[0001] The present invention relates to a cosmetic composition for enhancing exfoliation.
[0002] This application claims priority based on Korean Patent Application No. 10-2018-0083069 filed on July 17, 2018, and Korean Patent Application No. 10-2019-0063704 filed on May 30, 2019, and the contents disclosed in the specifications and drawings of those applications are incorporated herein in their entirety. [0003] The stratum corneum is the outermost layer of the skin and is composed of large, lamellar lipid layers containing keratinocytes and ceramides. The stratum corneum protects against external substances and functions as a barrier to prevent moisture loss from the interior. A normal stratum corneum consists of approximately 15 to 20 layers. The action of serine proteases activated on the outer surface of the stratum corneum causes the degradation of coniodesmosomes, eventually resulting in the stratum corneum's complete detachment from the skin. This process takes approximately 15 to 20 days for normal cells. In addition to genetic disorders such as ichthyosis, aging, psoriasis, and acne-prone skin can also cause the stratum corneum to thicken, resulting in keratinocyte stratification.
[0004] Stratification of the stratum corneum is generally known to be induced by factors such as a decrease in skin moisture retention, a decrease in the expression or activity of serine proteases, ultraviolet rays, and decreased cell activity. To resolve this stratification, methods such as physical exfoliation using scrubs, exfoliation using chemical preparations, and increased physiological cell activity are typically used. These exfoliation methods aim to resolve appearance problems such as rough skin and dullness by artificially removing outer keratinocytes and replacing the outermost stratum corneum with new keratinocytes from the inside, thereby eliminating keratinocyte stratification.
[0005] The reduction of stratum corneum stratification through enhanced exfoliation also provides additional benefits, such as the removal of fine wrinkles and dark spots and the improvement of rough skin, and is therefore utilized in a variety of products. Alpha hydroxy acid (AHA), beta hydroxy acid (BHA), and polyhydroxy acid (PHA) are typically used. However, the mode of action of AHA, BHA, and PHA is to penetrate the stratum corneum and release hydrogen ions, weakening coniodesmosome bonds and resulting in exfoliation. A low pH is essential for easy penetration into the stratum corneum and high hydrogen ion transfer when applied at low pH. Therefore, low pH can cause side effects such as stinging, itching, and erythema, and research is underway to overcome these side effects. However, when the pH is adjusted to a weakly acidic to neutral range to mitigate the side effects of such skin irritation, the activity is reduced by more than 80%. Furthermore, when the agent undergoes chemical transformation, the efficacy is offset, making it difficult to obtain the substantial benefits of exfoliation. Therefore, there is a growing need for new exfoliants that exhibit exfoliating efficacy similar to that of AHA under acidic conditions even at a weakly acidic or neutral range.
[0006] Therefore, the inventors of the present invention have identified a composition containing serine and carnitine in Korean Patent Application No. 10-2017-0165268 based on their previous research, which shows efficacy greater than that of AHA at the same concentration under neutral or weakly acidic conditions, and have confirmed that it causes relatively little or no irritation, and have provided it as an excellent new exfoliating material. [0008] The present invention aims to provide a cosmetic composition that selects a composition that maintains the accessibility of the exfoliating agent to keratinocyte-binding proteins, thereby fully demonstrating the added effect of enhancing exfoliation of keratin and causing no irritation.

[0019] To achieve the above object, the present invention provides a cosmetic composition for enhancing exfoliation of skin, which contains an exfoliating agent and is free of a polyol moisturizing agent or contains a polyol moisturizing agent in an amount of 10 wt% or less based on the total weight of the composition. The present invention will now be described in detail with reference to the drawings.
[0020] The present inventors attempted to produce commercially available cosmetics that have exfoliation-enhancing properties by incorporating an exfoliant into a cosmetic composition containing various additives such as surfactants, fragrances, preservatives, humectants, thickeners, and antioxidants. However, they found that the exfoliation-enhancing efficiency was significantly lower than expected depending on the concentration of the exfoliant. During their research, they discovered that the cause of the reduced exfoliation function was moisturizing ingredients among various cosmetic additives, particularly moisturizing ingredients with polyol groups, which reduced the accessibility of the exfoliating ingredients to the keratin-binding proteins. As a result, the exfoliation ability was reduced because the exfoliating ingredients did not affect the keratin-binding proteins. They proposed a solution to this problem, leading to the completion of the present invention.
[0021] The present invention provides a cosmetic composition that allows an exfoliant, particularly serine or carnitine, to maintain its effectiveness. More specifically, the present invention aims to provide a serine or carnitine cosmetic that provides superior exfoliation effects at a weakly acidic to neutral pH without irritation compared to existing ingredients.

Therefore, the present invention has found that the exfoliating effect of serine or carnitine is rapidly reduced when serine or carnitine contains a polyol-containing moisturizer, such as one or more polyols such as glycerin, dipropylene glycol (DPG), and 1,3-butylene glycol (BG), at a concentration above a certain level. Evaluation of exfoliation of pig skin Existing clinical evaluation methods are time-consuming and difficult to conduct experiments on all the different concentrations and pH conditions of various raw materials. In addition, it is difficult to conduct experiments on all the possible compositions that can be combined with each raw material. Therefore, there is a growing need for non-clinical methods for evaluating exfoliation.
[0043] Therefore, in the present invention, the keratin peeling evaluation method described in Journal of the European Academy of Dermatology and Venereology, 2014, 28, 415-423 was used. Specifically, pig skin was biopsied and the sample to be tested was added. The number of keratin layers that were removed was then measured using a cell counter, and the results were plotted using a sample containing only water as the negative control (0%) and 10% gluconolactone (pH 4) as the positive control (100%).

The results of the comparison of the reduction in exfoliating efficacy depending on the concentration of glycerin are shown in Figure 7. The results are calculated by using serine alone as the positive control (100%).
[0060] The experimental results showed that the exfoliating efficacy level decreased in a similar manner as the glycerin concentration increased in both the 0.5% serine experimental group (dashed line in Figure 7) and the 5% serine experimental group (solid line in Figure 7). [0074] Example 2. Method for evaluating the promotion of keratin turnover using the DHA staining method The DHA staining method for assessing keratin turnover rate was performed on 10 healthy male subjects aged 20-40 years using the method described in Dermatology 1993, 186, 133-137. Before application of the sample, the color of the inner lower and upper arm regions of the subjects was measured using a chroma meter. Approximately 0.4 ml of 10% dihydroxyacetone (DHA) was applied to a 1.5 x 1.5 cm gauze pad and applied to the inner upper and lower arm regions for 8 hours. After 48 hours, the color of the brown areas stained by DHA was measured and compared with the color before sample application. The sample was then applied twice daily, and the degree of depigmentation was measured using a chroma meter. The time required for the skin to return to its original color was also measured. TT (Turnover Time) 100% refers to the time it takes for the existing stratum corneum to be 100% replaced with new stratum corneum, and in this experiment, the time it takes for skin pigmented by DHA to return to its original state was calculated using a relative comparison analysis with regression analysis. Claims
1. A composition for enhancing exfoliation, comprising serine as an exfoliant, A composition comprising a polyol humectant in an amount of more than 0 to 2% by weight.
2. The composition according to claim 1 , except that the exfoliant contains one or more active ingredients selected from tropolone and its derivatives.
3. The composition of claim 1 , wherein the composition is a cosmetic composition.
4. 2. The composition according to claim 1, wherein the composition comprises a polyol humectant in an amount of more than 0 to 0.5% by weight.
5. 2. The composition according to claim 1, wherein the polyol moisturizer is at least one selected from the group consisting of glycerin, dipropylene glycol, butylene glycol, glycereth-26, methyl gluceth-20, and pentylene glycol.
6. 2. The composition according to claim 1, wherein the exfoliant is contained in an amount of 0.001 to 20% by weight based on the total weight of the composition.
7. The composition described in claim 1, characterized in that the cosmetic composition has a pH of 5 to 8.
8. A product comprising the composition of any one of claims 1 to 7.
9. A method for enhancing exfoliation, comprising applying to skin a cosmetic composition comprising serine as an exfoliant, The cosmetic composition comprises a polyol moisturizer in an amount of more than 0 and not more than 2% by weight.
10. The method according to claim 9 , except that the exfoliant contains one or more active ingredients selected from tropolone and its derivatives.
