Description

A fused tricyclic compound and its preparation method and application

[0001] Citation of Related Applications

[0002] This application claims priority to Chinese patent application No. 202310834844.5 filed on July 7, 2023 and Chinese patent application No. 202311370973.X filed on October 20, 2023, the contents of which are incorporated by reference into this application in their entirety and for all purposes. Technical Field

[0003] The present invention belongs to the field of medicine, and in particular relates to a fused tricyclic compound and a preparation method and application thereof. Background Art

[0004] Axon degeneration is closely related to the occurrence and development of neurological diseases. Wallerian degeneration is the main pathway of axon degeneration. Wallerian degeneration, also known as secondary degeneration, refers to the process in which the distal nerve fibers begin to degrade after the axons are broken due to trauma. This process involves two key proteins: nicotinamide adenine dinucleotide (NAD + ) synthase NMNAT2 and NAD + Hydrolase SARM1, and SARM1 (Sterile alpha and Toll / interleukin-1 receptor motif-containing 1) is the main execution molecule of this neurodegradation process. The regulation of its activity plays an important role in the treatment of corresponding neurological diseases.

[0005] In healthy neurons, NMNAT2 is transported along axons to maintain high levels of NAD. + When the level of nicotinamide mononucleotide (NMN) is low, SARM1 is maintained at a low activity level. When neurons are damaged, NMNAT2 transport along the axon stops and NMNAT2 is rapidly decomposed, causing NMN levels to begin to rise and NAD + The levels begin to decrease, and this period is called the axonal degeneration latency period. NMN and NAD + Changes in SARM1 lead to the activation of Ca 2+ Influx, calpain activation, and ATP depletion, along with the accumulation of ROS, lead to axonal degeneration. Numerous studies have reported that knocking out or inhibiting SARM1 has protective effects in various neurological disease models, such as peripheral neuropathy, traumatic brain injury, amyotrophic lateral sclerosis, Parkinson's disease, and glaucoma.

[0006] Treatment options for neurological diseases such as Alzheimer's disease, Parkinson's disease, multiple sclerosis, amyotrophic sclerosis, and Huntington's disease are very limited, with no effective treatments available. Currently, treatments are mostly symptomatic, but they cannot halt the progression of the disease, let alone cure it. Furthermore, with the aging population, the incidence of neurodegenerative diseases is also increasing. Therefore, there is an urgent need to develop more effective drugs to help slow disease progression, improve patients' quality of life, and even prevent disease progression in its early stages.

[0367] As used herein, the term "pharmaceutically acceptable salt" refers to a salt of a compound of the invention that is pharmaceutically acceptable and possesses the desired pharmacological activity of the parent compound. Such salts include acid addition salts formed with inorganic or organic acids, or coordination compounds formed by the substitution of acidic protons present in the parent compound with metal ions or organic bases.

[0368] As used herein, the term "stereoisomer" refers to an isomer formed due to at least one asymmetric center. In compounds with one or more (e.g., 1, 2, 3, or 4) asymmetric centers, racemic mixtures, single enantiomers, diastereomeric mixtures, and individual diastereomers can be produced. Specific individual molecules can also exist as geometric isomers (cis / trans). Similarly, the compounds of the present invention can exist as mixtures of two or more structurally different forms in rapid equilibrium (commonly referred to as tautomers). Representative examples of tautomers include keto-enol tautomers, phenol-ketone tautomers, nitroso-oxime tautomers, imine-enamine tautomers, etc. It is to be understood that the scope of this application encompasses all such isomers or mixtures thereof in any proportion (e.g., 60%, 65%, 70%, 75%, 80%, 85%, 90%, 95%, 96%, 97%, 98%, 99%).

[0369] Unless otherwise indicated, the compounds of the present invention are intended to exist as stereoisomers, including cis and trans isomers, optical isomers (e.g., R and S enantiomers), diastereomers, geometric isomers, rotational isomers, conformational isomers, atropisomers, and mixtures thereof.
