[Diagram 2] FIG. 2 is a front cross-sectional view that typically shows a blood collection container according to a second embodiment of the present invention. [Diagram 3] FIG. 3 is a front cross-sectional view that typically shows a blood collection container according to a third embodiment of the present invention. DETAILED DESCRIPTION OF THE PREFERRED EMBODIMENTS
[0028] The present invention will be described in detail below.
[0029] The blood collection container according to the present invention comprises a blood collection container body, a specific gravity liquid contained in the blood collection container body, and a blood separation material contained in the blood collection container body, wherein the specific gravity liquid contains an amino acid.
[0030] Since the blood collection container according to the present invention is provided with the above-mentioned configuration, it is possible to increase the amount of mononuclear cells collected. Since the specific gravity liquid in the blood collection container according to the present invention contains amino acids, it is possible to increase the amount of mononuclear cells collected compared to a conventional blood collection container in which the specific gravity liquid does not contain amino acids.
[0031] As a blood collection container, a sterilized blood collection container may be used. For example, blood collection containers used as medical equipment products must be sterilized. The present inventors have found that it is difficult to increase the amount of mononuclear cells collected in a conventional sterilized blood collection container, and that it is particularly difficult to increase the amount of mononuclear cells collected in a conventional blood collection container sterilized by gamma rays. The present inventors have conducted extensive research and found that 1) the reason why it is difficult to increase the amount of mononuclear cells collected in a conventional blood collection container is that precipitates are generated in the specific gravity liquid during sterilization, causing the properties of the specific gravity liquid to change, and 2) the use of a specific gravity liquid containing an amino acid makes it difficult for precipitates to be generated in the specific gravity liquid. The reason why precipitates are unlikely to be generated in the specific gravity liquid by using a specific gravity liquid containing an amino acid is presumed to be because radicals generated during sterilization are inactivated by the amino acid, but is not limited to this. The blood collection container of the present invention can suppress the occurrence of precipitates in the specific gravity liquid and effectively suppress changes in the properties of the specific gravity liquid, thereby enabling a large amount of mononuclear cells to be recovered even after sterilization.
[0032] Furthermore, the blood collection container according to the present invention allows easy separation of mononuclear cells from blood. For example, the blood collection container according to the present invention allows separation of mononuclear cells by collecting blood in the blood collection container and then centrifuging the blood collection container. By appropriately adjusting the specific gravity of the specific gravity liquid in the blood collection container according to the present invention, the mononuclear cells can be suspended in the specific gravity liquid layer after centrifugation, or a layer containing mononuclear cells can be formed above the specific gravity liquid layer.
[0033] The blood collection container according to the present invention will be described in detail below. In this specification, "(meth)acrylic" means either or both of "acrylic" and "methacrylic".
[0034] (specific gravity liquid) The blood collection container includes a specific gravity liquid contained in the blood collection container body. The specific gravity liquid may also be called a density gradient solution. The specific gravity liquid contains an amino acid. The specific gravity liquid preferably contains a component (X) that is solid at 25°C other than an amino acid. The specific gravity liquid preferably contains water.
[0035] <Amino acids> The specific gravity liquid contains an amino acid. The amino acid may be used alone or in combination of two or more kinds.
[0036] Examples of the amino acids include basic amino acids such as histidine and arginine; neutral amino acids such as α-alanine, β-alanine, proline, glycine, cysteine, serine, leucine, isoleucine, and phenylalanine; and acidic amino acids such as glutamic acid.
[0037] The amino acids preferably include basic amino acids or neutral amino acids, more preferably β-alanine, proline, histidine, or glycine, even more preferably β-alanine, and particularly preferably β-alanine. In this case, precipitation is less likely to occur in the specific gravity liquid during sterilization of the blood collection container, and therefore the amount of mononuclear cells recovered can be further increased.
[0038] In the 100% by weight of the specific gravity liquid, the content of the amino acid is preferably 0.001% by weight or more, more preferably 0.01% by weight or more, even more preferably 0.1% by weight or more, preferably 10% by weight or less, more preferably 8% by weight or less, even more preferably 5% by weight or less, even more preferably 3% by weight or less, and particularly preferably 2% by weight or less. When the content of the amino acid is above the lower limit and below the upper limit, precipitation is less likely to occur in the specific gravity liquid during sterilization of the blood collection container, and therefore the amount of mononuclear cells recovered can be further increased. In addition, in the 100% by weight of the specific gravity liquid, the content of the amino acid may be 2% by weight or more, or may be 3% by weight or more.
[0039] In the specific gravity liquid, the weight ratio of the content of the amino acid to the content of the component (X) (amino acid content / component (X) content) is preferably 0.001 or more, more preferably 0.005 or more, even more preferably 0.01 or more, preferably 10 or less, more preferably 5 or less, even more preferably 1 or less, even more preferably 0.5 or less, and particularly preferably 0.3 or less. When the weight ratio (amino acid content / component (X) content) is equal to or more than the lower limit and equal to or less than the upper limit, precipitation is less likely to occur in the specific gravity liquid during sterilization of the blood collection container, and therefore the amount of mononuclear cells recovered can be further increased.
[0040] <Component (X) that is solid at 25℃ and is different from amino acids> The specific gravity liquid preferably contains a component (X). The component (X) is a component that is solid at 25° C. and is different from amino acids. The component (X) is a component that is solid at 25° C. The component (X) is different from amino acids. The component (X) is a component that is solid at 25° C. As the component (X), a conventionally known component (a component that is solid at 25° C. and is different from amino acids) used in specific gravity liquids can be used. Only one type of the component (X) may be used, or two or more types may be used in combination.
[0041] In conventional specific gravity liquids containing component (X), when the blood collection container is sterilized, precipitates are generated in the specific gravity liquid, and the properties of the specific gravity liquid are easily changed. In contrast, in the blood collection container of the present invention, even when the specific gravity liquid contains component (X), the specific gravity liquid contains amino acids, so precipitates are unlikely to be generated in the specific gravity liquid. As a result, the blood collection container of the present invention can increase the recovery amount of mononuclear cells even after sterilization.
[0042] Examples of the component (X) include polysucrose, sodium diatrizoate, sodium metrizoate, silica microparticles, sodium chloride, sodium ascorbate, hydroxyethyl starch, sucrose, dextran, and iodixanol.
[0043] The component (X) is preferably polysucrose, sodium diatrizoate, sodium metrizoate, silica microparticles, sodium chloride, sodium ascorbate, hydroxyethyl starch, sucrose, dextran, or iodixanol, and more preferably polysucrose, sodium diatrizoate, silica microparticles, or sodium chloride, which can further increase the recovery amount of mononuclear cells.
[0044] Polyscrub: The component (X) preferably contains polysucrose. The specific gravity liquid preferably contains polysucrose. As the polysucrose, conventionally known polysucrose used in specific gravity liquids can be used. In conventional specific gravity liquids containing polysucrose, precipitation is likely to occur in the specific gravity liquid during sterilization of blood collection containers. However, in the present invention, even when the specific gravity liquid contains polysucrose, precipitation is unlikely to occur in the specific gravity liquid, and therefore the recovery amount of mononuclear cells can be further increased.
[0045] The number average molecular weight of the polysucrose is preferably 150,000 or more, more preferably 300,000 or more, and is preferably 700,000 or less, more preferably 550,000 or less.
[0046] The number average molecular weight means a number average molecular weight calculated as pullulan, measured by gel permeation chromatography (GPC).
[0047] In the specific gravity liquid (100% by weight), the content of the polysucrose is preferably 0.1% by weight or more, more preferably 1% by weight or more, even more preferably 5% by weight or more, particularly preferably 8% by weight or more, preferably 50% by weight or less, more preferably 30% by weight or less, and even more preferably 20% by weight or less. When the content of the polysucrose is equal to or more than the lower limit and equal to or less than the upper limit, the amount of mononuclear cells recovered can be further increased.
[0048] In the specific gravity liquid, the weight ratio of the amino acid content to the polysucrose content (amino acid content / polysucrose content) is preferably 0.001 or more, more preferably 0.005 or more, even more preferably 0.01 or more, preferably 5 or less, more preferably 1 or less, even more preferably 0.5 or less, and particularly preferably 0.1 or less. When the weight ratio (amino acid content / polysucrose content) is equal to or more than the lower limit and equal to or less than the upper limit, precipitation is less likely to occur in the specific gravity liquid during sterilization of the blood collection container, and therefore the amount of mononuclear cells recovered can be further increased.
[0049] In 100% by weight of the component (X), the content of the polysucrose is preferably 5% by weight or more, more preferably 15% by weight or more, even more preferably 25% by weight or more, particularly preferably 40% by weight or more, preferably 90% by weight or less, more preferably 80% by weight or less, and even more preferably 70% by weight or less. When the content of the polysucrose is equal to or more than the lower limit and equal to or less than the upper limit, the amount of mononuclear cells recovered can be further increased.
[0050] Sodium diatrizoate: The component (X) preferably contains sodium diatrizoate. The specific gravity liquid preferably contains sodium diatrizoate. In a conventional specific gravity liquid containing sodium diatrizoate, precipitation is likely to occur in the specific gravity liquid during sterilization of a blood collection container. However, in the present invention, even when the specific gravity liquid contains sodium diatrizoate, precipitation is unlikely to occur in the specific gravity liquid, and therefore, the recovery amount of mononuclear cells can be increased.
[0051] In the specific gravity liquid (100% by weight), the content of the sodium diatrizoate is preferably 0.5% by weight or more, more preferably 1% by weight or more, even more preferably 5% by weight or more, preferably 25% by weight or less, more preferably 20% by weight or less, and even more preferably 15% by weight or less. When the content of the sodium diatrizoate is equal to or more than the lower limit and equal to or less than the upper limit, the amount of mononuclear cells recovered can be further increased.
[0052] In the specific gravity liquid, the weight ratio of the content of the amino acid to the content of sodium diatrizoate (amino acid content / sodium diatrizoate content) is preferably 0.0001 or more, more preferably 0.001 or more, even more preferably 0.005 or more, preferably 5 or less, more preferably 1 or less, and even more preferably 0.5 or less. When the weight ratio (amino acid content / sodium diatrizoate content) is equal to or more than the lower limit and equal to or less than the upper limit, precipitation is less likely to occur in the specific gravity liquid during sterilization of the blood collection container, and therefore the amount of mononuclear cells recovered can be further increased.
[0053] In 100% by weight of the component (X), the content of the sodium diatrizoate is preferably 15% by weight or more, more preferably 25% by weight or more, even more preferably 35% by weight or more, preferably 95% by weight or less, more preferably 90% by weight or less, and even more preferably 85% by weight or less. When the content of the sodium diatrizoate is equal to or more than the lower limit and equal to or less than the upper limit, the recovery amount of mononuclear cells can be further increased.
[0054] It is more preferable that the above component (X) contains polysucrose and sodium diatrizoate. Conventional specific gravity liquids containing these components tend to produce precipitates during sterilization of blood collection containers, but the specific gravity liquid of the present invention is less likely to produce precipitates even when it contains these components, and therefore the recovery amount of mononuclear cells can be further increased.
[0055] In the specific gravity liquid (100% by weight), the total content of the polysucrose and the sodium diatrizoate is preferably 1% by weight or more, more preferably 5% by weight or more, even more preferably 10% by weight or more, preferably 80% by weight or less, more preferably 60% by weight or less, and even more preferably 40% by weight or less. When the total content is equal to or more than the lower limit and equal to or less than the upper limit, the amount of mononuclear cells recovered can be further increased.
[0056] In the specific gravity liquid, the weight ratio of the content of the amino acid to the total content of the polysucrose and the sodium diatrizoate (amino acid content / total content of polysucrose and sodium diatrizoate) is preferably 0.001 or more, more preferably 0.005 or more, even more preferably 0.01 or more, preferably 10 or less, more preferably 5 or less, even more preferably 1 or less, even more preferably 0.5 or less, and particularly preferably 0.1 or less. When the weight ratio (amino acid content / total content of polysucrose and sodium diatrizoate) is equal to or more than the lower limit and equal to or less than the upper limit, precipitation is less likely to occur in the specific gravity liquid during sterilization of the blood collection container, and therefore the amount of mononuclear cells recovered can be further increased.
[0057] In the specific gravity liquid, the content of the polysucrose is preferably greater than the content of the sodium diatrizoate, which can further increase the recovery amount of mononuclear cells.
[0058] In 100% by weight of the component (X), the total content of the polysucrose and the sodium diatrizoate is preferably 80% by weight or more, more preferably 90% by weight or more, even more preferably 95% by weight or more, particularly preferably 99% by weight or more, and most preferably 100% by weight. When the total content is equal to or more than the lower limit, the amount of mononuclear cells recovered can be further increased. In addition, in 100% by weight of the component (X), the total content of the polysucrose and the sodium diatrizoate may be 100% by weight or less, less than 100% by weight, or 99% by weight or less.
[0059] Silica Microparticles: The component (X) preferably contains silica fine particles. As the silica fine particles, conventionally known silica fine particles used in specific gravity liquids can be used. The silica fine particles are preferably silica fine particles coated with polyvinylpyrrolidone. [0060] The average particle size of the silica fine particles is preferably 5 nm or more, more preferably 10 nm or more, even more preferably 15 nm or more, and preferably 50 nm or less, more preferably 40 nm or less, even more preferably 30 nm or less. [0061] In the specific gravity liquid (100% by weight), the content of the silica fine particles is preferably 0.1% by weight or more, more preferably 1% by weight or more, even more preferably 10% by weight or more, preferably 65% by weight or less, more preferably 45% by weight or less, and even more preferably 25% by weight or less. When the content of the silica fine particles is equal to or more than the lower limit and equal to or less than the upper limit, the recovery amount of mononuclear cells can be further increased.

[0066] <Water> The specific gravity liquid preferably contains water. In the specific gravity liquid, the water serves as a solvent. The water may be contained in a commercially available product containing the component (X).
[0067] In 100% by weight of the specific gravity liquid, the content of the water is preferably 50% by weight or more, more preferably 60% by weight or more, even more preferably 70% by weight or more, particularly preferably 80% by weight or more, preferably 99% by weight or less, more preferably 95% by weight or less, and even more preferably 90% by weight or less.
[0068] <Other ingredients> The specific gravity liquid may contain other components in addition to the above-mentioned components (amino acids, component (X) and water). Examples of the other components include glycerol. Each of the other components may be used alone or in combination of two or more.
[0069] <Other details of specific gravity liquid> The specific gravity of the specific gravity liquid at 25° C. is preferably 1.060 or more, more preferably 1.065 or more, even more preferably 1.070 or more, particularly preferably 1.080 or more, preferably 1.098 or less, more preferably 1.095 or less, and even more preferably 1.090 or less. When the specific gravity of the specific gravity liquid at 25° C. is equal to or more than the lower limit and equal to or less than the upper limit, the recovery amount of mononuclear cells can be further increased.
[0070] The specific gravity of the specific gravity liquid at 25° C. is measured using a specific gravity meter (for example, “DA-130N” manufactured by Kyoto Electronics Manufacturing Co., Ltd.).

When the osmotic pressure of the specific gravity liquid is equal to or more than the lower limit and equal to or less than the upper limit, the amount of mononuclear cells recovered can be further increased.
[0073] The osmotic pressure of the specific gravity liquid is measured by the freezing point depression method using an osmometer (for example, "OM-6060" manufactured by Arkray).
[0074] The specific gravity liquid is preferably contained in the blood collection container body on the bottom side of the blood collection container body relative to the blood separation material. In the blood collection container, it is preferable that the blood separation material is located on the open end side of the blood collection container body, and the specific gravity liquid is located on the bottom side of the blood collection container body.

[0105] The inorganic fine powder is preferably fine silica, titanium oxide powder, calcium carbonate powder, zinc oxide powder, alumina powder, fine glass powder, talc powder, kaolin powder, bentonite powder, titania powder, or zirconium powder.
[0106] From the viewpoint of exerting the effects of the present invention more effectively, the inorganic fine powder preferably contains finely powdered silica. In the case of obtaining a blood separating composition having a specific gravity of 1.050 or more at 25°C, the inorganic fine powder more preferably contains finely powdered silica and an inorganic fine powder other than finely powdered silica (second inorganic fine powder). However, even if the blood separating composition has a specific gravity of 1.050 or more at 25°C, the inorganic fine powder may not contain the second inorganic fine powder. Also, even if the blood separating composition has a specific gravity of less than 1.050 at 25°C, the inorganic fine powder may contain the second inorganic fine powder. The inorganic fine powder, the finely powdered silica, and the second inorganic fine powder may each be used alone or in combination of two or more.
[0107] The fine silica powder includes natural silica and synthetic silica. The synthetic silica includes hydrophilic silica and hydrophobic silica. Hydrophilic silica, for example, imparts thixotropy to the blood separation composition by hydrogen bonding between hydroxyl groups on the particle surface, and has the effect of adjusting the specific gravity. On the other hand, hydrophobic silica has a smaller effect of imparting thixotropy than hydrophilic silica.
[0108] From the viewpoint of maintaining both the specific gravity and thixotropy of the blood separating composition within a suitable range, the fine powdered silica preferably contains hydrophilic silica, and more preferably contains hydrophilic silica and hydrophobic silica. The fine powdered silica preferably contains at least hydrophilic silica.
[0109] The second inorganic fine powder is preferably an inorganic fine powder having a higher specific gravity than fine silica powder, and more preferably an inorganic fine powder having a specific gravity of 3 or more, such as zinc oxide powder, titanium oxide powder, or alumina powder.
[0110] The specific gravity of the second inorganic fine powder is preferably 3 or more, more preferably 3.5 or more, and even more preferably 4 or more. The higher the specific gravity of the second inorganic fine powder, the better. When the specific gravity is equal to or higher than the lower limit, the specific gravity of the blood separating composition can be effectively increased.
[0111] The average particle size of the inorganic fine powder, the fine silica powder, and the second inorganic fine powder is not particularly limited. The average particle size of the inorganic fine powder, the fine silica powder, and the second inorganic fine powder may be 1 nm or more, 10 nm or more, 500 nm or less, or 100 nm or less.
[0112] The average particle diameters of the inorganic fine powder, the fine silica powder and the second inorganic fine powder are average diameters measured on a volume basis (volume average particle diameters), and are the values of the 50% median diameter (D50). The volume average particle diameter (D50) can be measured by a laser diffraction / scattering method, an image analysis method, the Coulter method, a centrifugal sedimentation method, and the like. The volume average particle diameter (D50) is preferably determined by a laser diffraction / scattering method or an image analysis method.
[0113] The specific surface area of the fine silica powder is not particularly limited. The specific surface area of the fine silica powder is 20 m 2 / g or more, 2 / g or more, 2 / g or less, and 2 / g or less.
[0114] The specific surface area of the finely powdered silica is measured by the BET method.
[0115] In 100% by weight of the blood separating composition, the content of the hydrophilic silica is preferably 0.01% by weight or more, more preferably 0.10% by weight or more, even more preferably 0.30% by weight or more, preferably 2.50% by weight or less, more preferably 2.00% by weight or less. When the content of the hydrophilic silica is equal to or more than the lower limit and equal to or less than the upper limit, both the specific gravity and thixotropy of the blood separating composition can be maintained within a more suitable range.
[0116] The content of the finely powdered silica in 100% by weight of the blood separating composition is preferably 0.1% by weight or more, more preferably 0.5% by weight or more, preferably 10% by weight or less, more preferably 7% by weight or less. When the content of the second inorganic fine powder is equal to or more than the lower limit and equal to or less than the upper limit, the specific gravity of the blood separating composition can be effectively increased.
[0118] In 100% by weight of the blood separating composition, the content of the inorganic fine powder is preferably 0.1% by weight or more, more preferably 0.5% by weight or more, preferably 10% by weight or less, more preferably 7% by weight or less. When the content of the inorganic fine powder is equal to or more than the lower limit and equal to or less than the upper limit, the specific gravity of the blood separating composition can be effectively increased.
[0119] Other Ingredients: The blood separating composition may contain other components in addition to the above-mentioned components, so long as the effects of the present invention are not impaired. Examples of the other components include organic gelling agents, thermoplastic elastomers, polyalkylene glycols, silicone oils, auxiliary solvents, antioxidants, colorants, and water. Only one type of each of the other components may be used, or two or more types may be used in combination.
[0120] The specific gravity of the blood separating composition at 25° C. is preferably 1.060 or more, more preferably 1.065 or more, even more preferably 1.070 or more, and preferably 1.095 or less, more preferably 1.090 or less, and even more preferably 1.085 or less. When the specific gravity of the blood separating composition at 25° C. is equal to or more than the above lower limit and equal to or less than the above upper limit, the effects of the present invention can be more effectively exhibited.
[0121] The specific gravity of the above-mentioned blood separating composition at 25°C is measured by dropping a drop of the blood separating composition into successively adjusted saline solutions at 25°C with specific gravities at intervals of 0.002, and observing the floating and sinking of the composition in the saline solutions.
[0122] The viscosity of the blood separating composition at 25° C. is preferably 100 Pa s or more, more preferably 150 Pa s or more, and preferably 500 Pa s or less, more preferably 400 Pa s or less. When the viscosity is equal to or more than the above lower limit and equal to or less than the above upper limit, the effects of the present invention can be more effectively exhibited.
[0123] The viscosity of the blood separating composition at 25° C. was measured using an E-type viscometer (for example, “TVE-35” manufactured by Toki Sangyo Co., Ltd.) at 25° C. and a shear rate of 1.0 sec -1 It is measured under the following conditions.
[0124] <Blood separation tool> The blood separation device is a device that moves between the specific gravity liquid layer and the blood cell layer during centrifugation to form a partition wall, and is used for the purpose of preventing component transfer between the specific gravity liquid layer and the blood cell layer.
[0125] The blood separation tool may be a conventionally known tool, for example, a mechanical separator as described in WO2010 / 132783A1.
[0126] Examples of materials for the blood separation tool include elastomers.
[0127] (Anticoagulant) The blood collection container preferably includes an anticoagulant contained in the blood collection container body. As the anticoagulant, a conventionally known anticoagulant can be used. The anticoagulant may be used alone or in combination of two or more kinds.
[0128] Examples of the anticoagulant include heparin, metal salts of heparin, ethylenediaminetetraacetic acid (EDTA), metal salts of EDTA, citric acid, and sodium citrate.
[0129] From the viewpoint of exerting good anticoagulant performance, the anticoagulant is preferably EDTA, a metal salt of EDTA, heparin, a metal salt of heparin, or sodium citrate.
[0130] The anticoagulant may be contained in the blood collection container body in a powder state or in a dissolved state in liquid. The anticoagulant may be present in the blood collection container body in both a powder state and a dissolved state in liquid.
[0131] The liquid includes water, alcohol, etc. The liquid is preferably water.
[0132] The anticoagulant is preferably disposed on the inner wall surface of the blood collection container body or on the surface of the blood separation material. The anticoagulant may be disposed on the inner wall surface of the blood collection container body, on the surface of the blood separation material, or on both the inner wall surface of the blood collection container body and the surface of the blood separation material.
[0133] When the anticoagulant is contained in a powdered state, the powdered anticoagulant is preferably attached to the inner wall surface of the blood collection container body or disposed on the surface of the blood separation material. For example, a mixture of anticoagulant and water can be sprayed onto the inner wall surface of the blood collection container body and then dried, thereby allowing the anticoagulant to adhere in a powdered state to the inner wall surface of the blood collection container body.
[0134] When the anticoagulant is contained in a dissolved state in a liquid, it is preferable that a mixture of the anticoagulant and the liquid (a liquid containing the anticoagulant) is placed on the surface of the blood separation material.
[0135] The amount of the anticoagulant contained in the blood collection container body is not particularly limited as long as it can exert anticoagulant properties.
[0136] In addition, when the blood collected in the blood collection container is blood to which an anticoagulant has been added, the blood collection container does not need to be provided with an anticoagulant.
[0137] (Blood collection container body) The shape of the blood collection container body is not particularly limited. The blood collection container body is preferably a tubular container with a bottom. The blood collection container body preferably has an open end at one end and a closed bottom (closed end) at the other end.
[0138] The material of the blood collection container body is not particularly limited. Examples of the material of the blood collection container body include thermoplastic resins such as polyethylene, polypropylene, polystyrene, polyethylene terephthalate, polymethyl methacrylate, and polyacrylonitrile; thermosetting resins such as unsaturated polyester resins, epoxy resins, and epoxy-acrylate resins; modified natural resins such as cellulose acetate, cellulose propionate, ethyl cellulose, and ethyl chitin; and glass such as silicate glass such as soda-lime glass, phosphosilicate glass, and borosilicate glass, and quartz glass. Only one type of material may be used for the blood collection container body, or two or more types may be used in combination.
[0139] The blood collection container body is preferably a resin container, more preferably a thermoplastic resin container, and even more preferably a polyethylene terephthalate container. When the blood collection container body is a resin container (particularly when it is a polyethylene terephthalate container), a sterilization method using gamma rays is likely to be adopted as a sterilization method for the blood collection container. In a conventional blood collection container, when sterilization is performed using gamma rays, precipitates are generated in the specific gravity liquid, and the amount of mononuclear cells recovered is likely to decrease. However, in the present invention, the generation of precipitates in the specific gravity liquid can be effectively suppressed, so that a resin container can be used as the blood collection container body without any problems. Therefore, when the blood collection container body is a resin container (particularly when it is a polyethylene terephthalate container), the effect of the present invention is even more effectively exhibited.
[0140] (stopper) The blood collection container preferably includes a stopper. The stopper is preferably attached to the open end of the blood collection container body. A conventionally known stopper can be used as the stopper. The stopper is preferably made of a material and has a shape that allows it to be attached to the open end of the blood collection container body in an airtight and liquidtight manner. The stopper is preferably configured to be pierced by a blood collection needle.
[0141] Examples of the stopper include a stopper having a shape that fits into the open end of the blood collection container body, a sheet-like seal stopper, and the like.
[0142] The stopper may be a stopper including a stopper body such as a rubber stopper and a cap member made of plastic, etc. In this case, the risk of blood coming into contact with the human body can be reduced when the stopper body is pulled out from the open end of the blood collection container body after blood collection.
[0143] Examples of the material of the stopper (or the stopper main body) include synthetic resin, elastomer, rubber, metal foil, etc. Examples of the rubber include butyl rubber and halogenated butyl rubber, etc. Examples of the metal foil include aluminum foil, etc. From the viewpoint of improving the sealing performance, the material of the stopper is preferably butyl rubber. The stopper (or the stopper main body) is preferably a butyl rubber stopper.
[0144] (Other details of blood collection container) The blood collection container is preferably used for separating mononuclear cells from blood, and more preferably used for separating peripheral blood mononuclear cells (PBMCs) from blood.
[0145] The amount of blood collected in the blood collection container is appropriately changed depending on the size and internal pressure of the blood collection container, etc. The amount of blood collected in the blood collection container may be 1 mL or more, 2 mL or more, 4 mL or more, 12 mL or less, 11 mL or less, or 10 mL or less.
[0146] The blood collection container is preferably a blood collection tube. The blood collection container body is preferably a blood collection tube body.
[0147] The internal pressure of the blood collection container is not particularly limited. The internal pressure of the blood collection container is preferably reduced. When the blood collection container is a reduced pressure container, a predetermined amount of blood can be easily collected in the blood collection container. The blood collection container can also be used as a vacuum blood collection tube, the inside of which is evacuated and sealed with the stopper. When it is a vacuum blood collection tube, a certain amount of blood can be easily collected regardless of the skill level of the blood collector.
[0148] From the viewpoint of preventing bacterial infection, it is preferable that the inside of the blood collection container is sterilized according to ISO or JIS standards. As a sterilization method, a conventionally known method can be used. Examples of the sterilization method include gamma ray sterilization, electron beam sterilization, and high-pressure steam sterilization.
[0149] The blood collection container is preferably a sterilized blood collection container, and more preferably a blood collection container sterilized by gamma rays. In conventional blood collection containers, sterilization by gamma rays makes it more likely that precipitates will form in the specific gravity liquid, which in turn makes it more likely that the amount of mononuclear cells recovered will decrease. However, in the present invention, the formation of precipitates in the specific gravity liquid can be effectively suppressed even when sterilization is performed by gamma rays. Therefore, when the blood collection container is a blood collection container sterilized by gamma rays, the effects of the present invention are even more effectively achieved.
[0150] The blood collection container can be manufactured, for example, as follows.
[0151] An amino acid and component (X) are dissolved in water to obtain a specific gravity liquid. The obtained specific gravity liquid is placed in a blood collection container body. Next, a blood separation material is placed in the blood collection container body. In addition, an anticoagulant is placed in the blood collection container body.
[0152] Hereinafter, specific embodiments of the present invention will be described with reference to the drawings. Note that in the following drawings, the size, thickness, shape, etc. may differ from the actual size, thickness, shape, etc. for convenience of illustration.

The liquid 5 containing an anticoagulant is disposed on the surface of the blood separating composition 4. In the blood collection container 1, the specific gravity liquid 3, the blood separating composition 4, and the liquid 5 containing an anticoagulant are disposed in this order from the bottom 2b side of the blood collection container body 2 toward the opening end 2a.

[0157] The blood collection container 1A shown in FIG. 2 includes a blood collection container body 2, a specific gravity liquid 3, a blood separating composition 4, a powdered anticoagulant 5A, and a plug 6. The blood collection container 1 shown in FIG. 1 and the blood collection container 1A shown in FIG. 2 differ in the form in which the anticoagulant is contained. That is, in the blood collection container 1 shown in FIG. 1, the anticoagulant is contained in the blood collection container body 2 in a state dissolved in liquid, whereas in the blood collection container 1A shown in FIG. 2, the anticoagulant is contained in the blood collection container body 2 in a powdered state. More specifically, in the blood collection container 1A shown in FIG. 2, the powdered anticoagulant 5A is disposed on the surface of the blood separating composition 4.
[0158] FIG. 3 is a front cross-sectional view that typically shows a blood collection container according to a third embodiment of the present invention.

The blood collection container 1 shown in FIG. 1 and the blood collection container 1B shown in FIG. 3 have different anticoagulant storage forms. That is, in the blood collection container 1 shown in FIG. 1, the anticoagulant is stored in a liquid-dissolved state in the blood collection container body 2, whereas in the blood collection container 1B shown in FIG. 3, the anticoagulant is stored in a powdered state on the inner wall surface of the blood collection container body 2. In the blood collection container 1B shown in FIG. 3, the anticoagulant 5B is arranged in layers.
[0160] (Method of isolating mononuclear cells) The blood collection container can be used to separate mononuclear cells from blood. The method for separating mononuclear cells preferably includes a step of collecting blood in the blood collection container, and a step of centrifuging the blood collection container in which the blood has been collected.
[0161] The centrifugation conditions in the centrifugation step are not particularly limited, and examples of the centrifugation conditions include centrifugation at 400 G or more and 4000 G or less for 5 minutes or more and 120 minutes or less.
[0162] After the centrifugation step, for example, red blood cell components are located below the partition formed by the blood separation material, a specific gravity liquid layer containing mononuclear cells is located on the partition formed by the blood separation material, and plasma is located above the specific gravity liquid layer containing mononuclear cells. Mononuclear cells can be obtained by recovering the specific gravity liquid layer containing mononuclear cells.
[0163] The method for isolating mononuclear cells is preferably a method for isolating peripheral blood mononuclear cells (PBMC).
[0164] The present invention will be described in more detail below with reference to examples, but the present invention is not limited to the following examples.
[0165] The following materials were prepared for the specific gravity liquid:
[0166] (Component (X)) Polysucrose (Cytiva "Ficoll TM PM 400" Sodium diatrizoate Silica microparticles (Cytiva's "Percoll") Sodium chloride
[0167] (amino acid) β-Alanine Histidine Proline glycine
[0168] water
[0169] The following materials were prepared as the blood separation composition.
[0170] (Materials made of organic components that are fluid at 25℃) (Meth)acrylic resin: 2-Ethylhexyl acrylate and butyl acrylate were radically polymerized by solution polymerization in the presence of an azo polymerization initiator to obtain a (meth)acrylic acid ester polymer having fluidity at 25° C. The specific gravity of the (meth)acrylic resin at 25° C. was 1.033.
[0171] (Inorganic fine powder) Hydrophilic silica (fine silica powder, "200CF" manufactured by Nippon Aerosil Co., Ltd.) Hydrophobic silica (fine silica powder, "RX200" manufactured by Nippon Aerosil Co., Ltd.) Calcium carbonate powder (IMERYS "Socal UP-G")
[0172] (Other Ingredients) Silicone oil (Toray Dow Corning "SF8410")
[0173] Preparation of blood separation composition: A blood separation composition was prepared by mixing 91.9 parts by weight of (meth)acrylic resin, 1.00 parts by weight of hydrophilic silica, 5.0 parts by weight of hydrophobic silica, 1.95 parts by weight of calcium carbonate powder, and 0.15 parts by weight of silicone oil. The specific gravity of the blood separation composition at 25°C was 1.077.
[0174] Example 1 Preparation of specific gravity solution: A specific gravity liquid was obtained by mixing the components shown in Table 1 in the mixing ratios shown in Table 1. The amounts of ingredients in Table 1 and Tables 2 to 5 described later are pure amounts.
[0175] Preparation of blood collection containers: A tubular polyethylene terephthalate container (PET bottomed tube) with a length of 100 mm and an inner diameter of 14 mm at the open end was prepared as the blood collection container body. 1.0 mL of the obtained specific gravity liquid was placed in the blood collection container body. Then, 1 mL of the obtained blood separation composition was placed in the blood collection container body. Then, 30 mg of a 24 wt% aqueous solution of EDTA2K dihydrate (anticoagulant-containing liquid) was applied to the inner wall surface of the blood collection container body and dried. Then, the inside of the blood collection container was decompressed so that the blood collection volume was 4 mL, and the container was sealed with a butyl rubber stopper. In this way, a blood collection container was prepared in which the specific gravity liquid, the blood separation composition, and the anticoagulant were placed in the blood collection container body. Then, the blood collection container was irradiated with gamma rays at a dose of 30 kGy to obtain a blood collection container sterilized by gamma rays. In the obtained blood collection container, the specific gravity liquid is contained in the blood collection container body closer to the bottom of the blood collection container body than the blood separation composition, and the blood separation composition is contained in the blood collection container body closer to the open end than the specific gravity liquid. The anticoagulant is disposed on the inner wall surface of the blood collection container body closer to the open end than the blood separation composition.
[0176] (Examples 2 to 8 and Comparative Examples 1 to 5) Except for changing the blending composition of the specific gravity liquid as shown in Tables 1 to 5, specific gravity liquids and blood collection containers sterilized by gamma rays were prepared in the same manner as in Example 1.
[0177] (evaluation) (1) Specific gravity of specific gravity liquid The specific gravity of the resulting specific gravity liquid at 25° C. was measured using a specific gravity meter (Kyoto Electronics Manufacturing Co., Ltd., “DA-130N”).
[0178] (2) Osmotic pressure of specific gravity liquid The osmotic pressure of the obtained specific gravity liquid was measured by the freezing point depression method using an osmometer ("OM-6060" manufactured by Arkray Corporation).
[0179] (3) Presence or absence of precipitate in specific gravity liquid The specific gravity liquid in the blood collection container sterilized by gamma radiation was visually observed to confirm whether or not a precipitate had formed in the specific gravity liquid. The presence or absence of a precipitate in the specific gravity liquid was evaluated according to the following criteria. Note that no precipitate had formed in the specific gravity liquid before sterilization by gamma radiation.
[0180] <Criteria for determining the presence or absence of precipitates in specific gravity liquid> A: No precipitates have formed in the specific gravity liquid. B: Precipitation has occurred in the specific gravity liquid
[0181] (4) Recovery of mononuclear cells (PBMC) Blood from two individuals (donor A and donor B) was prepared, and the following procedure was carried out for each. 4 mL of blood was collected into the obtained blood collection container (a blood collection container sterilized by gamma radiation). The blood collection container into which the blood had been collected was then centrifuged at 20°C and 1700×g for 20 minutes. After centrifugation, the red blood cell layer was located below the partition formed by the blood separation composition. In addition, the specific gravity liquid layer was located above the partition formed by the blood separation composition, and the plasma layer was located above the specific gravity liquid layer. In addition, mononuclear cells were suspended in the specific gravity liquid layer.
[0182] The specific gravity liquid layer was sucked up with a pipette and transferred to a centrifuge tube. Then, 1 mL of phosphate buffered saline was added to the centrifuge tube. Then, the centrifuge tube was centrifuged at room temperature and 500×g for 10 minutes to precipitate the cells. After removing the supernatant, the precipitated cells were suspended in 1 mL of plasma to obtain a mononuclear cell suspension. The mononuclear cell suspension was analyzed using a multi-item automated blood cell analyzer (Sysmex Corporation, "XE-5000") to measure the number of mononuclear cells in the mononuclear cell suspension, which was taken as the amount of mononuclear cells recovered. The average of the amount of mononuclear cells recovered from donor A and the amount of mononuclear cells recovered from donor B was taken as the average amount of recovery (Ave).
[0183] The increase (ΔAve) in the amount of mononuclear cells recovered due to the presence or absence of amino acids in the specific gravity liquid was determined for blood collection containers containing the same type and amount of component (X). That is, the increase (ΔAve) is the value obtained by subtracting the average recovery amount (Ave) in the reference blood collection container from the average recovery amount (Ave) in the blood collection container to be evaluated. More specifically, the increase (ΔAve) is the value calculated by the following formula. The combinations of the blood collection container to be evaluated and the reference blood collection container are as follows:
[0184] ΔAve = [Average recovery amount (Ave) from the blood collection container under evaluation] - [Average recovery amount (Ave) from the reference blood collection container]
[0185] More specifically, it is as follows.
[0186] ΔAve=[average recovery amount (Ave) in the blood collection container of Example 1)]−[average recovery amount (Ave) in the blood collection container of Comparative Example 1] ΔAve=[average recovery amount (Ave) in the blood collection containers of Examples 2 to 5]−[average recovery amount (Ave) in the blood collection container of Comparative Example 2] ΔAve=[average recovery amount (Ave) in the blood collection container of Example 6]−[average recovery amount (Ave) in the blood collection container of Comparative Example 3] ΔAve=[average recovery amount (Ave) in the blood collection container of Example 7]−[average recovery amount (Ave) in the blood collection container of Comparative Example 4] ΔAve=[average recovery amount (Ave) in the blood collection container of Example 8)]−[average recovery amount (Ave) in the blood collection container of Comparative Example 5]
[0187] <Criteria for determining mononuclear cell recovery amount> A: ΔAve is 500,000 or more B: ΔAve is greater than 200,000 and less than 500,000 C: ΔAve is less than 200,000
[0188] The configuration and results are shown in Tables 1 to 5 below.
