Abstract
A novel spectrophotometric chemical solution for evaluating enzymatic activity after treatment with a cellulase-type enzyme on a cellulosic textile. This solution is based on the chemical reaction that occurs between sodium picric acid hydroxide and a glucose solution. The color change of this mixture indicates the presence of glucose, and the intensity of the color reflects the amount of glucose released. First, the textile material is cleaned at 45°C using a detergent and an anti-depositing agent. Then, it undergoes an enzymatic treatment with a cellulase-type enzyme, and a small amount of the final enzyme bath is collected. The actual assay step then begins by adding a predetermined volume of sodium hydroxide picric acid and the final enzyme bath to a test tube and heating it at 100°C for 20 minutes. Finally, after cooling, the mixture is analyzed with a spectrophotometer, the absorbance is measured, and the exact rate of glucose released is calculated. This method has proven its effectiveness and applicability in the textile industry, using a cellulosic base and a cellulase-type enzyme.
