meant that the pH of an aqueous solution containing a suitable amount of pH buffer does not change appreciably when a small amount of strong acid or strong base is added. In the context of the present invention, non-edible pH buffers are excluded. In the context of the present invention, "edible" refers to a composition having a food grade or feed grade. A food grade composition is one intended for human consumption. Also excluded are pH buffers that do not reduce the degradation of vitamin B9 of the extruded strands when stored in a sealed aluminum bag at 30° C. and 65% humidity for 3 months. The pH buffer of the present invention is preferably a mixture of (i) ascorbic acid and (ii) an edible salt of ascorbic acid, such as sodium ascorbate. Less preferred pH buffers are: - a mixture of citric acid and edible citrates, - Carbonic acid and mixtures of edible carbonates It is.
[0019] In a preferred embodiment of the invention, the extruded strands of the invention are obtained by extruding a composition comprising vitamin B9, at least one pH buffer, at least one excipient, at least one lubricant and water, said at least one pH buffer being a mixture of (i) ascorbic acid and (ii) an edible salt of ascorbic acid. In an even more preferred embodiment of the invention, the extruded strands of the invention are obtained by extruding a composition comprising vitamin B9, vitamin B1, vitamin B2, at least one pH buffer, at least one excipient, at least one lubricant and water, said at least one pH buffer being a mixture of (i) ascorbic acid and (ii) an edible salt of ascorbic acid.
[0020] The extruded strands comprise at least one "excipient". In a preferred embodiment of the invention, the excipient is (i) edible and (ii) water soluble or water dispersible. The extruded strands preferably comprise at least 50% by weight, more preferably at least 60% by weight, and most preferably at least 70% by weight, of at least one excipient, based on the total weight of the extruded strands and not including water. Preferred excipients are starch (such as wheat starch) and semolina (such as durum wheat semolina). Preferably, a mixture of starch and semolina is used as the excipient.
[0021] Thus, the extruded strands are preferably obtained by extruding a composition comprising vitamin B9, at least one pH buffer, at least one lubricant, water and at least 50% by weight of at least one excipient, based on the total weight of the extruded strand and excluding water, said at least one excipient being preferably a mixture of starch and semolina, such as a mixture of wheat starch and durum wheat semolina. In an even more preferred embodiment of the present invention, the extruded strands are preferably obtained by extruding a composition comprising vitamin B9, vitamin B1, vitamin B2, at least one pH buffer, water, at least one lubricant and at least 50% by weight of at least one excipient, based on the total weight of the extruded strand and excluding water, said at least one excipient being preferably a mixture of starch and semolina, such as a mixture of wheat starch and durum wheat semolina.
[0022] Preferably, the extruded strands comprise at least one lubricant. Preferably, said at least one lubricant is liquid at room temperature. Preferred lubricants are edible oils such as medium chain triglycerides (MCT), corn oil, vegetable oil and coconut oil. MCT is the most preferred lubricant. Thus, in the most preferred embodiment of the present invention, the extruded strands of the present invention are obtained by extruding a composition comprising vitamin B9, vitamin B1, vitamin B2, at least one pH buffer, at least one excipient, at least one lubricant such as MCT and water, said at least one pH buffer being a mixture of (i) ascorbic acid and (ii) an edible salt of ascorbic acid.
[0023] The diameter of the extruded strand depends on the extruder die. The extruder die has at least one hole. In most cases, the extruder die has multiple holes, each of which has the same diameter. The diameter of the extruder hole is preferably 0.1 mm to 2 mm, more preferably 0.3 mm to 1.2 mm, and most preferably 0.4 mm to 0.8 mm.
[0024] The shaping of the composition into strands is only possible if the composition to be shaped has a certain degree of plasticity. According to the invention, the plasticity of the extruded composition can be adjusted by the amount of water. The extruded strands preferably contain 15% to 60% by weight of water, more preferably 20% to 55% by weight of water, and most preferably 25% to 45% by weight of water, based on the total weight of the extruded strand.
[0025] The plasticity of the extruded composition can also be adjusted by increasing the extrusion temperature. The temperature of the extruder die is preferably 50° C. to 120° C., more preferably 55° C. to 90° C., most preferably 60° C. to 80° C. Temperatures above 120° C. are not preferred, as such high temperatures may have a negative effect on the vitamins.
[0026] The length of the extruded strands can vary. In a continuous extrusion process, the strands do not have a definite length. Moreover, the length of the extruded strands depends on the cutting process. Usually, the strands are cut at the die face. Instead of cutting at the die face, cutting can also be done downstream.
[0027] [Multiple extrusions] Plurality means more than one. Thus, the expression "plurality of extrudates" refers to two or more extrudates. Preferably, the multiple extrudates of the present invention are powders having a mass between 1 g and 10 kg.
[0028] The extrudates of the present invention are preferably water-soluble or water-dispersible. Therefore, the extrudates are preferably essentially free of fat, essentially free of fat-soluble proteins, and / or essentially free of organic solvents. In this context, the expression "essentially free" means less than 1% by weight, preferably less than 0.8% by weight, more preferably less than 0.5% by weight, and most preferably less than 0.2% by weight, based on the total weight of the extrudates. In the context of the present invention, the term "fat" does not include liquids such as liquid lubricants or any other oils.
[0029] The multiple extrudates of the present invention can be obtained by cutting the extrudate strands described herein. The size of the individual extrudates is therefore preferably determined by the diameter of the extruder hole. In a less preferred embodiment of the present invention, the processing conditions are selected such that the extrudate expands as it leaves the die. The size of the individual extrudates is also determined by the cutting step. Preferably, the cutting is performed such that the length of the extrudate approximately corresponds to the diameter of the extruder hole.
[0030] After cutting, the individual extrudates are cylindrical in shape. To increase the flowability of the multiple extrudates, the still wet extrudates can optionally be shaped in a spheronizer.
[0031] Preferably, the extrudate is at least partially dried after cutting and optional spheronization. A fluidized bed dryer or any other suitable device may be used for drying. Typically, drying is performed at a temperature of 40°C to 80°C for 30 to 90 minutes. After drying, the extrudate may still contain some residual moisture. The extrudate of the multiple extrudates preferably contains less than 10% by weight of water, more preferably less than 8% by weight, most preferably less than 5% by weight, based on the total weight of the extrudate.
[0032] The extrudates of the multiple extrudates contain at least one pH buffer. Thus, when a predetermined amount of the extrudates is dissolved or dispersed in deionized water, a certain pH is reached. The pH of the liquid obtainable by dispersing 1 g of the multiple extrudates in 40 ml of deionized water is preferably 4.6-6, more preferably 4.65-5.2, most preferably 4.7-5 at room temperature (i.e. 23° C.). Preferably, said 1 g is measured after a drying step has been performed.
[0033] Without wishing to be bound by theory, it is believed that the pH ranges indicated above reduce the degradation of vitamin B9, vitamin B1 and vitamin B2 during storage in extrudates containing the three B vitamins. The pH ranges indicated above can be achieved with a mixture of (i) ascorbic acid and (ii) an edible salt of ascorbic acid. However, the pH ranges can also be achieved with other edible pH buffers, such as citric acid or calcium carbonate based pH buffers. pH buffers that are not suitable to reach the pH ranges indicated above are excluded.
[0034] The amount and type of pH buffer is selected to provide suitable storage stability even in the presence of riboflavin and / or thiamine.
[0035] The vitamin B9 content of the extrudates of the plurality of extrudates, after 3 months of storage in sealed aluminum bags at 30°C and 65% humidity, is preferably at least 70%, more preferably at least 75%, and most preferably at least 80% of the vitamin B9 content of the extrudates before storage.
[0036] [Use of pH buffers] The present invention also relates to the use of a pH buffer to increase the shelf life of an extrudate comprising vitamin B9. In a preferred embodiment, the present invention relates to the use of a pH buffer to increase the shelf life of an extrudate comprising vitamin B9 and at least one further water-soluble vitamin, preferably selected from the group consisting of vitamin B1 and vitamin B2. Most preferably, the present invention relates to the use of a pH buffer to increase the shelf life of an extrudate comprising vitamin B9, vitamin B1 and vitamin B2.
[0037] Surprisingly, when the pH buffer of the present invention is included in an extrudate obtainable by extruding a composition comprising vitamin B9, vitamin B1, vitamin B2, at least one excipient and water, not only vitamin B9 but also vitamin B1 and vitamin B2 are significantly less degraded during storage.
[0038] The pH buffers described herein are preferably solid at room temperature and standard atmosphere. More preferably, the pH buffers described herein are a mixture of (i) ascorbic acid and (ii) an edible salt of ascorbic acid. Most preferably, the pH buffers described herein are a mixture of (i) ascorbic acid and (ii) sodium ascorbate. In this embodiment, the weight ratio between ascorbic acid and sodium ascorbate is preferably 1:1 to 1:5, more preferably 1:1 to 1:4, and most preferably 1:2.5 to 1:3.5.
[0039] Therefore, a preferred embodiment of the present invention relates to the use of a pH buffer for increasing the shelf life of an extrudate containing vitamin B9, said pH buffer being a mixture of (i) ascorbic acid and (ii) sodium ascorbate, wherein the weight ratio between ascorbic acid and sodium ascorbate is preferably 1:1 to 1:5, more preferably 1:1 to 1:4, and most preferably 1:2.5 to 1:3.5. [Brief description of the drawings]
[0040] [Figure 1] Results of Example 3 are shown. The relative content of vitamin B9 in the extrudates of Example 1 was measured after extrusion (time point: 0 months) and after storage periods of 1 month, 2 months and 3 months. On the y-axis, the measured vitamin B9 content is shown as a percentage of the calculated vitamin B9 content. Example: 90% vitamin B9 organic stability means that 10% of the vitamin B9 used to make the extrudate is no longer present. Extrudate #132 has a higher vitamin B9 organic stability than the other extrudates. Extrudate #132 contains a pH buffer of the present invention. [Diagram 2] Figure 2 shows the extrudates after drying and not spheronized. The individual extrudates have a diameter of 0.5mm to 2mm. The extrudates are like a coarse powder and are flowable.
[0041] [Example] Example 1 (Production of extrudate containing folic acid) Extrudate #132 was prepared as follows.
[0042] Powders containing folic acid, pH buffer (ascorbic acid and sodium ascorbate), vitamin mix (vitamins B1, B2, B6, B3 and B12), excipients (wheat starch and durum wheat semolina) and optionally lubricants were gravimetrically fed into the first barrel of a lab-scale co-rotating twin screw extruder (Thermo Fisher Scientific, HAAKE Polylab OS equipped with a PTW16 / 40 OS twin screw extruder) (Brabender Technologie). The extruder consisted of six (electrically heated and water cooled) barrels and a die head (12 or 21 x 0.5 mm), the diameter of the screw was 16 mm and the length to diameter ratio was 40. Deionized water was injected into the second barrel. The temperatures of barrels 2 to 6 and the die head were such that adiabatic extrusion conditions were reached. The powder premix was fed at 500 g / h and 60 g / h of water was added. The strands were then extruded. To obtain multiple extrudates, the extrudate strands were cut directly at the die face by two rotating knives of the die head. After cutting at the die face, the solid extrudates were dried in a laboratory-scale fluidized bed dryer (Retsch TG 200) at 50° C. for 60 minutes to obtain a final moisture content of 6 wt. % based on the total weight of the extrudate.
[0043] Extrudates #133, #131 and #124 were prepared similarly to extrudate #132. The exact compositions of all extrudates are shown in Table 1 below.
[0044] [Table 1]
[0045] In Table 1 above, weight percent refers to the total weight of each extrudate, not including any residual moisture. The vitamin mix was the same for all extrudates and included thiamine mononitrate (vitamin B1), riboflavin (vitamin B2), pyridoxine hydrochloride (vitamin B6), niacinamide (vitamin B3) and crystalline vitamin B12. Excipients and optional lubricants were also the same for all extrudates.
[0046] [Example 2 (pH measurement)] The pH of extrudate #132 was measured as follows.
[0047] A number of extrudates #132 with a total mass of 1 g were added to 40 ml of deionized water at room temperature and stirred for 1 h to ensure that all extrudates were dispersed. The pH was then measured using a Metrohm 744 pH meter.
[0048] The pH of extrudates #133, #131 and #124 were measured, as was the pH of extrudate #132. The results of these measurements are shown in Table 2 below.
[0049] [Table 2]
[0050] [Example 3 (storage stability)] The relative content of vitamin B9 in the extrudate of Example 1 was measured after extrusion (time point: 0 months) and after storage periods of 1 month, 2 months and 3 months. Storage conditions: 30°C, 65% humidity in sealed aluminum bags.
[0051] The following method was used.
[0052] 2 g of each extrudate was weighed into a volumetric flask and dissolved in mobile phase at pH 7. After a given time, an aliquot (10 μl) was injected onto a Nucleosil 100-7 C18 (Macherey & Nagel) column connected to an Agilent HPLC system equipped with a UV / VIS detector. The mobile phase consisted of 13.5% acetonitrile and 86.5% 0.25N pentasulfonic acid / 0.25N Na heptasulfonic acid, and was diluted in 1.4 ml portions. -1 The flow rate was kept constant at 290 nm. Vitamin B9 was detected at a wavelength of 290 nm, and the signal area was calculated using a standard graph.
[0053] The results of the measurements are shown in Figure 1. Figure 1 shows that vitamin B9 is not significantly degraded during storage when the extruded composition is supplemented with the pH buffer system of the present invention.
