Description

[Technical Field]

[0001] The present invention relates to the use of diethylamino hydroxybenzoyl hexyl benzoate in anti-aging care compositions containing at least one fragrance. [Background technology]

[0002] Ultraviolet (UV) radiation causes harmful effects on human skin. In addition to the acute effect of sunburn on the skin, UV radiation is also known to increase the risk of skin cancer. Furthermore, prolonged exposure to UV-A and UV-B rays can cause phototoxic and photoallergic reactions on the skin, which may accelerate skin aging. Therefore, UV filters are suitable ingredients for anti-aging care compositions.

[0003] To protect human skin from ultraviolet rays, various sunscreen UV filters (also called UV absorbers) exist, including UV-A filters, UV-B filters, and broadband filters. These filters are added to sunscreens or cosmetic compositions, for example. UV filters are either organic or inorganic, particulate or non-particulate compounds, all of which have high absorption effects within the UV light range. Generally, UV light can be divided into UV-A rays (320-400 nm) and UV-B rays (280-320 nm). Depending on the position of the absorption maximum, UV filters are divided into UV-A filters and UV-B filters. When a UV filter absorbs both UV-A and UV-B rays, it is called a broadband absorber.

[0004] Since 2006, the European Commission has recommended that all sunscreens or cosmetic compositions should have a UV-A protection factor of at least one-third of the labeled sun protection factor (SPF), where the sun protection factor primarily refers to UV-B protection.

[0005] 2-(4'-Diethylamino-2'-hydroxybenzoyl)benzoic acid hexyl ester (also known as diethylaminohydroxybenzoylhexyl benzoate or DHHB) is an effective UV-A filter and is used to protect the skin from ultraviolet rays (e.g., in sunscreens). 2-(4'-Diethylamino-2'-hydroxybenzoyl)benzoic acid hexyl ester is represented below and is also referred to below as the compound of formula (I):

[0006] [ka]

[0007] EP2276452 and EP2276449 relate to cosmetic preparations containing DHHB and at least one fragrance to test for odor stability.

[0008] Anti-aging care compositions often contain additional fragrances that contribute to a pleasant odor. However, fragrances tend to react with free radicals when exposed to UV light. This can cause radical chain reactions that destabilize the formulation itself. Therefore, when considering protection against UV radiation, the effectiveness of UV filters in such unstable formulations is also reduced. Furthermore, the presence of hydroperoxides and other peroxide free radicals is known to induce contact skin diseases and severe allergic reactions (Bjoerkman et al., Contact Dermatitis, 70, 2014, 129-138). [0010] [Non-Patent Document 1] Bjoerkman et al., Contact Dermatitis, 70, 2014, 129-138 Summary of the Invention

[0011] Since anti-aging care compositions are often applied to body parts directly exposed to ultraviolet light, there is a continuing need to provide an anti-aging care composition comprising at least one fragrance that can be safely applied to body parts exposed to ultraviolet light. In this regard, the object of the present invention was to provide an anti-aging care composition comprising at least one fragrance that reduces the formation of free radicals.Furthermore, the object of the present invention was to provide an anti-aging care composition comprising at least one fragrance that reduces lipid peroxidation. Finally, the object of the present invention was to provide a body care composition for use in a method for preventing skin reactions caused by fragrances.

[0012] It has surprisingly been found that at least one of the above objectives can be achieved by the use of diethylaminohydroxybenzoylhexyl benzoate in a body care composition comprising at least one fragrance.

Unless otherwise defined, all technical and scientific terms used herein have the same meaning as commonly understood by one of ordinary skill in the art.

[0035] The term "compound of formula (I)" refers to 2-(4'-diethylamino-2'-hydroxybenzoyl)benzoic acid hexyl ester (also known as diethylaminohydroxybenzoylhexyl benzoate or DHHB).

[0036] The term "body care composition" refers to any product suitable for application to the human body, such as anti-aging care compositions, sunscreen compositions, bath and shower products, preparations containing fragrances and odoriferous substances, hair care products, dentifrices, decorative preparations, and cosmetic formulations containing active ingredients. Sunscreen compositions are preferred.

[0037] The term "sunscreen composition" or "sunscreen agent" refers to any topical product that reflects and / or absorbs a specific portion of ultraviolet radiation. Therefore, the term "sunscreen composition" should be understood to include not only sunscreen compositions but also any cosmetic composition that provides UV protection. The term "topical product" refers to a product that is applied to the skin and may be, for example, a spray, lotion, cream, oil, foam, powder, or gel. According to the present invention, the sunscreen composition may contain one or more active agents, such as organic UV filters, as well as other ingredients or additives, such as emulsifiers, emollients, viscosity modifiers, stabilizers, preservatives, or fragrances.

[0038] The term "anti-aging care composition" refers to any topical product that can absorb, and further reflect and scatter a certain portion of ultraviolet radiation, and is used as a daily care product for the human body, for example, face, body, or hair.Anti-aging care compositions can contain one or more active agents, such as organic or inorganic UV filters, and other ingredients or additives, such as emulsifiers, emollients, viscosity adjusters, stabilizers, preservatives, or fragrances. It should be understood that the anti-aging care composition according to the present invention provides a reduction in oxidative degeneration and limits the skin aging process induced by free radicals.

[0046] The term "emollient" relates to cosmetic preparations used to protect, moisturize and lubricate the skin. The word emollient comes from the Latin "mollire" (to soften). Generally, emollients prevent the evaporation of water from the skin by forming an occlusive coating. They can be divided into different groups according to their polarity index.

[0047] The term "photostability" refers to the ability of a UV filter or any other molecule exposed to sunlight to remain stable upon irradiation. In particular, this means that the compound does not undergo decomposition processes upon UV irradiation.

[0048] The term "critical wavelength" is defined as the wavelength at which the area under the UV protection curve (% protection vs. wavelength) is 90% of the total area under the curve in the UV region (280-400 nm). For example, a critical wavelength of 370 nm indicates that the protection of a sunscreen composition is not limited to UV-B wavelengths, i.e., wavelengths between 280 and 320 nm, but extends up to 370 nm, such that 90% of the total area under the protection curve in the UV region reaches 370 nm.

[0049] The term "ultraviolet filter" or "UV filter" as used herein refers to an organic or inorganic compound that can absorb and / or reflect ultraviolet radiation generated by sunlight. UV filters can be classified as UV-A filters, UV-B filters, or broadband filters based on their UV protection curves. In the context of this application, broadband filters can also be referred to as UV-A filters, since they also provide UV-A protection. In other words, preferred UV-A filters also include broadband filters.

[0050] The definition of "broadband" protection (also called broad spectrum or broad-spectrum protection) is based on "critical wavelengths." Broad-spectrum coverage must provide UV-B and UV-A protection. According to U.S. requirements, a critical wavelength of at least 370 nm is required to achieve broad-spectrum protection. Furthermore, the European Commission recommends that all sunscreens or cosmetic compositions should have a UVA protection factor of at least one-third of the labeled sun protection factor (SPF); for example, if a sunscreen composition has an SPF of 30, the UVA protection factor must be at least 10.

[0073] To measure free radical generation in the formulation after UV irradiation, the concentration of photostable spin markers added to the test samples was followed by electron spin resonance (ESR) spectroscopy before and after UV irradiation of the test samples. The photostable spin markers react readily with free radicals generated in the formulation after UV irradiation.

[0074] The test fragrance is incorporated into a formulation at 0.1% (w / w). The formulation is then diluted with distilled water (1:10 dilution ratio) and an aqueous solution of the spin marker PCA (2,2,5,5-tetramethylpyrrolidine N-oxyl) is added to obtain a spin marker concentration of 0.01 mM. Knowing the concentration of the spin trap PCA in the formulation, the amount of PCA lost can be calculated (Herrling et al., SOFW-Journal, 5-2014, 140, 10-16). The sample is inserted into a capillary quartz tube and ESR measurements are performed 20 minutes before and after UV irradiation. The irradiance as an integral over the spectral range of the solar simulator is, for example, E = 23.5 W / m for UVB. 2 , 180W / m for UVA 2 It could be.

[0075] The percentage of UV-induced free radicals in the formulation containing the tested fragrance serves as an indicator of the sensitivity of said fragrance.According to the above test, the formulation containing the sensitive fragrance contains at least 1%, preferably at least 2%, more preferably at least 3%, even more preferably at least 4%, even more preferably at least 5%, even more preferably at least 10%, even more preferably at least 15%, even more preferably at least 20%, particularly at least 50% of UV-induced free radicals after 20 minutes of UV irradiation.

[0104] With regard to the above-described preferred embodiment of the anti-aging care composition, particularly with regard to the UV filters DHHB and BMDBM, the present inventors surprisingly discovered that BMDBM remains stable in the composition. This diketone is highly susceptible to photodecomposition, which leads to the decomposition of BMDBM. Therefore, compositions containing BMDBM require the addition of a stabilizer, and photostable UV absorbers such as ethylhexyl-2-cyano-3,3-diphenylacrylate (INCI octocrylene) are commonly used. Therefore, the present inventors surprisingly discovered that BMDBM can be stabilized in octocrylene-free sunscreen compositions by using the UV filter DHHB, even in the presence of at least one fragrance.

It is understood that a photostable body care composition remains stable when exposed to ultraviolet light, and in particular, the compounds contained therein do not undergo degradation processes when exposed to ultraviolet light.

[0128] The present invention is further illustrated by the following examples. [Example]

[0129] method Electron spin resonance (ESR) measurements were carried out using a highly sensitive X-band benchtop electron spin resonance spectrometer MiniScope MS300 supplied by Magnettech GmbH (Berlin, Germany).

[0130] UV irradiation of the samples was carried out using a xenon arc lamp solar simulator, Newport-ORIEL product line 81260 (Newport Solar Simulator - Product Specifications, USA), equipped with a 300 W xenon lamp.

[0131] 2,2,5,5-Tetramethylpyrrolidine N-oxyl was purchased from Merck. Synthetic sebum was purchased from wfk Testgewebe GmbH (Brügen, Germany).

[0132] Example 1: Quantification of free radicals generated in a formulation containing fragrance

[0133] To measure free radical generation in the formulations after UV irradiation, the concentration of a photostable spin marker added to the test samples was followed by electron spin resonance (ESR) spectroscopy before and after UV irradiation of the test samples. This photostable spin marker reacts rapidly with the free radicals generated in the formulations after UV irradiation.

[0134] The formulation incorporated 0.1% (w / w) of the fragrance to be tested (compare Table 1). The formulation was then diluted with distilled water (1:10 dilution factor) and an aqueous solution of the spin marker PCA (2,2,5,5-tetramethylpyrrolidine N-oxyl) was added to obtain a spin marker concentration of 0.01 mM. Knowing the concentration of spin trap PCA in the formulation, the amount of PCA lost was calculated. Samples were inserted into capillary quartz tubes and ESR measurements were performed before and after 20 minutes of UV irradiation. The irradiance as an integral over the spectral range of the solar simulator was E = 23.5 W / m for UVB; 2 , 180W / m for UVA 2 It was.

[0135] The percentage of UV-induced free radicals in formulations containing the test fragrances is shown in Table 1.

[0136] [Table 1]

[0137] Example 2: Effect of UV filter systems to reduce fragrance-induced sebum oxidation

[0138] Squalene in sebum is highly susceptible to oxidation. Squalene by-products in the form of peroxides affect cellular and tissue physiology. In this lipid oxidative degradation, free radicals steal electrons from lipids. Lipid peroxidation forms reactive aldehydes, such as malondialdehyde (MDA), as natural by-products, which can be used as markers of lipid peroxidation to assess oxidative damage.

[0139] Based on this principle, the following in vitro method was developed to evaluate sebum lipid oxidation.

[0140] The test formulations were prepared as follows.

[0141] The ingredients for Part A and the ingredients for Part B provided in Tables 2.1, 2.2, 2.3, and 2.4 below for each test sunscreen composition were combined and heated to 80°C, whereupon Part A was added to Part B under stirring and homogenized. The compositions were then cooled to room temperature under stirring. Parts C, D, and E, if present, were added under stirring, and the pH of each composition was adjusted to 5.5-7.00 with NaOH (30%), as needed.

[0142] The remaining fragrances in Table 1 were incorporated into formulations according to Table 2.1.

[0143] [Table 2-1] [Table 2-2] [Table 2-3] [Table 2-4] [Table 2-5] [Table 2-6]

[0144] The test formulation was applied at 2 mg / cm2 onto an artificial sebum film in a Petri dish. 2 The formulation film was applied in an amount of 1.25 mW / cm. A sebum film was first prepared by adding 1 g of synthetic sebum to 100 mL of heptane to obtain a sebum solution. 1 mL of this sebum solution was deposited on a Petri dish (6 cm diameter) and placed in a nitrogen chamber until the solvent had completely evaporated. The formulation film was dried in the nitrogen chamber and then irradiated with 1.25 mW / cm. 2 The samples were exposed to UVA for 2 hours.

[0145] Sebum was collected by adding heptane to a Petri dish and dissolving the sebum film using an ultrasonic bath. Finally, the MDA level, which corresponds to the oxidation product derived from sebum, was determined using the TBARS (thiobarbituric acid reactive substances) method. MDA in the sample was reacted with thiobarbituric acid (TBA) to produce the MDA-TBA adduct. The MDA-TBA adduct was quantified colorimetrically at 532 nm.

[0146] In this regard, 0.3 mL of sebum sample solution, 0.2 mL of 8 weight % (w%) SDS solution, 1.5 mL of 20 volume % (v%) acetic acid, 1.5 mL of TBA solution, and 0.5 mL of DI water were added to a tub, and the mixture was then heated at 95°C for 60 minutes, cooled to room temperature in an ice bath for 15 minutes, and centrifuged at 3000 rpm for 5 minutes. The absorbance of the reaction solution was measured at 532 nm using a UV spectrophotometer to detect the presence of TBA-MDA adducts.

[0147] [Table 3] TIFF0007819107000010.tif96170

[0148] As can be seen from Table 3, the lowest MDA levels are obtained when the formulation contains DHHB.
