Abstract

The present invention provides a method for improving glycemic control in a patient in need thereof by administering tirzepatide or a pharmaceutically acceptable salt thereof. The present invention also provides a method for improving weight management in a patient in need thereof by administering tirzepatide or a pharmaceutically acceptable salt thereof. Furthermore, the present invention also provides a method for treating a condition selected from atherosclerosis, chronic kidney disease, NAFLD, and NASH. Furthermore, a method for preventing diabetes or inducing remission of diabetes, comprising administering tirzepatide or a pharmaceutically acceptable salt thereof, is also provided. Technical Field

[0001] The present invention provides a method of using a novel dosage of a glucose-dependent insulinotropic polypeptide (GIP) / glucagon-like peptide-1 (GLP1) dual agonist peptide, tirzepatide, or a pharmaceutically acceptable salt thereof for treating type 2 diabetes (T2D). The present invention also provides a method of using a novel dosing regimen of a GIP / GLP1 dual agonist peptide, tirzepatide, or a pharmaceutically acceptable salt thereof for treating type 2 diabetes. Furthermore, the present invention provides a novel medical use of tirzepatide, or a pharmaceutically acceptable salt thereof. More specifically, the present invention provides a method for treating a condition selected from the group consisting of chronic kidney disease, atherosclerosis, non-alcoholic fatty liver disease ("NAFLD"), and non-alcoholic steatohepatitis ("NASH"). In another embodiment, the present invention provides a novel dosing regimen comprising an escalating dose (i.e., a dose lower than the desired maintenance dose) and a maintenance dose. In another embodiment, the present invention provides a novel dosing regimen comprising one or more escalating doses and one or more maintenance doses. The present invention provides a novel dosing regimen comprising administering at least one escalating dose approximately once a week for at least about 4 weeks, and thereafter administering at least one maintenance dose approximately once a week for at least about 4 weeks. Claims

1. A pharmaceutical composition comprising tildepagliflozin or a pharmaceutically acceptable salt thereof for use in treating type 2 diabetes in patients in need of treatment for type 2 diabetes, wherein an escalating dose for at least one administration is administered once a week for at least 4 weeks, and after said escalating dose, a maintenance dose for at least one administration is administered once a week for at least 4 weeks, characterized in that the escalating dose is selected from the group consisting of 2.5 mg, 7.5 mg and 12.5 mg of tildepagliflozin or a pharmaceutically acceptable salt thereof, the first escalating dose administered is 2.5 mg, the maintenance dose after the escalating dose is an increment of 2.5 mg, and the maintenance dose is selected from the group consisting of 5.0 mg, 10.0 mg and 15.0 mg of tildepagliflozin or a pharmaceutically acceptable salt thereof.

2. The composition according to claim 1, wherein the escalating dose is 2.5 mg and the maintenance dose is 5.0 mg.

3. The composition according to claim 1, wherein the escalating dose is 7.5 mg and the maintenance dose is 10.0 mg.

A pharmaceutical composition comprising tildepazide or a pharmaceutically acceptable salt thereof for use in preventing diabetes in patients in need of diabetes prevention, wherein at least one escalating dose is administered once a week for at least 4 weeks, and after said escalating dose, at least one maintenance dose is administered once a week for at least 4 weeks, characterized in that the escalating dose is selected from the group consisting of 2.5 mg, 7.5 mg and 12.5 mg of tildepazide or a pharmaceutically acceptable salt thereof, the first escalating dose administered is 2.5 mg, the maintenance dose after the escalating dose is an increment of 2.5 mg, and the maintenance dose is selected from the group consisting of 5.0 mg, 10.0 mg and 15.0 mg of tildepazide or a pharmaceutically acceptable salt thereof.
