ANTI-CD25 ANTIBODY AND USES THEREOFTechnical Field

[0001] The present invention generally relates to antibodies and antigen-binding fragments thereof for targeting CD25 and selectively depleting Tregs. In addition, the present invention relates to nucleic acid molecules encoding such antibodies and antigen-binding fragments, and vectors and host cells comprising such nucleic acid molecules. The invention further relates to methods for producing the antibodies and antigen-binding fragments thereof, and to uses thereof in the treatment of infectious, autoimmune and cancer diseases.Background Art

[0002] Regulatory T cells (Treg) are defined by their expression of CD4, persistently high expression of CD25, and intracellular expression of the transcription factor FoxP3. These cells play an important role in regulation of innate and adaptive immune responses. The suppressive activity of Tregs may account for poor immune response to cancer and be associated with uncontrolled pathogen growth and infectious disease progression.

[0003] CD25 is a component of interleukin-2 receptor. The IL-2 receptor (IL-2R) exists in two functional forms, the high-affinity trimeric IL-2R composed of IL-2Rα (CD25) , IL-2Rβ (CD122) and IL2Rγ (CD132) , and the intermediate -affinity dimeric IL-2R composed of IL-2Rβ and IL2Rγ. The high-affinity IL-2R assembles when IL-2 is captured by the IL-2Rα (CD25) subunit that in turn drives additional IL-2 binding and subsequent signaling by the IL-2Rβ (CD122) and IL-2Rγ (CD132) subunits; this IL-2R is found at high levels on Tregs but lower levels on T effector cells (Teff cells) . The intermediate-affinity IL-2R occurs when IL-2 binds directly to CD122 and CD132, where these subunits are found on many Ag-experienced CD8+ T cells and most NK cells. (See, e.g., IL-2 / CD25: A Long-Acting Fusion Protein That Promotes Immune Tolerance by Selectively Targeting the IL-2 Receptor on Regulatory T Cells, https:  / / doi. org / 10.4049 / jimmunol. 1800907) .

[0004] CD25 is constitutively expressed on Tregs and is absent on naive T effectors (Teff) . CD25 has been proposed as a promising target for inhibiting or eliminating Tregs. Clinical studies have been conducted to explore the use of daclizumab, a humanized IgG1 anti-human CD25 antibody, or LMB-2, a recombinant fusion protein combining anti-human CD25 Fv and a fragment of Pseudomonas exotoxin A (PE) , which displayed a variable impact on the number of circulating Tregs. However, there is no clear evidence that Tregs in the tumor microenvironment were effectively reduced, and antitumor activity has been disappointing, with no survival benefit.

[0005] There is still a need in the art to develop therapies that selectively interfere with and / or deplete Tregs and enhance the immune response in patients, such as those having infectious diseases and cancers.Summary of Invention

[0006] The present invention provides anti-CD25 antibodies, which are capable of specifically binding to CD25 without blocking the IL-2 binding to CD25 or the IL-2 signaling via CD25, and capable of selectively depleting Tregs over T effector cells, so as to enhance the immune response.

[0266] C57B / L6 mice were immunized subcutaneously and intraperitoneally using Fc-tagged human CD25 (Acro biosystem) emulsified in Complete Freund's Adjuvant (Pierce) at a 1: 1 ratio. After initial immunization, the recombinant CD25 protein was given every two weeks as a booster to the inoculated animals. After immunizations, anti-serum from the mice was collected for an ELISA experiment to determine the anti-hCD25 titer.

[0267] Four days following the last boost, the animals were killed and spleen cells were separated for hybridoma fusion. Electro-fusion was employed to fuse the spleen cells with mouse myeloma cell line SP2-0 cells. The resultant hybridoma cells were grown in hypoxanthine-aminopterin-thymidine-containing DMEM medium. Hybridoma supernatants were obtained for antigen binding screening after a 10-day culture.

[0273] To test the binding activity to CD25-expressing cells, SU-DHL-1 cell line with high expression of human CD25 at the cell surface was employed. In a 96-V bottom plate, 60, 000 SU-DHL-1 cells were plated per well. After a wash, anti-CD25 antibodies (serially diluted in the assay buffer, starting from the concentration of 10 μg / ml) were added into the wells of the plate and incubated for 1 hour at 4℃. After two washes, anti-mouse IgG antibody (FITC) (1: 500 dilution) was added to the wells, and then incubated at 4 ℃ for one hour. The plate was ready for detection by Beckman cytoflex FACS device after two washings. Ab 19 and PC-61 (a reference anti-mouse CD25 antibody, also depicted in US20200010554A1) were used as control in the FACS assay. In contrast to anti-mouse PC-61, the human CD25 specific antibodies, Ab 19 and clone 75G1A5C10, were capable of binding to SU-DHL-1 cells (Figure 2) .

[0275] The CD25 protein with his tag (Acro biosystem) was diluted to 0.5 μg / mL with a coating buffer having a high pH (0.16%Na2CO3, 0.3%NaHCO3, pH9.8) , and used to coat a ELISA plate at 100 μl / well by incubation overnight at 4℃. The plate was then washed one time on an automatic plate washer using a washing buffer (PBS +0.1%Tween 20) . 200 μl per well of a blocking buffer (PBS+1%BSA+1%Goat serum+0.05%Tween 20) was added and incubated for 2 hours at room temperature. The blocking buffer was then aspirated off, and 100 μl per well of anti-CD25 antibody serially diluted in a dilution buffer (PBS+1%BSA+1%Goat serum+0.01%Tween 20) was transferred to the wells of the ELISA plate and allowed to incubate for 60 min at room temperature. After washing again, 100 μl per well of 0.1 μg / ml biotinylated IL-2 (R&D) in the dilution buffer , and then 100 μl / well of Neutravidin-HRP (Thermo) diluted in the dilution buffer were added to the wells of the plate. The ELISA plate was incubated for 20 min at room temperature and then washed 3 times with 250 μl / well washing buffer. Finally, 100 μl / well of TMB substrate was added. After the reaction was terminated using 0.64M H2SO4, the plate was read on a Thermo Multiscan FC at 450nm. Anti-human CD25 antibodies, Ab19 and Daclizumab (in-house generated based on the sequences under DrugBank Accession Number DB00111, which can block IL-2 binding to CD25) , were included as control in the assay. Hybridoma 75G1A5C10 did not block the IL-2 binding to human CD25, while daclizumab that was set as positive control showed the blocking activity (Figure 3) .

[0277] V-gene cloning and sequencing of hybridoma antibodies

[0278] The lead antibody 75G1A5C10 with a desired activity profile was selected for V-gene cloning. The sequences of the light chain and heavy chain variable regions of the murine anti-human CD25 antibody were obtained by the polymerase chain reaction (PCR) amplification technique. Total RNA was extracted from hybridoma cells using the MiniBest Universal RNA Extraction Kit (TaKaRa) , and the cDNA was synthesized using the 1st Strand cDNA Synthesis Kit (TaKaRa) with Oligo (dT) primer. The variable regions of mouse IgG genes were amplified by PCR using primers for different isotypes of heavy chains and primers for Kappa light chains. The PCR products were subcloned into a TA cloning vector. For each variable gene construct, more than 10 single colonies were used for DNA sequencing by Synbio Technologies (Suzhou, China) . The amino acid sequences of Vh and Vk were derived from the DNA sequencing results.

[0287] His-tagged full-length CD25 proteins, including human CD25-His (Acro biosystem) , cynomolgus monkey CD25-His (Sino Biological) , rat CD25-His (Sino Biological) , mouse CD25-His (Acro biosystem) , were used to detect the binding activity of the chimeric antibody. An ELISA plate was coated by adding 100 μl per well of 0.5 μg / ml a CD25-His protein into the wells. After blocking the plate with PBS containing 1%BSA + 1%normal goat serum + 0.05%Tween20, the chimeric antibody was added to the plate for 1hr. The binding activity of the chimeric anti-human CD25 antibody (75G1-chi) was detected with an HRP-linked anti-human IgG antibody. 100 μl / well of TMB was added to each well, and after the reaction was terminated using 0.64M H2SO4, the plate was read on a Thermo Multiscan FC at 450nm. Ab19 was used as control. Chimeric 75G1 antibody retained the human CD25 binding activity (Figure 4) , and also bound to cynomolgus CD25 (Figure 5) .

[0289] The binding activity was also confirmed with Treg cells. CD4+ T cells were isolated using the human  CD4+ T cell isolation kit II (Miltenyi Biotech) according to manufacturer’s instructions. CD4+ T cells were adjusted to a cell density of 1×106 cells / ml, and activated by CD3 / CD28 T Cell Activator (Gibco) for 6 days in RPMI 1640 medium mainly containing 10%FBS, 1%Glutamax, 500 U / ml IL2, 109 nM rapamycin and 10 ng / ml TGFβ1 to produce iTreg cells. The iTreg cells (induced Treg cells) were stained by anti-human CD25 antibody with a serial dilution from 11.11111nM. An anti-human IgG antibody with PE (1: 200 dilution) was used as a secondary antibody. Cells were detected and analyzed by Beckman CytoFLEX flow cytometry. Similar to SU-DHL-1, chimeric 75G1 antibody showed the binding activity to iTreg cells (Figure 7) .

[0291] The CD25 protein with his tag (Acro biosystem) was diluted to 0.5 μg / ml with a coating buffer having a high pH (0.16%Na2CO3, 0.3%NaHCO3, pH9.8) , and was added to an ELISA plate at 100 μl / well and incubated overnight at 4℃ for coating the plate. The coated plate was washed one time on an automatic plate washer using a washing buffer (PBS +0.1%Tween 20 (Sigma) ) . 200 μl of a blocking buffer (PBS+1%BSA+1%Goat serum+0.05%Tween 20) was added per well and incubated for 2 hours at room temperature. The blocking buffer was then aspirated off and 100 μl per well of anti-CD25 antibody serially diluted in dilution buffer (PBS+1%BSA+1%Goat serum+0.01%Tween 20) was transferred into the wells of the ELISA plate and allowed to incubate for 60 min at room temperature. After repeating the washing, the 100 μl / well of 0.1 μg / ml biotinylated IL-2 (R&D) in the dilution buffer was added, and then 100 μl / well of Neutravidin-HRP (Thermo) diluted in the dilution buffer was added to the wells of the plate. The ELISA plate was incubated for 20 min at room temperature, and then washed 3 times with 250 μl / well washing buffer. Finally, 100 μl / well of TMB was added to each well and after the reaction was terminated using 0.64M H2SO4, the plate was read on a Thermo Multiscan FC at 450nm. Daclizumab and Ab19 were included as control. As shown in Figure 8, chimeric 75G1 antibody did not block the IL-2 binding to human CD25 while daclizumab that was set as positive control showed the blocking activity (Figure 8) .

[0292] SU-DHL-1 based assay was also used to detect the blocking activity of the chimeric antibody. 200, 000 SU-DHL-1 cells were plated per well in a 96-V bottom plate. Anti-CD25 antibodies (serially diluted in the assay buffer at 1: 3 from 10 μg / ml ) were added and incubated at 4℃ for 1h. After washing 2 times, 0.2 μg / ml of an IL-2-biotin dilution was added per well. Then, PE-strep-avidin (1: 100) was added into the wells and incubated at 4 ℃ for 1h. After washing 2 times, the plate was ready for detection by Beckman cytoflex FACS device. The chimeric 75G1 antibody, in contrast to Daclizumab, did not block the binding of IL-2 to human CD25 on SU-DHL-1 cells (Figure 9) .

[0294] The chimeric anti-human CD25 antibody was analyzed in an antibody-dependent cell-mediated cytotoxicity assay (ADCC assays) , by utilizing Jurkat-NFAT-Luc-CD16a cells as the source of effector cells and SU-DHL-1 human cell line as target cells. The reporter cell line Jurkat-NFAT-Luc-CD16a has been engineered to stably express CD16a and contain a luciferase reporter gene driven by a NFAT-response element. In a 96-well white tissue culture-treated plate, 25 μl of the target cells at 10000 cells / well, 25 μl of serially diluted antibodies, and 50 μl of the Jurkat cells at 60000 cells / well were sequentially added to the wells and incubated for 5 hours. Ab19 was used as a positive control. The Promega ONE-Glo Luciferase Assay Buffer / Substrate was applied, and the reaction was allowed for 15 minutes, and then the value of luminescence was measured via the microplate reader. Strong ADCC activity was exhibited by chimeric 75G1 antibody (Figure 10) .

[0297] A humanization process enabling the generation of a highly optimized mAb consists of five steps. The first step is to select acceptor human antibodies for their antigen binding activity, immunogenicity, expression, stability, and pharmacokinetics. The second step is to generate CDR-grafted mAbs by grafting the CDRs of a murine antibody into the frameworks (FRs) of the human antibodies selected in the first step, then modelling the three-dimensional (3D) Fv structures of the parental murine antibody and the CDR-grafted mAbs, and under the guidance of the structures, the FR amino acids , which are close to the CDR regions and located at the interface of the VH and VL in the structures, are back-mutated to the corresponding amino acids in the FRs of the murine antibody. The third step is to prepare expression vectors for the multiple versions of humanized antibodies. The fourth step is to express and purify the humanized antibodies. The last step is to multidimensionally evaluate the humanized antibodies.

[0310] The humanized antibodies were analyzed in an ADCC assay, wherein SU-DHL-1 human cells were used as target cells , and Jurkat-NFAT-Luc-CD16a cells were used as effector cells. The reporter cells Jurkat-NFAT-Luc-CD16a have been engineered to stably express CD16a and contain a luciferase report gene driven by the NFAT element. In a 96-well White Tissue Culture-treated plate, 25 μl of target cells at 10000 cells / well, 25 μl of serially diluted antibodies, and 50 μl of Jurkat cells at 60000 cells / well were sequentially added to  the plate and incubated for 5 hours. The Promega ONE-Glo Luciferase Assay Buffer / Substrate was applied, and the reaction was allowed for 15 minutes. The value of luminescence was measured using a microplate reader. The variants displayed divergent ADCC activities. For instance, Hu75G1 HeL0 exhibited strong ADCC activity comparable to that of chimeric 75G1 antibody, while Hu75G1-HgLc lost the ADCC activity (Table 5 and Figures 13 and Figures 14) .

[0312] Example 4 Affinity maturation of humanized antibody candidate

[0313] Affinity maturation screening

[0314] Hu75G1-HeL0 was subjected to affinity maturation. The CDR regions in the Hu75G1-HeL0 sequences were defined by the Chithia scheme. To construct a CDR mutant library, codon-based primers for each of the CDRs (including HCDR1, HCDR2, HCDR3, LCDR1, LCDR2 and LCDR3) were used so that six individual affinity maturation libraries were established for hu75G1-HeL0. The diversified CDR fragments were amplified and the products were transformed into competent TG1 cells by electroporation to construct scFv libraries for screen. The diversity of the HCDR1, HCDR2, HCDR3, LCDR1, LCDR2 and LCDR3 libraries was greater than 1×10^8. The library screen was performed with four rounds of magnetic bead-based panning by means of DynabeadsTM M-280 Streptavidin (InvitrogenTM, 11205D) . In order to obtaining an affinity-improved antibody, affinity pressures were applied for these selections, either by titrating down biotinylated monomeric antigen or by preincubating the biotinylated antigen with parental IgG for 30 min and then applying that precomplexed mixture to the phage library for a length of time which would allow the selection to reach an equilibrium. The higher affinity antibodies were then able to be sorted.

[0315] The affinity of the maturated antibodies was higher than that of the parent antibody. H5L0 and H8L6 showing the best affinity were expressed as a full IgG format with the constant regions of human IgG1 and kappa (shown in table 7) . The variable region sequences of H5L0 and H8L6 are shown in Table 6.

[0327] A STAT5 phosphorylation assay was used to evaluate IL-2-blocking, in which IL-2 signaling was investigated. Prior to adding IL-2 (R&D) at 100 ng / ml in RPMI 1640 (Thermo) containing 10%FBS, previously frozen PBMCs (Saily, China) were cultured in a 96-U-bottom well plate in the presence of 10 μg / ml an anti-CD25 antibody for 30 minutes. IL-2 induced STAT5 phosphorylation was stopped when the cells were fixed and permeabilized with the eBioscienceTM Foxp3 / Transcription Factor Staining Buffer Set (Invitrogen) and treated with the BD Phosflow Perm Buffer III (BD Biosciences) . Following that, cells were then simultaneously stained with surface and intracellular fluorochrome labelled antibodies (pSTAT5-FITC, CD3-PerCP, FoxP3-PE, BD Bioscience) and samples were acquired and analyzed on the Flow Cytometer (Beckman Cytoflex) . The population of pSTAT5+ / CD3+ T cells or pSTAT5+ / FOXP3+ T cells was collected for statistical analysis. Graphs and statistical analysis were performed using GraphPad Prism v9 (Figures 21 and 22) . Daclizumab was set as a positive control. Comparing with daclizumab, H5L0 had no effects on downstream Stat5 phosphorylation in the IL-2 signaling events .

[0329] Generation of Afucosylated antibody

[0330] The plasmid expressing the light chain and the heavy chain of the H5L0 antibody was transfected into CHOK1SV GS-KO cells by Maxcyte electroporator according to the protocol recommended by the manufacturer. Stable cell pools were obtained by electrotransfection and subsequent selective screening (CD CHO +25uM MSX) . The stable cell pools were assessed in a fed-batch culture. The pooled cells were inoculated at 9×105 cells / mL in basal medium Dynamis containing 4 mM L-Glutamine with different concentrations of 2-fluorofucose (2FF) (0, 10, 20, 30, 50 μM) . 2FF can decrease the fucosylation of Fc domain. The feeding media were Cellboost 7a (CB7a) and Cellboost 7b (CB7b) (purchased from GE Healthcare) , and fed on day3, day6, day8, day10, and day12, respectively. Glucose was also fed on the same feeding day to maintain a minimum concentration of 2g / L based on the glucose consumption rate. All the fed-batch cultures were carried out in shake flasks in a shaker incubator with 26 mm orbital diameter, at 37℃, 5%CO2, and 140 RPM. The culture temperature was shifted to 33℃ when the viable cell density (VCD) reached 15×106 cells / ml. The fed-batch cultures were harvested on day13. After centrifugation and filtration, the supernatants were loaded onto protein A column and then purified with the GE AKTA purification system. After washing, antibodies were eluted with citric acid (pH3.5) . Detection of fucosylation level was then performed and the data are summarized in Table 8.

[0332] SU-DHL-1 cells or iTreg cells were used for cell-based binding assay. iTreg induction was performed using the procedure outlined in Example 2.100, 000 cells were plated in each well of a 96-V bottom plate before a wash and a 1-hour incubation at 4℃ with H5L0 and afucosylated antibodies (serially diluted from 5 μg / ml with the assay buffer) . After two washes, an anti-human IgG antibody (PE, 1: 200) was added to a well, and incubated at 4 ℃ for 1 hour. After two washes, the plate was ready for detection by Beckman cytoflex FACS device. In comparison to anti-HEL Human IgG1 isotype control (Anti-HEL-huIgG1) , H5L0 and afucosylated antibodies showed increased binding activity to SU-DHL-1 or iTreg (Figures 23 and 24)

[0333] In Vitro ADCC reporter Assay

[0334] Anti-human CD25 antibodies were characterized in ADCC assay using SU-DHL-1 human cell line or iTreg cells as target cells and Jurkat-NFAT-Luc-CD16a cells as the source of effector cells. The iTreg cells were generated as described in the Example 2. The Jurkat-NFAT-Luc-CD16a cells as described above were used. 25 μl target cells at 10000 cells / well, 25 μl serially diluted antibodies and 50 μl Jurkat cells at 60000 cells / well were sequentially added to a White Tissue Culture-treated 96 well plate and incubated in a cell incubator for 5 h. ONE-Glo Luciferase Assay Buffer / Substrate (Promega) was applied, and the reaction was allowed for 15 min and then the value of luminescence was read on the microplate reader. The results are shown in Figures 25 and 26. Compared to control IgG1 antibody and H5L0, afucosylated H5L0 antibodies demonstrated an improved ADCC activity as the level of 2-FF substrate in the medium increased and the fucose level of the antibody was reduced in a dose-dependent manner. The afucosylated versions produced at 30 μM and 50 μM of 2-FF, H5L0-30 μM and H5L0-50 μM, were chose for further development. The performances of the antibodies on the two kinds of target cells, SU-DHL-1 and iTreg, were very comparable.

[0335] The ADCC activity of the antibodies was assessed on CD8+T cells, which have been described to express a significant level of CD25. CD8+ T cells were isolated from PBMC (Saily, China) and were then activated for 72 hours with CD3 / CD28 activator beads (Gibco) . The cells were collected and plated in a White Tissue Culture-treated 96-well plate in 25 μl medium with 10, 000 cells per well. 25 μl serially diluted antibodies and 50 μl Jurkat cells at 60000 cells / well were sequentially added into the plate and incubated for 5 hours. The Promega ONE-Glo Luciferase Assay Buffer / Substrate was applied, and the reaction was allowed for 15 minutes, and then the value of luminescence was measured on the microplate reader. CD8+T cells also express CD25. As a result, afucosylated H5L0 showed ADCC activity on CD8+ T cells in a dose dependent manner. Due to lower CD25 expression on CD8+ T cells, as revealed in the ADCC assay, the maximum RLU values of afucosylated H5L0 antibodies on CD8+ T cells were lower than on SU-DHL-1 and iTreg (Figure 27) .

[0336] In vitro NK cytotoxicity assay

[0337] The following procedures were used to generate activated human NK cells. NK cells were isolated from resting human PBMCs (Saily, China) using the human NK cell isolation kit in accordance with the manufacturer's instructions (Stemcell) . The isolated NK cells were grown at 37℃ and 5%CO2 in Culture medium CTSTM AIM VTM SFM (Thermo) +10ng / ml IL-2 (R&D) + 50ng / ml IL-15 (R&D) . The entire culture  medium was replaced every two days for 10~14 days. To confirm the culture was NK cell population, cell proliferation and cell surface markers (CD56-APC, CD3-PC5.5, CD16-PE, BD bioscience) were detected by FACS. The NK cells were then divided into aliquots and stored at -80℃ for subsequent use.

[0338] Anti-human CD25 antibodies were characterized utilizing ADCC assay with activated NK cells serving as the source of effector cells and iTreg or CD8+T cells as target cells. Following the manufacturer's instructions, the cells were collected, labeled with CFSE, and seeded into each well with 5000 cells in 25 μl medium in a U-bottom 96-well cell culture plate. 25 μl serially diluted antibodies and 50 μl Jurkat cells at 40000 cells  / well were sequentially added into the wells of the plate and incubated in a cell incubator for 16 hours. 10ul / per well of propidium iodide (PI) dye was added. After 10 minutes of incubation, the plate was transferred to Beckman Cytoflex for FACS analysis. The live cells identified as being CFSE positive and PI negative were collected and the cytotoxicity was determined. As expected, the cytotoxicity induced by afucosylated antibodies on iTreg cells with higher CD25 expression was much greater than on CD8+ T cells. Expression data was not shown. The afucosylated versions produced at 30 μM and 50 μM of 2-FF, H5L0-30 μM and H5L0-50 μM, displayed a similar cytotoxicity profile on iTreg cells. It appears that the potency of the afucosylated antibody with about 20%fucose level remaining is enough to deplete iTregs effectively. (Figures 28 and 29) .

[0339] Example 6 In vivo efficacy of anti-CD25 antibodies

[0340] To test the in vivo efficacy of anti-CD25 antibodies, we evaluated anti-CD25 antibody on MC38 / hPD-L1 tumor utilizing hCD25 KI mice. Briefly, 120 female human CD25 knock-in mice of 6~7 weeks age (purchased from Biocytogen company) were inoculated with 2*10^6 MC38 / hPD-L1 cells. The animals were randomized when the tumor was around 60~80mm^3, and were treated with control IgG1 at 10 mg / kg (mpk) , or benchmark RG6292-analog (which is a afucosylated benchmark from Roche, having the same sequence as 686-m3) at 10mpk, or protein-engineered version of H5L0, H5L0_VLPYLL (H5L0 with the Fc region engineered to contain VLPYLL mutation) at 10mpk, or glycol-engineered version of H5L0, H5L0_30% (H5L0 with 38%fucose) at 10mpk or afucosylated H5L0 (H5L0 with about 0.2%fucose) at 10mpk, by Intraperitoneal injection (IP) twice a week for 4 weeks. The results (Figures 30A and Figures 30B) indicate that 10mg / kg engineered anti-CD25 antibodies significantly inhibited tumor growth and had similar anti-tumor activity with benchmark (RG6292-analog) .

[0341] In a companion experiment with the same treatment, peripheral and tumor lymphocytes were collected weekly after dosing for biomarker analysis. The results (Figures 30C) showed that, relative to control IgG1, the anti-CD25 antibodies significantly increased CD8+ / Treg ratio in peripheral blood during the 4-week treatment, and that each of the engineered versions of anti-CD25 antibody H5L0 showed a higher ratio than benchmark. The results (Figures 30D) showed that, relative to control IgG1, the CD8+ / Treg ratio in tumor was also elevated significantly after 2-week treatment.
